Abstract
本发明属于微生物饲料领域,特别涉及一种利用复合微生物菌共发酵甘蔗渣生产饲料的方法,包括如下步骤:(1)制备菌株种子液:(2)选取甘蔗渣;(3)发酵制备饲料。本申请利用一次压榨得到的新鲜甘蔗渣为原料,通过白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳杆菌等多菌种共同发酵生产得到高蛋白、高纤维饲料,既能解决甘蔗渣的处理问题,减少环境污染,又能变废为宝,实现甘蔗渣的资源化利用,缓解畜牧业发展与饲料短缺的矛盾问题。
Description
技术领域
本发明属于微生物饲料领域,特别涉及一种利用复合微生物菌共发酵甘蔗渣生产饲料的方法。
背景技术
随着我国人口的增长,对粮食和畜肉、奶产品的需求量迅猛增长,给国家农业生产带来了巨大压力。而饲料是畜牧业赖以发展的基础。目前我国的畜牧业发展除大量发展养猪、养鸡、鸭、鹅外,正加大速度发展肉牛、奶牛、肉羊甚至奶羊事业,因此反刍家畜的发展进入一个快速发展阶段。随着畜牧业的发展,饲料方面的生产出现了瓶颈问题,即畜牧业的发展与饲料短缺的矛盾。这就要求我们既要充分地合理开发农作物秸秆等粗饲料资源,又要开发利用工业废物等生产短缺的蛋白质饲料资源。
新鲜甘蔗经过一次压榨得到的蔗渣,含水量为45-50%,其中蔗糖的相对百分含量高达4%。同时蔗渣含有较高的木质纤维素类物质,主要是纤维素、半纤维素和木质素,其中纤维素达40%,半纤维素达18%。蔗渣含粗蛋白低,约2%-4%。一次压榨得到的甘蔗渣直接喂养动物难消化且营养不足,目前这些甘蔗渣大部分拿来烧锅炉或造纸用,如能借助多种微生物共同作用,改变甘蔗渣的木质纤维结构状况,增加营养物质的含量,制成牛羊养殖饲料,将能更好利用自然资源,给饲料工业带来新资源。
发明内容
本发明的目的在于提供一种利用复合微生物菌共发酵甘蔗渣生产饲料的方法,以实现甘蔗渣的资源化利用,缓解畜牧业发展与饲料短缺的矛盾问题。
利用复合微生物菌共发酵甘蔗渣生产饲料的方法,包括如下步骤:
(1)制备菌株种子液:将新鲜压榨的甘蔗汁浓度分别调整至15-30 oBX,作为微生物菌种的液体培养基;选择低浓度的甘蔗汁,并使其起始pH5.0-7.0,分别接入白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳杆菌,并分别制成四种菌种悬浮液;将四种菌种悬浮液混合并用高浓度的甘蔗汁逐级扩大培养,培养温度为25-30℃,培养3-5天,制成微生物发酵新鲜甘蔗渣的专用菌株种子液;
(2)选取甘蔗渣:选择甘蔗经过一次压榨后得到的新鲜甘蔗渣;所述甘蔗渣含水量为45-50%,其中蔗糖的质量含量为3-5%;
(3)发酵制备饲料:将步骤(1)制备的菌株种子液均匀喷淋到新鲜甘蔗渣上,搅拌均匀,进行厌氧和好氧发酵,发酵温度25-45℃,发酵时间为7-10天,即可制成富含蛋白质、氨基酸、消化酶、抗生素、乳酸、双歧因子、抗氧化物质、免疫小肽等多种活性物质的饲料。其中,蛋白质质量含量占干物质的18-23%。
白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳酸杆菌均为我国农业部公布允许使用的饲料级微生物。(1)白腐菌是一种霉菌,在生长过程中主要产生纤维素酶,具有很强的分解纤维素能力,破坏甘蔗渣纤维素结构并降解甘蔗渣纤维素。(2)枯草芽孢杆菌是一种需氧菌,可利用蛋白质、多种糖类及淀粉,在培养过程中产生枯草素、多粘菌素、制霉菌素、短杆菌肽等活性物质,这些活性物质对于致病菌或内源性感染的条件致病菌有明显的一致作用;同时枯草芽孢杆菌可迅速消耗环境游离的氧,造成动物肠道缺氧,促进有益的厌氧菌生长,并产生乳酸等有机酸类,降低肠道pH值,间接抑制其他致病菌的生长;枯草芽孢杆菌能合成α-淀粉酶、蛋白酶和纤维素酶等酶类,在消化道内与动物体内的消化酶类共同发挥作用;也能合成维生素B1、B2、B6、烟酸等多种B族维生素,提高动物体内干扰素和巨噬细胞的活性,刺激动物免疫器官的生长发育,提高免疫球蛋白和抗体水平。(3)产朊假丝酵母其蛋白质和维生素B的含量都比啤酒酵母高,能以尿素和硝酸作为氮源,充分利用五碳糖和六碳糖,在培养基中不需要加入任何生长因子即可生长。既能利用造纸工业的亚硫酸废液,还能利用糖蜜、木材水解液等生产出人畜可食用的蛋白质。酵母菌体蛋白的营养价值很高,饲料酵母干物质中蛋白质含量可高达50%,其赖氨酸含量比大豆还高,接近动物蛋白,色氨酸含量比大豆高7倍以上,还含有B族维生素、矿物质以及其他生理活性物质,一直作为单细胞蛋白被广泛使用。活酵母可分泌多种酶,如淀粉酶、蛋白酶、纤维素酶等。使用产朊假丝酵母饲料可加强对营养物质的消化利用,促进畜禽生长,增进其食欲,增强抵抗疾病和抗应激的能力。(4)植物乳杆菌是一种活性很高的乳酸菌,能产生大量乳酸而使环境的pH值下降,而且其产出的酸性物质能降解重金属。植物乳酸菌为兼性好氧的厌氧细菌,在繁殖过程中能产出一种特有生物型的防腐剂—乳酸杆菌素。
白腐菌、枯草芽孢杆菌、产朊假丝酵母菌和植物乳杆菌等多种菌种组成复合发酵体系,利用新鲜甘蔗朗汁营养成分共同培养成为发酵甘蔗渣的微生物菌种子液,喷淋转接到新鲜甘蔗渣上,经过发酵培养,各种菌株相互作用、相互促进,形成共生共存的关系,转化新鲜甘蔗渣中的糖分,降解甘蔗渣纤维素,生产出富含蛋白质、氨基酸、消化酶、抗生素、乳酸、双歧因子、抗氧化物质、免疫小肽等活性物质的蛋白饲料。通过多菌种发酵处理,提高了新鲜甘蔗渣的综合利用,降低了饲料的成本,改善了饲料的品质。促进畜牧业和以工业废弃物为原料生产饲料的工业化。
优选的,步骤(1)中,白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳杆菌的用量为1:1:1:1。即取相同质量的四种菌种,再用相同浓度相同体积的甘蔗汁分别溶解菌种,制备出四种菌种悬浮液。
优选的,步骤(1)中,将新鲜压榨的甘蔗汁浓度分别调整至15 oBX、20 oBX、30 oBX,15 oBX的甘蔗汁用于制备菌种悬浮液,20 oBX和30 oBX的甘蔗汁分别用于菌种悬浮液逐级扩大培养。
优选的,四种不同的微生物菌种分别接入含有15oBX甘蔗汁的斜面培养基,在25-30℃的条件下培养时间3-4天,各自进行活化并驯化菌种,并分别制成四种悬浮液;把四种不同菌种悬浮液以10%的接种量转接入装有20oBX甘蔗汁的容器种进行种子培养,在25-30℃的条件下培养时间3-4天,把培养好的四种不同菌种种子液等量混合制成混合菌液,按浓度15%的量将混合菌液接入装有30oBX甘蔗汁的培养容器中,在25-30℃的条件下培养时间3-4天,扩大培养成甘蔗渣发酵培养的多菌株种子液。
优选的,步骤(2)中,所述甘蔗渣中粗蛋白的质量含量为2-4%,纤维素的质量含量为35-45%,半纤维素的质量含量为15-20%。
优选的,步骤(3)中,菌株种子液的喷淋量为:控制甘蔗渣含水量为60%-70%。
优选的,步骤(3)中,甘蔗渣喷淋菌株种子液后分装打包密封后进行厌氧发酵。
本申请的另一技术方案,将上述制备好的饲料代替牧草用于牛或羊的喂养。
本申请的又一技术方案,往上述制备好的饲料中添加8%的玉米粉,制成颗粒饲料后用于家禽家畜或水产鱼的喂养。采用市面上常用的饲料加工方法进行饲料的造粒。
与现有的技术相比,本发明具有如下有益效果:
1、本申请充分利用工业制糖或甘蔗饮品生产等剩余的甘蔗渣,通过喷淋自配的复合微生物菌液进行发酵,改变甘蔗渣的木质纤维结构状况,增加营养物质的含量,制成牛羊养殖饲料,既能解决甘蔗渣的处理问题,减少环境污染,又能变废为宝,实现甘蔗渣的资源化利用,缓解畜牧业发展与饲料短缺的矛盾问题。
2、本申请利用新鲜甘蔗汁来培养制备菌株种子液,充分利用工业生产中的产品,无需额外采购和添加其他辅料,且菌种的培养基主要成分与饲料发酵生产的相似,有利于饲料的发酵生产,在提高发酵效率的基础上能更好地转化甘蔗渣中的营养成分,制备出来的饲料富含蛋白质、氨基酸、消化酶、抗生素、乳酸、双歧因子、抗氧化物质、免疫小肽等多种活性物质,其中蛋白质质量含量占干物质的18-23%,可直接用于牛羊等反刍家畜的喂养。
具体实施方式
下面对本发明的具体实施方式进行详细描述,但应当理解本发明的保护范围并不受具体实施方式的限制。
实施例1
利用复合微生物菌共发酵甘蔗渣生产饲料的方法,包括如下步骤:
(1)制备菌株种子液:新鲜甘蔗汁加水将浓度调整为15oBX、20oBX和30 oBX,分别制成浓度为15oBX的斜面培养基及浓度为20oBX和30 oBX的液体培养基。取等量的白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳杆菌,并将四种不同的微生物菌种分别接入含有15oBX糟液的斜面培养基,25℃,培养时间3天,各自进行活化并驯化菌种,并分别制成悬浮液。把四种不同菌种悬浮液以10%的接种量转接入20oBX新鲜甘蔗汁的三角瓶进行种子培养,25℃,培养时间3天,把培养好的四种不同菌种种子液等量混合制成混合菌液,按浓度15%的量接入30oBX新鲜甘蔗汁培养基种子罐中,25℃,培养时间3天,扩大培养成甘蔗渣发酵培养的多菌株种子液。
(2)选取甘蔗渣:选择甘蔗经过一次压榨后得到的新鲜甘蔗渣;所述甘蔗渣中蔗糖的质量含量为3-5%;粗蛋白的质量含量为2-4%,纤维素的质量含量为35-45%,半纤维素的质量含量为15-20%。
(3)发酵制备饲料:将制好的菌株种子液喷淋到新鲜甘蔗渣上,边喷淋边搅拌均匀,喷淋量控制甘蔗渣含水量为60%为宜。然后分装打包密封厌氧发酵,发酵时间7天,制成富含蛋白质、氨基酸、消化酶、抗生素、乳酸、双歧因子、抗氧化物质、免疫小肽等活性物质的饲料。经检测蛋白含量可达18.48%,可以代替牛羊的牧草喂养。也可添加8%的玉米粉,可制备成颗粒饲料,应用于家禽家畜和水产鱼饲料。
实施例2
利用复合微生物菌共发酵甘蔗渣生产饲料的方法,包括如下步骤:
(1)制备菌株种子液:将压榨得到的新鲜蔗汁用纯水将浓度调整为15oBX、22oBX和30oBX,分别制成浓度为15oBX的斜面培养基及浓度为22oBX和30 oBX的液体培养基。取等量的白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳杆菌,并将四种不同的微生物菌种分别接入含有15oBX糟液的斜面培养基,30℃,培养时间5天,各自进行活化并驯化菌种,并分别制成悬浮液。把四种不同菌种悬浮液以10%的接种量转接入22oBX新鲜甘蔗汁的三角瓶进行种子培养,30℃,培养时间5天,把培养好的四种不同菌种种子液等量混合制成混合菌液,按浓度15%的量接入30oBX新鲜甘蔗汁培养基种子罐中,30℃,培养时间5天,扩大培养成甘蔗渣发酵培养的多菌株种子液。
(2)选取甘蔗渣:选择甘蔗经过一次压榨后得到的新鲜甘蔗渣;所述甘蔗渣中蔗糖的质量含量为3-5%;粗蛋白的质量含量为2-4%,纤维素的质量含量为35-45%,半纤维素的质量含量为15-20%。
(3)发酵制备饲料:将制好的菌株种子液喷淋到新鲜甘蔗渣上,边喷淋边搅拌均匀,喷淋量控制甘蔗渣含水量为70%为宜。后分装打包密封厌氧发酵,发酵时间10天,制成富含蛋白质、氨基酸、消化酶、抗生素、乳酸、双歧因子、抗氧化物质、免疫小肽等活性物质的饲料。检测蛋白含量高达22.41%,可完全替代牛羊的牧草喂养。也可添加8%的麦麸,可制备成颗粒饲料,应用于家禽家畜和水产鱼饲料。
对比实施例1
本实施例的方法与实施例1的相似,其区别在于:对比实施例1只利用单一菌株产朊假丝酵母喷淋甘蔗渣进行发酵,结果蛋白质含量只有10.21%。
因为没有白腐菌和枯草芽孢杆菌分泌纤维素酶降解甘蔗渣的纤维素和木质素等,同时造成产阮假丝酵母生产培养基糖分不足,最终造成单细胞蛋白质含量偏低,仅达到10.21%,而甘蔗渣纤维素和木质素降解没有多大变化,甘蔗渣表观为较为坚硬,得到的甘蔗渣饲料适口性差。无法与实施例1,实施例2相比。在实施例1和实施例2中经过多菌株联合发酵后,营养成分有所提高,尤其是粗蛋白含量。妨碍动物营养消化的纤维素和木质素有所下降,加上植物乳酸菌的发酵,增加了动物的适口性。甘蔗渣中的糖类、降解纤维素和木质素所生成的水解物经过微生物代谢作用并转化为蛋白质和脂类等,改变了甘蔗渣的结构和成分。
对比实施例2
本实施例的方法与实施例2的相似,其区别在于:对比实施例2只利用单一菌株白腐菌喷淋甘蔗渣进行发酵,结果蛋白质含量只有5.89%,只是甘蔗渣纤维素和木质素降解有了较大的变化,纤维素降解率为55.69,木质素降解率为32.58%;甘蔗渣表观为较为柔软。但是由于没有接种产阮假丝酵母菌造成单细胞蛋白质含量偏低,仅为5.89%,得到的甘蔗渣饲料营养成分低。无法与实施例1,实施例2相比。在实施例1和实施例2中经过多菌株联合发酵后,营养成分有所提高,尤其是粗蛋白含量。妨碍动物营养消化的纤维素和木质素有所下降,加上植物乳酸菌的发酵,增加了动物的适口性。甘蔗渣中的糖类、降解纤维素和木质素所生成的水解物经过微生物代谢作用并转化为蛋白质和脂类等,改变了甘蔗渣的结构和成分。
实施例3
将实施例2制备的饲料用于羊的喂养,并与用牧草青贮饲料喂养的做对照,喂养6个月,测量羊的体重和料肉比,结果见下表。
由上表可见,本申请方法制备的饲料可以代替常规的牧草青贮饲料来喂养羊,且还可节约饲料的喂养量,降低饲料成本。缓解畜牧业发展与饲料短缺的矛盾问题。

Claims (9)
1.利用复合微生物菌共发酵甘蔗渣生产饲料的方法,其特征在于,包括如下步骤:
(1)制备菌株种子液:将新鲜压榨的甘蔗汁浓度分别调整至15-30 oBX,作为微生物菌种的液体培养基;选择低浓度的甘蔗汁,并使其起始pH5.0-7.0,分别接入白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳杆菌,并分别制成四种菌种悬浮液;将四种菌种悬浮液混合,并用高浓度的甘蔗汁逐级扩大培养,培养温度为25-30℃,培养3-5天,制成微生物发酵新鲜甘蔗渣的专用菌株种子液;
(2)选取甘蔗渣:选择甘蔗经过一次压榨后得到的新鲜甘蔗渣;所述甘蔗渣含水量为40-50%,其中蔗糖的质量含量为3-5%;
(3)发酵制备饲料:将步骤(1)制备的菌株种子液均匀喷淋到新鲜甘蔗渣上,搅拌均匀,进行厌氧和好氧发酵,发酵温度25-45℃,发酵时间为7-10天,即可制成富含多种活性物质的饲料。
2.根据权利要求1所述的利用复合微生物菌共发酵甘蔗渣生产饲料的方法,其特征在于,步骤(1)中,白腐菌、枯草芽孢杆菌、产朊假丝酵母和植物乳杆菌的用量为1:1:1:1。
3. 根据权利要求1所述的利用复合微生物菌共发酵甘蔗渣生产饲料的方法,其特征在于,步骤(1)中,将新鲜压榨的甘蔗汁浓度分别调整至15 oBX、20 oBX、30 oBX,15 oBX的甘蔗汁用于制备菌种悬浮液,20 oBX和30 oBX的甘蔗汁分别用于菌种悬浮液逐级扩大培养。
4.根据权利要求3所述的利用复合微生物菌共发酵甘蔗渣生产饲料的方法,其特征在于,四种不同的微生物菌种分别接入含有15oBX甘蔗汁的斜面培养基,在25-30℃的条件下培养时间3-4天,各自进行活化并驯化菌种,并分别制成四种悬浮液;把四种不同菌种悬浮液以10%的接种量转接入装有20oBX甘蔗汁的容器种进行种子培养,在25-30℃的条件下培养时间3-4天,把培养好的四种不同菌种种子液等量混合制成混合菌液,按浓度15%的量将混合菌液接入装有30oBX甘蔗汁的培养容器中,在25-30℃的条件下培养时间3-4天,扩大培养成甘蔗渣发酵培养的多菌株种子液。
5.根据权利要求1所述的利用复合微生物菌共发酵甘蔗渣生产饲料的方法,其特征在于,步骤(2)中,所述甘蔗渣中粗蛋白的质量含量为2-4%,纤维素的质量含量为35-45%,半纤维素的质量含量为15-20%。
6.根据权利要求1所述的利用复合微生物菌共发酵甘蔗渣生产饲料的方法,其特征在于,步骤(3)中,菌株种子液的喷淋量为:控制甘蔗渣含水量为60%-70%。
7.根据权利要求1所述的利用复合微生物菌共发酵甘蔗渣生产饲料的方法,其特征在于,步骤(3)中,甘蔗渣喷淋菌株种子液后分装打包密封后进行厌氧发酵。
8.权利要求1-7任一项所述的饲料的应用,其特征在于,制备的饲料用于牛或羊的喂养。
9.权利要求1-7任一项所述的饲料的应用,其特征在于,往制备好的饲料中添加8%的玉米粉,制成颗粒饲料后用于家禽家畜或水产鱼的喂养。
