技术领域
本发明涉及培养基技术领域,具体的说是一种大肠杆菌高密度培养基。
背景技术
大肠杆菌是工业生产上应用最为广泛的菌种,用来生产各种诸如蛋白质、核酸类以及小肽类等产品。大肠杆菌在生长发育过程中,需要各种营养物,通常由培养基中的成分来提供。
现有技术中,大肠杆菌发酵培养基配方中通常有有机碳、有机氮、无机盐、生长因子和水等,当存在含量较高的有机氮时,首先发酵培养成本较高,其次由于杂蛋白的产生,增加了非特异性蛋白对后续纯化的影响,增加了后期纯化难度;且难以进一步提高高密度可达到菌体湿重200g/L以上。
发明内容
本发明所要解决的技术问题在于提供一种配方改良的大肠杆菌高密度培养基。仅添加少量的有机氮源,即可实现大肠杆菌的高密度发酵以及高表达。
本发明是通过以下技术方案实现的:
设计一种大肠杆菌高密度培养基,其特征在于,包含发酵培养基和补料培养基,
发酵培养基包括以下浓度组分:

进一步的,所述发酵培养基与补料培养基的体积比为2:1。
进一步的,发酵培养基pH7.3。
本发明的有益效果在于:经过M9配方改良,蛋白胨和酵母粉含量极低,降低成本的同时,减少了非特异性蛋白对后续纯化的影响;高密度可达到菌体湿重200g/L以上。添加少量有机氮源,实现高密度发酵。实现大肠杆菌高表达,同时降低生产成本;减少杂蛋白的产生,降低后期纯化难度。
