Abstract
The invention belongs to the field of plant extraction and daily chemicals, and particularly relates to an application of a gorse extract, wherein the gorse extract is used as an anti-aging active ingredient in a skin external preparation, particularly a skin repair active ingredient, and/or a skin structure stabilizing active ingredient, and/or a skin elasticity active ingredient, and has the effects of improving cell repair/proliferation, stabilizing a basement membrane structure and function, promoting extracellular matrix secretion, and keeping skin elasticity. The broom extract has high activity, can generate obvious effect under the condition of extremely low content, is natural, safe and effective, and does not irritate the skin.

Technical Field
The invention belongs to the field of plant extraction and daily chemicals, and particularly relates to an application of a gorse extract.
Background
The skin covers the whole human body surface and is in direct contact with the external environment, so that the mirror not only protects the body from being damaged by various external factors, but also reflects the health state of the body. The skin is composed of epidermis, dermis, and subcutaneous tissue, and contains skin appendages, abundant nerves, blood vessels, lymphatic vessels, and muscles. The connecting area of the dermis and the epidermis is called as a basement membrane zone, the basement membrane is a supporting pad of epithelial cells, has supporting and connecting effects between epithelial cell tissues and connective tissues, and is also a semipermeable membrane with material permeability, so that the material exchange is facilitated. The basement membrane not only serves as a layer of structural model for supporting epidermal cells, but also serves as a selective passing barrier of cells, prevents connective tissue cells from contacting epithelial cells on the basement membrane, and allows lymphocytes, macrophages and neuronal synapses to pass through; many vital phenomena such as cell morphology, cell polarity, cell metabolism, plasma membrane protein distribution, cell survival, cell proliferation and differentiation, and cell migration are closely related to the basement membrane.
The dermis is composed of cells and extracellular matrix, fibroblasts are main cells of the dermis, and the normal proliferation and differentiation of the fibroblasts maintain the structure of the dermis of the skin to form a normal structure and physiological function; extracellular matrix is a kind of macromolecular substance synthesized and secreted by cells and interacting around the cells, and many life phenomena such as proliferation and differentiation of cells, cell migration and the like are closely related to the extracellular matrix. The extracellular matrix is composed of four major classes of molecules: collagen, elastin, glycoprotein and proteoglycan. The extracellular matrix is finely and skillfully entangled and interwoven through different molecules to form various exquisite three-dimensional structures, so that the skin is endowed with elasticity and tensile strength, and looks plump and full of vitality.
The longevity gene is a gene capable of keeping active for a long time, and can greatly improve the health of organisms and prolong the life of the organisms. When a person encounters severe cold, heat or water and food deficient environments, these genes can activate the self-defense system in the human body, making the cell repair capacity powerful.
SIRT1 (sirtuin 1): one of the famous longevity genes. SIRT1 is a highly conserved protein deacetylase, is involved in regulating longevity caused by energy restriction, is called a longevity gene, has complex physiological actions, and is involved in regulating glycolipid metabolism, inflammation, oxidative stress, cell senescence, apoptosis and the like.
The HBEGF (heparin binding EGF like growth factor) gene encodes a synthetic heparin binding epidermal growth factor, which is an EGF family member that binds to and activates EGF receptors. hbegf plays a key role in pathological and physiological processes, including cancer. After the skin part of the human body is damaged, the epidermal growth factor is produced, the proliferation and the homing of cells (comprising keratinocytes, dermal cells and basement membrane cells) are promoted, and the healing of skin wounds is promoted. The up-regulation of the gene expression is helpful for promoting the repair of skin and delaying skin aging.
The col4 alpha 1 (collagen type IV alpha 1 chain) gene codes and synthesizes type IV collagen alpha 1 chain, the type IV collagen is the main component forming basement membrane, the up-regulation of the gene expression can promote the integrity of the basement membrane (also called as dermoepidermal junction, DEJ) and delay skin aging.
The IGF1 (insulin like growth factor 1) gene encodes a synthetic insulin-like growth factor 1, a protein similar in function and structure to insulin, a member of a family of proteins involved in the regulation of growth and development. Insulin-like growth factor 1 promotes keratinocyte migration and re-epithelialization of the wound surface.
The ELN (elastin) gene encodes synthetic elastin, which is the main component of elastic fibers (elastic fibers), and is rich in hydrophobic amino acids such as glycine and proline, and elastin molecules are cross-linked with each other through covalent bonds between lysine residues, and the formed cross-linked network can generate elasticity through configuration change. Elastin is an important extracellular matrix in skin and is closely related to dermal aging, which is a ring of importance in causing skin aging. Elastin acts as a rubber band in the skin, allowing the skin to stretch and fold, and functions like a spring in a mattress to maintain and support the elasticity of the skin. Therefore, elastin plays an important role in maintaining skin elasticity.
Therefore, the research on how to improve the expression of genes related to key parts and functions of the skin has important significance on improving the skin repair capacity, keeping the structural stability and delaying the skin aging.
Genistis Caragana (Latin Wen: parochetus communis) also known: the sparrow flower is creeping herb of Paspalum of Leguminosae, with a height of 10-20 cm, and is sparsely covered and soft. The rootstock is filiform, the root is rooted on the node, and the root nodule is formed. The tip of the petal is slightly sharp, and the side of the petal is divided into two petals, the trend is like flying sparrow, the color is golden yellow, so the petal is named as 'gold sparrow'. It is born on woodland grassland, hillside and roadside wasteland, and has an altitude of 2000-3000 m. Sichuan, yunnan and Tibet. India, nipal, plumbum preparatium, srilanka, myanmar, thailand, malaysia and eastern Africa are also distributed.
Xiguang, it usually occurs on hillside and faces the sun. The root system is developed, has root nodules, resists drought and barren and can grow in gaps of the hills and stones. Root: tonify and strengthen body, promote blood circulation and regulate menstruation, dispel wind and remove dampness. Can be used for treating hypertension, dizziness, tinnitus, dim eyesight, asthenia, menoxenia, leucorrhea, galactostasis, rheumatic arthralgia, and traumatic injury. Flower: dispel wind, activate blood, relieve cough and resolve phlegm. Can be used for treating dizziness, tinnitus, cough due to lung deficiency, and infantile dyspepsia.
The flos Caraganae Sinicae contains protein, fat, carbohydrate, vitamins, minerals, etc. The wild herb broom cypress flower has mild sweet taste, and has the effects of nourishing yin, regulating blood and tonifying spleen. It can be used for treating cough due to excessive heat, dizziness, soreness of waist, asthma, leucorrhea, infantile malnutrition, acute mastitis, and traumatic injury. The book of plant experiments records "nourishing yin and tonifying yang". Shanghai commonly used Chinese herbal medicine carries the functions of promoting blood circulation, dispelling wind, relieving cough and strengthening body. It can be used for treating dizziness, headache, tinnitus, dim eyesight, lung deficiency, chronic cough, and infantile malnutrition. The nature and flavor of the herbs are sour, astringent and warm.
The invention proves that the broom extract can improve the expression of cell repair related genes, basement membrane stability related genes and anti-aging related genes, improve the cell repair activity, stabilize the structure and function of the basement membrane, promote the secretion of extracellular matrix and delay skin aging for the first time, and is suitable for preparing skin external preparations.
Disclosure of Invention
The invention aims to provide application of a gorse extract.
The flos Caraganae Sinicae extract has effects of improving skin repairing activity, stabilizing skin structure, promoting extracellular matrix secretion, maintaining skin elasticity, and delaying skin aging, and is suitable for preparing skin external preparation.
The sparrow flower extract enhances the cell repair activity by improving the expression of cell repair/proliferation related genes, so as to improve the skin repair activity and delay skin aging; the broom cypress extract enhances the structure and the function of the basement membrane by improving the expression of the genes related to basement membrane stability so as to stabilize the structure of the skin and delay skin aging; the caragana sinica franch enhances the cell repair and/or proliferation activity, and/or basement membrane structure and function, and/or extracellular matrix secretion, and/or maintains the skin elasticity and delays the skin aging by improving the expression of anti-aging related genes.
The broom extract can be used for preparing skin external preparations, is used as an anti-aging active ingredient in the skin external preparations, and particularly is used as a skin repair active ingredient and/or a skin structure stabilizing active ingredient and/or an active ingredient for maintaining skin elasticity to realize an anti-aging effect.
The broom extract is used as an active component for promoting cell repair and/or proliferation, and can enhance skin repair activity by promoting cell proliferation and repair, especially dermal cells or epidermal cells, so that the skin repair function is realized, and skin aging is delayed.
The broom extract is used as a basement membrane structure and functional stabilizing active ingredient, stabilizes the skin structure and delays skin aging.
The broom extract is used as an extracellular matrix secretion active component to maintain or enhance skin elasticity and delay skin aging.
The broom extract is used as an active ingredient for improving the expression of cell repair and/or proliferation related genes, and/or improving the expression of basement membrane related genes, and/or improving the expression of skin elasticity related genes. Preferably, the cell repair and/or proliferation related gene is SIRT1 gene, HBEGF gene or IGF1 gene, the basement membrane related gene is col4 α 1 gene, and the skin elasticity related gene is ELN gene.
The broom extract enhances cell repair and/or proliferation activity by increasing the expression of SIRT1 gene to promote synthesis of histone deacetylase, by increasing the expression of HBEGF gene to promote synthesis of heparin-binding epidermal growth factor, and by increasing the expression of IGF1 gene to promote synthesis of insulin-like growth factor 1.
The gorse extract can improve the expression of col4 alpha 1 gene, promote the synthesis of IV type collagen alpha 1 chain, and enhance the structure and function of basement membrane.
The sparrow extract promotes elastin synthesis by improving ELN gene expression, thereby promoting extracellular matrix secretion and maintaining skin elasticity.
The broom extract refers to extract containing effective components obtained by extracting in any reasonable manner.
The preparation method of the broom extract comprises the following steps: taking the broom as a raw material, extracting with a solvent, and taking a filtrate. The extraction solvent comprises water, alcohol or aqueous alcohol solution, preferably 60-85% (by volume) ethanol, and more preferably 70% ethanol.
Extracting for 1-3 times, preferably 2 times; the extraction time is 20 to 80 minutes, preferably 30 minutes.
The extraction method comprises ultrasonic method, leaching method, supercritical method, percolation method, reflux method or distillation method, wherein the ultrasonic method, leaching method and percolation method are carried out at room temperature. The extraction method is preferably ultrasonic extraction.
The dosage ratio of the broom and the solvent is 1 kg/5-20L, and the preferred dosage ratio is 1kg/10L.
The filtrate may be further concentrated or concentrated to dryness.
The concentration method is vacuum concentration.
The concentrating and drying method is vacuum drying or freeze drying.
The maidenhair is pulverized into particles with a particle size of 20-300 meshes, more preferably 50-200 meshes before extraction. Preferably, the broom is dried to a moisture content of less than 10wt%.
The broom refers to the flower, whole grass, root and rhizome of the broom or a mixture thereof, and is preferably the flower of the broom.
In a preferred embodiment of the present invention, the extract of broom is prepared by mixing pulverized broom with 70% ethanol (volume concentration) at a ratio of 1kg/10L, and ultrasonically extracting at room temperature for 30min for 2 times; the filtrates were combined, the solvent removed and dried.
The extract of the broom (canary flower) can be used for preparing a skin external preparation, and is used as an anti-aging active ingredient in the skin external preparation, in particular an active ingredient for repairing skin, an active ingredient for stabilizing skin structure or an active ingredient for maintaining skin elasticity.
The broom extract can be used for preparing skin external preparations for enhancing SIRT1 gene expression and promoting synthesis of histone deacetylase, for enhancing HBEGF gene expression and promoting synthesis of heparin-binding epidermal growth factor, for enhancing IGF1 gene expression and promoting synthesis of insulin-like growth factor 1, and for enhancing cell repair and/or proliferation activity.
The sparrow extract can be used for preparing a skin external agent for enhancing col4 alpha 1 gene expression and promoting IV type collagen alpha 1 chain synthesis, and can enhance basement membrane structure and function.
The flos Caraganae Sinicae extract can be used for preparing skin external preparation for enhancing ELN gene expression, promoting elastin synthesis, promoting extracellular matrix secretion, and maintaining skin elasticity.
The broom extract can promote skin repair and delay skin aging by improving the expression of cell repair related genes.
The sparrow extract stabilizes a basement membrane structure and enhances true epidermis connection on one hand by enhancing the expression of basement membrane related genes (such as col4 alpha 1 genes), thereby stabilizing the skin structure and realizing anti-aging; on the other hand, the composition promotes the generation and the secretion of extracellular matrix, strengthens and maintains the elasticity of the skin, thereby delaying the skin aging.
A skin external preparation has skin antiaging activity, especially skin repairing activity, and/or skin function stabilizing activity, and/or skin elasticity improving activity, and contains flos Caraganae Sinicae extract. Wherein, the content of the broom extract is 0.0001wt% -10 wt%, preferably 0.0005wt% -10 wt%; more preferably 0.001wt% to 5wt%.
Furthermore, when the extract is used for improving the expression of SIRT1 gene, HBEGF gene or col4 alpha 1 gene, the content of the broom extract is 0.0005wt% -10 wt%, preferably 0.001wt% -5 wt%; when the extract is used for improving the expression of IGF1 gene or ELN gene, the content of the broom extract is 0.005wt% -10 wt%, preferably 0.01wt% -5 wt%.
The skin external agent refers to a product which is spread on any part of the surface of the human body by smearing, spraying or the like, so as to clean, maintain, beautify, modify and change the appearance, or modify the smell of the human body and keep the good state. The skin external preparation can be basic cosmetics, facial cosmetics, head care products, and body care products for cleaning, caring, beautifying or caring skin.
The formulation of the external preparation for skin is not particularly limited, and may be determined according to the use, such as cleansing cream, essence, lotion, cream, lotion (such as smoothing toner, toner), mask, makeup remover, gel, aerosol, spray, or the like.

The skin external preparation further comprises one or more of thickener, solubilizer, cosolvent, antiseptic, surfactant, stabilizer, softener, aerosol solvent, aromatic, toner, penetration enhancer, controlled release agent, sustained release agent, humectant, emulsifier, shaping agent or pearling agent.
Compared with the prior art, the invention has the following advantages:
(1) The broom extract can improve the expression of cell repair and proliferation related genes, basement membrane stability related genes and skin elasticity related genes, has the effects of improving cell repair/proliferation activity, stabilizing basement membrane structure and function, promoting extracellular matrix secretion, keeping skin elasticity and the like, and can repair skin function, stabilize skin structure, enhance and maintain skin elasticity, thereby delaying skin aging. The broom extract has anti-aging effect, and has wide application prospect in skin external preparation, especially in cosmetics.
(2) The preparation process of the broom extract is simple, the process conditions are easy to control, and the preparation cost is low.
(3) The broom extract has high activity, can generate obvious effect under the condition of extremely low content, is natural, safe and effective, and does not irritate the skin.

Detailed Description
Example 1 preparation of a broom extract
10g of broom (flower) which is crushed to 50-200 meshes and dried until the water content is lower than 5% is put into 70% ethanol, and the mixture is ultrasonically extracted for 2 times at 500W, wherein 100mL of 70% ethanol is added each time and the extraction is carried out for 30min.
The two extraction filtrates were combined and the filtrate was rotary evaporated to dryness to give about 0.8g of the extract of maidenhair.
Example 2 efficacy verification of Genistis canadensis extract
1. Preparation of skin model
Human primary fibroblasts (NHDF) and human primary keratinocytes (NHEK) were extracted from skin tissue, respectively. Reviving human primary fibroblasts, mixing the human primary fibroblasts with collagen after culture and digestion, performing dermal stage culture for 3-4 days, digesting and inoculating human primary keratinocytes cultured by using a feeder layer to the surface of a dermal stage skin model, and performing epidermal immersion culture and gas-liquid phase culture in sequence for 18 days to obtain a 3D skin model for detection.
During the period from the surface culture of the skin model at the dermal-seeded stage of human primary keratinocytes to the 3D skin model formation days 3 to 18, the groups were divided according to the medium used, and a blank control (NT) group, a gorgonian extract (10 μ g/mL, about 0.001 wt%) group, and a gorgonian extract (100 μ g/mL, about 0.01 wt%) group were formed.
Wherein, the sparrow extract (10 μ g/mL) means that the amount of the obtained sparrow extract in a unit volume (mL) of the culture medium is 10 μ g, based on the dry weight of the sparrow extract, and is about 0.001wt%.
The extract of Genistis sinica (100 μ g/mL) refers to the amount of the obtained extract of Genistis sinica (100 μ g) in culture medium per unit volume (mL), and is about 0.01wt% based on the dry weight of the extract of Genistis sinica (L).

Clipping 3D skin model tissue about 2 x 3mm, and placing in a 2mL EP tube; adding 1mL of Trizol, adding 2 steel balls, homogenizing by using a tissue disruptor, and carrying out 50Hz ultrasonic treatment for 5min; centrifuging at 9000rpm and 4 deg.C for 1min, and transferring the supernatant to a new 1.5mL EP tube; adding 200 mu L of chloroform, slightly reversing for several times, uniformly mixing, and standing for 5 minutes at room temperature; centrifuging at 9000rpm and 4 deg.C for 15min; transferring the upper aqueous phase (about 400 μ L) into a new 1.5mL EP tube, adding isopropanol with the same volume, mixing, and standing at room temperature for 10min; centrifuging at 9000rpm and 4 deg.C for 10min; discarding the supernatant, washing the precipitate with pre-cooled 70% anhydrous ethanol for 1 time, and centrifuging at 8000rpm and 4 deg.C for 5min; discarding the supernatant, air-drying the precipitate for 5-10 min, and dissolving in 30 μ L of DEPC (diethyl cokenate) water; the RNA concentration was determined spectrophotometrically.
3. Analysis of Gene expression
RNA purification and quantitative analysis of genes were performed according to the QuantiGene R2.0 Plex Assay kit, and gene expression was analyzed using the luminex platform.
As can be seen from the results of the gene expression test, the expressions of the SIRT1, HB-EGF, col4 alpha 1, IGF1 and ELN genes were significantly increased after the addition of the extract of Gentiana malachite of the present invention, relative to the blank control group (NT). The histone deacetylase coded and synthesized by the SIRT1 gene can improve the cell repair activity, enhance the cell resistance and delay skin aging; the heparin-binding epidermal growth factor coded and synthesized by the HB-EGF gene can induce cell proliferation, promote wound repair and delay skin aging; the collagen type IV alpha 1 chain synthesized by the coding of the col4 alpha 1 gene can stabilize the structure and the function of a basement membrane and delay skin aging; IGF1 gene coded and synthesized insulin-like growth factor 1 promotes the proliferation and growth of cells, maintains the activity of the cells and delays the skin aging; the ELN gene encodes and synthesizes elastin which promotes the generation of extracellular matrix, maintains the elasticity of skin and delays skin aging. Therefore, the broom extract can improve the skin repair activity, stabilize the skin structure, promote the secretion of extracellular matrix and delay the skin aging.
(1) Increasing expression of cell repair/proliferation-related genes (SIRT 1 gene, HB-EGF gene and IGF1 gene)
As shown in FIG. 1, the expression of SIRT1 gene involved in histone deacetylase synthesis was up-regulated by 66% when the extract of Genistis sinica (10. Mu.g/mL) was added, compared to the control blank (NT). The histone deacetylase coded and synthesized by the gene can interact with a plurality of proteins, and can enhance the cell activity, improve the cell self-repairing activity, especially the wound repairing activity, and improve the cell viability in the process of resisting the stress of the external adverse environment. The gorse extract of the invention enhances SIRT1 gene expression, enhances the resistance and activity of skin, improves the skin repair activity and delays skin aging.
As seen from FIG. 2, the expression of HB-EGF gene associated with heparin-binding epidermal growth factor synthesis was up-regulated by 187% after adding the Genistein extract (10. Mu.g/mL) compared to the blank control group (NT). After the heparin-binding epidermal growth factor coded and synthesized by the gene is combined with an epidermal growth factor receptor, cell proliferation is induced, and wound repair and healing are promoted. The gorse extract disclosed by the invention can enhance HB-EGF gene expression, enhance skin repair activity and delay skin aging.
As can be seen from fig. 4, the expression of IGF1 gene associated with insulin-like growth factor 1 synthesis was up-regulated by 53% when the maidenhair extract (100 μ g/mL) was added compared to the blank control group (NT). The gene coded and synthesized insulin-like growth factor 1 can promote the proliferation and growth of cell, maintain the activity of cell and delay skin senility. The broom extract of the invention enhances IGF1 gene expression, and is helpful for delaying skin aging.
(2) Increasing expression of basement membrane associated gene (col 4 alpha 1 gene)
As shown in fig. 3, when the genistein flower extract (10 μ g/mL) was added, the expression of col4 α 1 gene involved in the synthesis of type IV collagen α 1 chain was up-regulated by 44% compared to the blank control group (NT). The alpha 1 chain of the IV type collagen synthesized by the gene coding is the main structural component of the basement membrane, thereby ensuring the structural stability of the basement membrane and maintaining the functional integrity of the basement membrane. The broom extract disclosed by the invention can enhance the expression of the col4 alpha 1 gene, stabilize the structure and function of a basement membrane and delay skin aging.
(3) Increasing expression of genes associated with skin elasticity maintenance (ELN genes)
As shown in fig. 5, the expression of ELN gene involved in elastin synthesis was up-regulated by 62% when the genistein flower extract (100 μ g/mL) was added compared to the blank control group (NT). The elastin coded and synthesized by the gene is an important component of extracellular matrix, promotes the generation of the extracellular matrix and maintains the skin elasticity. The broom extract disclosed by the invention can be used for improving the ELN gene expression, and is beneficial to enhancing and maintaining the skin elasticity and delaying the skin aging.
Claims (2)
1. The application of the broom extract as an active ingredient for improving the expression of cell repair and/or proliferation related genes, and/or improving the expression of basement membrane related genes, and/or improving the expression of skin elasticity related genes in the aspect of preparing skin external preparations is characterized in that the cell repair and/or proliferation related genes are SIRT1 genes, HBEGF or IGF1 genes, the basement membrane related genes are col4 alpha 1 genes, and the skin elasticity related genes are ELN genes.
2. The use according to claim 1, wherein the preparation method of the broom extract comprises: taking the gorse as a raw material, extracting with a solvent, and taking a filtrate; the extraction solvent comprises water, alcohol or aqueous alcohol solution.
