本发明涉及植物应用研究领域,具体为一种小分子簕菜提取物的制备方法及其应用,先提取簕菜提取物,再利用酶解反应对其进行小分子改造,使其从不易吸收的大分子物质转化为易吸收的小分子物质,最后过滤杀菌灌装,簕菜活性成分有抗氧化、抗菌、抗衰老、抗肿瘤等活性,通过小分子改性,将蛋白质、多肽和糖类进行酶解,成为小分子物质,使其更易被肌肤吸收,显著地提高了簕菜活性成分的吸收率,减少了有效成分的流失;低温酶解,控制酶解速度,在酶解的同时不破坏其他有效成分;中和反应后得到的水杨酸钠和柠檬酸钠也是一种对人体有效物质,不会影响人体对其吸收和利用;加入的复合酶最后被过滤掉,不会保留在成品中。

背景技术
近年来,科研人员对簕菜化学成分进行了深入的研宄。目前,许多国内外学者己经从簕菜茎叶中分离得到了超过30种单体化合物,其中既包括多糖、挥发油、酸类、黄酮、阜苷,也含有蛋白质、维生素、铁、锌等多种营养成分。
中国专利库中公开了一种簕菜提取物及其制备方法和应用(CN201210274788.6),首先称取簕菜全草,用乙醇浸泡,经旋转蒸发浓缩后得到水溶液,然后分别用油醚、乙酸乙酯、正丁醇依次萃取水溶液;萃取液经减压蒸馏后得到膏体,即得到石油醚萃取物,乙酸乙酯萃取物,正丁醇萃取物,水溶物;通过抗自由基氧化实验,巨噬细胞生长实验、NF-kB核转录因子的抑制实验和Erk1/2的激活作用实验,证实簕菜提取物具有抗氧化和抗炎能力,添加了簕菜及其提取物的治疗炎症的药物或抗氧化的化妆品或食品组合物具有抗炎和抗氧化作用,而可用于防治炎症或衰老的化妆品、药物、植物性饮料及保健食品。
现有技术中,簕菜提取物已经应用到化妆品、保健品和食品等领域,但直接添加簕菜提取物,由于其含有较多的大分子物质(蛋白质、多肽),这会导致其吸收不佳,大量的有效成分流失,如何对其进行小分子改造是值得研究的课题。
发明内容
针对上述问题,本发明的目的是提供一种小分子簕菜提取物的制备方法及其应用,先提取簕菜提取物,再利用酶解反应对其进行小分子改造,使其从不易吸收的大分子物质转化为易吸收的小分子物质,最后过滤杀菌灌装,通过对簕菜提取物提取物的小分子改造,使其运用于化妆品等产品时更容易被吸收。
为了实现上述目的,本发明采用以下方案
一种小分子簕菜提取物的制备方法,其包括以下步骤:
S1簕菜提取物粗提,对新鲜簕菜进行活性成分提取,得到簕菜粗提物;
S2极速冻融,将簕菜粗提物快速降温至零度以下,冻融15-30Min;
S3酶解,将冻融后的簕菜粗提物解冻并升温至15-20℃,将其调节为酸性环境,pH为4.5-5.5,加入复合酶进行酶解,得到簕菜酶解液;
S4酸碱中和,加入碱性溶液调节簕菜酶解液的pH为7.0-7.5;
S5过滤灌装,将中和处理后的勒菜酶解液进行过滤,之后根据需要实际需要制备成各种形态的小分子簕菜提取物,最后杀菌灌装。
优选地,簕菜提取物的粗提方法,包括以下步骤:
A1粉碎,使用粉碎机将簕菜进行粉碎,制成簕菜微粒,粒径为20-50筛目;
A2制混悬液,将簕菜微粒和溶剂混合制成混悬液,将混悬液采用超声波处理1-3h,簕菜微粒和溶剂的质量比为1:8-15,温度为18-30℃,超声频率 200-800KHz,超声强度3-8W/cm2;
A3雾化浸提,使用雾化器将超声波处理后的混悬液雾化并喷洒出去,液雾自然沉降恢复成混悬液,收集混悬液,保持温度为18-50℃;
A4过滤分离,将步骤A3得到混悬液进行过滤,得到清液和簕菜残渣;
A5二次浸提,将簕菜残渣和溶剂混合制成混悬液,簕菜微粒和溶剂的质量比为1:5-8,温度为18-30℃;使用雾化器将混悬液雾化并喷洒出去,液雾自然沉降恢复成混悬液,收集混悬液,保持温度为18-50℃;
A6二次过滤,将步骤A5得到混悬液进行过滤,得到清液和簕菜残渣;
A7溶剂分离,将步骤A4和步骤A6得到的清液混合并加热,分离出溶剂,得到簕菜提取液。
优选地,步骤S3中,加入的酸性物质为质量比为1:(2-4)的水杨酸和柠檬酸,水杨酸的浓度为5%,柠檬酸的浓度为8%。
优选地,步骤S3中,复合酶包括蛋白酶、肽酶和麦芽糖酶。
优选地,步骤S3中,蛋白酶、肽酶和麦芽糖酶的质量比为(3-5):(2-4): 2。
优选地,步骤S3中,加入复合酶后进行慢速搅拌,搅拌速度为50-100rpm,时间10-15小时。
优选地,步骤S4中,碱性溶液为6%含量的氢氧化钠溶液。

优选地,将小分子簕菜提取物作为一种原料添加到润肤美白、防晒补水、抗氧化、去皱抗衰老类化妆品中。
本发明的有益效果是:簕菜活性成分有抗氧化、抗菌、抗衰老、抗肿瘤等活性,通过小分子改性,将蛋白质、多肽和糖类进行酶解,成为小分子物质,使其更易被肌肤吸收,显著地提高了簕菜活性成分的吸收率,减少了有效成分的流失;低温酶解,控制酶解速度,在酶解的同时不破坏其他有效成分;中和反应后得到的水杨酸钠和柠檬酸钠也是一种对人体有效物质,不会影响人体对其吸收和利用;加入的复合酶最后被过滤掉,不会保留在成品中。

为了使本技术领域的人员更好的理解本申请中的技术方案,下面将结合实施例来对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本申请的一部分实施例,基于本申请中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都应当属于本申请保护的范围。
簕菜活性成分
近年来,科研人员对簕菜化学成分进行了深入的研宄。目前,许多国内外学者己经从簕菜茎叶中分离得到了超过30种单体化合物,其中既包括多糖、挥发油、酸类、黄酮、阜苷,也含有蛋白质、维生素、铁、锌等多种营养成分。现对其主要成分按以下分类进行具体阐述。
1.簕菜多糖
多糖是一种通过糖苷键连结10个以上的单糖组成的聚合糖,具有抗炎、降血糖、清除机体自由基、抗肿瘤、提高免疫力和抗衰老的作用。蔡凌云等经多糖确证性实验首次证明了四川南充市产地的簕菜中含有多糖成分且不同部位的多糖含量不同,其中茎皮中多糖含量最高。研究还表明茎皮在微波辅助提取最佳条件下,簕菜总多糖的含量为6.28mg/g。
2.簕菜挥发油
纳智通过GC-MS联用技术首次研究了云南西双版纳地区簕菜叶中挥发油的化学成分。研究结果表明,其中含有大量的萜烯、萜醇类化合物及少量的长链脂肪族和芳香族化合物。其中主要的单萜类化合物α-蒎烯、β-水芹烯、 D-柠檬烯和β-蒎烯分别具有明显的镇咳、祛痰、抗真菌、抗炎和祛痛等作用。法国和越南对越南产簕菜的联合研究显示,越南产簕菜的香精油/挥发油成分中,已有超过60种,总量为97.4%的成分被确定,主要成分为:α-蒎烯(23.9%)、香桧烯(14.9%)、松油烯-4-醇(9.0%)、(β-蒎烯(7.7%)以及p-伞花烃(5.8%),此外也含有少量假虎刺酮、桉叶烃衍生物。
3.簕菜酚类
酚类是芳香烃中苯环上的氢原子被羟基取代所生成的化合物,根据其分子所含的羟基数目可分为一元酚和多元酚。通过高效液相色谱法,陈智毅等次测定了广东恩平地区簕菜中的酚类成分,并检验出了13种酚类成分:绿原酸、安息香酸、声丁、龙胆酸、阿魏酸、丁香酸、白藜芦醇、槲皮素、表儿茶素、咖啡酸、没食子酸、儿茶素、水杨酸。由此可见,簕菜中含有较多的多酚类化合物。
4.簕菜黄酮
国内研究人员分别采用超声辅助法微波辅助法或聚酰胺树脂纯化法M等方法对簕菜中黄酮的提取工艺进行了优化。研究表明簕菜叶中含有槲皮素为苷元的黄醇。初步的抑菌实验也证明簕菜叶总黄酮提取液对大肠杆菌有较强的抑制作用,对金黄色葡萄球菌也有一定的抑制作用,而对枯草杆菌的抑制作用不明显泰国研究团队通过HPLC分析了不同季节的簕菜叶的黄酮类化合物,研究表明一年中冬季的一月和十一月的黄酮类含量较其他月份更高。
5.簕菜阜苷
国内高侠等以簕菜叶总阜苷提取率为评价标准,优化了超声波法提取簕菜叶总阜苷的工艺,在最佳工艺下提取率为16.32%。该课题组又采用微波辅助法提取白簕叶中的阜苷类化合物,通过正交实验确定了最佳提取工艺条件下簕菜总阜苷含量为84.36mg/g中国台湾地区产簕菜中分离到一个新的三蔽苷。
国外产簕菜的研究主要集中于阜苷类成分结构鉴定。PhanVanKienm等从簕菜叶中分离出两种新的羽扇烷三萜羧酸和新的24-反-羽扇烷-糖苷, Sang-YongPark等从日本刺五加叶中分离得到两种新的反齐墩果烯型三萜糖苷。越南地区簕菜中分离得到多种三萜类化合物及其糖苷,还有少量的二萜化合物、苯丙素苷和黄酮苷。
不同品种簕菜提取物的提取得率
不同品种簕菜提取物真空干燥后称重,计算得率为:青梗(32.8±1.8)%、白梗(26.5±1.1)%、紫柄(24.1±0.6)%、大叶大刺(26.8±0.5)%、红梗 (30.4±1.1)%,其中青梗和红梗簕菜的提取率稍高于白梗和大叶大刺簕菜,紫柄簕菜的提取率最低。
实施例1
一种小分子簕菜提取物的制备方法,其包括以下步骤:
S1簕菜提取物粗提,对新鲜簕菜进行活性成分提取,得到簕菜粗提物;
S2极速冻融,将簕菜粗提物快速降温至零度以下,冻融15Min;
S3酶解,将冻融后的簕菜粗提物解冻并升温至15℃,加入质量比为1:2的水杨酸和柠檬酸,水杨酸的浓度为5%,柠檬酸的浓度为8%,将其调节为酸性环境,pH为4.5-5.5,加入质量比为3:2:2的蛋白酶、肽酶和麦芽糖酶组成的复合酶进行酶解,酶解过程中慢速搅拌,搅拌速度为50rpm,时间15小时,得到簕菜酶解液;
S4酸碱中和,加入适量6%的氢氧化钠溶液,调节簕菜酶解液的pH为7.0-7.5;
S5过滤灌装,将中和处理后的勒菜酶解液进行过滤,采用平板超滤设备对簕菜酶解液进行过滤,选用截留相对分子质量为10000的膜,压力范围为0.3MPa;之后根据需要实际需要制备成各种形态的小分子簕菜提取物,最后杀菌灌装。
实施例2
一种小分子簕菜提取物的制备方法,其包括以下步骤:
S1簕菜提取物粗提,对新鲜簕菜进行活性成分提取,得到簕菜粗提物;
S2极速冻融,将簕菜粗提物快速降温至零度以下,冻融22Min;
S3酶解,将冻融后的簕菜粗提物解冻并升温至18℃,加入质量比为1:3的水杨酸和柠檬酸,水杨酸的浓度为5%,柠檬酸的浓度为8%,将其调节为酸性环境,pH为4.5-5.5,加入质量比为4:3:2的蛋白酶、肽酶和麦芽糖酶组成的复合酶进行酶解,酶解过程中慢速搅拌,搅拌速度为80rpm,时间12小时,得到簕菜酶解液;
S4酸碱中和,加入适量6%的氢氧化钠溶液,调节簕菜酶解液的pH为7.0-7.5;
S5过滤灌装,将中和处理后的勒菜酶解液进行过滤,采用平板超滤设备对簕菜酶解液进行过滤,选用截留相对分子质量为10000的膜,压力范围为0.4MPa;之后根据需要实际需要制备成各种形态的小分子簕菜提取物,最后杀菌灌装。
实施例3
一种小分子簕菜提取物的制备方法,其包括以下步骤:
S1簕菜提取物粗提,对新鲜簕菜进行活性成分提取,得到簕菜粗提物;
S2极速冻融,将簕菜粗提物快速降温至零度以下,冻融30Min;
S3酶解,将冻融后的簕菜粗提物解冻并升温至20℃,加入质量比为1:4的水杨酸和柠檬酸,水杨酸的浓度为5%,柠檬酸的浓度为8%,将其调节为酸性环境,pH为4.5-5.5,加入质量比为5:4:2的蛋白酶、肽酶和麦芽糖酶组成的复合酶进行酶解,酶解过程中慢速搅拌,搅拌速度为100rpm,时间10小时,得到簕菜酶解液;
S4酸碱中和,加入适量6%的氢氧化钠溶液,调节簕菜酶解液的pH为7.0-7.5;
S5过滤灌装,将中和处理后的勒菜酶解液进行过滤,采用平板超滤设备对簕菜酶解液进行过滤,选用截留相对分子质量为10000的膜,压力范围为0.5MPa;之后根据需要实际需要制备成各种形态的小分子簕菜提取物,最后杀菌灌装。
对比例1
一种簕菜提取物的制备方法,其包括以下步骤:
A1粉碎,使用粉碎机将簕菜进行粉碎,制成簕菜微粒,粒径为30筛目;
A2制混悬液,将簕菜微粒和溶剂混合制成混悬液,将混悬液采用超声波处理2h,簕菜微粒和溶剂的质量比为1:11,温度为25℃,超声频率500KHz,超声强度5.5W/cm2;
A3雾化浸提,使用雾化器将超声波处理后的混悬液雾化并喷洒出去,液雾自然沉降恢复成混悬液,收集混悬液,保持温度为40℃;
A4过滤分离,将步骤A3得到混悬液进行过滤,得到清液和簕菜残渣;
A5二次浸提,将簕菜残渣和溶剂混合制成混悬液,簕菜微粒和溶剂的质量比为1:7,温度为25℃;使用雾化器将混悬液雾化并喷洒出去,液雾自然沉降恢复成混悬液,收集混悬液,保持温度为35℃;
A6二次过滤,将步骤A5得到混悬液进行过滤,得到清液和簕菜残渣;
A7溶剂分离,将步骤A4和步骤A6得到的清液混合并加热,分离出溶剂,得到簕菜提取液。
试验例1
分别取用等量的实施例1-3制备的小分子簕菜提取物和对比例1制备的簕菜提取液制备美白类化妆品,在美白类化妆品中,其他成分的重量和比例都相同,分别记作,实施例S1、试验例S2、试验例S3和对比例D1,随机选择20-35 岁女性分成四组,每组五人,分别涂抹等量的四种美白类化妆品于脸部和颈部,在涂抹0.1小时、0.5小时、1.0小时和2小时后,分别测量脸部皮肤上化妆品残留物中多肽的含量,并推算出肌肤的吸收率,试验结果如表一。

通过上述数据说明,检测以簕菜多肽为代表的勒菜活性成分的吸收率可以得出小分子簕菜提取物的吸收率明显比普通的勒菜提取液的吸收率更高,由此可得出,小分子勒菜提取物更容易被吸收,吸收率提升后,化妆品的使用效果也可以得到提升。
试验例2
分别取用等量的实施例1-3制备的小分子簕菜提取物和对比例1制备的簕菜提取液制备食品,分别喂养四组小白鼠,每组四只,小白鼠选用远交系KM小鼠,体重约25g,误差小于1g,分别记作实施例A1、试验例A2、试验例A3和对比例B1,分别在喂养后1小时、2小时、3小时和4小时后提取白鼠胃内残留物,检测其蛋白质含量,并推断出吸收率,试验结果如表二。

通过上述数据说明,作为食品,小分子化后的簕菜提取物比普通的勒菜提取液的吸收速度更快,其有效物质更能有效快速地被人体吸收。

Claims (9)
1.一种小分子簕菜提取物的制备方法,其特征在于,包括以下步骤:
S1簕菜提取物粗提,对新鲜簕菜进行活性成分提取,得到簕菜粗提物;
S2极速冻融,将簕菜粗提物快速降温至零度以下,冻融15-30Min;
S3酶解,将冻融后的簕菜粗提物解冻并升温至15-20℃,将其调节为酸性环境,pH为4.5-5.5,加入复合酶进行酶解,得到簕菜酶解液;
S4酸碱中和,加入碱性溶液调节簕菜酶解液的pH为7.0-7.5;
S5过滤灌装,将中和处理后的勒菜酶解液进行过滤,之后根据需要实际需要制备成各种形态的小分子簕菜提取物,最后杀菌灌装。
2.根据权利要求1所述的一种小分子簕菜提取物的制备方法,其特征在于,簕菜提取物的粗提方法,包括以下步骤:
A1粉碎,使用粉碎机将簕菜进行粉碎,制成簕菜微粒,粒径为20-50筛目;
A2制混悬液,将簕菜微粒和溶剂混合制成混悬液,将混悬液采用超声波处理1-3h,簕菜微粒和溶剂的质量比为1:8-15,温度为18-30℃,超声频率200-800KHz,超声强度3-8W/cm2;
A3雾化浸提,使用雾化器将超声波处理后的混悬液雾化并喷洒出去,液雾自然沉降恢复成混悬液,收集混悬液,保持温度为18-50℃;
A4过滤分离,将步骤A3得到混悬液进行过滤,得到清液和簕菜残渣;
A5二次浸提,将簕菜残渣和溶剂混合制成混悬液,簕菜微粒和溶剂的质量比为1:5-8,温度为18-30℃;使用雾化器将混悬液雾化并喷洒出去,液雾自然沉降恢复成混悬液,收集混悬液,保持温度为18-50℃;
A6二次过滤,将步骤A5得到混悬液进行过滤,得到清液和簕菜残渣;
A7溶剂分离,将步骤A4和步骤A6得到的清液混合并加热,分离出溶剂,得到簕菜提取液。
3.根据权利要求1所述的一种小分子簕菜提取物的制备方法,其特征在于,步骤S3中,加入的酸性物质为质量比为1:(2-4)的水杨酸和柠檬酸,水杨酸的浓度为5%,柠檬酸的浓度为8%。
4.根据权利要求1所述的一种小分子簕菜提取物的制备方法,其特征在于,步骤S3中,复合酶包括蛋白酶、肽酶和麦芽糖酶。
5.根据权利要求4所述的一种小分子簕菜提取物的制备方法,其特征在于,步骤S3中,蛋白酶、肽酶和麦芽糖酶的质量比为(3-5):(2-4):2。
6.根据权利要求1所述的一种小分子簕菜提取物的制备方法,其特征在于,步骤S3中,加入复合酶后进行慢速搅拌,搅拌速度为50-100rpm,时间10-15小时。
7.根据权利要求1所述的一种小分子簕菜提取物的制备方法,其特征在于,步骤S4中,碱性溶液为6%含量的氢氧化钠溶液。
8.根据权利要求1所述的一种小分子簕菜提取物的制备方法,其特征在于,步骤S5中,采用平板超滤设备对簕菜酶解液进行过滤,选用截留相对分子质量为10000的膜,压力范围为0.3-0.5MPa。
9.一种小分子簕菜提取物的应用,其特征在于,将实施例1-8任一所述的小分子簕菜提取物作为一种原料添加到化妆品、保健品、药品和食品中。
