本发明公开了一种荧光传感器及其制备方法和应用,所述荧光传感器是由氨基化碳量子点(N‑CQDs)与对氨基苯硫酚修饰的银纳米颗粒(N‑AgNPs)组成的纳米复合物构成;制备时,首先制备氨基化碳量子点,再制备对氨基苯硫酚修饰的银纳米颗粒,最后将氨基化碳量子点与对氨基苯硫酚修饰的银纳米颗粒制备得到N‑CQDs/N‑AgNPs纳米复合物荧光传感器,本发明的荧光传感器可用于检测血清中多巴胺的含量;本发明制备的荧光传感器可以方便、简洁地对血清样品中多巴胺含量进行检测,该方法具有价廉、简单、灵敏度、选择性好、线性范围宽,检测时间短等优点,在生物分析或疾病研究过程中的DA检测有较大的应用前景。

技术领域
本发明涉及荧光传感技术领域,具体是一种荧光传感器及其制备方法和应用。
背景技术
多巴胺(DA)是儿茶酚胺和苯乙胺类药物中的一种重要的激素和神经递质,它在人类大脑中的起到许多调节作用并控制许多生理功能,如动机、情绪、内分泌调节和运动;许多疾病,例如帕金森氏病、精神分裂症和厌食症都与DA水平的异常有关。为研究生命过程中DA的生物功能,建立测试DA的方法一直是分析领域的热点之一。然而,DA在生物系统中的正常浓度的范围为0.1mM到1.0mM,这是一个非常低的浓度范围。因此,对DA的灵敏性检测仍然具有挑战性。
到目前为止,检测DA的方法有高效液相色谱、质谱、分光光度法和电化学方法等。上述方法均存在缺点,例如,色谱和质谱方法依赖昂贵的设备和特定的样品预处理程序;分光光度法灵敏度相对较低,而电化学方法通常因抗坏血酸的干扰而产生相似的氧化电位,因此上述方法的应用均受到了极大地限制。荧光分析方法由于其成本低、操作简单,灵敏度高,实用性强,选择性好等突出特点,成为检测DA更好的替代方法,而且荧光方法大多拥有在复杂生物基质中没有分离程序就可以直接检测DA的优势。当前,一些荧光探针,如有机染料和单臂碳纳米管被开发作为选择性、灵敏性的直接检测神经递质DA的方法。然而有机染料荧光探针不可避免的因为毒性和复杂的合成步骤而受到一定的限制,与此同时,已有的一些利用不同荧光探针测试神经递质DA的方法中,大多采用猝灭的方法来达到检测的目的。众所周知,荧光方法最大的不足就是容易猝灭,而引起猝灭的因素太多,导致方法的选择性受阻。因此开发出一种无毒且具有高选择性和高灵敏度的检测DA的荧光传感器具有十分重要的意义。
在荧光分析方法中,碳量子点(CQDs)已被证明是特别合适作为荧光探针的一种纳米材料。碳量子点作为一种新型的碳基零维材料,具有优秀的光学性质,良好的水溶性、低毒性、环境友好、原料来源广、成本低、生物相容性好等诸多优点,并且碳量子点的应用广泛,在医学成像技术、环境监测、化学分析、催化剂制备、能源开发等许多的领域都有较好的应用前景。
发明内容
本发明的目的就是针对目前的检测血清中DA的方法存在试验设备昂贵、检测灵敏度低等问题,提供一种用于检测血清中多巴胺的荧光传感器及其制备方法和应用,本发明制备的荧光传感器可以方便、简洁地对血清样品中多巴胺含量进行检测,该方法具有价廉、简单、灵敏度、选择性好、线性范围宽,检测时间短等优点,在生物分析或疾病研究过程中的DA检测有较大的应用前景。
本发明的一种荧光传感器,所述荧光传感器是由氨基化碳量子点(N-CQDs)与对氨基苯硫酚修饰的银纳米颗粒(N-AgNPs)组成的纳米复合物构成。
本发明的一种荧光传感器的制备方法,包括以下步骤:
(1)氨基化碳量子点(N-CQDs)的制备
称取0.72g葡萄糖置于圆底烧瓶中,向其中加入25mL超纯水溶解,超声10min后放入真空干燥箱中,在200℃下加热3h,将反应结束得到的橙黄色粘稠液体自然冷却至室温,离心、过滤后用超纯水定容至25mL,得到的黄色溶液即为CQDs母液;取CQDs母液5mL至50mL圆底烧瓶中,向其中加入100μL乙二胺,超声混匀后放入真空干燥箱中,在200℃下加热2h,将反应得到的棕褐色粘稠液体自然冷却至室温,离心、过滤后用超纯水定容至100mL,即得到氨基化碳量子点(N-CQDs)溶液;
(2)对氨基苯硫酚(PATP)修饰的银纳米颗粒(N-AgNPs)的制备
将100mL浓度为2.0mM的NaBH4溶液添加到25mL浓度为1.0mM AgNO3溶液中,在冰水浴下混合均匀,再向其中加入100μL浓度为0.1M柠檬酸钠和200μL浓度为0.1mM PATP,剧烈搅拌30min,得到黄棕色的N-AgNPs溶液;再用分子量为10kDa的半透膜在去离子水中透析3次,即可获得N-AgNPs贮备液,将N-AgNPs贮备液置于4℃下保存备用;
(3)N-CQDs/N-AgNPs纳米复合物荧光传感器的制备
等体积量取步骤(1)制备的氨基化碳量子点(N-CQDs)溶液和步骤(2)制备的N-AgNPs贮备液,室温下将二者溶解于浓度为10nM pH=8.0的PBS缓冲液中,混合搅拌6min后即得到N-CQDs/N-AgNPs纳米复合物荧光传感器,将N-CQDs/N-AgNPs纳米复合物置于4.0℃下保存备用。
本发明还提供了一种荧光传感器在检测血清中多巴胺的应用。
本发明的一种荧光传感器在检测血清中多巴胺的应用,包括以下步骤:
(1)标准溶液中多巴胺含量的检测
取200μL的荧光传感器5份,分别添加1.0mL浓度梯度为1.65、10、50、100、150、250和325nM的多巴胺标准溶液样品,用浓度为10nM pH=8.0的PBS缓冲液定容至5.0mL,反应12min后,在401nm激发下,用荧光分光光度计检测标准溶液样品的荧光光谱,根据标准溶液样品的浓度与荧光光谱F/F0的关系建立标准工作直线;其中F0代表荧光传感器自身的原始荧光,F代表加入不同浓度的多巴胺后发出的荧光光谱;
(2)样品中多巴胺含量的检测
取临床血清样品1.0mL,将血清样品通过3500rpm离心后,得到上层清液,取20μL上层清液向其中加入200μL的荧光传感器,用浓度为10nM pH=8.0的PBS缓冲液定容至5.0mL,反应12min后,在401nm激发下,用荧光分光光度检测样品溶液的荧光光谱,根据得到的标准工作直线计算样品溶液中多巴胺的含量。
本发明的工作原理是:乙二胺修饰的N-CQDs具有稳定而强的荧光信号,而对氨基苯硫酚(PATP)功能化的AgNPs形成的(N-AgNPs)没有荧光信号,由于二者表面均具有丰富的胺基,容易通过氢键作用形成新的纳米复合物(N-CQDs/N-AgNPs),并伴随着N-AgNPs与N-CQDs之间的内滤效应,削弱N-CQDs的荧光,即二者复合后N-CQDs的荧光大大减弱。然而,在该纳米复合物体系中加入DA后,带有丰富胺基和羟基的DA也能与N-CQDs形成氢键作用,能引发N-CQDs/N-AgNPs纳米复合物分解,使N-CQDs/N-AgNPs纳米复合物中的N-AgNPs解离一部分,削弱二者间的内滤效应,使荧光信号激活和荧光恢复。因此,利用该原理构建了一个可激活的荧光检测器实现血清中DA选择性检测,且该方法的灵敏度极高。
本发明的优点是:本发明制备的荧光传感器可以方便、简洁地对血清样品中多巴胺含量进行检测。不同于荧光猝灭机理(能引起荧光探针猝灭的因素很多),本发明是利用荧光激活的方式实现血清中DA的检测,无需任何样品分离或前处理技术,方法具有价廉、简单、灵敏度、选择性好、线性范围宽,检测时间短等优点,在生物分析或疾病研究过程中的DA检测有较大的应用前景。

具体实施方式
实施例1
本实施例的一种荧光传感器,所述荧光传感器是由氨基化的碳量子点(N-CQDs)与对氨基苯硫酚修饰的银纳米颗粒(N-AgNPs)组成的纳米复合物构成。
本实施例的一种荧光传感器的制备方法,包括以下步骤:
(1)氨基化碳量子点(N-CQDs)的制备
称取0.72g葡萄糖置于圆底烧瓶中,向其中加入25mL超纯水溶解,超声10min后放入真空干燥箱中,在200℃下加热3h,将反应结束得到的橙黄色粘稠液体自然冷却至室温,离心、过滤后用超纯水定容至25mL,得到的黄色溶液即为CQDs母液;取CQDs母液5mL至50mL圆底烧瓶中,向其中加入100μL乙二胺,超声混匀后放入真空干燥箱中,在200℃下加热2h,将反应得到的棕褐色粘稠液体自然冷却至室温,离心、过滤后用超纯水定容至100mL,即得到氨基化碳量子点(N-CQDs)溶液;
(2)对氨基苯硫酚(PATP)修饰的银纳米颗粒(N-AgNPs)的制备
将100mL浓度为2.0mM的NaBH4溶液添加到25mL浓度为1.0mM AgNO3溶液中,在冰水浴下混合均匀,再向其中加入100μL浓度为0.1M柠檬酸钠和200μL浓度为0.1mM PATP,剧烈搅拌30min,得到黄棕色的N-AgNPs溶液;再用分子量为10kDa的半透膜在去离子水中透析3次,即可获得N-AgNPs贮备液,将N-AgNPs贮备液置于4℃下保存备用;
(3)N-CQDs/N-AgNPs纳米复合物荧光传感器的制备
取100μL氨基化碳量子点(N-CQDs)溶液和100μL N-AgNPs储备液,室温下溶解于625μL浓度为10nM pH=8.0的PBS缓冲液中,混合搅拌6min后得到N-CQDs/N-AgNPs纳米复合物,即为荧光传感器,将N-CQDs/N-AgNPs纳米复合物置于4.0℃下保存备用。
为得到性能优良的碳量子点及氨基化的碳量子点,分别对碳量子点的合成时间和氨基化的时间进行了优化。
首先让葡萄糖溶液在200℃的真空干燥箱中分别加热1.5h、2.0h、2.5h、3.0h、3.5h、4.0h、4.5h,得到不同反应时间的碳量子点。稀释同样的倍数后,分别对它们进行了荧光检测,如图2。并在此基础上,再让加有乙二胺的碳量子点溶液在200℃下的真空干燥箱中分别加热0.5h、1.0h、1.5h、2.0h、2.5h、3.0h、3.5h、4.0h,得到不同反应时间的氨基化碳量子点,稀释同样的倍数后,对它们进行荧光检测,如图3。
实验结果表明,从图2中发现加热3.0h和3.5h得到的碳量子点荧光强度最高,且两者之间相差不大。因此,选择加热3.0h来合成碳量子点。从图3中发现加热2.0h得到的氨基化碳量子点荧光强度最强,故选择2.0h为氨基化的最优时间。
为了验证本实施例的氨基化碳量子点(N-CQDs)的结构组成,我们用红外光谱仪对乙二胺、碳量子点、氨基化碳量子点(N-CQDs)进行了表征,试验结果如图4所示。
由图4可看出,碳量子点在3420cm-1、2920cm-1、1648cm-1、1405cm-1和1028cm-1处均有明显的峰,分别代表:-OH的伸缩振动、=C-H的伸缩振动、C=O的伸缩振动、C-H的弯曲振动和C-O-C的伸缩振动。而碳量子点表面大量的羟基和羧基,也正好说明了碳量子点良好的亲水性。而氨基化的碳量子点并没有出现C-O-C的伸缩振动,反而在1397cm-1处出现了C-N伸缩振动。这证明,碳量子点已被氨基化,生成了氨基化碳量子点。
为了使N-CQDs/N-AgNPs纳米复合物的组装过程达到最佳效果,对N-CQDs/N-AgNPs纳米复合物的组装条件,如N-AgNPs的浓度以及向N-CQDs中加入N-AgNPs后的猝灭时间进行了优化,试验结果如图5和6所示。
图5的结果表明,向N-CQDs中加入不同浓度的AgNPs,在487nm处,145μM N-CQDs的荧光信号逐渐降低。在60nM AgNPs的存在下,N-CQDs的最大猝灭效率约为70%。从图6中可以发现荧光猝灭过程非常快,在6min时获得的最大猝灭效率并在6min、8min、10min处稳定,形成一个猝灭平台,意味着N-CQDs和N-AgNPs之间的结合速度快且强。因此,AgNPs浓度优化为60nM,猝灭时间选择6min。
另外,我们对碳量子点和氨基化碳量子点进行了紫外表征,得到碳量子点和氨基化碳量子点的紫外-可见吸收光谱图,参见图7-8。
从图7和图8可知,碳量子点有两个明显的紫外吸收峰,分别位于222nm和280nm处,而氨基化碳量子点只在260nm处有一个明显的紫外吸收峰。
为了确定碳量子点和氨基化碳量子点的荧光激发波长和最大发射波长,对碳量子点和氨基化碳量子点进行了荧光表征,得到的荧光光谱如图9和图10。
图9的实验结果表明,碳量子点的荧光激发波长为380nm,在该波长的激发下,得到的最大发射波长为514nm;图10表明,氨基化碳量子点的荧光激发波长为401nm,在该波长激发下,得到的最大发射波长为487nm。
实施例2
本实施例的一种荧光传感器在检测血清中多巴胺的应用,包括以下步骤:
(1)标准溶液中多巴胺含量的检测
取200μL实施例1中制备好的荧光传感器5份,分别添加1.0mL浓度梯度为1.65nM、10nM、50nM、100nM、150nM、250nM和325nM的多巴胺标准溶液样品,用浓度为10nM pH=8.0的PBS缓冲液定容至5.0mL,反应12min后,在401nm激发下,用荧光分光光度计检测多巴胺标准溶液样品的荧光光谱,根据标准溶液样品的浓度与荧光光谱F/F0的关系建立标准工作直线;其中F0代表荧光传感器自身的原始荧光,F代表加入不同浓度的多巴胺后发出的荧光光谱,实验结果见下表1。

(2)样品中多巴胺含量的检测
取临床血清样品1.0mL,将血清样品通过3500rpm离心后,得到上层清液,取20μL上层清液向其中加入200μL的荧光传感器,用浓度为10nM pH=8.0的PBS缓冲液定容至5.0mL,反应12min后,在401nm激发下,用荧光分光光度检测样品溶液的荧光光谱,根据得到的标准工作直线计算样品溶液中多巴胺的含量。
经检测,本实施例的血清样品中多巴胺的含量为0.81ng/mL。
实施例3
为了进一步研究N-CQDs/N-AgNPs纳米复合物的实际应用,分别向4份临床血清样品中加入了多巴胺标准样品溶液,利用制备的荧光传感器对加标后血清中的DA的含量进行了检测,试验结果见表2。

表2的结果表明,回收率为98.8-109.8%,相对标准偏差(RSD)小于3.0%,具有较好的准确度。基于上述结果,说明N-CQDs/N-AgNPs纳米复合物荧光传感器可以很好地应用于血清中的DA检测,这为医学临床分析提供了更好的应用潜力。
实施例4
为了探究本发明的荧光传感器的工作原理,对AgNPs猝灭N-CQDs荧光的机理就行了研究。如图11所示,从图11可得知N-CQDs的荧光也随着N-CQDs/N-AgNPs纳米复合物的组装而发生荧光猝灭。此外,我们发现了AgNPs吸收谱图与N-CQDs荧光激发谱图重叠,参见图12,这一现象表明发生在N-CQDs/N-AgNPs纳米复合物之间的荧光猝灭过程可能是由于内滤效应(IFE)。因此,我们首先利用Stern-Volmer方程探究了QDs在加入不同浓度的AgNPs后分别在不同温度(288K,313K,323K)下的荧光猝灭过程,5min后,在401nm的激发波长下收集487nm处发射峰的荧光强度。实验是通过一个循环水系统进行的,该系统可以有效地调节恒温培养温度。因此,通过Stern-Volmer方程,探讨了温度对猝灭过程的影响:
在这个实验中,F0和F分别代表N-CQDs在加入N-AgNPs作为猝灭剂前后的荧光强度。[Q]代表AgNPs的浓度(L/mol),Ksv代表Stern-Volmer猝灭常数(L/mol),由F0/F与[Q]在3种不同温度(288k、313k、323k)下的线性拟合曲线可得。结果显示荧光猝灭常数Ksv随着温度的升高而下降,表明静态猝灭机理在N-CQDs猝灭过程中占主导地位,参见图13和表3。同时,我们测量了在N-CQDs与N-AgNPs混合前后的荧光寿命(τ0,τ)。结果如图14所示,荧光寿命并未发生明显的变化,因此可以排除动态猝灭过程。综上所述,我们推测静态猝灭过程是荧光猝灭的主导过程,并且由于氢键作用形成了N-CQDs/N-AgNPs纳米复合物的装配结构,从而伴随内滤效应。

与此同时,我们还研究了N-CQDs/N-AgNPs纳米复合物在PBS缓冲液(pH=8.0)中对DA的荧光响应。N-CQDs/N-AgNPs纳米复合物在401nm处表现出非常弱的荧光发射,加入65nMDA后,在487nm处观察到荧光发射随着时间(0-15min)逐渐激活并恢复,参见图15。在401nm激发波长下,由DA引起的N-CQDs/N-AgNPs纳米复合物的最大活化荧光是在12min处,荧光恢复到最大并形成一个稳定的平台,参见图16。
Claims (3)
1.一种荧光传感器,其特征在于:所述荧光传感器是由氨基化碳量子点与对氨基苯硫酚修饰的银纳米颗粒组成的纳米复合物构成;
制备时,包括以下步骤:
(1)氨基化碳量子点的制备
称取0.72g葡萄糖置于圆底烧瓶中,向其中加入25mL超纯水溶解,超声10min后放入真空干燥箱中,在200℃下加热3h,将反应结束得到的橙黄色粘稠液体自然冷却至室温,离心、过滤后用超纯水定容至25mL,得到的黄色溶液即为CQDs母液;取CQDs母液5mL至50mL圆底烧瓶中,向其中加入100μL乙二胺,超声混匀后放入真空干燥箱中,在200℃下加热2h,将反应得到的棕褐色粘稠液体自然冷却至室温,离心、过滤后用超纯水定容至100mL,即得到氨基化碳量子点(N-CQDs)溶液;
(2)对氨基苯硫酚修饰的银纳米颗粒的制备
将100mL浓度为2.0mM的NaBH4溶液添加到25mL浓度为1.0mM AgNO3溶液中,在冰水浴下混合均匀,再向其中加入100µL浓度为0.1M柠檬酸钠和200µL浓度为0.1mM PATP,剧烈搅拌30min,得到黄棕色的对氨基苯硫酚修饰的银纳米颗粒(N-AgNPs)溶液;再用分子量为10kDa的半透膜在去离子水中透析3次,即可获得对氨基苯硫酚修饰的银纳米颗粒贮备液,将对氨基苯硫酚修饰的银纳米颗粒贮备液置于4℃下保存备用;
(3)N-CQDs/N-AgNPs纳米复合物荧光传感器的制备
等体积量取步骤(1)制备的氨基化碳量子点(N-CQDs)溶液和步骤(2)制备的对氨基苯硫酚修饰的银纳米颗粒(N-AgNPs)贮备液,室温下将二者溶解于浓度为10nM pH=8.0的PBS缓冲液中,混合搅拌6min后即得到N-CQDs/N-AgNPs纳米复合物荧光传感器,将N-CQDs/N-AgNPs纳米复合物置于4.0℃下保存备用。
2.如权利要求1所述的一种荧光传感器在检测血清中多巴胺的应用。
3.如权利要求2所述的一种荧光传感器在检测血清中多巴胺的应用,其特征在于包括以下步骤:
(1)标准溶液中多巴胺含量的检测
取200µL的荧光传感器5份,分别添加浓度梯度为1.65nM、10nM、50nM、100nM、150nM、250nM和325nM的多巴胺标准溶液样品,用浓度为10nM pH=8.0的PBS缓冲液定容至5.0mL,反应12min后,在401nm激发下,用荧光分光光度计检测标准溶液样品的荧光光谱,根据标准溶液样品的浓度与荧光光谱F/F0的关系建立标准工作直线;其中F0代表荧光传感器自身的原始荧光,F代表加入不同浓度的多巴胺后发出的荧光;
(2)样品中多巴胺含量的检测
取临床血清样品1.0mL,将血清样品通过3500rpm离心后,得到上层清液,取20µL上层清液向其中加入200µL的荧光传感器,用浓度为10nM pH=8.0的PBS缓冲液定容至5.0mL,反应12min后,在401nm激发下,用荧光分光光度计检测样品溶液的荧光光谱,根据得到的标准工作直线计算样品溶液中多巴胺的含量。
