Abstract

现有技术
防腐剂用于保护多种产品免受微生物污染和腐败。近年来,由于毒理学问题,一系列防腐剂已经声名狼藉,其使用受到法律的部分限制。在这方面,然而,没有受到影响的防腐剂具有其他缺点,例如它们仅在有限的pH范围内有效或者它们对整个微生物范围没有效用。在这方面,提供例如食品、化妆品和其他产品的安全保存则变得越来越困难,并且需要以多种方式有效且可用的产品。

发明详述
现已令人惊讶地发现,向苯甲酸和/或山梨酸中加入糖脂基本上可以中和这些酸的令人不愉快的味道。此外,可以扩大这些防腐剂有效的pH范围。尽管苯甲酸/山梨酸或者糖脂在所用浓度下都没有显示出足够的防腐效果,但所述混合物在微生物生长方面产生了良好的稳定性。
本发明涉及包含苯甲酸和/或山梨酸及糖脂的组合物。
本发明的一个优点是这些防腐剂可以在较宽的pH范围和产品范围内用于相应的混合物中。例如,它们可用于改善牙齿护理产品(诸如漱口水和牙膏)的味道和微生物稳定性,也可用于稳定和改善护理产品(洗浴添加剂、口红等)的味道。另一个优点是因此较少的香水或香料需要被使用以掩盖令人不愉快的味道。
另一个优点是在较低的pH范围内不稳定的产品也可以因此被保存。另一个优点是可以减少所需防腐剂的量。

存在于本发明组合物中的糖脂由于给定的pH而可以至少部分地作为盐而存在。
在本发明优选的组合物中,存在的糖脂盐的阳离子选自包含Li+、Na+、K+、Mg2+、Ca2 +、Al3+、NH4 +、伯铵离子、仲铵离子、叔铵离子和季铵离子的组选自优选由Li+、Na+、K+、Mg2+、Ca2 +、Al3+、NH4 +、伯铵离子、仲铵离子、叔铵离子和季铵离子组成的组。
合适的铵离子的示例性代表是四甲基铵、四乙基铵、四丙基铵、四丁基铵和[(2-羟乙基)三甲基铵](胆碱),以及2-氨基乙醇(乙醇胺,MEA)、二乙醇胺(DEA)、2,2',2'’–次氨基三乙醇(三乙醇胺,TEA)、1-氨基丙-2-醇(单异丙醇胺)、乙二胺、二亚乙基三胺、三亚乙基四胺、四亚乙基五胺、1,4-二乙二胺(哌嗪)、氨基乙基哌嗪和氨基乙基乙醇胺的阳离子。
根据本发明,上述阳离子的混合物也可以作为存在的糖脂盐的阳离子而存在。
特别优选的阳离子优选选自包含Na+、K+、NH4 +、和三乙醇铵阳离子的组优选由Na+、K+、NH4 +、和三乙醇铵阳离子组成的组。
上述阳离子的总量优选占组合物中存在的除H+和H3O+外的所有阳离子的50-99重量%,特别优选70-90重量%。
本发明的优选组合物包含50-99重量%,优选70-95重量%,特别优选85-90重量%的糖脂阴离子,其中所述糖脂阴离子优选选自鼠李糖脂阴离子和槐糖脂阴离子,尤其是鼠李糖脂阴离子,其中重量%参考组合物中存在的除OH-外的所有阴离子。
在本发明的特别优选的组合物中,总的干质量包含40-98重量%,优选50-95重量%,特别优选60-90重量%的糖脂,其中所述糖脂优选选自鼠李糖脂和槐糖脂,特别是鼠李糖脂,其中重量百分比参考总的干质量。
在本发明优选的组合物中,至少60重量%,优选至少80重量%,特别优选至少95重量%的优选选自鼠李糖脂和槐糖脂特别是鼠李糖脂的糖脂以溶解形式存在,其中重量百分比参考优选选自鼠李糖脂和槐糖脂特别是鼠李糖脂的糖脂的总量。
这通过经过0.2μm针筒式滤器进行过滤之前和之后的总糖脂的HPLC分析进行测量,其中洗脱液中糖脂的量对应于溶解的糖脂的量。
根据本发明优选的是,所述组合物包含51-95重量%,优选70-90重量%,特别优选75-85重量%的diRL-C10C10,其中重量百分比参考存在的所有鼠李糖脂的总和。
根据本发明优选的是,所述组合物包含0.5-9重量%,优选0.5-3重量%,特别优选0.5-2重量%的monoRL-C10C10,其中重量百分比参考存在的所有鼠李糖脂的总和。

还优选,根据本发明的组合物仅包含少量的式monoRL-CX或diRL-CX的鼠李糖脂。具体而言,根据本发明的组合物优选包含0-5重量%,优选0-3重量%,特别优选0.1-1重量%的diRLC10,其中重量百分比参考存在的所有鼠李糖脂的总和,并且,术语“0重量%”应理解为无法检测到的量。
根据本发明优选的是,本发明的组合物基本上不含脂肪油(20℃下的酰基甘油液体),因此特别包含基于总组合物小于0.5重量%,特别是小于0.1重量%,特别优选无法检测量的脂肪油。
本发明的优选组合物包含槐糖脂作为糖脂,其中内酯形式与酸形式的重量比在20:80至80:20的范围内,特别优选在30:70至40:60的范围内。
本发明进一步涉及糖脂在改善选自山梨酸、苯甲酸和上述酸的盐的防腐剂抗微生物特别是细菌的防腐作用的用途,其中所述糖脂优选选自槐糖脂和鼠李糖脂,特别是鼠李糖脂。
本发明还涉及糖脂在降低选自山梨酸、苯甲酸和上述酸的盐的防腐剂的苦味的用途,其中所述糖脂优选选自槐糖脂和鼠李糖脂,特别是鼠李糖脂。
本发明还涉及本发明的组合物作为食品、化妆品、家用清洁剂、洗涤剂和漂洗剂、牙齿护理产品和医药产品中的防腐剂的用途,特别是用于食品和牙齿护理产品中的防腐剂的用途。
下面引用的实施例将以实施例的形式对本发明进行描述,但并不意图将本发明限于实施例中所指定的实施方案,本发明的应用范围由整个说明书和权利要求是显而易见的。
具体实施方式
实施例
实施例1:通过添加鼠李糖脂而遮蔽苯甲酸和山梨酸的味道
如EP3023431A1(浓缩的低粘度鼠李糖脂组合物)中所述而制备了用KOH部分中和的鼠李糖脂,并且进行分析。得到干质量含量为40%且pH为6的鼠李糖脂溶液。鼠李糖脂及其盐的比例基于干质量为>90重量%。各种鼠李糖脂同系物相对于所有鼠李糖脂总和的以重量百分比计的相对比例列于下表中。这里,所述比率是指HPLC分析所定量的酸形式。
表1:使用的鼠李糖脂的组成。基于所有鼠李糖脂(作为酸形式)的总和的各个同系物(作为酸形式)的重量%的数据。
蛋白质含量通过光度二联喹啉测定(photometric bicinchoninic assay,BCA测定,ThermaFisher Scientific)进行测定,并且基于鼠李糖脂的干质量<1重量%。
将得到的高浓度鼠李糖脂溶液进行稀释,并加入苯甲酸钠或山梨酸钠(SigmaAldrich)。制备了具有下表所述的组合物的溶液。将溶液调节至pH=6。

然后对所述混合物进行感官评价。为此,通过小组(10名参与者)品尝各5ml并且描述味道印象。对于不含添加剂的山梨酸或苯甲酸,描述了令人不愉快、燃烧和略带涩味的味道,其不能通过添加山梨醇而掩盖。在鼠李糖脂的存在下,几乎没有感觉到这种味道印象,而是描述了仅略甜的椰子状的味道。
实施例2:鼠李糖脂与苯甲酸或山梨酸的协同作用
通过向实施例1中所述的高浓度鼠李糖脂溶液中加入苯甲酸钾和/或山梨酸钾,制备了下表中所述的组合物并将pH调节至5.8。

然后根据欧洲药典2011年第7版第5.1.3段进行微生物污染测试。为此,用给定的多种微生物的细菌计数对表3中的组合物进行接种,并将接种的样品在室温下储存。在固定的时间点,测量细菌计数。用于微生物污染测试的微生物是大肠杆菌(Escherichia coli)、铜绿假单胞菌(Pseudomonas aeruginosa)、金黄色葡萄球菌(Staphylococcus aureus)、白色念珠菌(Candida albicans)和巴西曲霉(Aspergillus brasiliensis)。
苯甲酸钾(M7)在该pH下没有作用,并且细菌数并未减少。山梨酸钾(M9)仅有很小的作用。苯甲酸钾和山梨酸钾的组合(M12)或单独的鼠李糖脂(M11)也只有很小的作用。特别是,酵母和真菌的细菌数没有显著减少。然而,山梨酸钾(M10),苯甲酸钾(M8)或其混合物(M13)与鼠李糖脂的组合对所研究的所有微生物提供了非常好的下降。特别是革兰氏细菌和酵母菌可以更快地显著降低,并且甚至在2天(第一个测试时间点)之后,不再能够检测到活的微生物。
实施例2b:鼠李糖脂和苯甲酸的协同作用
通过向实施例1中所述的高浓度鼠李糖脂溶液中加入苯甲酸钠而制备了下表中所述的组合物,并将pH调节至5.8。

然后根据欧洲药典2011年第7版第5.1.3段进行微生物污染测试。为此,用给定的多种微生物的细菌计数对表4中的组合物进行接种,并将接种的样品在室温下储存。在固定的时间点,测量细菌计数。
用于微生物污染测试的微生物是白色念珠菌(Candida albicans)和巴西曲霉(Aspergillus brasiliensis)。
结果示于图1和2中,并显示出两种组分的协同效应。
实施例3:槐糖脂与苯甲酸和山梨酸的混合物的协同作用
将市购槐糖脂(SL446)稀释至槐糖脂含量为10%。将0.1%山梨酸和0.1%苯甲酸加入一样品中,并且另一样品不进行防腐。将两个样品的pH调节至6.2。还制备了0.1%山梨酸和0.1%苯甲酸的水溶液,并将pH调节至6.2。然后如实施例2中所述,用巴西曲霉对三个样品进行微生物污染试验。仅在槐糖脂与防腐剂的组合中,观察到随着时间细菌数显著减少。
实施例4:鼠李糖脂与苯甲酸和山梨酸的混合物的协同作用
在本例中,根据基于DIN58940-8的微稀释方法测定了多种组合物对白色念珠菌(Candida albicans)的最小抑制浓度。
将来自原始培养物的白色念珠菌DSM1386接种在Sab.琼脂板上并在30℃下孵育2天。由该预培养,将Sab.琼脂斜面管(Sab.agar slant tube)进行接种,然后在30℃下培养两天。
为了制备测试接种物,用pH 6的7ml Mueller-Hinton培养基冲洗所述琼脂斜面管,将冲洗掉的微生物悬浮液装入含有5g玻璃珠的100ml烧瓶中,并将烧瓶置于定轨振荡器上3分钟。
将这些微生物悬浮液以1:100稀释,以在测试接种物中达到106CFU/ml的细菌计数。
借助螺旋计而测定接种物的确切细菌数。
将100μl Mueller-Hinton培养基中待测试的物质的等分试样的合适稀释序列置于微量滴定板中,并提供100μl的上述制备的白色念珠菌培养物,并将板在30℃下培养2-3天。。
基于在各个测试物质的浓度下不存在或存在细胞生长而测定最小抑制浓度。
基于苯甲酸钠与山梨酸钾的重量的2:1混合物在所述实验设置中显示出0.5-1%的最小抑制浓度,而实施例1的鼠李糖脂中在最高浓度为10%进行测试时,没有观察到抑制作用。
对于基于苯甲酸钠与山梨酸钾与鼠李糖脂的重量为2:1:130的混合物,最小抑制浓度(MIC)分别降至0.5(基于苯甲酸钠和山梨酸钾)和10(基于鼠李糖脂):

Claims (11)
1.一种组合物,其包含:
5-70重量%,优选6-60重量,特别优选10-55重量%,特别优选20-50重量%的至少一种优选选自槐糖脂和鼠李糖脂,特别是鼠李糖脂的糖脂,和
0.1-10重量%,优选0.2-5重量%,特别优选0.4-1重量%的至少一种选自山梨酸、苯甲酸和上述酸的盐的防腐剂,
其中所述重量百分比参考总的组合物,
其特征在于所述组合物在25℃的pH在3.5-9,优选5.6-7,特别优选5.6-6.6的范围内。
2.权利要求1的组合物,其特征在于:
存在的糖脂与存在的防腐剂的重量比在1000:1至1:1,优选500:1至10:1,特别优选100:1至30:1的范围内,其中存在的糖脂优选选自槐糖脂和鼠李糖脂,特别是鼠李糖脂,存在的防腐剂选自山梨酸、苯甲酸和上述酸的盐。
3.权利要求1或2的组合物,其特征在于:
存在至少一种选自异噻唑啉酮、苯氧乙醇、苯甲醇、对羟基苯甲酸酯类、抗微生物肽、萜烯、抗微生物脂肪酸、甲醛释放剂和醇类的其他防腐剂。
4.前述权利要求至少之一的组合物,其特征在于所述组合物包含51-95重量%的diRL-C10C10,其中所述重量百分比参考存在的所有鼠李糖脂的总和。
5.前述权利要求至少之一的组合物,其特征在于所述组合物包含0.5-9重量%的monoRL-C10C10,其中所述重量百分比参考存在的所有鼠李糖脂的总和。
6.前述权利要求至少之一的组合物,其特征在于存在的所有二-鼠李糖脂与存在的所有单-鼠李糖脂的重量比大于51:49,特别大于91:9,优选大于97:3,特别优选大于98:2。
7.前述权利要求至少之一的组合物,其特征在于所述组合物包含0.5-25重量%的diRL-C10C12,其中所述重量百分比参考存在的所有鼠李糖脂的总和。
8.前述权利要求至少之一的组合物,其特征在于所述组合物包含:
0.1-5重量%的monoRL-C10C12,和/或,优选和,
0.1-5重量%的monoRL-C10C12:1,
其中所述重量百分比参考存在的所有鼠李糖脂的总和。
9.糖脂改善防腐剂的抗微生物防腐作用,特别是细菌防腐作用的用途,其中所述糖脂优选选自槐糖脂和鼠李糖脂,特别是鼠李糖脂,所述防腐剂选自山梨酸、苯甲酸和上述酸的盐。
10.糖脂降低防腐剂的苦味的用途,其中所述糖脂优选选自槐糖脂和鼠李糖脂,特别是鼠李糖脂,所述防腐剂选自山梨酸、苯甲酸和上述酸的盐。
11.根据权利要求1-8中至少之一的组合物作为食品、化妆品、家用清洁剂、洗涤剂和漂洗剂、牙齿护理产品和医药产品中的防腐剂的用途,特别是用于食品和牙齿护理产品中的防腐剂的用途。
