Description
本発明にかかるヒトリンパ球は、少なくともインターロイキン2およびインターロイキン18によって刺激されてなるヒトリンパ球であって、CD56の発現強度が102以上であり、かつ、CD11cの発現強度が101以上102以下であり、上記発現強度はフローサイトメトリーによって測定したものである。
本発明にかかるヒトリンパ球の製造方法は、少なくともインターロイキン2およびインターロイキン18によって、CD3の発現強度が100以上102.3以下であり(つまり、CD3の発現が陰性であり)、かつ、CD11cの発現強度が101以上102以下であるヒトリンパ球(以下、被刺激リンパ球とも称する)を刺激する工程を含む方法である。
本発明にかかるγδT細胞の増殖方法は、本発明にかかるヒトリンパ球と、γδT細胞とをインターロイキン2、インターロイキン18およびリン酸化合物を用いて刺激する工程を含む方法である。
以下の実施例において用いた試薬類および実験方法は下記のとおりである。
組換えヒトIL−18は、Glaxo-SmithKline社製、IL−2はR&D Systems社製、ゾレドロネートはNovartis Pharma社製のものを用いた。
ヒト末梢血単核球(PBMCs)は、健康なドナー(25歳〜40歳)の血液から、室温(摂氏25℃)で、スイングローターTS−7(トミー精工)を用い、Ficoll−Hypaque密度勾配遠心法によって分離した。PBMCsは、ストレプトマイシン、ペニシリン、グルタミン酸、および5%AB血清を添加したAlyS505N−O培地(Iscov MEMをベースにした無血清培地。(株)細胞科学研究所製)中で、37℃、5%CO2雰囲気下で培養した。
CD3、γδ−T細胞レセプター、Vδ2、CD11a、CD18、CD11c、CD25、CD28、CD40、CD40L、CD56、CD80、CD83、CD86、CD122、HLA−DR、NKG2D、IL−18Rα、IL−18Rβのそれぞれに対する、FITC、PE、APCおよびビオチンによって標識した抗体によって、細胞を4℃で20分間染色し、フローサイトメーター(使用装置: BECTON DICKINSON, FACS Calibur(登録商標))を用いて解析した。データはCell Questソフトウェア(BDバイオサイエンス製)を用いて解析した。
細胞障害活性は、DELFIA EuTDA細胞障害活性試薬(登録商標、PerkinElmer製)を用いて測定した。該試薬は、蛍光を増強するリガンドであるBATDAを加水分解させ、親水性リガンドであるTDAを細胞内部に形成させる試薬である。ターゲット細胞としては、NK細胞のターゲット細胞として知られるK562細胞を用いた。K562細胞は、フェノールレッドを含有せず、2%FCSを含有するRPMI1640培地中で、1×104cells/wellとなるように96穴プレートに播種した。エフェクター細胞は、後述する実施例3で用いた「CD3陽性細胞を除去したPBMCs」2.5×105個/ml〜1×106個/mlを、IL−2(10ng/ml)およびIL−18(50ng/ml)によって刺激することによって誘導した。エフェクター細胞は、エフェクター細胞の細胞数とターゲット細胞の細胞数との割合が3:1、10:1、または30:1となるように培養系に添加し、さらに3時間培養した。
<サイトカインのアッセイ>
IFN−γ、TNF−α、CCL21は、ELISA法を用いてアッセイした(使用装置:BioLegend製ELISAキット)。
データは、平均値±標準偏差で表し、スチューデントのT検定またはボンフェローニの多重比較を用いて解析した。p<0.05で有意差ありとした。
<PBMCs中のγδT細胞の増殖および本発明にかかるヒトリンパ球の増殖に対するIL−18の効果>
5%のヒトAB型血清を含有するPRMI1640培地にゾレドロネートまたは2M3BPPを最終濃度1μMとなるように加え、IL-2(最終濃度5ng/ml)、または、IL−2およびIL−18(IL−2の最終濃度5ng/ml、IL−18の最終濃度50ng/ml)を添加した培養液に、健康なドナーから調製したPBMCsを2.5×105〜1×106個/mlとなるようにサスペンドした。図3の(a)に示すように、γδT細胞は上記PBMCs中に約2%含まれている。
<CD56陽性、CD11c陽性、またはCD14陽性のヒトリンパ球を除去した場合の、本発明にかかるヒトリンパ球の増殖およびγδT細胞の増殖>
本実施例では、γδT細胞の増殖に対するアクセサリー細胞の関与について検討した。γδT細胞の数は加齢に伴い減少するので、40歳以下のヒトから得たPBMCsを実験に用いた。
<(3)CD3陽性細胞を除去したPBMCsにおける、本発明にかかるヒトリンパ球のIL−18による増殖>
上記ヒトリンパ球Aの細胞数がγδT細胞の細胞数と並行して増加する現象(図5)は、一方の細胞が他方の増殖を助けているか、相互に増殖を促進しあっているか、ということを示唆しているため、以下の実験において検討した。
<本発明にかかるヒトリンパ球の特徴に関する検討>
本実施例では、上記ヒトリンパ球Aの特徴を調べた。図12は、CD3陽性細胞を除去したPBMCsをIL−2、または、IL−2およびIL−18で10日間刺激し、表面分子の発現の変化について検討した結果を示すものである。
<本発明にかかるヒトリンパ球によって生産されたサイトカインおよびケモカイン、ならびに本発明にかかるヒトリンパ球の細胞障害活性に関する検討>
実施例3で用いた「CD3陽性細胞を除去したPBMCs」を、GM−CSFおよびIL−4、IL−2、IL−2およびIL−18、IL−2および抗IL−18Rα抗体またはIL−18の存在下で6日間培養し、培養上清中のサイトカインおよびケモカインのプロファイルを検討した。IL−2およびIL−18の存在下で培養した上記PBMCsは、本発明にかかるヒトリンパ球を60%含有していた。
<γδT細胞の増殖に対する本発明にかかるヒトリンパ球の効果>
γδT細胞に対する本発明にかかるヒトリンパ球の直接的な作用について検討した。図14はγδT細胞上の共刺激分子の発現を示すものである。γδT細胞の表面分子は、フローサイトメトリーによって解析した。
なお、自発的に移動したγδT細胞の数は、下流側のチャンバーに何もいれず、上流側のチャンバーに上記γδT細胞を入れて同じ実験を行い、上流側のチャンバーから減少した細胞数を数えることによって求めることができる。
Claims (9)
- CD3の発現強度が100以上102.3以下であり、かつ、CD11cの発現強度が101以上102以下であるヒトPBMCsを、インターロイキン2およびインターロイキン18を含む培地中で培養してなる細胞群に含有されるヒトリンパ球であって、
CD56の発現強度が102以上、かつ、CD11cの発現強度が101以上102以下であり、
上記CD3、CD11cおよびCD56の発現強度は、CD3、CD11cおよびCD56のそれぞれに対する、FITC、PE、APCおよびビオチンによって標識した抗体によって、ヒトリンパ球を4℃で20分間染色し、フローサイトメーターを用いて解析したものであることを特徴とするヒトリンパ球。 - 上記ヒトリンパ球の80%以上は、CD25の発現強度が102以上であり、CD25の発現強度は、CD25に対する、FITC、PE、APCおよびビオチンによって標識した抗体によって、上記ヒトリンパ球を4℃で20分間染色し、フローサイトメーターを用いて解析したものであることを特徴とする請求項1に記載のヒトリンパ球。
- 上記ヒトリンパ球の80%以上は、HLA−DRの発現強度が102以上であり、HLA−DRの発現強度は、HLA−DRに対する、FITC、PE、APCおよびビオチンによって標識した抗体によって、上記ヒトリンパ球を4℃で20分間染色し、フローサイトメーターを用いて解析したものであることを特徴とする請求項1または2に記載のヒトリンパ球。
- 上記ヒトリンパ球の80%以上は、CD86の発現強度が102以上であり、CD86の発現強度は、CD86に対する、FITC、PE、APCおよびビオチンによって標識した抗体によって、上記ヒトリンパ球を4℃で20分間染色し、フローサイトメーターを用いて解析したものであることを特徴とする請求項1から3のいずれか1項に記載のヒトリンパ球。
- 上記ヒトリンパ球は、CD122を発現しているヒトリンパ球の数が、全細胞数の10%未満であることを特徴とする請求項1から4のいずれか1項に記載のヒトリンパ球。
- 請求項1から5のいずれか1項に記載のヒトリンパ球を製造する方法であって、
CD3の発現強度が100以上102.3以下であり、かつ、CD11cの発現強度が101以上102以下であるヒトPBMCsを、インターロイキン2およびインターロイキン18を含む培地中で培養する工程を含み、
上記CD3およびCD11cの発現強度は、CD3およびCD11cのそれぞれに対する、FITC、PE、APCおよびビオチンによって標識した抗体によって、上記ヒトリンパ球を4℃で20分間染色し、フローサイトメーターを用いて解析したものであることを特徴とする方法。 - 請求項1から5のいずれか1項に記載のヒトリンパ球と、γδT細胞とを、インターロイキン2、インターロイキン18およびリン酸化合物を用いて刺激する工程を含むことを特徴とする、γδT細胞の増殖方法。
- 上記ヒトリンパ球がケモカインを分泌しており、γδT細胞がケモカインレセプターを発現していることを特徴とする、請求項7に記載のγδT細胞の増殖方法。
- 上記リン酸化合物は、ゾレドロネートであることを特徴とする、請求項7または8に記載のγδT細胞の増殖方法。
