Abstract

技术领域

背景技术
类沸石咪唑骨架材料(ZIFs)由金属离子与咪唑类配体连接而成,其具有类似沸石拓扑结构的三维四面体结构,表现出很好的热稳定性和化学稳定性而广泛用于吸收、催化和电化学传感等领域。ZIF-8是由硝酸锌和2-甲基咪唑配合而成,通过进一步聚集形成方钠石拓扑结构。以ZIF-8为前驱体,经煅烧后形成氮掺杂多孔碳(NPC),其具有以下的优异特性:一是多孔碳材料中氮原子的孤对电子作为载流子能使多孔碳材料周围的电荷密度增大,从而提高多孔碳材料的导电性;二是氮原子的掺入使得多孔碳材料中的缺陷位增长,使得多孔碳材料中的活性位点数量增加;三是氮原子的存在能够增加多孔碳材料作为电极材料与电解液的亲和性和生物相容性;四是氮原子的存在能够改善多孔碳材料与金属离子的结合能力,提高金属在多孔碳材料表面的负载能力。故本方法利用ZIF-8为前驱体制备NPC后,进一步与氮掺杂石墨烯(NGR)混合制备氮掺杂石墨烯-氮掺杂多孔碳(NGR-NPC)复合纳米材料。这种具有富氮成分的复合纳米材料是一种优良的电极修饰材料,它与电解液具有良好的亲和性,电解液中的小分子能与电极修饰材料快速进行电子转移。

发明内容
为解决上述现有技术存在的问题,本发明的目的在于提供一种氮掺杂石墨烯-氮掺杂多孔碳(NGR-NPC)的制备方法并用于制备化学修饰电极,用于生物样品中多巴胺、尿酸和抗坏血酸含量的检测,旨在通过简单的电化学检测方法,实现生物样品中多巴胺、尿酸和抗坏血酸含量的快速、有效测定。
电化学传感器的灵敏度和选择性主要取决于电极上的换能器(信号转换元件)和识别器(敏感元件),其中换能器的界面传感材料需要有高电子传导性、高比表面积与良好的催化活性等特征,以提高换能器对敏感材料的吸附性能及生物相容性、降低氧化还原反应过电势、提高生物传感器的选择性与灵敏度。因此,本发明利用ZIF-8为前驱体制备NPC后,进一步与NGR混合制备NGR-NPC纳米复合材料。这种具有富氮成分的纳米复合材料作为电极修饰材料与电解液具有良好的亲和性,能加快电解液中小分子与电极表面电子传递速率,从而构建快速有效的电化学传感器件,实现生物样品中多巴胺、尿酸和抗坏血酸含量的同时测定。
为达到上述目的,本发明的技术方案为:NGR-NPC纳米复合材料用于化学修饰电极的制备,进一步建立生物样品中多巴胺、尿酸和抗坏血酸含量同时检测的方法,步骤如下。

通过溶剂热法合成ZIF-8,其形貌为十二面体;在氮气气氛中高温碳化,形成NPC;取1.0 mg NPC和1.0 mg NGR在旋涡震荡仪上震荡20分钟后,形成NGR-NPC纳米复合材料,其形貌表现为NPC附着在NGR表面或NPC被NGR包裹。NGR中氮原子的存在可以使氮孤对电子与石墨烯的π轨道共扼,进而表现出优良的电子和机械性能;NPC中氮原子的孤对电子作为载流子能使多孔碳材料周围的电荷密度增大,因此能有效提高多孔碳材料的导电性。二者混合产生协同作用,其比表面积和表面的附着作用力都优于其单独存在时的大小,从而获得高电子传导、高比表面积的电极界面修饰材料(NGR-NPC)。
步骤二、基于NGR-NPC复合纳米材料修饰电极检测多巴胺、尿酸和抗坏血酸。
取1.0 mg NGR-NPC固体粉末和100 μL Nafion乙醇溶液,加入1.0 mL超纯水并超声分散2~3 h后得到Nafion-NGR-NPC分散液。移取6.0 µL Nafion-NGR-NPC分散液滴涂于玻碳电极(d=3 mm, GCE)表面,静置晾干,得工作电极(Nafion-NGR-NPC/GCE)。
将构建好的Nafion-NGR-NPC/GCE用于生物样品中多巴胺、尿酸和抗坏血酸的快速同时检测,其中磷酸盐缓冲溶液的最佳pH为3.0。在最佳的实验条件下,1)考察了扫描速度分别对多巴胺、尿酸和抗坏血酸电化学行为的影响,根据扫描速度与氧化峰电流值线性拟合得出的方程说明修饰电极对多巴胺、尿酸和抗坏血酸主要是吸附控制;2)采用差分脉冲伏安法对多巴胺、尿酸和抗坏血酸进行检测,根据浓度和氧化峰电流值的线性关系得出该修饰电极对多巴胺、尿酸和抗坏血酸的线性范围和检测限;3)修饰电极在检测多巴胺、尿酸和抗坏血酸时显示出优良的抗干扰能力,通过标准曲线法对生物样品中的多巴胺、尿酸和抗坏血酸进行检测,得出该修饰电极对生物药品中的多巴胺、尿酸和抗坏血酸具有良好的响应,可用于对生物样品的检测。
相对于现有技术,本发明的有益效果有以下几点。
本发明是一种基于NGR-NPC复合纳米材料修饰电极检测多巴胺、尿酸和抗坏血酸含量的方法,采用本方法制备化学修饰电极是一种快速而又简单的方法,检测方法操作简便、快速和成本低廉,可以有效、快速地对生物样品中的多巴胺、尿酸和抗坏血酸进行检测。同时本发明方法可以消除样品准备、检测样品复杂、耗时的缺陷,该检测分析方法简单便捷、重现性好且灵敏度高。

具体实施方式
下面结合附图和具体实施方式对本发明技术方案做进一步详细描述:
试验例:
1、NPC的合成。
将16 mmol/L 2-甲基咪唑溶解在7.5 mL甲醇中并记为溶液A;将4 mmol/L硝酸锌分散在15 mL甲醇中记为溶液B。将两种溶液分别超声处理15分钟,向溶液B缓慢滴加溶液A,并在室温下磁力搅拌60分钟。将混合物移入100 mL聚四氟乙烯内胆的反应釜中,保持120℃反应4小时得到白色浑浊液。离心分离并分别用甲醇和N,N-二甲基甲酰胺洗涤两次,在70℃下真空干燥12小时得到ZIF-8白色粉末。将ZIF-8放置于瓷舟中并转移到管式炉,在氮气保护下升温至900 ℃(升温速率为5 ℃/min),保持3小时后梯度降温(降温速率为5 ℃/min)。ZIF-8在氮气气氛900℃下热解后,转化为NPC。
2、NGR-NPC复合纳米材料修饰电极的制备。
取1.0 mg NGR-NPC固体粉末和100 μL Nafion乙醇溶液,加入1.0 mL超纯水并超声分散2~3 h后得到Nafion-NGR-NPC分散液。移取6.0 µL Nafion-NGR-NPC分散液滴涂于玻碳电极(d=3 mm, GCE)表面,静置晾干,得工作电极(Nafion-NGR-NPC/GCE)。
结果与讨论。
3、基于NGR-NPC复合纳米材料电化学传感器同时检测多巴胺、尿酸和抗坏血酸。
(1)修饰电极对多巴胺、尿酸和抗坏血酸的电化学行为。
分别配制1.0 mmol/L多巴胺、1.0 mmol/L尿酸和100 mmol/L抗坏血酸标准溶液,用0.1 mol/L的磷酸盐缓冲溶液,配制含10 µmol/L多巴胺、10 μmol/L尿酸和100 μmol/L抗坏血酸的pH为3.0的磷酸缓冲混合溶液。扫描范围为-0.2~1.0 V,扫描速度为100 mV/s。
为了研究Nafion-NGR-NPC/GCE同时检测多巴胺、尿酸和抗坏血酸的灵敏度和选择性,采用循环伏安法分别以Nafion/GCE(a)、Nafion-NGR/GCE(b)、Nafion-NPC/GCE(c)和Nafion-NGR-NPC/GCE(d)为工作电极进行电化学检测(图1)。Nafion-NGR/GCE的峰电流较Nafion/GCE的高;但在Nafion/GCE和Nafion-NGR/GCE上有一个氧化峰,表明在Nafion/GCE和Nafion-NGR/GCE上多巴胺、尿酸和抗坏血酸的氧化峰发生重叠,选择性和灵敏度低。由于NPC具有大的比表面积和高导电能力,在Nafion-NPC/GCE上对多巴胺、尿酸和抗坏血酸的响应显著增加且氧化峰相互分开;当NPC-NGR混合材料修饰玻碳电极时,由于氮掺杂石墨烯与芳核结构之间的相互作用,Nafion-NGR-NPC/GCE形成三个明显分离的多巴胺、尿酸和抗坏血酸氧化峰,氧化峰电位分别在0.40 V、0.58 V和0.15 V。所有氧化峰的产生原理是由于抗坏血酸中的呋喃环羟基氧化成羰基,多巴胺中的卡托醇氧化成邻醌和尿酸中的桥连羟基氧化成双键。
(2)不同pH对同时检测多巴胺、尿酸和抗坏血酸的影响。
在修饰电极上多巴胺、尿酸和抗坏血酸的电化学信号受缓冲溶液pH值的影响。在2.0至8.0的不同pH值下研究多巴胺、尿酸和抗坏血酸的氧化峰电流。结果表明多巴胺、尿酸和抗坏血酸的氧化峰电流随着pH值从2.0到3.0的增加而增大,在pH 3.0时达到最大值,然后随着pH从3.0到8.0的增大而降低。当PBS为中性或碱性后,尿酸和抗坏血酸的氧化峰消失。因此pH值对多巴胺、尿酸和抗坏血酸的测定有显著影响,当pH值为3.0时,得到三种物质的最大氧化峰电流值。因此采用pH 3.0的磷酸缓冲溶液作为进一步试验的最佳pH。
(3)扫描速度对多巴胺、尿酸和抗坏血酸电化学行为的影响。
为了更好地阐明多巴胺、尿酸和抗坏血酸的电化学反应机理,本发明试验了扫速对三种物质电化学响应的影响。结果如图2所示,随着扫描速度的增加,氧化还原峰电流逐渐增加(图2为10 μmol/L尿酸(A)、10 μmol/L多巴胺(B)和100 μmol/L抗坏血酸(C)在0.1mol/L pH 3.0磷酸缓冲溶液中不同扫速下的循环伏安图;其中尿酸和抗坏血酸的扫速在20~80 mV/s范围内;多巴胺的扫速在50~600 mV/s范围内)。在含10 μmol/L尿酸的磷酸缓冲溶液中,氧化峰电流与扫速的线性关系为Ipa/μA=-4.52v (V/s)-0.818 (R2=0.998);在含10μmol/多巴胺的磷酸缓冲溶液中,峰电流与扫速的线性关系为Ipa/μA=-59.9v (V/s)-0.429(R2=0.998)和Ipc/μA=51.3v (V/s)+0.893 (R2=0.999);在含有100 μmol/L抗坏血酸的磷酸缓冲溶液中,氧化峰电流与扫速的线性关系为Ipa/μA=-15.0v (V/s)-0.227 (R2=0.985)。三种物质的氧化峰电流与扫速之间呈良好的线性关系,表明在修饰电极上多巴胺、尿酸和抗坏血酸的电极反应是吸附控制过程,这与所采用的NPC和NGR具有多孔、大比表面积和高导电性的特征有关。
(4)微分脉冲伏安法同时检测多巴胺、尿酸和抗坏血酸。
将Nafion-NGR-NPC/GCE用于多巴胺、尿酸和抗坏血酸的电化学测定,通过绘制标准工作曲线得到线性范围和检测限。不同浓度的多巴胺、尿酸和抗坏血酸在磷酸缓冲溶液中共存的微分脉冲伏安响应如图3A所示,可以看出多巴胺、尿酸和抗坏血酸的氧化峰电流值随着浓度的增加而增加,且同时测定多巴胺、尿酸和抗坏血酸的浓度时,三种物质的峰电位保持稳定。标准曲线在尿酸(UA)浓度为3.0~48.0 μmol/L,多巴胺(DA)浓度为5.0~90.0 μmol/L和抗坏血酸(AA)浓度为200~2000 μmol/L的范围内呈线性关系,检测限分别为0.045μmol/L、0.11 μmol/L和46.24 μmol/L(S/N=3)。对三种物质进行电化学测定的线性回归方程可表示如下:
Ip UA/μA=-0.159C(μmol/L)-0.141 (R2=0.995) (如图3B);
Ip DA/μA=-0.321C(μmol/L)-3.00 (R2=0.994) (如图3C);
Ip AA/μA=-0.00516C(μmol/L)-2.70 (R2=0.9959) (如图3D)。
(5)修饰电极的选择性、稳定性和重现性。
为了考察Nafion-NGR-NPC/GCE测定时的选择性,研究了实际样品中常见的各种干扰物对50 μmol/L尿酸,10 μmol/L多巴胺和500 μmol/L抗坏血酸检测的影响。结果表明,Cl-、SO4 2-、Na+、K+、Ca2+、Zn2+、Fe3+、NO3 -和NO2 -等无机离子的耐受极限为1.0 mmol/L,葡萄糖和腺嘌呤等物质的可耐受浓度均为2.0 mmol/L。考察了Nafion-NGR-NPC/GCE的稳定性,将Nafion-NGR-NPC/GCE在常温下储存10天后,测定三种物质的氧化峰电流值没有明显变化,UA、DA和AA的峰值电流强度分别仅下降7.9%,0.90%和1.3%。进一步考察修饰电极的重现性,平行制备6支修饰电极用于50 μmol/L尿酸,10 μmol/L多巴胺和500 μmol/L抗坏血酸的检测,其RSD分别为1.40%,1.09%和3.30%。因此,本方法构建的电化学传感器对多巴胺、尿酸和抗坏血酸的同时检测显示出高选择性、良好的稳定性和重现性。
(6)修饰电极对生物样品的检测。
为了验证该方法在实际样品中同时检测多巴胺(DA)、尿酸(UA)和抗坏血酸(AA)的可靠性,使用标准加入法对健康人尿样进行分析。将尿液样品以5000 rpm离心15分钟并收集上清液,用磷酸缓冲溶液(pH 3.0)稀释100倍作为样品溶液。结果如表1所示,加标样品的回收率范围为100.2%~102.8%。

总结。
设计了一种基于氮掺杂石墨烯-氮掺杂多孔碳修饰电极的电化学传感器,用于高灵敏度和选择性同时测定多巴胺、尿酸和抗坏血酸,并成功应用于生物样品中的检测。所制备的具有丰富氮元素的石墨烯-多孔碳材料表现出优异的电催化活性,可同时检测多巴胺、尿酸和抗坏血酸;所构建的传感器具有检测浓度范围宽、检测限低的优点;检测限分别为0.045 μmol/L、0.11 μmol/L和46.24 μmol/L(S/N=3)。Nafion-NGR-NPC/GCE具有良好的稳定性,进一步用于检测尿液样品中的尿酸、多巴胺和抗坏血酸含量的测定,结果令人满意。以上结果表明,本方法成功开发了一种灵敏和可靠的电化学传感器,对生物样品中药物分子含量的检测具有较大的潜在价值。
以上所述,仅为本发明的具体实施方式,但本发明的保护范围并不局限于此,任何不经过创造性劳动想到的变化或替换,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应该以权利要求书所限定的保护范围为准。
Claims (5)
1.一种氮掺杂石墨烯-氮掺杂多孔碳复合纳米材料修饰电极同时测定多巴胺、尿酸和抗坏血酸的方法。
2.根据权利要求1所述的一种氮掺杂石墨烯-氮掺杂多孔碳复合纳米材料修饰电极同时测定多巴胺、尿酸和抗坏血酸的方法,其特征在于,氮掺杂多孔碳-氮掺杂石墨烯复合纳米材料的制备方法为:采用溶剂热法制备锌基沸石咪唑酯骨架材料,以锌基沸石咪唑酯骨架材料为前驱体,煅烧后形成氮掺杂多孔碳,进一步与氮掺杂石墨烯通过直接混合法制备氮掺杂多孔碳-氮掺杂石墨烯复合材料。
3.根据权利要求1所述的一种氮掺杂多孔碳-氮掺杂石墨烯纳米复合材料修饰电极同时测定多巴胺、尿酸和抗坏血酸的方法,其特征在于,其构建的氮掺杂石墨烯-氮掺杂多孔碳复合纳米材料修饰电极为工作电极,可用于分别单独对多巴胺、尿酸和抗坏血酸进行电化学检测;也可同时测定多巴胺、尿酸和抗坏血酸的含量;通过标准曲线法对生物样品中的多巴胺、尿酸和抗坏血酸含量进行检测。
4.根据权利要求2所述的氮掺杂石墨烯-氮掺杂多孔碳复合纳米材料的制备方法,其特征在于,所述中氮掺杂石墨烯/氮掺杂多孔碳的质量比为1:1。
5.根据权利要求3所述的通过标准曲线法对生物样品中的多巴胺、尿酸和抗坏血酸含量进行检测,其特征在于,所述中的生物样品为人体尿液。
