Description

Yangsheng snow chrysanthemum sleeping water and preparation method thereof
Technical Field
The invention relates to the technical field of composition formulas, in particular to a nerve-soothing preparation, and especially relates to a male-lotus-snow chrysanthemum sleeping water and a preparation method thereof.
Background
Insomnia refers to a subjective experience that a patient does not meet the sleep time and/or quality and affects day-to-day social functions, and the patient has the following clinical manifestations: (1) sleep process disorders such as difficulty in falling asleep, decreased sleep quality, and decreased sleep time; (2) daytime cognitive dysfunction such as memory decline and attention decline; (3) peripheral brain systems such as cardiovascular system and motor system and peripheral vegetative nerve functional disturbance; (4) immune system, endocrine system, and other system symptoms.
The causes of insomnia are multifaceted and often a composite effect of multiple causes. Modern medicine considers that the cause of the insomnia mainly comprises environmental insomnia, psychological behavior disorder, mental diseases, physical diseases, medicament induction and the like; the theory of traditional Chinese medicine considers that the causes of insomnia mainly include external six excesses, internal seven emotions, overeating and deficiency of qi and blood, which are basically consistent with the understanding of modern medicine on the causes of insomnia. The traditional Chinese medicine theory is used for analyzing the insomnia pathology, the insomnia is mainly divided into insomnia caused by yin-yang disharmony, insomnia caused by ying-wei disharmony, insomnia caused by deficiency of qi and blood and insomnia caused by viscera dysfunction, the insomnia treatment mainly aims at coordinating yin and yang and harmonizing ying-wei, aims at tonifying deficiency of viscera and eliminating excess of pathogenic factors, adopts a non-pathogenic factor (pathogenic factors are not obvious) to treat the root according to the specific conditions of a patient, and has a pathogenic factor to treat the root and the root.
The prescriptions for treating insomnia throughout the generations and the existing preparations for treating insomnia are directed at deficiency syndromes such as deficiency of heart-qi, liver-blood, heart-yin and heart-yang, tangible and intangible pathogens, and deficiency-excess complications such as disharmony between heart and kidney, liver depression, obstruction of triple energizer, and disharmony between liver and spleen. Wherein the incoordination between liver and spleen is insomnia caused by liver depression, blood deficiency, fire, and spleen qi deficiency, the traditional treatment formula or preparation is DANZHIXIAOYAO powder, and bupleuri radix is used for dispersing stagnated liver qi and relieving qi stagnation; angelica and white peony root have the functions of enriching blood and nourishing liver; poria, Atractylodis rhizoma, and radix Glycyrrhizae Preparata for invigorating spleen; cortex moutan and fructus Gardeniae clear heat and purge fire. The medicines are used together to play the effects of soothing liver, nourishing blood, strengthening spleen, tonifying qi, clearing heat and purging fire.
in the theory of mutual generation and restriction of five elements in traditional Chinese medicine, there is a mutual relationship between five zang organs.
in the relationship of mutual restriction, the liver qi can remove the stagnation of spleen dampness, which is named as mu ke soil; when the restriction of the liver and spleen is too strong, the liver-qi will transverse to the stomach or pass the spleen, resulting in the syndrome of disharmony between the liver and stomach (or) the liver and spleen, which is called wood-hyperactivity over earth. For therapeutic reasons, the method of soothing liver and calming liver should be used to strengthen the body resistance, while the method of tonifying spleen qi should be used to strengthen the body resistance.
In the relationship of generation, the liver stores blood, the heart governs blood vessels, and the liver has normal functions of storing blood and regulating blood volume, which is helpful for normal exertion of the functions of heart governing blood vessels, namely, the liver wood can be used for treating heart fire, namely, wood can be used for generating fire; when liver blood is deficient, it can not nourish heart, resulting in endogenous deficient heat and malnutrition of heart-mind; for therapeutic principle, tonify blood, nourish heart, clear heat and purge fire should be used.
The prescription or the preparation conforms to the theory of mutual generation and mutual inhibition of five elements in the traditional Chinese medicine, treats both symptoms and root causes, and has certain efficacy on treating insomnia caused by liver depression, blood deficiency, fire transformation and spleen qi deficiency. However, for deficiency of liver blood, the prescription or preparation only uses angelica and white peony root to enrich and nourish the liver, and does not nourish the liver from the mutual relationship of five elements, because the kidney can store essence, the kidney essence can generate liver blood, namely kidney water nourishes liver wood, namely water grows wood, and when the liver blood is deficient, the kidney also needs to be nourished to nourish the liver, namely a water nourishing wood method; most of the prescription or preparation is traditional Chinese medicinal materials, the medicinal properties are strong, and the prescription for recuperating both food and medicine is needed to promote sleep and improve insomnia symptoms caused by liver depression, blood deficiency, fire transformation and spleen qi deficiency.

(1) 56 KM mice, weighing 18-22 g, half male and half female, were randomly divided into blank groups, examples 1-5, and 8 mice per group. In examples 1-5, the sleep water with the crude drug amount of 16g/kg is administrated in a stomach irrigation mode, the diazepam water solution with the dose of 0.002g/kg is administrated in an intraperitoneal injection way after the administration of the 1% sodium carboxymethylcellulose solution for 20min in a positive control group, the 1% sodium carboxymethylcellulose solution is administrated in a blank group, the administration volume is 0.025ml/g (body weight), the administration is carried out for 1 time every day, and the administration lasts for 7 days; pentobarbital sodium is injected into the abdominal cavity to affect sleep, and the number and time of sleep are observed. The experimental result shows that the sleep water has the effect of improving abnormal sleep of the experimental mouse caused by the sodium pentobarbital, and the sleep water of each embodiment can obviously prolong the sleep time of the experimental mouse (table 1), which shows that the sleep water has the nerve soothing effect.
(2) 56 KM mice, weighing 18-22 g, half male and half female, were randomly assigned to the blank group, the positive control group, examples 1-5, and 8 mice per group. Before the gavage, the mice are put into a 40cm/24cm/30cm organic glass box to adapt for 3min, and then the spontaneous activity time and the lifting times of forelimbs of the mice within 2min are respectively measured to be used as normal activity reaction indexes. In examples 1-5, the crude drug amount of 16g/kg of sleep water was administered by gavage, the positive control group was administered with a 1% sodium carboxymethylcellulose solution for 20min, and then administered with a 0.002g/kg dose of diazepam aqueous solution by intraperitoneal injection, and the blank group was administered with a 1% sodium carboxymethylcellulose solution, and the administration volume was 0.025ml/g (body weight) 1 time per day for 7 consecutive days. The above indexes are retested 30min and 60min after the last gastric lavage. The experimental result shows that the sleep water has the function of improving and regulating the spontaneous activity of the mice, and the sleep water of each embodiment can obviously reduce the number of times of forelimb erection and the spontaneous activity time of the experimental mice (table 2), which shows that the sleep water has the function of soothing the nerves.
