The bottle seedlings which survive are domesticated and transplanted into a greenhouse for conventional cultivation management, and chrysanthemum plants with high drought resistance are obtained.

Technical Field

Background
With global warming, environmental deterioration and frequent drought, drought stress becomes one of the bottlenecks limiting chrysanthemum production and application. Therefore, the breeding of the new chrysanthemum variety with stronger drought resistance has important significance.
The chrysanthemum has bright color and rich flower types, and is deeply loved by people. At present, more than 3000 chrysanthemum varieties exist in China, and the cultivation area reaches 8900 hectare. The chrysanthemum is mainly used for landscaping, ground cover, interior decoration, tea, medicine and the like. Chrysanthemum belongs to a herbaceous plant with stronger drought resistance, but can grow badly in water-deficient areas, thereby influencing the sustainable development of the chrysanthemum industry. Therefore, the improvement of the drought resistance of the chrysanthemum is very important. In the aspect of chrysanthemum drought resistance research, researchers carry out a great deal of research work. The existing research shows that in the case of the American iso-transgenic DREB1A gene ground-cover chrysanthemum seedling as the experimental material, the potted seedling is naturally water-loss stressed under different temperature conditions, and the water loss condition of the transgenic ground-cover chrysanthemum seedling under the two environmental stresses of air and water drought is analyzed, and the results show that 5 transgenic chrysanthemum strains have better drought tolerance and water conservation than the wild type plants. The DREB gene is transferred into ground-cover chrysanthemum 'rain flower exciting chapter' by the Zuixinli et al, and the drought tolerance of the obtained transgenic line is detected, so that the drought tolerance of the transgenic line is improved to different degrees. Research on chrysanthemum varieties 'medal of flowers' of CgDREBa genes, such as Chensu plum and the like, shows that the transgenic chrysanthemum has higher survival rate and fresh weight preservation rate under drought stress compared with wild species. Also, studies show that drought stress affects the normal growth and development of chrysanthemum by inhibiting the photosynthesis of chrysanthemum, thereby affecting the yield and quality of chrysanthemum. However, there has been no report on obtaining chrysanthemum plants with strong drought resistance by in vitro culture using PEG. Therefore, the chrysanthemum bottle seedlings with strong tolerance and survival are screened in a mode of adding PEG into a culture medium in the process of chrysanthemum in vitro culture, and then chrysanthemum plants with strong drought resistance are obtained through domestication and screening.
