(1) sterilization test
The experimental method comprises the following steps:
preparing experimental bacteria liquid: respectively inoculating staphylococcus aureus, streptococcus agalactiae, streptococcus dysgalactiae and escherichia coli to a nutrient agar culture medium for culturing for 24 hours, then inoculating the staphylococcus aureus to a nutrient broth culture medium, the streptococcus agalactiae and the streptococcus dysgalactiae to a fresh blood agar culture medium, inoculating the escherichia coli to an LB culture medium, and culturing for 18-24 hours at 37 ℃ for later use;
the bacterial liquid is divided into 4 groups, which are respectively as follows: a group of staphylococcus aureus experimental group, B group of streptococcus agalactiae experimental group, C group of streptococcus dysgalactiae experimental group and D group of escherichia coli experimental group; each group of experiments is provided with 6 reaction tubes, and 1mL of corresponding bacterial liquid is added into each tube of each group; after 1-6 test samples are sequentially and respectively added into each group of 6 reaction tubes, 1mL of bacterial liquid of 4 groups of 26 reaction tubes is taken at 5 th, 10 th, 15 th, 20 th, 25 th and 30 th s, 9mL of neutralizing agent is added, shaking is carried out uniformly, after neutralization for 10min, 1mL of the bacterial liquid is taken out, 9mL of corresponding liquid culture medium is added, after shaking is carried out uniformly, the bacterial liquid is placed into a constant temperature box for culturing at 37 ℃ for 24h, the growth condition of bacteria in each tube is observed, and the results are shown in Table 1:
according to the experimental results, the technical scheme of the invention can kill staphylococcus aureus, streptococcus agalactiae, streptococcus dysgalactiae and escherichia coli more easily and rapidly, has quick sterilization, is used for diminishing inflammation and sterilizing the nipples of the dairy cows, and can achieve the aim more easily;
(2) anti-dripping property test
The experimental method comprises the following steps: taking 6 identical glass rods and 6 identical beakers, sequentially marking the glass rods as 1-6 and the beakers as 1-6, forming a group of the glass rods and the beakers with the same number to obtain experimental groups A-F, respectively inserting the 6 glass rods into the experimental samples 1-6 at the same time to dip the samples, staying for 5s, simultaneously removing the glass rods, hanging the glass rods above the corresponding beakers, recording the dripping time of each sample drop by using a stopwatch, and finally removing the average dripping time;
average drop time = total elapsed time/total number of drops after last drop time
And calculating the retention rate of each group of effective components:
effective component storage rate = (dipping mass-dripping mass)/dipping mass × 100%
Wherein, the dipping mass = the mass of the experimental sample-the mass of the experimental sample after dipping;
drop mass = beaker mass after test-empty beaker mass;
the results of the experiment are shown in table 2:
the experimental results in table 2 show that the adhesiveness of the bath lotion is smaller than that of the bath lotion obtained by the prior art, but the average dripping time and the retention rate of active ingredients are far greater than those of the prior art, which indicates that the bath lotion has small viscosity, saves the using amount, has excellent anti-dripping performance, can stay on the surface of an object for a long time, and achieves the effects of resisting bacteria, sterilizing and protecting skin for a long time;
(3) freezing point test of bath liquid
The measuring instrument is as follows: freezing point tester
The test method comprises the following steps: aligning a refraction prism of the freezing point tester to the bright direction, adjusting the ocular diopter ring until the marked line is clear, and calibrating the tester to ensure that the blue-white boundary in the ocular visual field coincides with the reference line. During testing, 3 drops of a sample to be tested are dropped on the dioptric prism, the cover plate is covered and slightly pressed, and the scale corresponding to the blue-white boundary is read out to be a measurement result;
the results of the experiment are shown in table 3:
according to the test results, the freezing point of the bath lotion is far lower than that of the bath lotion prepared by the prior art, which shows that the bath lotion has low freezing point, the system is not easily affected by the external environment and the use is convenient;
(4) test for moisture Retention Performance
The test instrument: skin moisture tester
Dividing the back of the hand into 6 areas with the same size, cleaning the areas, and keeping the moisture value of the test value unchanged; the same amount of the experimental samples 1-6 is respectively coated on 6 areas, timing is started, the probe of the tester is vertically pressed on the surface of the skin to be tested every 30 minutes, and the skin moisture value is read and recorded. The test was performed 3 times and the mean values were taken for the corresponding time points. After each test, the skin is washed clean by water and naturally laid for 3 hours. The results are reported in table 4:
as can be seen from the data of the humidity value in the table 4, the test samples 1-3 of the bath lotion are far higher than the test samples 4-6 of other bath lotions, and the humidity value is not obviously reduced within 180min, which shows that the bath lotion has good moisturizing performance and long duration;
(5) clinical experiments
Selecting 120 cows with non-difference body conditions as test objects in a certain pasture in Shaanxi, dividing the 120 cows into 6 groups A-F, 20 cows in each group, milking twice a day, after milking each time, sequentially smearing test samples 1-6 medicinal bath liquid on the teats of the 6 groups A-F of cows respectively, measuring the number of somatic cells in the milk sample after milking each time, and stopping the medicinal bath liquid after the experiment lasts for one month; the teat condition of the cows was recorded during the experiment and the somatic cell count and incidence of stealth mastitis were determined in milk samples taken one month and one week off using the bath solution, respectively, and the results are shown in table 5:
from the clinical test results, the nipple medicated bath liquid of the technical scheme has better wetting effect after being tried for one month; the incidence of subclinical mastitis is lower; the number of somatic cells is obviously reduced compared with that of a control group, which shows that the nipple medicinal bath liquid has better effect of repairing mammary tissues. After the medicine is stopped for one week, the nipple of the milk cow is chapped or turned outwards, and the incidence rate of the recessive mastitis is increased in the medicine bath liquid obtained by the prior art, but the nipple of the milk cow has no obvious abnormality after the medicine is stopped for one week, and the incidence rate of the recessive mastitis is still low, which shows that the medicine bath liquid has an obvious effect on improving the self-defense function of the milk cow;
the experimental results show that the compound cow nipple medicated bath liquid is superior to the medicated bath liquid obtained in the prior art in sterilization, anti-dripping, moisturizing and low-temperature resistance, and the clinical experimental results are superior to the prior art in skin care, antibiosis, prevention and treatment of inflammation recurrence and the like.
