Abstract
本发明涉及一种抗氧化含片,由以下重量份组分组成:SOD0.5‑1%,淀粉浆10‑12%、润滑剂1‑2%、微晶纤维素含量45‑50%、以及下列组份中的一种罗汉果提取物1‑1.5%、木瓜提取物6‑8%、玛卡提取物1‑1.5%、苹果提取物1‑1.5%、枸杞提取物0.4‑0.6%、沙棘提取物1.5‑2%、余量矫味剂。两种抗氧化组份协同作用,提高抗氧化能力,并且减少SOD使用量,从而降低成本。含片溶解时间在18.53‑25.60min之间,既可以保证抗氧化剂通过口腔黏膜的吸收时间,又不会时间过久,使用不方便。

背景技术
抗氧化(Anti-Oxidant)是抗氧化自由基的简称。机体内有一整套完整的抗氧化系统,正常生理条件下,机体在氧化代谢的同时,会不断产生自由基,又不断清除自由基,自由基和自由基清除在体内处于一种相对动态平衡状态。由于年龄增长、不合理膳食、压力等原因机体自由基清除剂生成减少,过量的自由基会对机体产生有害作用,过量的自由基会进攻生物膜脂质双分子层中的不饱和脂肪酸引起脂质过氧化,导致生物膜结构和功能改变,造成细胞坏死,组织器官的功能受到影响而老化。自由基攻击蛋白,会使蛋白失去功能,引起病变。如果发生在胶原组织,会令皮肤失去弹性。自由基攻击DNA,会造成DNA股的切断或碱基改变,会使遗传发生错误导致癌变可能。合理的膳食营养补充及外源性抗氧化营养素的摄入,可阻止自由基对细胞的损伤,延缓衰老并预防相关疾病。
超氧化物歧化酶(Superoxide dismutase, SOD)是生物体系中抗氧化酶系的重要组成成员,广泛分布在微生物、植物和动物体内。它能够高效清除体内的自由基。体内多余自由基会引起细胞损伤以及色素沉着,人体衰老是由于自由基堆积和清除障碍的后果。补充外源SOD可以美白肌肤、延缓衰老。

CN103191070A提供了一种超氧化物歧化酶口服含片及制备方法,含片包括超氧化物歧化酶和食品添加剂,每克的口服含片中含有1.4~4.5万U活性的超氧化物歧化酶。制备时采用适量的活性为10U/g~50万U/g的超氧化物歧化酶干粉,与食品添加剂混合后,压片成型。本发明含片在服用时,SOD通过唾液的缓慢溶解而逐步释放其活性,从而避免了SOD进入胃部而在胃酸环境下失活的现象。且本发明中的SOD干粉是从玉米中提取,在最大限度的保留活性外,大大降低了SOD干粉的成本。
发明内容
本发明的目的在于提供一种抗氧化含片。
为实现本发明的上述目的,采用以下技术方案:含片由以下重量份的组分组成:SOD 0.5-1%,淀粉浆10-12%、润滑剂1-2%、微晶纤维含量45-50%、以及下列组份中的一种罗汉果提取物1-1.5%、木瓜提取物6-8%、玛卡提取物1-1.5%、苹果提取物1-1.5%、枸杞提取物0.4-0.6%、沙棘提取物1.5-2%、余量矫味剂。
本发明部分原料购买信息如下:
SOD,购自武汉远程科技发展有限公司;
罗汉果提取物、木瓜提取物、玛卡提取物、苹果提取物、枸杞提取物、沙棘提取物均购自陕西百草萃生物科技有限公司。
制备包括以下步骤:
步骤一:称取SOD、微晶纤维素、矫味剂、另一种抗氧化组份过100目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过20目筛进行造粒;
步骤三:干燥,干燥温度30℃;
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加润滑剂,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射15min,进行泡罩包装。
具体的,本发明中用到的SOD可清除生物体新陈代谢产生的超氧阴离子自由基及羟基自由基,同时,也可作为超氧阴离子的底物诱导酶,保护机体细胞免受有氧代谢物的损害。SOD作为一种良好的自由基清除剂,在提高机体免疫力,延缓衰老等方面有着很好的功效。补充外源性SOD,可以提高体内SOD的含量和活性,清除体内自由基,维持人体器官、组织的正常运行。
罗汉果有清热、润肺、滑肠通便的功效。罗汉果是传统中药材和天然甜味剂。罗汉果多糖是糖尿病、肥胖等不宜吃糖者的理想替代品,罗汉果多糖还具有抗癌、提高免疫、抗病毒、抗炎、降血糖、降血脂、抗氧化、抗衰老和促进骨折愈合等功能。
《本草纲目》中记载木瓜性温、具有平肝和胃、舒经络、降血压等功效。木瓜富含皂甙、黄酮类、木瓜酚、还原糖、过氧化物酶、木瓜黄酮等多种营养成分。木瓜黄酮具有抗氧化活性,可以清除超氧阴离子和羟基自由基、抗癌、降压、降血脂等。

沙棘又名醋柳、沙枣、酸刺等,我国华北、西北和四川均有分布。沙棘具有良好的抗氧化性、抗溃疡性、抑菌和抗癌作用,也可作为肝脏和心脏保护药物。沙棘黄酮可以清除超氧阴离子、过氧化氢和羟基自由基等。
与现有技术相比,本发明的有益效果为:
本发明抗氧化含片,通过添加SOD及以下抗氧化组份中的一种:罗汉果提取物、木瓜提取物、玛卡提取物、苹果提取物、枸杞提取物、沙棘提取物,两种抗氧化组份协同作用,提高抗氧化能力,并且减少SOD使用量,从而降低成本。含片溶解时间在18.53-25.60min之间,既可以保证抗氧化剂通过口腔黏膜的吸收时间,又不会时间过久,使用不方便。

具体实施方式
以下通过具体实施例进一步描述本发明,但本发明不仅限于此。
实施例一
配方:SOD 1%、淀粉浆12%、硬脂酸镁2%、微晶纤维素50%、罗汉果提取物1%、甘露醇34%。本实施例溶解时间18.53min。
制备过程如下:
步骤一:称取SOD、微晶纤维素、甘露醇、罗汉果提取物过100目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过20目筛进行造粒;
步骤三:干燥,干燥温度30℃。
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加硬脂酸镁,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射15min,进行泡罩包装。
实施例二
配方:SOD 0.8%、淀粉浆10%、滑石粉1%、微晶纤维素45%、木瓜提取物8%、山梨醇35.2%。本实施例溶解时间19.30min。
制备过程如下:
步骤一:称取SOD、微晶纤维素、山梨醇、木瓜提取物过100目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过20目筛进行造粒;
步骤三:干燥,干燥温度30℃。
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加滑石粉,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射15min,进行泡罩包装。
实施例三
配方:SOD 1%、淀粉浆11%、聚乙二醇-4000 1%、微晶纤维素48%、玛卡提取物1.5%、葡萄糖37.5%。本实施例溶解时间20.18min。
制备过程如下:
步骤一:称取SOD、微晶纤维素、葡萄糖、玛卡提取物过100目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过20目筛进行造粒;
步骤三:干燥,干燥温度30℃。
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加聚乙二醇-4000,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射15min,进行泡罩包装。
实施例四
配方:SOD 0.5%、淀粉浆10%、聚乙二醇-4000 2%、微晶纤维素48%、苹果提取物1.5%、乳糖38%。本实施例溶解时间21.37min。
制备过程如下:
步骤一:称取SOD、微晶纤维素、乳糖、苹果提取物提取物过100目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过20目筛进行造粒;
步骤三:干燥,干燥温度30℃。
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加聚乙二醇-4000,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射15min,进行泡罩包装。
实施例五
配方:SOD 0.7%、淀粉浆12%、硬脂酸镁1%、微晶纤维素50%、沙棘提取物2%、乳糖34.3%。本实施例溶解时间25.60min。
步骤一:称取SOD、微晶纤维素、乳糖、沙棘提取物提取物过100目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过20目筛进行造粒;
步骤三:干燥,干燥温度30℃。
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加硬脂酸镁,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射15min,进行泡罩包装。
实施例六
配方:SOD 1%、淀粉浆10%、聚乙二醇-4000 2%、微晶纤维素48%、枸杞提取物0.6%、乳糖38.4%。本实施例溶解时间22.60min。
步骤一:称取SOD、微晶纤维素、乳糖、枸杞提取物提取物过100目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过20目筛进行造粒;
步骤三:干燥,干燥温度30℃。
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加聚乙二醇-4000,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射15min,进行泡罩包装。
体外抗氧化活性实验
实验分为12组:SOD组、罗汉果多糖、木瓜黄酮、玛卡多糖、苹果多酚、枸杞多糖、沙棘多糖、实施例一到实施例六组。
实验仪器及试剂:BSA223S-G分析天平(上海精天电子仪器公司)、DR6000紫外-可见光分光光度计(美国哈希公司)、SIGMA3-18K冷冻离心机(北京博励行仪器)、智能数显恒温水浴锅(江苏金怡仪器科技有限公司)、超氧阴离子试剂盒、羟基自由基试剂盒均购自南京建成生物工程研究所。
实验方法:参照试剂盒说明书进行测定。
最终实验结果如图1、2、3、4所示。
根据实验结果可以看出SOD较低浓度时就表现出很高的超氧阴离子自由基清除率和羟基自由基清除率。罗汉果多糖超氧阴离子和羟基自由基清除率较低,分别为44.97%和35.05%。单组份超氧阴离子清除率为90.11%、羟基自由基最高清除率均为90.08%。通过单组份自由基清除实验以及各组分协同作用,合理配置各抗氧化组分,组合后超氧阴离子清除率最高到98.97%、羟基自由基清除率最高到99.01%,其中实施例六的效果最好。
Claims (10)
1.一种抗氧化含片,其特征在于,由以下重量份组分组成:SOD 0.5-1%,淀粉浆 10-12%、润滑剂 1-2%、微晶纤维含量 45-50%、以及下列组份中的一种罗汉果提取物 1-1.5%、木瓜提取物 6-8%、玛卡提取物 1-1.5%、苹果提取物 1-1.5%、枸杞提取物 0.4-0.6%、沙棘提取物 1.5-2%、余量矫味剂。
2.一种抗氧化含片,其特征在于,由以下重量份组分组成:SOD 1%,淀粉浆 10%、聚乙二醇-4000 2%、微晶纤维 48%、枸杞提取物 0.6%、乳糖 38.4%。
3.根据权利要求 2 所述的一种抗氧化含片,其特征在于,在口中的溶化时间为22.60min。
4.根据权利要求 1 或 2 所述的一种抗氧化含片,其特征在于,SOD 为从植物中提取的,
SOD 的酶活为 100000U/g。
5.根据权利要求 1 所述的一种抗氧化含片,其特征在于,润滑剂为硬脂酸镁、滑石粉、聚
乙二醇-4000 中的一种。
6.根据权利要求 1 所述的一种抗氧化含片,其特征在于,矫味剂为甘露醇、山梨醇、葡萄
糖、乳糖中的一种。
7.根据权利要求 1 所述的一种抗氧化含片,其特征在于,罗汉果提取物中罗汉果多糖的含
量≥50%;木瓜提取物中木瓜黄酮含量≥5%;玛卡提取物中玛卡多糖含量≥50%;苹果提取
物中苹果多酚的含量≥50%;枸杞提取物中枸杞多糖含量≥70%;沙棘提取物中沙棘黄酮含
量≥30%。
8.根据权利要求 2 所述的一种抗氧化含片,其特征在于,枸杞提取物中枸杞多糖含量≥
70%。
9.根据权利要求 1 或 2 所述的一种抗氧化含片的制备方法,其特征在于,制备过程包括以下步骤:
步骤一:称取 SOD、微晶纤维素、矫味剂、另一种抗氧化组份过 100 目筛后混匀;
步骤二:制粒,加入淀粉浆制成软材,通过 20 目筛进行造粒;
步骤三:干燥,干燥温度 30℃;
步骤四:整粒和压片,整粒后为使含片表面光滑,更易脱模,添加润滑剂,混合均匀,送入压片机压片;
步骤四:灭菌和包装,将片剂在紫外线下照射 15min,进行泡罩包装。
10.根据权利要求 1 所述的一种抗氧化含片,其特征在于,在口中的溶化时间为18.53- 25.60min。
