本发明涉及一种改善人体肠道健康的乳酸菌固体饮料及其制备方法。
背景技术
中医认为:脾为后天之本,是生命之源,主运化、主升清、主统血。脾在五行学说中属土,在八卦中属坤卦,在九宫属中宫。脾在病理和生理中处于非常重要的位置,人体的一切功能活动,都需要脾来协助完成,历代医家都很重视脾的作用,医圣张仲景所有方剂都重视顾护脾胃,有针对脾胃的建中汤,理中丸。金元时期李东垣更是把脾胃学说发扬到极致,创立补中益气丸,造福人类。而中医里的脾正是肠道,可见肠道健康的重要性。
最新发现,肠道是人体最大的免疫器官,分布着全身70%的淋巴组织和85%的免疫细胞,肠道健康与否决定着机体免疫力高低,而肠道健康是由肠道微生物所决定。人体肠道分布着几百种微生物,细胞数量约为百万亿,微生物包含的基因数量约为200~500万,从细胞数量和基因数量都远远超过人体自身,所以科学家形象的把肠道微生物称为人体新的器官,把肠道微生物的基因组称为人体第二基因组。根据对人体利害,把肠道微生物分为有益菌、中性菌和有害菌,有益菌能帮助人体消化吸收、产生大量维生素、抑制病原菌,对健康有益;有害菌会生成大量毒素、炎症因子等,对健康有害。如果肠道菌群平衡被破坏,导致肠道不健康,就会引发各种慢性疾病,如肠道疾病、代谢性疾病、神经性疾病、免疫性疾病等。而饮食不健康、生活不规律、滥用抗生素、环境条件的影响等都会对肠道微生物造成破坏,进而引发各种慢性病。
据中关村新智源健康管理研究院、中南大学健康管理研究中心与社会科学文献出版社共同发布的《健康管理蓝皮书:中国健康管理与健康产业发展报告(2018)》的数据显示:我国慢性病发病人数在3亿左右。

为了克服上述缺陷,本发明提供了一种改善人体肠道健康的乳酸菌固体饮料及其制备方法,从人体的肠道微生物出发,通过维护和帮助恢复肠道菌群平衡,改善人体肠道健康。
本发明的有益效果是:本发明通过筛选功能性乳酸菌,进行科学配伍,成功研制出一种改善人体肠道健康的乳酸菌固体饮料,通过维护和帮助恢复肠道菌群平衡,改善肠道健康。

一种改善人体肠道健康的乳酸菌固体饮料,包括以下质量份数的组分:嗜酸乳杆菌2~5份、植物乳杆菌2~5份、干酪乳杆菌2~5份、两歧双歧杆菌2~5份、青春双歧杆菌2~5份、长双歧杆菌2~5份、鼠李糖乳杆菌2~5份、菊粉8份、低聚木糖6份、低聚异麦芽糖8份。
上述改善人体肠道健康的乳酸菌固体饮料的制备方法,包括以下步骤:
步骤1,培养基制作:
首先,将蛋白胨4份、葡萄糖5份、酵母粉1份、脱脂乳1份、乙酸钠0.5份、轻质碳酸钙0.3份、柠檬酸铵0.3份、吐温0.2份、硫酸镁0.02份、硫酸锰0.01份、蒸馏水75~85份,加热至70~73℃左右,搅拌使各原料溶解;然后,以20%的氢氧化钠溶液调pH为7.0~7.2,完成配料;再以120~125℃灭菌20分钟后,冷却至37℃左右,获得灭菌培养基;
步骤2,菌种的活化及扩大培养:
将冷藏的嗜酸乳杆菌接种于100ml上述灭菌培养基进行一次活化,温度为37~39℃,30小时后检测,检测达标后将100ml菌液全部接入500ml上述灭菌培养基进行二次活化,温度为37~39℃,30小时后检测达标后,在上述灭菌培养基进行扩大培养72小时;
植物乳杆菌的活化及扩大培养同上述嗜酸乳杆菌;
干酪乳杆菌的活化及扩大培养同上述嗜酸乳杆菌;
两歧双歧杆菌的活化及扩大培养同上述嗜酸乳杆菌;
青春双歧杆菌的活化及扩大培养同上述嗜酸乳杆菌;
长双歧杆菌的活化及扩大培养同上述嗜酸乳杆菌;
鼠李糖乳杆菌的活化及扩大培养同上述嗜酸乳杆菌;
步骤3,菌泥的收集:
用离心机对发酵后的嗜酸乳杆菌液进行离心,并收集菌泥;
植物乳杆菌的菌泥收集方法同上述嗜酸乳杆菌;
干酪乳杆菌的菌泥收集方法同上述嗜酸乳杆菌;
两歧双歧杆菌的菌泥收集方法同上述嗜酸乳杆菌;

步骤4,菌泥的冷冻干燥:
以包埋菌粉处理工艺将步骤3中所收集的嗜酸乳杆菌泥进行冷冻干燥制备,具体步骤为:a.菌泥加入10%比例的葡萄糖、10%比例的脱脂乳粉、5%比例的低聚木糖,混匀;b.盛入冷冻平皿在-30℃冷冻50分钟;c.在-80℃冷冻60分钟;d.在冷冻真空干燥机上冷冻干燥15小时;e.冻干粉密闭保存-4~-6℃冰箱中;

取步骤4中制备的嗜酸乳杆菌冻干粉2~5份、植物乳杆菌冻干粉2~5份、干酪乳杆菌冻干粉2~5份、两歧双歧杆菌冻干粉2~5份、青春双歧杆菌冻干粉2~5份、长双歧杆菌冻干粉2~5份、鼠李糖乳杆菌冻干粉2~5份,另取菊粉8份、低聚木糖6份、低聚异麦芽糖8份,在无菌状态下混匀后装袋,获得一种改善人体肠道健康的乳酸菌固体饮料,有效活菌≥5.0×109CFU/g。
本发明的作用原理为:1.通过筛选和培育功能性乳酸菌(嗜酸乳杆菌、植物乳杆菌、干酪乳杆菌、两歧双歧杆菌、青春双歧杆菌、长双歧杆菌、鼠李糖乳杆菌),耐胃酸、耐胆盐和胰液,顺利到达肠道后,乳酸菌生成各种胞外酶帮助消化分解食物中的营养(蛋白质、淀粉、脂肪)、生成各种维生素、促进矿物质和微量元素的吸收,供给机体各系统充足和均衡的营养;2.乳酸菌代谢生成短链脂肪酸(乙酸、丙酸和丁酸等),抑制病原菌、刺激肠道隐窝干细胞分化、修复肠道上皮,保持肠道上皮细胞、内分泌细胞、潘氏细胞等正常工作,改善腹泻;3.短链脂肪酸(乙酸、丙酸和丁酸),可以刺激肠道平滑肌蠕动,促进肠道杯状细胞正常分泌粘液,改善便秘。以下结合实施例对本发明的效果做详细说明。
1、实验准备
1)实验人员:年龄在10~80岁之间的腹泻患者;年龄在10~80岁之间便秘患者;
2)实验产品:本发明的乳酸菌固体饮料;
2、实验设计
1)实验人员分为实验组(服用本发明的产品)和对照组(未服用本发明的产品);
2)本发明的产品服用方法为:一天2次,一次2克;腹泻患者服用2天;便秘患者服用7天。
3)腹泻患者的改善以粪便成形状为标准;便秘患者的改善以粪便松软为标准。
4)腹泻患者和便秘患者服用的产品为下述的实施例1~4。其中,实施例1~4分别依据表1中所示的原料配方及本发明所述工艺步骤获取。

4、实验结论
通过实施例1、实施例2、实施例3和实施例4的实验结果数据可以看出:腹泻患者实验组中改善率最低为94%,最高为100%,不良反应为0例;对照组中腹泻患者改善率为20%。实验组中便秘患者改善率最低为80%,最高为86%,不良反应为0例;对照组中便秘患者改善率为5%。实验组对腹泻患者和便秘患者改善率远远高于对照组。
综上所述,本发明的乳酸菌固体饮料在改善人体肠道健康方面,效果显著。

1.一种改善人体肠道健康的乳酸菌固体饮料,其特征在于,包括以下质量份数的组分:嗜酸乳杆菌2~5份、植物乳杆菌2~5份、干酪乳杆菌2~5份、两歧双歧杆菌2~5份、青春双歧杆菌2~5份、长双歧杆菌2~5份、鼠李糖乳杆菌2~5份、菊粉8份、低聚木糖6份、低聚异麦芽糖8份。
2.一种如权利要求1所述的改善人体肠道健康的乳酸菌固体饮料的制备方法,其特征在于,包括以下步骤:
步骤1,培养基制作:
首先,将蛋白胨4份、葡萄糖5份、酵母粉1份、脱脂乳1份、乙酸钠0.5份、轻质碳酸钙0.3份、柠檬酸铵0.3份、吐温0.2份、硫酸镁0.02份、硫酸锰0.01份、蒸馏水75~85份,加热至70~73℃左右,搅拌使各原料溶解;然后,以20%的氢氧化钠溶液调pH为7.0~7.2,完成配料;再以120~125℃灭菌20分钟后,冷却至37℃左右,获得灭菌培养基;

以包被菌粉处理工艺将步骤3中所收集的嗜酸乳杆菌泥进行冷冻干燥制备,具体步骤为:a.菌泥加入10%比例的葡萄糖、10%比例的脱脂乳粉、5%比例的低聚木糖,混匀;b.盛入冷冻平皿在-30℃冷冻50分钟;c.在-80℃冷冻60分钟;d.在冷冻真空干燥机上冷冻干燥15小时;e.冻干粉密闭保存-4~-6℃冰箱中;

干酪乳杆菌的菌泥冷冻干燥方法同上述嗜酸乳杆菌;
两歧双歧杆菌的菌泥冷冻干燥方法同上述嗜酸乳杆菌;
青春双歧杆菌的菌泥冷冻干燥方法同上述嗜酸乳杆菌;
长双歧杆菌的菌泥冷冻干燥方法同上述嗜酸乳杆菌;
鼠李糖乳杆菌的菌泥冷冻干燥方法同上述嗜酸乳杆菌;
步骤5,成品制备:
取步骤4中制备的嗜酸乳杆菌冻干粉2~5份、植物乳杆菌冻干粉2~5份、干酪乳杆菌冻干粉2~5份、两歧双歧杆菌冻干粉2~5份、青春双歧杆菌冻干粉2~5份、长双歧杆菌冻干粉2~5份、鼠李糖乳杆菌冻干粉2~5份,另取菊粉8份、低聚木糖6份、低聚异麦芽糖8份,在无菌状态下混匀后装袋,获得一种改善人体肠道健康的乳酸菌固体饮料,有效活菌≥5.0×109CFU/g。
