发明内容

具体实施方式
发明人已经基于使用人工microRNA(amiR)减少NKCC1表达开发了RNA干扰(RNAi)策略。通过用针对hNKCC1的反义靶向序列替换天然存在的初级microRNA(pri-miRNA)支架(scaffold)(例如小鼠microRNA155;基因库登录号:NR_029565.1)的21-22个核苷酸的反义靶向序列(所谓的引导链)来构建人工microRNA。特别地,发明人已设计了一种系统,该系统通过使用人突触素启动子驱动转基因表达来实现针对NKCC1的特定amiR的神经元特异性表达。
因此,在第一方面,本发明涉及用于减少有需要的受试者中NKCC1表达的治疗方法的载体,该载体包含编码能够降低NKCC1表达的amiR的多核苷酸,其中amiR包含选自由SEQID NO:4、8、15、16、18、23、25、39、40、44、45、53、57、64、66、67、68、69、82和88组成的组的至少一个核酸序列。

如本文所用,用于“Na-K-Cl协同转运蛋白”的NKCC表示有助于钠、钾和氯化物主动转运进出细胞的蛋白质。该膜转运蛋白有几种变体或同种型,特别是NKCC1和NKCC2。NKCC1由SLC12A2基因(基因ID 6558)编码,并广泛分布于整个身体,但也分布在大脑中,且尤其是在发育中的动物和人脑中。它起到增加神经元中细胞内氯化物的作用,并从而使GABA更具兴奋性。大量研究表明,阻断NKCC1减少了细胞内的氯化物,从而增强GABA的抑制作用。示例性的人核酸和氨基酸序列分别由NCBI参考序列NM_001046.2和NP_001037.1表示。

如本文所用,术语“雷特综合征”具有其在本领域中的一般含义,并且是指X染色体连锁的神经发育障碍,其导致发育逆转,特别是在表达语言和手使用方面。临床特征包括手脚小和头部生长速度减慢,某些情况下包括小头畸形。雷特综合征与树突棘的神经病理学有关,特别是在患有雷特综合征的患者中发现了海马锥体神经元中树突棘密度的降低((Chapleau CA et al,Neurobiol Dis 2009,35(2):219-33)。

在一些实施方式中,有效量是治疗有效量。“治疗有效量”是指以合理的获益/风险比治疗疾病的足够量的载体。应该理解,载体的每日总用量将由主治医师在合理的医学判断范围内决定。任何特定患者的具体治疗有效剂量水平将取决于多种因素,包括患者的年龄、体重、总体健康状况、性别和饮食;给药时间,给药途径和所用特定化合物的排泄速率;治疗的持续时间;与所使用的特定多肽组合或同时使用的药物;以及医学领域众所周知的类似因素。例如,本领域技术人员中公知的是,以低于获得期望的治疗效果所需的水平开始所述化合物的剂量,并逐渐增加剂量直至获得所需效果。因此,可以根据疾病状况、受试者(例如,根据他的体重、新陈代谢等)、治疗方案等来调整载体的剂量。在本发明的上下文中优选的有效剂量是允许最佳转导的剂量。通常,在小鼠中每剂量给药108至1012个病毒基因组(vg)。通常,要在人类中给药的AAV载体的剂量可以在1010至1014vg的范围内。
本发明的载体被配制成药物组合物。除了载体以外,这些组合物还可包含药学上可接受的赋形剂、载体、缓冲剂、稳定剂或本领域技术人员众所周知的其他材料。这样的材料应该是无毒的并且不应干扰活性成分(即,本发明的载体)的功效。载体或其他材料的精确性质可以由技术人员根据给药途径来确定。药物组合物通常为液体形式。液体药物组合物通常包括液体载体,诸如水、石油、动物或植物油、矿物油或合成油。可以包括生理盐水溶液、氯化镁、右旋糖或其他糖溶液或诸如乙二醇、丙二醇或聚乙二醇的二醇。对于注射,活性成分将为水溶液形式,其不含热原且具有合适的pH、等渗性和稳定性。本领域相关技术人员能够很好地使用例如等渗媒介物(诸如氯化钠注射液、林格氏注射液、乳酸林格氏注射液)来制备合适的溶液。根据需要可以包括防腐剂、稳定剂、缓冲剂、抗氧化剂和/或其他添加剂。为了延迟释放,可以将载体包含在药物组合物中,该药物组合物根据本领域已知的方法被配制为用于缓慢释放,诸如在由生物相容性聚合物形成的微胶囊中或在脂质体载体系统中。通常,本发明的药物组合物在预填充注射器中提供。“即用型注射器”或“预填充注射器”是以填充状态提供的注射器,即,待给药的药物组合物已经存在于注射器中并准备好给药。与分别提供的注射器和小瓶相比,预填充注射器具有许多优势,诸如改善的便利性、可负担性、准确性、无菌性和安全性。预填充注射器的使用可提高剂量精度,减少从小瓶中吸取药物时可能发生的针刺伤害的可能性,以预先确定的剂量减少因需要重构药物和/或将药物吸入至注射器而导致的配量错误,以及减少注射器的过量填充以通过最小化药物浪费来降低成本。在一些实施方式中,本发明的液体药物组合物的pH在5.0至7.0、5.1至6.9、5.2至6.8、5.3至6.7或5.4至6.6的范围内。

实施例1
在Ts65Dn神经元中,细胞内氯化物浓度升高,导致GABA的去极化和的可能的兴奋作用。我们用amiR设计了一种RNAi策略,以减少神经元中NKCC1的表达。使用唐氏综合症的著名小鼠模型Ts65Dn小鼠的原代神经元培养物,我们发现我们的使用病毒载体的amiR策略能够减少NKCC1蛋白的表达。通过amiR降低NKCC1蛋白表达恢复了Ts65Dn神经元中GABA的抑制作用,与野生型(WT)神经元相似。此外,我们发现在Ts65Dn小鼠海马中amiR分子的体内病毒递送在四种不同的学习和记忆的行为测试中挽救了认知缺陷。因此,该方法在体外和体内均能有效恢复DS中的生理活性和行为参数。
实施例2
通过使用在线工具BLOCK-iTTMRNAi Designer(算法1,表A),均基于人NKCC1(hNKCC1)DNA序列(NM_001046.2)预测了表B中报告的amiR序列。通过使用8种其他算法(表A中列出的算法2-9),我们预测了一些其他RNAi序列,这些序列在一些情况下与使用算法1获得的表B中报道的序列相对应。根据这些序列±4个核苷酸被算法2-9预测的次数,对由算法1预测的每个序列(表B)进行评分。因此,表B中列出的序列具有结合和沉默NKCC1的最高概率。
实施例3
用双荧光素酶hNKCC1传感器评价了表B中报告的89个amiR序列中的78个对hNKCC1的沉默效力(图6和7)。人H293T肾细胞在96孔板中生长,并用Lipofectamnine2000(Thermo)转染每个amiR序列以及hNKCC1双荧光素酶质粒传感器。48小时之后,在Victor3多板读板仪(PerkinElmer)上用双荧光素酶测定法(Promega)测量海肾荧光素酶(RLuc)和萤火虫荧光素酶(FLuc)之间的比率。对于每个转染的amiR序列,相对于用对照amiR质粒转染的样品,RLuc/FLuc比率的降低表明了hNKCC1敲落的程度。表C中报告了不同amiR序列对hNKCC1的沉默活性,表示为平均RLuc/Fluc比率(从4次独立转染中获得),以及通过相对于用对照amiR质粒转染的样品的学生t检验获得的相应P值。
实施例4
还评价了表B中报道的针对hNKCC1的不同amiR序列的可能的脱靶基因的数量,其由可自由获取的microRNA预测工具TargetScan预测(Agarwal et al.,Elife.12:4(2015))。针对所有注释的人类基因(ENSEMBL版本GRCh38)的全长转录物序列(编码序列加上3'非翻译区)评价了脱靶预测。表D示出了每个amiR序列可能的脱靶基因的数目。该列表从可能的脱靶基因的最低数量到最高数量进行排序,并且还从表C报告了相应的沉默活性。因此,表D顶部列出的序列具有最高的活性,以最低的脱靶机率使NKCC1沉默。

表D:amiR可能的脱靶基因的预测。使用TargetScan软件评价了每个amiR序列在所有注释的人类转录物上脱靶基因的可能数目。该表还示出了表C中的沉默活性。该表从检测到的可能的脱靶基因的最低数量到最高数量进行排序。
基于表D中报告的数据,SEQ ID No:25、39、45、53、64、66和82由于它们显示出最低数目的可能的脱靶基因而被鉴定为用于本发明的最优选的序列。

Claims (15)
1.一种包含编码能够减少受试者中NKCC1表达的人工microRNA(amiR)的多核苷酸的载体,用于在有需要的受试者中降低NKCC1表达的治疗处理方法的用途,其特征在于,所述amiR包含选自由SEQ ID NO:4、8、15、16、18、23、25、39、40、44、45、53、57、64、66、67、68、69、82和88组成的组的至少一种核酸序列。
2.根据权利要求1所述的用途的载体,其中,所述受试者患有选自由以下组成的组的疾病:唐氏综合症、脆性X综合征、雷特综合征、结节性硬化症、外伤性脑损伤、癫痫病、自闭症、精神分裂症、帕金森氏病和高血压。
3.根据权利要求1或2所述的用途的载体,其中,所述amiR由以下组成:源自天然miRNA的5'侧翼区域,TGCT核苷酸突出端,5'G+源自hNKCC1的选自由SEQ ID NO:4、8、15、16、18、23、25、39、40、44、45、53、57、64、66、67、68、69、82和88组成的组的短21个核苷酸反义序列,环形成序列,源自hNKCC1的2个核苷酸被除去(Δ2)以产生内部环的短19个核苷酸有义序列,CAGG突出端以及源自天然miRNA的3'侧翼区域。
4.根据权利要求1至3中任一项所述的用途的载体,其中,所述源自天然miRNA的5'侧翼区域是SEQ ID NO:90。
5.根据权利要求1至4中任一项所述的用途的载体,其中,所述环形成序列是SEQ IDNO:91。
6.根据权利要求1至5中任一项所述的用途的载体,其中,所述源自天然miRNA的3'侧翼区域为SEQ ID NO:92。
7.根据权利要求1至6中任一项所述的用途的载体,其中,所述载体是非病毒载体或病毒载体。
8.根据权利要求7所述的用途的载体,其中,所述病毒载体是AAV载体。
9.根据权利要求1至8中任一项所述的用途的载体,其中,所述载体包含神经元特异性启动子。
10.根据权利要求9所述的用途的载体,其中,所述神经元特异性启动子选自由突触蛋白-1(Syn)启动子、神经元特异性烯醇化酶(NSE)启动子、神经丝轻链基因启动子以及神经元特异性vgf基因启动子组成的组。
11.根据权利要求1至10中任一项所述的用途的载体,其中,所述治疗处理方法包括将所述载体递送至所述受试者的海马。
12.根据权利要求1至10中任一项所述的用途的载体,其中,所述治疗处理方法包括通过静脉内注射将所述载体递送至所述受试者。
13.根据权利要求1至10中任一项所述的用途的载体,其中,所述治疗处理方法包括通过鞘内给药将所述载体递送至所述受试者。
14.一种药物组合物,用于减少有需要的受试者中NKCC1表达的治疗处理方法的用途,所述组合物包含根据权利要求1至10中任一项所述的载体。
15.一种减少有需要的受试者中NKCC1表达的方法,包括向所述受试者递送治疗有效量的根据权利要求1至10中任一项所述的载体。
