Abstract
本发明提供细胞毒活性更高的NK细胞。本发明的课题在于提供可以期待高效果的用于NK细胞疗法的药物组合物。本发明提供具备下述(1)及(2)的特征的NK细胞或其群体:(1)CD16阳性、CD56高表达、且CD57阴性;(2)NKG2C阳性、NKG2A阴性~低表达、及CD94阳性。本发明还提供药物组合物,其包含这样的NK细胞的群体、及治疗上有效量的抗体。
Description
技术领域
本发明涉及具有高细胞毒活性的自然杀伤细胞(NK细胞)及其应用。
背景技术
自然杀伤细胞(NK细胞)为作为自然免疫的主要因子工作的细胞毒性的淋巴细胞。已知人外周血NK细胞为CD16阳性,以低水平表达CD56,和CD57阳性(非专利文献1、2)。
作为基于NK细胞细胞毒性的机制之一,已知抗体依赖性细胞毒性(Antibodydependent cellular cytotoxicity,ADCC)。NK细胞在其细胞表面上具有Fc受体(CD16),而ADCC为NK细胞经由Fc受体与结合在靶标细胞的抗体进行结合,进行靶标细胞伤害的机制。
NK细胞在肿瘤细胞、病毒感染细胞的排斥中是重要的。将从患者自身提取的NK细胞,通过体外的培养使其增殖几百倍~数千倍,使其活化,输回患者的NK细胞疗法,作为副作用比较少的治疗方法而受到关注。通常,从正常的成人的外周血进行1次单采血液成分术(apheresis)可以回收约1×1010个单核细胞,如果使外周血单核细胞中的NK细胞的构成比率为约7%,则可得到7×108个的NK细胞。另一方面,如果设患者的体重为60kg,则也就是说需要6×106个~4.8×109个NK细胞。为此,推进了将从供体得到的NK细胞进行培养扩增,而得到对使靶标细胞被杀灭而言足够的NK细胞的技术的开发。例如,专利文献1提出了NK细胞的扩增方法,其特征在于包括:制备包含NK细胞的细胞群体的步骤,和从包含NK细胞的细胞群体中除去T细胞的步骤,和对进行除去后剩余的细胞利用作为细胞因子仅包含2500IU/mL至2813IU/mL的IL-2的培养基,而不使用饲养层细胞而进行培养的步骤。

发明内容
发明所要解决的问题
基于本发明人等的研究,则从外周血得到的初代NK细胞为CD16高表达,但在将NK细胞作为效应细胞(E)、K562细胞作为靶标细胞(T)而以混合比(E∶T)1∶1进行共培养的情况下,细胞毒活性为10~20%左右。希望获得细胞毒活性更高的NK细胞。
另外,对众多抗体药物而言,基于NK细胞的ADCC活性为其作用机制之一。为了NK细胞发挥ADCC活性,需要存在于NK细胞表面的CD16抗体与能诱导抗体依赖性细胞毒性的抗体进行结合。然而,至今仍没有对于关于将抗体药物和NK细胞疗法有效地组合的报告。

具体实施方式
[NK细胞、NK细胞群体]
本发明提供具备下述(1)的特征的NK细胞或其群体:
(1)CD16阳性、CD56高表达、且CD57阴性。
上述NK细胞或其群体还具备下述(2)的特征:
(2)NKG2C阳性、NKG2A阴性~低表达、及CD94阳性。
上述NK细胞或其群体可以进一步具备下述(3)的特征:
(3)在将该NK细胞作为效应细胞(E)、将K562细胞作为靶标细胞(T)以混合比(E∶T)1∶1进行共培养的情况下的细胞毒活性为50%以上。
NK细胞为不表达T细胞受体(TCR)、作为T细胞普遍的标记的CD3、及作为膜免疫球蛋白的B细胞受体的大型颗粒性淋巴细胞,通常在人中为CD16阳性、且CD56阳性。是否为NK细胞,本领域技术人员可以基于细胞表面标记的表达模式等容易地判断。NK细胞具有细胞毒活性,该细胞毒活性的有无、程度可以通过公知的各种方法测定。在称为NK细胞时,除了有特殊记载的情况以外,包括:外周血NK细胞、初代NK细胞、培养NK细胞、活化NK细胞、根据本发明得到的NK细胞等各种NK细胞。
<细胞群体>
细胞群体是指多个细胞,例如由1×105细胞以上的细胞构成的一群。NK细胞的群体为以超过50%的纯度(纯度(%)=NK细胞的个数/全部细胞的个数×100)包含NK细胞的群体。NK细胞在人的末梢血中通常占据淋巴细胞的10~30%。即,显然,外周血中的纯度为50%以下。因此,NK细胞的群体可以说在天然中是不存在的。NK细胞的群体还可以制备成各种细胞密度。可以制备为例如1×105细胞/mL以上。
根据本发明提供的NK细胞的群体中的NK细胞的纯度优选为60%以上、更优选为70%以上、进一步优选为80%以上。NK细胞的群体中包含的NK细胞的数量优选为1×106细胞以上、更优选为5×106细胞以上、进一步优选为1×107细胞以上。NK细胞的群体中包含的NK细胞的数量还能够使为适于对人的施用的1×106细胞~1×1010细胞。另外,NK细胞的群体中的细胞密度为1×105细胞/mL以上、优选为2×105细胞/mL以上、更优选为5×105细胞/mL以上、进一步优选为1×106细胞/mL以上。可以使细胞密度的上限值为例如1×1010细胞/mL以下。NK细胞的群体中的细胞密度还能够使为适于培养、冻存的1×106~1×108细胞/mL,还能够使为适于对人的施用的1×105~1×109细胞/mL。
<特征(1)>
根据本发明提供的NK细胞或其群体为CD16高表达、或CD16低表达。另外,根据本发明提供的NK细胞的群体可以包含为CD16高表达的NK细胞的群体,及为CD16低表达的NK细胞的群体中的两者。包含两者的细胞群体在利用流式细胞术对于CD16进行分析时,为双峰性。另外,根据本发明提供的NK细胞或其群体为CD56高表达。已知CD56为作为NK细胞的标记而有用的抗原。进一步在根据本发明提供的NK细胞或其群体中,未发现CD57的表达。CD57作为阶段5(活化型或成熟型)的标记而已知。
对于CD16等标记,为阳性有时用+表示,为阴性有时用-表示。例如,有时将CD16阳性表示为CD16+,将CD16阴性表示为CD16-。阳性包括为高表达的情况和为低表达的情况。为高表达有时用高、亮表示。例如,CD16高表达,有时表示为CD16高、CD16亮。低表达有时用低、暗表示。例如,CD16低表达,有时表示为CD16低、CD16暗。
阳性、阴性、高表达、低表达能够基于根据流式细胞术得到的图表而判断。在图表中出现的位置,可能根据仪器的电压设定、灵敏度设定、使用的抗体克隆、染色条件、使用的染料等不同而变动,但本领域技术人员,可以在得到的图表中,以将被认为是一组的细胞群体不分开的方式适当进行划线。
对目标的标记的表达而言,在为阳性还是为阴性的判断中,可以将使用了同种型对照(Isotype control)抗体的情况用作阴性对照而判断。同种型对照抗体为与特定的抗原不反应的抗体。一般而言,在使用了抗体的实验中,存在由于与靶标以外的蛋白质的非特异性结合、与细胞表面上的Fc受体的结合而产生背景的可能性。通过与使用了作为阴性对照的抗体的体系进行比较,由此使得明确相对于目标的抗原的一次抗体的反应为特异性。另外,能够排除背景的影响,准确解释信号的强度。
目标的标记的表达的程度(为低表达、或为高表达),可以通过与在相同条件下测定的对照细胞的结果的比较来判断。对照细胞的例子为如在本申请说明书的实施例的项中记载的初代(primary)NK那样的,从外周血得到的没有进行实质的培养的NK细胞。
例如,在某NK细胞的群体中的CD16的表达的程度能够通过以下进行判断:使用流式细胞术,将该细胞群体中的CD16表达量与从外周血得到的没有进行实质的培养的NK细胞的群体(对照,已知为CD16高表达)中的CD16表达量相比,将发现与对照为同等表达的情况判断为高表达,将比对照细胞表达低的情况判断为低表达。
另外,关于CD56,一般理解为阶段4的NK细胞(从干细胞起经过了pro-NK、pre-NK、不成熟NK的阶段的年轻NK细胞)为CD56亮,阶段5的NK细胞(如果在阶段4的NK细胞中转录因子MCM4发生活化,则NK细胞分化为阶段5)为CD56暗。因此,某NK细胞的群体中的CD56的表达的程度,也可以通过与阶段4的NK细胞的群体,及阶段5的NK细胞的群体中的至少一者相比而进行判断。
将至今已知的NK细胞和根据本发明得到的NK细胞的特征汇集于以下(参考非专利文献1、2)。

从外周血得到的初代的(primary)NK细胞为CD16高表达。另外,已知一般为CD16高表达的NK细胞为CD56暗/CD57+。至今尚无报告CD16高/CD56高/CD57-的NK细胞。
<特征(2)>
根据本发明提供的NK细胞还为NKG2C阳性、NKG2A阴性~低表达、及CD94阳性。NKG2A阴性~低表达、CD94的表达可在NK细胞的大部分观察到。CD94与NKG2家族的分子之一进行二硫键合,形成针对MHC类I分子的受体。已知NKG2C主要在NK细胞中表达,参与NK细胞的活化,CD94/NKG2C异二聚体作为活化受体发挥功能。CD94/NKG2A异二聚体为针对MHC类I的抑制性受体。在NKG2A阴性~低表达中,包括NKG2A为阴性的情况、有NKG2A的非常弱表达的情况、及NKG2A为低表达的情况。
对根据本发明提供的NK细胞或其群体而言,根据本发明人等的研究,在与由非专利文献1报告的NK细胞(参考上述a~d)的比较中,为CD94++,KIR+,且穿孔素+++。
根据本发明提供的NK细胞或其群体,可以除了(2)的特征以外,或代替(2)的特征而根据下述(2-1)与现有的NK细胞或其群体进行区别。
(2-1)KIR(s)(杀伤细胞免疫球蛋白样受体(Killer cell Immunoglobulin-likeReceptor(s)))阳性。
KIR(s)阳性是指选自由KIR2DL1、KIR2DL2、KIR2DL3、KIR2DL4、KIR2DL5、KIR3DL1、KIR3DL2、KIR3DL3、KIR2DS1、KIR2DS2、KIR2DS3、KIR2DS4、KIR2DS5、及KIR3DS1组成的组中的至少一个为阳性。
根据本发明提供的NK细胞或其群体,可以除了(2)的特征以外,或代替(2)的特征而根据下述(2-2)与现有的NK细胞或其群体进行区别。
(2-2)满足选自由NKp44阳性、NKp30高表达、及NKp46高表达组成的组中的任一项、优选为任2项、优选为全部。
NKp44通常在初代的NK细胞中为阴性。NKp30为诱发非MHC限制性的细胞伤害的NK特异性目标受体,与NKp46、NKp44一起为至今已鉴定的天然细胞毒性受体(NaturalCytotoxicity Receptor,NCR)之一。已知NKp30的表达与NKp46同时进行,为NKp46亮的NK细胞为NKp30亮。另外,已知NKp30亮与细胞伤害活性高相关。对于根据本发明提供的NK细胞或其群体利用(2-2)进行特征表征而言,可以参考本申请说明书的实施例1及图1。
需要说明的是,关于NK细胞的每个分化阶段表面标记的表达模式,可以参考以下文献。

<特征(3)>
根据本发明提供的NK细胞或其群体可发挥高细胞毒活性。除了有特殊记载的情况以外,细胞毒活性是指对象细胞(效应细胞,E)的针对靶标细胞(T)的溶解能力。细胞毒活性能够利用由于效应细胞而导致死亡的靶标细胞的百分率(%)表示,根据下式求出。
(与效应细胞共培养的情况下的细胞死亡-自然细胞死亡(阴性对照))/(最大细胞死亡(阳性对照)-自然细胞死亡(阴性对照))×100
在细胞毒活性的测定的时候,通常,根据效应细胞的细胞毒活性的程度等,对效应细胞和靶标细胞的混合比(E∶T)、效应细胞和靶标细胞的共培养的时间而言,可以根据使用的细胞种类、活性的强度而适当制定。NK细胞作为效应细胞时,存在靶标细胞为K562细胞、急性粒细胞白血病细胞、慢性粒细胞白血病细胞的情况,但不限于这些。效应细胞和靶标细胞、活细胞和死细胞可以利用被放射性物质、荧光染料等标记了的抗体等试剂进行区别以及定量。将NK细胞作为效应细胞时的细胞毒活性,可以通过例如将K562细胞作为靶标细胞,利用使E∶T=1∶0.05~10、优选为1∶0.1~5,使培养时间为0.5~18小时、优选为1~12小时的条件进行测定。
对根据本发明提供的NK细胞或其群体而言,在将靶标细胞作为K562细胞,利用E∶T=1∶1进行混合,在进行共培养1~3小时,更特定则为2小时的情况下的细胞毒活性为50%以上、优选为60%以上、更优选为70%以上。
从外周血得到的初代的NK细胞的细胞毒活性较低。至今尚无报告CD16高/CD56高/CD57-,且细胞毒活性高的NK细胞。
[NK细胞或其群体的制备方法]
本发明提供上述NK细胞或其群体的制备方法,其包括下述:
将初代NK细胞的群体利用包含IL-2的培养基进行培养的工序。
<初代NK细胞>
在本发明的制备方法中,初代NK细胞的群体可以通过从受试者提取的血细胞中分离单核细胞的工序得到。对血细胞而言,存在从外周血、脐带血、骨髓和/或淋巴结提取的情况。对血细胞而言存在从外周血利用单采血液成分术(apheresis)提取的情况。
在本发明的制备方法中,初代NK细胞的群体存在从选自由:源自选自由胚胎干细胞、成体干细胞及人工多功能性干(iPS)细胞组成的组中的任意干细胞的造血干细胞、源自脐带血的造血干细胞、源自外周血的造血干细胞、源自骨髓血的造血干细胞、脐带血单核细胞、外周血单核细胞组成的组中的至少1种细胞制备的情况。作为初代NK细胞群体的供体的受试者,存在源自为作为接受者的患者自身、或该患者的近亲属、或与患者没有血缘关系的人的情况。对受试者而言,存在健康的人和罹患疾病的患者的情况。对NK细胞而言,存在源自与接受者的主要组织适合性抗原(MHC)和杀伤免疫球蛋白样受体(KillerImmunoglobulin-like Receptor:KIR)不一致的供体的情况。
在本发明的制备方法中,也可以从初代NK细胞的群体中除去T细胞。对T细胞的除去而言,存在通过除去CD3阳性细胞的工序实现的情况。
在本发明的NK细胞的制备方法中存在包括从初代NK细胞的群体中除去造血祖细胞的工序的情况。从包含NK细胞的细胞群体中除去造血祖细胞的工序,存在通过将CD34阳性细胞除去的工序而实现的情况。
初代NK细胞的群体可以使用本领域技术人员已知的各种顺序进行制备。例如,在从如脐带血及外周血那样的血液中回收单核细胞的时候,可以使用密度离心法。另外,NK细胞可以使用免疫磁珠提取。进一步,初代NK细胞的群体,可以利用针对细胞表面标记的特异性的抗体进行免疫荧光染色,使用FACS(荧光激活细胞分选仪(Fluorescence activatedcell sorter))或流式细胞仪进行分离、鉴定。另外,初代NK细胞的群体也可以使用包括但不限于从Invitrogen公司销售的Dynal公司制Dynabeads(商标)、Miltenyi Biotech公司的CliniMACS(商标)的免疫磁珠,将表达细胞表面抗原CD3和/或CD34的细胞分离除去而制备。另外,也存在利用针对T细胞和/或造血祖细胞的特异性的结合配偶体,使T细胞和/或造血祖细胞选择性受伤或被杀灭的情况。需要说明的是,除去T细胞的工序,也可以为将其它细胞类型,例如:造血祖细胞、B细胞和/或NKT细胞与T细胞一起除去的工序。除去造血祖细胞的工序也可以为将其它细胞类型,例如:T细胞、B细胞和/或NKT细胞与造血祖细胞一起除去的工序。
<培养基>
用于培养初代NK细胞的群体而使用的细胞培养用培养基,包括:KBM501培养基(kohjin-bio株式会社,包含IL-2-1,750JRU/ml,人NK细胞原代培养用)、CellGro SCGM培养基(CellGenix,岩井化学药品株式会社)、X-VIVO15培养基(lonza,TAKARABio株式会社)、Cosmedium008(Cosmo Bio,包含IL-2-1,750JRU/ml,人NK细胞原代培养用)、CTS AIM VMedium GibcoTM CTSTMAIM VTM Medium(Thermo Fisher Scientific。用于进行T细胞及树突细胞增殖和操作的已知组成的无血清培养基)、CTS OpTmizer T Cell Expansion BasalMedium(Thermo Fisher Scientific,人T淋巴细胞的生长及增殖用)、IMDM、MEM、DMEM、RPMI-1640等,但不限于这些。
存在在培养基中利用能够实现本发明的目标的浓度添加白介素-2(IL-2)的情况。IL-2的浓度存在为2500IU/mL~2813IU/mL的情况。IL-2优选具有人的氨基酸序列,从安全方面优选利用重组DNA技术进行生产。IL-2的浓度存在以日本国内标准单位(JRU)及国际单位(IU)表示的情况。由于1IU为约0.622JRU,因此现有的培养基的1750JRU/mL相当于约2813IU/mL。
存在在培养基中,添加受试者的自体血清、从BioWhittaker公司以外的能够获取的人AB型血清、从日本红十字会能够获取的献血人血清白蛋白的情况。自体血清及人AB型血清优选为以1至10%的浓度添加,献血人血清白蛋白优选为以1至10%的浓度添加。
在培养基中,存在以不破坏NK细胞的扩增效果为条件而包含适当的蛋白质、细胞因子、抗体、化合物、其它成分的情况。细胞因子存在为:白介素3(IL-3)、白介素7(IL-7)、白介素12(IL-12)、白介素-15(IL-15)、白介素-21(IL-21)、干细胞因子(SCF)、和/或FMS样酪氨酸激酶3配体(Flt3L)的情况。IL-3、IL-7、IL-12、IL-15、IL-21、SCF及Flt3L优选具有人的氨基酸序列,从安全方面优选利用重组DNA技术进行生产。
培养基优选为无血清培养基。无血清培养基优选包含血清白蛋白、转铁蛋白及胰岛素。用于培养淋巴细胞的无血清培养基已经开发、市售,在本发明中可以利用它们。无血清培养基的优选例之一为在基础培养基中作为支持人T细胞的增殖的组成而添加了市售的CTS Immune Cell SR(Thermo Fisher Scientific)而成的那些。
对培养基的交换而言,以能够得到期望的NK细胞的细胞数作为条件,可以在培养开始之后任何时候进行,优选为每3~5天。
培养时使用的培养容器包括能够商业获取的平皿、烧瓶、板、多孔板,但不限于这些。对培养条件而言,以不破坏NK细胞的扩增效果为条件,没有特别限定,通常为在37℃,5%CO2及饱和水蒸气气体氛围中的培养条件。由于利用培养基进行培养的时期越长就能够得到更多的NK细胞,因而有利。对培养时期而言,以将NK细胞扩增至期望的细胞数为止作为条件,没有特别限定。
对通过培养得到的指定的NK细胞的群体而言,存在除了目标的NK细胞以外,包含NK细胞前体、T细胞、NKT细胞、造血祖细胞等的情况。培养之后,存在使用例如密度离心法、免疫磁珠、FACS、流式细胞术等进行选择的情况。存在使用例如:抗CD3抗体、抗CD16抗体、抗CD34抗体、抗CD56抗体、抗CD69抗体、抗CD94抗体、抗CD107a抗体、抗KIR3DL1抗体、抗KIR3DL2抗体、抗KIR2DL3抗体、抗KIR2DL1抗体、抗KIR2DS1抗体、抗KIR2DL5抗体、抗NKp46抗体、抗NKp30抗体、抗NKG2D抗体等,将目标的NK细胞或其群体选择性地分离的情况。存在抗体为单克隆抗体、多克隆抗体等的情况。目标的NK细胞或其群体的选择,存在将T细胞、NKT细胞、造血祖细胞、其它细胞选择性除去而进行的情况。
[NK细胞或其群体的应用]
本发明提供药物组合物,其包含NK细胞的群体、及药学上允许的添加物。本发明还提供药物组合物,其包含NK细胞的群体、及治疗上有效量的抗体。作为药学上允许的添加物,可例示例如:等渗剂、pH调节剂、缓冲剂、稳定剂、冷冻保护剂、抗生素等。具体而言,可列举:水、乙醇、氯化钠、葡萄糖、白蛋白等。本发明的药物组合物中包含的NK细胞的群体优选为上述根据本发明提供的NK细胞的群体。另外,本发明的药物组合物中包含的NK细胞的群体可以为CD16高表达,在任一情况下,均更优选为细胞毒活性高的NK细胞的群体。
本发明的药物组合物中使用的抗体优选为能够作为抗体药物使用的那些。另外,优选为能够诱导抗体依赖性细胞毒性(Antibody-Dependent-Cellular-Cytotoxicity,ADCC)的那些。在本发明的药物组合物中使用的抗体可以为小鼠抗体、嵌合抗体、人源化抗体、人抗体中的任一种,优选为人源化抗体或人抗体。使用的抗体还可以被修饰,可以为利用Potelligent技术(除去IgG的Fc区域的岩藻糖)制作而成的那些。

在本发明的药物组合物中使用的抗体优选为与CD16亲和性高的那些。另外,在本发明的药物组合物中,优选抗体的至少一部分与NK细胞结合。基于本发明人等的研究,在作为抗体使用了采用Potelligent技术制作而成的莫加利珠单抗(mogamulizumab)的情况下,在NK细胞与靶标细胞的共培养中存在抗体的体系,以及预先将NK细胞与抗体混合,在与靶标细胞进行共培养前将NK细胞洗涤后(不与细胞结合的抗体被除去)进行共培养的体系中,在后者中观察到了更高的细胞毒活性。其一方面,在同条件下使用了达妥昔单抗的情况下,没有观察到由预先将NK细胞与抗体混合带来的效果。这提示的是,与CD16亲和性高的抗体可通过预先与NK细胞混合而在NK细胞上进行抗体结合,由此发挥高ADCC活性。需要说明的是,认为如果对条件进行研究,达妥昔单抗也存在得到同样的效果的可能性。
本发明的药物组合物存在在使用时进行制备的情况。例如,本发明的药物组合物可以以将NK细胞的群体与抗体保持于不同容器的状态,在即将进行对对象的施用前至数小时前通过混合而制备。
本发明的药物组合物可以在将NK细胞群体与抗体混合后,经过将不与NK细胞结合的抗体除去的工序而制备。即,本发明的药物组合物的优选实施方式之一,为包含NK细胞和抗体,其中,抗体与NK细胞结合,实质上不包含与NK细胞不结合的抗体。
认为抗体药物多为在静脉内施用之后显示基于ADCC活性的抗肿瘤效果。其一方面,在发挥ADCC活性的时候,除了NK以外,单核细胞/巨噬细胞、中性粒细胞也被动员。并且,除了NK细胞以外的效应子对正常细胞和癌细胞无区别地显示ADCC活性,认为其也与副作用的发生相关联。在本发明中,通过将NK细胞与抗体在施用前进行混合,由此使抗体预先装备至NK细胞。因此,认为效应子被限定于NK细胞,能够减少施用的抗体的量,另外,对副作用的减少也极其有效。
对本发明的药物组合物而言,存在对具有与根据本发明的制备方法制备的NK细胞不同的HLA基因型的患者进行施用的情况。
药物组合物典型的为将NK细胞悬浊于溶液而成的形态。对用于悬浊NK细胞的溶液而言,例如:包含DMSO的冷冻用保护液、生理盐水、磷酸缓冲生理盐水(PBS)、培养基、血清等是通常的。溶液存在包含作为药物及准药而在药学上允许的载体的情况。
本发明的药物组合物存在用于治疗传染病或癌症而使用的情况。本发明的药物组合物另外可以适用于对NK细胞具有敏感性的各种疾病的治疗和/或预防。疾病例如包括:口腔癌、胆囊癌、胆管癌、肺癌、肝癌、结肠癌、肾癌、膀胱癌、白血病、基于病毒、细菌等的传染病,但不限于这些。本发明的细胞疗法存在单独,或与外科疗法、化学疗法、放射疗法、抗体药物等组合而实施的情况。在使用了本发明的药物组合物的细胞疗法中,存在NK细胞被施用向例如:静脉、动脉、皮下、腹膜内等的情况。
本发明的药物组合物的制造,优选根据适合于药物及准药的制造管理及品质管理规定的条件(良好操作规范,good manufacturing practice,GMP)及再生医疗等产品的制造管理及品质管理的基准(良好的基因、细胞和基于组织的产品制造实践(Good Gene,Cellular,and Tissue-based Products Manufacturing Practice),GCTP)而实施。
在本发明中,对于脐带血及外周血的全血的提取、和自体血清的制备、和从全血的单核细胞的制备、和该单核细胞的培养前后的细胞数的测定、和培养前后的单核细胞中的NK细胞、T细胞、造血祖细胞其它细胞类型的构成比率的测定、和NK细胞的扩增倍率的算出、和测定误差、有统计学意义的统计分析而言,可以采用本领域技术人员众所周知的任何方法实施。
以下进行说明的本发明的实施例仅为例示的目的,并不意在对本发明的技术范围进行限定。本发明的技术范围仅由利用权利要求书的记载而限定。在不脱离本发明的主旨的条件下,能够进行本发明的变更,例如,本发明的构成条件的追加、删除及取代。
实施例
[实施例1]
从健康人志愿者实施采血,使用Ficoll(GE Healthcare,17144002)分取得到的外周血单核细胞。向得到的外周血单核细胞添加CD3珠※1并悬浊,于4℃,培养15分钟之后,加入分离缓冲液1※2充分悬浊,于300xg进行离心10分钟。除去上清,悬浊于1mL的分离缓冲液中,添加至预先添加分离缓冲液2mL而润湿的LD柱(Miltenyi Biotech,130-042-901),回收了来自LD柱的洗脱液。进一步向LD柱添加分离缓冲液1mL,回收了洗脱液。之后将柱洗涤2次,计数回收的液中的细胞数,算出总细胞数。于500xg进行离心5分钟,除去上清之后,以使得为5x105细胞/mL的方式悬浊于NK培养基I※3中。将一部分的细胞进行回收作流式细胞仪测定用(将在此回收的细胞表示为“初代NK”),其余的细胞进行培养。培养使用6孔板(ThermoFisher Scientific,140675)或T-75烧瓶(Thermo Fisher Scientific,156499)利用CO2培养箱进行(37℃,5%CO2)。在培养第5天及第9天取培养液的一部分对细胞数进行计数,以使得细胞密度为5x105细胞/mL的方式添加了NK培养基I。在第14天回收细胞(将在此回收的细胞表示为“第14天”),使用一部分的细胞利用流式细胞仪进行了细胞表面抗原的测定。其余的细胞在悬浊于STEM-CELLBANKER GMP级(TAKARABio,CB045)中后,于-80℃保存。
将回收到的细胞利用以下抗体进行了染色:
将BV421标记的抗人CD56抗体(Biolegend,318328)、PerCP/C 5.5标记的抗人CD3抗体(Biolegend,300430)、APC标记的抗人NKp30抗体(Biolegend,325209)、PE标记的抗人NKp44抗体(Biolegend,325107)、FITC标记的抗人NKp46抗体(Biolegend,331921)、PE-Cy7标记的抗人NKG2D抗体(Biolegend,320811)、PE-Cy7标记的抗人CD16抗体(Biolegend,302015)、APC标记的抗人CD69抗体(Biolegend,310910)、FITC标记的抗人CD94抗体(Biolegend,305504)、FITC标记的抗人CD14抗体(Biolegend,325604)、PE标记的抗人NKG2C抗体(R&D,FAB138P)、APC标记的抗人NKG2A抗体(Milteni Biotec,130-098-809)、PerCPCy5.5标记的抗人CD94抗体(Biolegend,305514)、APC标记的抗人CD57抗体(Biolegend,359610)以1μg/mL的浓度于4℃,染色30分钟之后,离心(500xg,5分钟,4℃),除去上清,悬浊于PBS(和光纯药工业)中后,使用流式细胞仪(BD LSRFortessa,BD Bioscience公司)进行了测定,利用FlowJo软件(TreeStar)进行了分析。
※1:CliniMACS CD3,Miltenyi Biotech公司,产品目录编号130-017-601(每1x107细胞5μ)
※2:包含0.5%人AB型血清(Cosmo Bio,12181301,于56℃经30分钟的灭活处理而成)、2mM EDTA(Thermo Fisher Scientific,15575-020)的PBS
※3:添加了5%人AB型血清(Cosmo Bio,12181301,于56℃经30分钟的灭活处理而成)的Cosmedium 008(Cosmo Bio,COS-008)
将结果示于图1。第14天的NK细胞呈现强表达NKp46、NKp30,NKG2D等活化型受体,即所谓高活化NK细胞的面貌。其一方面,CD16的表达为双峰性。
[实施例2]
利用实施例1中记载的方法进行了细胞的制备。将培养开始时的细胞和培养结束时(培养第14天)的细胞的一部分回收,利用流式细胞仪进行了细胞表面抗原的测定。
将回收的约1x105个的细胞利用BV421标记的抗人CD56抗体、PerCP/C 5.5标记的抗人CD3抗体、PE-Cy7标记的抗人CD16抗体、APC标记的抗人CD69抗体、FITC标记的抗人CD14抗体以1μg/mL的浓度于4℃,染色30分钟之后,离心(500xg,5分钟,4℃)除去上清,悬浊于适量的PBS中之后,使用流式细胞仪(BD LSRFortessa,BD Bioscience公司)进行了测定,利用FlowJo软件进行了分析。将CD3阴性CD56阳性的细胞作为NK细胞,利用CD16和CD56进行展开而分析了NK细胞上的CD16的表达。
将结果示于图2A~C。在第14天的NK细胞中,包含CD56高/CD16高NK细胞((i)群体)、和CD56高/CD16低NK细胞((ii)群体)(图2A),另外,没有发现通常被作为阶段5(活化型或成熟型)的标记的CD57的表达(图2B)。发现第14天的NK细胞进一步具备为NKG2C阳性、NKG2A阴性~低表达、CD94阳性的特征(图2C)。
[实施例3]
对实施例1中记载的方法进行一部分改变而进行了细胞的制备。具体而言,使用分离缓冲液2※4,使用NK培养基II※5进行了培养。将培养开始时的细胞和培养结束时(培养第14天)的细胞的一部分回收,通过实施例2中记载的方法进行了测定及分析。
※4:包含0.5%CTSTM Immune Cell SR(Thermo Fisher Scientific,A25961-01,下文作为CTS)、2mM EDTA的PBS
※5:添加了5%的CTS的Cosmedium 008培养基(Cosmo Bio,COS-008)
将结果示于图3。利用添加了CTS的无血清培养基,成功将(i)群体占全部NK细胞的比例提高了。
[实施例4]
<K562(靶标细胞)的制备>
将K562细胞(人慢性粒细胞白血病细胞株)利用包含10%FBS(NichireiBioscience,171012-500ML)及100单位的青霉素、100μg/mL的链霉素(Nacalai Tesque,26253-84)的RPMI1640培养基(和光纯药工业,189-02025)(下文作为10%FBS/RPMI1640))以1x106细胞/ml的浓度进行了制备。向制备的K562细胞以使得终浓度为30μM的方式添加了DiOC18(Sigma,D4292-20MG),于37℃使其反应了10分钟。在离心(400xg,5分钟,室温)后,除去上清,添加10%FBS/RPMI1640并悬浊。将该洗涤工序重复3次,最终使用10%FBS/RPMI1640以使得为2x106细胞/mL的方式进行了制备。
<NK细胞(效应细胞)的制备>
将从健康人志愿者按照实施例1中记载的方法得到的细胞回收洗涤之后,利用10%FBS/RPMI1640进行悬浊,利用同样的培养基以1x106细胞/ml的浓度进行了制备。将制备的细胞悬浊液利用PE-Cy7标记的抗人CD16抗体(1μg/mL)于4℃,染色30分钟之后,离心(500xg,5分钟,4℃),除去上清。将细胞利用10%FBS/RPMI1640进行悬浊之后,以2x107细胞/ml的浓度使用同样的培养基进行制备,使用流式细胞仪(细胞分选仪SH800,SONY)进行了细胞表面抗原的测定,确认了CD16的表达程度。将CD16高NK细胞和CD16暗NK细胞利用同样的细胞分选仪进行分选,分别回收至填充了10%FBS/RPMI1640 7ml的15ml锥形管中。回收之后,离心(500xg,5分钟,4℃),除去上清之后,利用10%FBS/RPMI1640进行悬浊,利用同样的培养基以2x106细胞/ml的浓度进行了制备。
<细胞毒活性的测定>
也准备了没有进行分选的NK细胞和K562细胞的组、CD16高NK细胞和K562细胞的组、CD16暗NK细胞和K562细胞的组、各自的仅NK细胞计6组,以及作为阴性对照的仅K562细胞的组,作为阳性对照将K562细胞利用10%福尔马林进行了固定的组。将NK细胞和K562细胞以使得以细胞比计为1∶1的方式添加至96孔板,混合,于37℃共培养了2小时。此时,首先将K562细胞添加至板,接着将200μg/ml的APC标记的抗人CD107a抗体※6(Biolegend,328620)以使得为终浓度1μg/ml的方式添加,最后添加了各NK细胞。培养之后,离心(500xg,5分钟,4℃),除去上清之后,添加利用PBS稀释的7-AAD溶液(Beckman Coulter,A07704)并悬浊,于室温培养10分钟。使用流式细胞仪(BD LSR Fortessa,BD Bioscience公司)进行了测定,利用FlowJo软件进行了分析。
细胞毒活性利用下式进行计算。需要说明的是,在以下实施例中也同样地计算。
(在与效应细胞培养的情况下的靶标细胞的细胞死亡-自然细胞死亡(阴性对照))/(最大细胞死亡(阳性对照)-自然细胞死亡(阴性对照))×100
※6:由于CD107a存在于NK细胞内颗粒,在脱颗粒(释放穿孔素、颗粒酶)时向细胞膜表面上转移,因此CD107a为阳性的情况间接地表示NK攻击了对象。
<结果>
将结果示于图4。(i)组、(ii)组细胞毒活性均超过了50%。
[实施例5]
<IMR32细胞(靶标细胞)的制备>
将IMR32细胞(人MYCN扩增的神经母细胞瘤细胞系)利用10%FBS/RPMI1640而制备成1x106细胞/ml的浓度。向制备的K562细胞以使得终浓度为30μM的方式添加了DiOC18(Sigma,D4292-20MG),于37℃使其反应了10分钟。在离心(400xg,5分钟,室温)后,除去上清,添加10%FBS/RPMI1640并悬浊。将该洗涤工序重复3次,最终使用10%FBS/RPMI1640以使得为2x106细胞/mL的方式进行了制备。
<NK细胞(效应细胞)的制备>
将从健康人志愿者按照实施例1中记载的方法得到的细胞回收洗涤之后,利用10%FBS/RPMI1640进行悬浊,利用同样的培养基制备成1x106细胞/ml的浓度。将制备液利用PE-Cy7标记的抗人CD16抗体(1μg/mL)于4℃,染色30分钟之后,离心(500g,5分钟,4℃),除去上清。将细胞利用10%FBS/RPMI1640进行悬浊之后,利用同样的培养基制备为2x107细胞/ml的浓度,利用流式细胞仪(细胞分选仪SH800,SONY)进行了细胞表面抗原的测定,确认了CD16的表达程度。将CD16高NK细胞和CD16暗NK细胞利用同样的细胞分选仪进行分选,分别回收至填充了10%FBS/RPMI1640 7ml的15ml锥形管中。回收之后,离心(500g,5分钟,4℃),除去上清之后,利用10%FBS/RPMI1640进行悬浊,利用同样的培养基制备成2x106细胞/ml的浓度。
<细胞毒活性的测定>
也准备了没有进行分选的NK细胞和K562细胞的组、CD16高NK细胞和K562细胞的组、CD16暗NK细胞和K562细胞的组、各自的仅NK细胞计6组,以及作为阴性对照的仅K562细胞的组,作为阳性对照将K562细胞利用10%福尔马林进行了固定的组。将NK细胞和K562细胞以使得以细胞比计为1∶1的方式添加至96孔板,混合,于37℃共培养了2小时。
此时,首先将IMR32细胞添加至板,接着将利用PBS制备成100μg/ml的达妥昔单抗(商品名Unituxin,United Therapeutics公司)各添加1μl(达妥昔单抗添加组),接着以使得终浓度为1μg/ml的方式添加了200μg/ml的APC标记的抗人CD107a抗体,最后添加了各NK细胞。共培养之后,离心(500g,5分钟,4℃),除去上清之后,添加利用PBS稀释的7-AAD溶液(Beckman Coulter,A07704)并悬浊,于室温培养10分钟。使用流式细胞仪(BD LSRFortessa,BD Bioscience公司)进行了测定,利用FlowJo软件进行了分析。
<结果>
将结果示于图5。在达妥昔单抗添加组中发现了更高的细胞毒活性。
[实施例6]
<细胞的制备>
将利用实施例1的方法得到的NK细胞悬浊于包含4μg/mL的浓度的莫加利珠单抗(Poteligeo(注册商标)点滴静脉注射20mg,协和发酵kirin)的NK培养基I中,于37℃培养20分钟。离心(500xg,5分钟,室温)之后,除去上清,添加10%FBS/RPMI1640并悬浊之后,离心(500xg,5分钟,室温),除去上清,利用10%FBS/RPMI1640以使得为2x106细胞/mL的方式进行制备,制成了NK+莫加利珠单抗(预混合)。
向Hut78细胞(人T淋巴瘤细胞株)中以使得终浓度为1μm的方式添加了DiOC18(Sigma,D4292-20MG),于37℃使其反应10分钟。离心(500xg,5分钟,室温)之后,除去上清,添加10%FBS/RPMI1640并悬浊。将该洗涤工序重复3次,最终使用10%FBS/RPMI1640以使得为2x106细胞/mL的方式进行了制备。
<细胞毒活性的测定>
准备了相对于NK细胞1使Hut 78细胞为5(细胞数比)的组、相对于NK细胞1使Hut78细胞为5(细胞数比)的基础上以使得终浓度为4μg/mL的方式添加了莫加利珠单抗的组、相对于NK+莫加利珠单抗(预混合)1使Hut 78细胞为5(细胞数比)的组,计3组。以细胞比1∶5添加至96孔板,混合,于37℃共培养了2小时。也准备了作为阴性对照的仅Hut 78细胞的组,作为阳性对照将Hut 78利用10%福尔马林进行了固定的组。在2小时的共培养之后,离心(500xg,5分钟,室温),除去上清,以60μL/孔添加了利用PBS稀释为6倍的7-AAD溶液(Beckman Coulter,A07704)并悬浊,于培养室温10分钟。使用流式细胞仪(BDLSRFortessa,BD Bioscience公司)进行了测定,利用FlowJo软件进行了分析。
<IMR32细胞的制备、细胞毒活性的测定>
按照上述Hut78的实验进行。条件不同的是:使用的细胞株为IMR32的方面,使用的抗体为达妥昔单抗的方面,NK细胞与肿瘤细胞的细胞数比为1∶10的方面。
<结果>
将结果示于图6。关于莫加利珠单抗,在NK+莫加利珠单抗(预混合)组中发现了最高的细胞毒活性。虽然众多抗体药物将NK细胞的抗体依赖性细胞毒性(ADCC)活性作为其作用机制之一,但提示了预先将NK细胞与抗体药物混合的有效性。
[实施例7]
将利用实施例1中记载的方法得到的NK细胞使用STEM-CELLBANKER GMP级(TAKARABio,CB045)进行冷冻之后,进行解冻而使用NK培养基II利用37℃的CO2培养箱进行了过夜培养。将细胞利用包含1mM EDTA(Thermo Fisher Scientific,15575-020)的PBS剥落,向圆底的96孔板(IWAKI)中各分别注入1x105个,离心(500xg,5分钟,室温)之后,除去上清,添加100μL的莫加利珠单抗溶液(终浓度:0μg/mL、1μg/mL、10μg/mL、100μg/mL、1,000μg/mL)并悬浊,于室温培养1小时。离心(500xg,5分钟,室温)之后,除去上清,利用300μL的PBS进行了3次洗涤操作(500xg,5分钟,室温)。接着,添加染色用的抗体溶液(BV421标记的抗人CD56抗体、PerCP/C 5.5标记的抗人CD3抗体、PE-Cy7标记的抗人CD16、PE标记的抗人IgG Fc抗体(Southern Biotech,2043-09)并悬浊,于4℃使其反应30分钟。离心(500xg,5分钟,室温)之后,除去上清,添加100μL的PBS并悬浊,使用流式细胞仪(BD LSRFortessa,BD Bioscience公司)进行了测定,利用FlowJo软件进行了分析。
将结果示于图7。确认了NK细胞与莫加利珠单抗结合。不仅(i)组,在莫加利珠单抗为高浓度的条件下,也使(ii)组与莫加利珠单抗结合。
Claims (12)
1.NK细胞或其群体,其具备下述(1)及(2)的特征:
(1)CD16阳性、CD56高表达、且CD57阴性;
(2)NKG2C阳性、NKG2A阴性~低表达、及CD94阳性。
2.根据权利要求1所述的群体,其包含CD16高表达的NK细胞的群体,及CD16低表达的NK细胞的群体。
3.根据权利要求1所述的NK细胞或其群体,其中CD16高表达。
4.根据权利要求1所述的NK细胞或其群体,其中CD16低表达。
5.根据权利要求1~4中任一项所述的NK细胞或其群体,其进一步具备下述(3)的特征:
(3)将该NK细胞作为效应细胞(E)、将K562细胞作为靶标细胞(T)以混合比(E∶T)1∶1进行共培养的情况下的细胞毒活性为50%以上。
6.根据权利要求1~5中任一项所述的NK细胞或其群体的制备方法,其包括下述:
将初代NK细胞的群体利用包含IL-2的培养基、或无血清培养基进行培养的工序。
7.根据权利要求6所述的制备方法,其中,初代NK细胞的群体为经过除去CD3阳性细胞的工序的群体。
8.药物组合物,其包含权利要求1~5中任一项所述的NK细胞的群体、及药学上允许的添加物。
9.药物组合物,其包含NK细胞的群体、及治疗上有效量的抗体。
10.根据权利要求9所述的药物组合物,其中,NK细胞的群体为权利要求1~5中任一项所述的NK细胞的群体。
11.根据权利要求9或10所述的药物组合物,其中,抗体为能诱导抗体依赖性细胞毒性(ADCC)的抗体。
12.根据权利要求9~11中任一项所述的药物组合物,其中,抗体的至少一部分与NK细胞结合。
