本发明涉及含有金属离子的口腔组合物。
背景技术
亚锡离子,尤其是亚锡盐诸如氟化亚锡为在洁齿剂中用作预防菌斑的试剂的已知抗微生物剂。然而,亚锡盐的一些缺点包括不稳定性、有使牙齿染色的倾向、收敛性、以及对使用者而言令人不快的味道。
锌离子,尤其是锌盐也为有时掺入洁齿剂制剂中的已知抗微生物剂。各种锌盐的使用常常受到锌的溶解度、当利用较高水平锌时出现的不希望的消费者收敛性、以及一旦锌离子可用于反应时锌的反应性(即,锌离子有时引起制剂内的不良反应)的限制。
多磷酸盐和离子活性成分在洁齿剂中用于促进口腔健康。多磷酸盐为有助于延迟牙结石形成的已知抗牙垢剂。
虽然此类成分先前已用于洁齿剂中,出于若干原因,已证实将这些成分一起提供在具有满意的化学和物理稳定性的制剂中具有挑战性。
提供稳定洁齿剂组合物的一种尝试为减少存在于组合物中的水的量。然而,如美国专利申请公布2012-0207686的背景中所述的,减少水含量并任选地用保湿剂替换一些或所有去除的水,会在组合物中获得可接受的流变性和增稠特性方面产生问题。
当去除作为高极性溶剂的水时,一些常见增稠剂往往不适当胶凝。减少洁齿剂组合物中的水含量的尝试包括例如EP 0 638 307 B1;美国专利号4,647,451;以及美国专利号5,670,137中所述的洁齿剂。已显示此类已知制剂随着时间表现出逐渐的增稠,这延长了时间段或甚至阻止洁齿剂达到流变性稳定状态。理想地,洁齿剂制剂需要在两周内达到消费者接受的稳定性状态。如果制剂的粘度按惯例随着时间而增加,则制剂的分配将变得困难,这可能会导致消费者不满意。

在本领域中需要提供低水单相洁齿剂组合物,其具有改进的流变性特性并且具体地具有足够粘度的稳定流变性,该流变性有效减少或消除组合物随着时间的逐渐增稠,这进而提供了可在其保质期时段期间有效分配的组合物。
发明内容
本发明涉及一种口腔护理组合物,所述组合物包含:
(a)亚锡离子源;
(b)锌离子源;
(c)多磷酸盐;以及
(d)增稠剂,其包含:
(i)1重量%至3.5重量%聚乙烯吡咯烷酮,
(ii)0.2重量%至0.45重量%多糖胶,以及
(iii)0.05重量%至0.3重量%羧甲基纤维素。
在本发明的另一方面,所述口腔护理组合物在所述组合物制备后七天在25℃温度下表现出相对于初始粘度小于70%的粘度增加。在本发明的另一个实施方案中,所述口腔护理组合物在所述组合物制备后七天在25℃温度下表现出相对于初始粘度小于60%的粘度增加。在本发明的另一实施方案中,所述口腔护理组合物在所述组合物制备后七天在25℃下表现出相对于初始粘度40%-60%之间的粘度增加(粘度测量,粘度在25℃下使用布鲁克费尔德Model RVT粘度计Spindle V74在1RPM下测量;粘度以厘泊(cP)计)。

各种实施方案的口腔护理组合物优选地为洁齿剂形式。如贯穿本说明书的术语“洁齿剂”表示糊剂或凝胶制剂。洁齿剂组合物为一种产品,该产品在普通施用过程中不能出于全身性施用特定治疗剂的目的而故意吞咽,而是出于口腔活性而保留在口腔中,持续足以接触基本上所有牙齿表面和/或口腔组织的时间。洁齿剂可为任何希望的形式,诸如深条纹、浅条纹、多层、糊剂周围有凝胶或其任何组合。

锌离子源于以有效量存在于洁齿剂组合物中的锌化合物,即锌离子源。锌离子的有效量被限定为至少1000ppm锌离子,优选地2,000ppm至15,000ppm。更优选地,锌离子以3,000ppm至13,000ppm以及甚至更优选地4,000ppm至10,000ppm的量存在。这是存在于组合物中以用于递送至牙齿表面的锌离子总量。用于本发明的口腔组合物中的锌化合物(锌源)的量可在基于所述组合物总量0.5重量%至5.0重量%之间变化,通常在基于所述口腔护理组合物总量1重量%至4重量%之间变化。
组合物还包含提供亚锡离子源的亚锡化合物。适合亚锡离子化合物或亚锡离子源包括但不限于,氟化亚锡、其他亚锡卤化物诸如氯化亚锡二水合物、焦磷酸亚锡、有机羧酸亚锡盐诸如甲酸亚锡、乙酸亚锡、葡糖酸亚锡、乳酸亚锡、酒石酸亚锡、草酸亚锡、丙二酸亚锡和柠檬酸亚锡、亚乙基果绿定亚锡(stannous ethylene glyoxide)等。一种或多种亚锡离子源任选地且说明性地以0.01%至10%,例如0.1%至3%、0.3%至0.7%、以及0.4%至0.6%的总重量存在。
合并的金属离子源(亚锡和锌)将以0.25重量%至11重量%的量存在于最终组合物中。优选地,金属离子源以0.4%至7%、更优选1%至5%或2.%至3.25%的量存在。
洁齿剂组合物包含提供具有所需流变学特性的洁齿剂的增稠剂,以使得所述洁齿剂可保存在分配容器中一段时间并且此后由所述使用者从中稳定分配。洁齿剂优选地应具有不仅能分配而且在刷牙期间在口腔内表现出可接受的稠度的正确粘度。本发明人已发现使用含有至少三种组分的增稠剂为所述组合物提供改进的流变学特性。三种组分为聚乙烯吡咯烷酮(PVP)、多糖胶和羧甲基纤维素(CMC)。
PVP可为交联的并且可包含N-乙烯基-2-吡咯烷酮。聚乙烯吡咯烷酮可构成所述组合物的1重量%至3.5重量%,在一个实施方案中,可构成所述组合物的2.75重量%至3.25重量%。并且在另一个实施方案中,构成1%-1.5%。
本发明的洁齿剂组合物具有有效于提供具有所需流变学特性的洁齿剂的粘度和屈服应力,以使得所述洁齿剂可保存在分配容器中一段时间并且此后由所述使用者从中稳定分配。洁齿剂优选地应具有不仅能分配而且在刷牙期间在口腔内表现出可接受的稠度的正确粘度。所述组合物具有有效减少或消除所述组合物随着时间的逐渐增稠的稳定流变性,从而提供了可在其保质期时段期间有效分配的组合物。粘度和屈服应力为随着时间基本上恒定的,例如在两周、六周、六个月、一年等时间内。在一个实施方案中,洁齿剂组合物在两周内达到消费者可接受的稳定状态。如果制剂的粘度按惯例随着时间而增加,则制剂的分配将变得困难,这可能会导致消费者不满意。如本文所用,“屈服应力”为物质在7天后变形时的应力值。本发明的组合物在25℃下通过布鲁克费尔德粘度计测量的屈服应力为15帕斯卡(Pa)至70Pa,在一个实施方案中为20Pa至50Pa。本发明的组合物在7天后在25℃下的最大布鲁克费尔德粘度通常为150,000厘泊(cP)至1,000,000cP,在另一个实施方案中为200,000cP至700,000cP。
CMC和多糖胶沿着其主链含有羧酸基团。虽然两种物质均为带电荷多糖,但是沿着主链的带电荷基团的密度为不同的,并且通常CMC的密度远高于许多多糖胶的密度。例如,一种已知商业上可获得的CMC即CMC 2000S(从CPKelco获得)的取代度为每个糖残基0.9个羧酸基团。其他CMC的取代度为每个糖残基0.7、1.0或1.2个羧酸基团。相比之下,优选的多糖胶黄原酸胶的取代度为<每个糖残基0.4个羧酸基团。CMC和多糖胶一起的量通常为所述组合物的0.5重量%或更少。
在某些实施方案中,多糖胶增稠剂沿着多糖主链具有每个糖残基单元至多0.5个带电荷基团。对于另外的信息,参见2009年4月2日提交的PCT申请号PCT/US2009/039268,该专利以引用的方式并入本文。
多糖胶的实例包括黄原酸胶、阿拉伯树胶、瓜尔豆胶、刺槐豆胶、黄蓍胶、结冷胶以及他拉胶。多糖胶例如黄原酸胶以基于所述组合物0.2重量%至0.45重量%的量存在在一个实施方案中,以所述组合物的0.35重量%至0.45重量%的量存在,并且在另一个实施方案中,以所述组合物的0.2重量%至0.3重量%的量存在。
除了本文所述的三种组分之外,增稠剂可任选地含有最少量的另外的增稠剂,例如角叉菜胶、淀粉、羟乙基丙基纤维素、羟丁基甲基纤维素、羟丙基甲基纤维素、羟乙基纤维素、以及胶体二氧化硅。还可使用泊洛沙姆。泊洛沙姆为合成的环氧乙烷和环氧丙烷的嵌段共聚物。它以多种类型可用。在本文中,泊洛沙姆407为优选的。它可部分溶解在水中。当温度高于65℃时,它可溶解在甘油中。POLOXAMER例如可获自BASF CORPORATION,NewJersey,USA。还可使用卡波姆。增稠剂还可包括另外的无机增稠剂诸如胶状硅酸铝镁或细碎二氧化硅,以进一步改进纹理。
在一个实施方案中,增稠剂的总量的范围为基于所述组合物重量的1重量%至5重量%。在另一个实施方案中,增稠剂的量为基于所述组合物重量的1.5重量%-3.75重量%。
口腔组合物还含有多磷酸盐。多磷酸盐已知有助于延迟牙结石形成。通常认为多磷酸盐由主要以线性构型排列的两个或更多个磷酸盐分子组成,尽管也可存在一些环形衍生物。适合的多磷酸盐为无机多磷酸盐,其优选地为碱土金属盐。在一些实施方案中,多磷酸盐的平均链长度为4或更小或者3或更小。典型实例包括三聚磷酸盐或焦磷酸盐。
在一个实施方案中,多磷酸盐为焦磷酸盐的碱金属盐。多磷酸盐的实例包括焦磷酸四钠(TSPP)和三聚磷酸钠(STPP)以及酸式焦磷酸钠(SAPP)。可使用一种或多种多磷酸盐的混合物。
多磷酸盐的有效量为总洁齿剂组合物的0.1重量%至30重量%、或2重量%至20重量%、或1重量%至10重量%、或3重量%至7重量%。
本发明的组合物含有口腔可接受的载体。贯穿本说明书所使用的表达“载体”或“水性载体”表示在本文中使用的任何安全且有效的物质,而不是亚锡离子源、锌离子源、多磷酸盐以及增稠剂。此类物质包括例如水、保湿剂、离子活性成分、缓冲剂、抗齿垢剂、研磨抛光剂、过氧化物源、碱金属碳酸氢盐、表面活化剂、二氧化钛、着色剂、调味剂系统、甜味剂、抗微生物剂、草药试剂、脱敏剂、减污剂、以及其混合物。此类物质为本领域已熟知的并且容易由本领域技术人员基于对于所制备的组合物需要的物理特性和美学特性来选择。载体通常构成所述洁齿剂组合物的40重量%至99重量%、优选70重量%至98重量%、以及更优选90重量%至95重量%。
所述组合物还可包含至少一种保湿剂。保湿剂用于保持牙膏组合物以免于在暴露于空气后硬化并且某些保湿剂还可赋予牙膏组合物希望的甜味。用于本发明中的适合保湿剂包括甘油、山梨醇、聚乙二醇、丙二醇、木糖醇、以及其他可食用的多元醇类。优选的是甘油、聚乙二醇、聚丙二醇、以及其混合物,尤其是其混合物。保湿剂通常构成所述组合物的0.1重量%至70重量%、优选1重量%至60重量%、以及更优选15重量%至55重量%。
本文所述的组合物还可含有缓冲剂。缓冲系统优选地适于螯合在所述组合物中的亚锡离子。如本文所用的缓冲剂是指可用于将组合物的pH调整至范围pH 3.0至pH 10,在一些实施方案中调节至pH 4至pH 7、pH 4.5至pH 6、或pH 5至pH 5.5的试剂。
缓冲剂包括碱金属氢氧化物、氢氧化铵、有机铵化合物、碳酸盐、倍半碳酸盐、硼酸盐、硅酸盐、磷酸盐、咪唑、以及其混合物。特定缓冲剂包括磷酸一钠、磷酸三钠、苯甲酸钠、苯甲酸、氢氧化钠、氢氧化钾、碱金属碳酸盐、碳酸钠、咪唑、焦磷酸盐、柠檬酸、以及柠檬酸钠。在一些实施方案中,缓冲系统可包含有机酸或其碱金属盐中的至少一种,所述有机酸优选地为柠檬酸。缓冲系统可包含柠檬酸和柠檬酸三钠的混合物。缓冲剂以本发明的重量的0.1%至30%、在一个实施方案中为0.1%至10%、以及在另一个实施方案中为1%至5%、以及在另一个实施方案中为0.3%至3%的水平使用。
所述组合物可含有氟离子源。本文中氟离子源为能够提供游离氟离子的氟源并且通常为可溶的。氟离子源包括氟化亚锡、氟化钠、氟化钾、氟化铟、氟化锌、单氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺(例如,N′-十八烷基三亚甲基二胺-N,N,N′-三(2-乙醇)-二氢氟化物)、氟化铵、氟化钛、六氟硫酸盐、以及其组合;
氟化钠和氟化亚锡为优选的可溶性氟离子源。Norris等的美国专利号2,946,725、以及美国专利号3,678,154和美国专利号6,190,644公开了此类氟离子源以及其他氟离子源。
本发明组合物可含有能够提供50ppm至3500ppm、以及优选500ppm至3000ppm的游离氟离子的可溶性氟离子源。为递送希望的量的氟离子,氟离子源可以总洁齿剂组合物的0.1重量%至5重量%、优选0.2重量%至1重量%、以及更优选0.3重量%至0.6重量%的量存在于总洁齿剂组合物中。
相应地,本发明还提供了一种用于制造洁齿剂组合物的方法,所述方法包括提供亚锡离子源、将所述亚锡离子源与适于螯合由水性缓冲系统形成的预混合物中的亚锡离子的所述水性缓冲系统混合、以及将所述预混合物与至少一种活性组分和洁齿剂组合物的口腔可接受的媒介物组合。
本发明的组合物可含有水。用于制备商业上适合的口腔组合物的水应优选地具有低离子含量并且不含有机杂质。在洁齿剂组合物中,水通常构成本文中的组合物的小于15重量%,在一个实施方案中小于10重量%,在另一个实施方案中为0.1重量%至10重量%,并且在另一个实施方案中为0重量%至6重量%。水的量包括添加的游离水和与其他物质诸如二氧化硅、表面活性剂溶液和/或颜色溶液一起引入的游离水。
所述组合物还可采用合成阴离子聚合物[包括聚丙烯酸酯和马来酸酐或马来酸与甲基乙烯基醚的共聚物(例如,),例如,如Gaffar等的美国专利号4,627,977所述的;以及例如聚氨基丙烷磺酸(AMPS)]、三水合柠檬酸锌、二磷酸盐(例如,EHDP;AHP)、多肽(诸如聚天冬氨酸和聚谷氨酸)、以及其混合物。
在牙膏组合物中还可包含研磨或微粒抛光物质。本发明的组合物中预期使用的研磨抛光物质可为并不过分磨损牙质的任何物质。典型研磨抛光物质包括二氧化硅,包括凝胶型、沉淀型和水合二氧化硅;矾土;氧化铝;氧化铁;珍珠岩;塑料颗粒,例如聚乙烯;磷酸盐,包括正磷酸盐、聚偏磷酸盐和焦磷酸盐。特定实例包括碳酸氢钠、正磷酸二钙二水合物、焦磷酸钙、磷酸三钙、聚偏磷酸钙、不溶性聚偏磷酸钠、水合氧化铝、β焦磷酸钙、碳酸钙、磷酸钙(例如,磷酸二钙二水合物)、硫酸钙、沉淀的碳酸钙、以及树脂性研磨物质(诸如尿素和甲醛的颗粒状缩合产品)、以及其他实例,诸如在美国专利号3,070,510中公开的。可使用两种或更多种研磨剂的组合。
各种类型的二氧化硅牙齿研磨剂为适合的,因为它们具有优异牙齿清洁和抛光性能的独特益处而不过度磨损牙釉或牙质。本文中的二氧化硅研磨抛光物质以及其他研磨剂通常具有范围在0.1至30微米之间并且优选在5至15微米之间的平均粒度。研磨剂可为沉淀的二氧化硅或二氧化硅凝胶,诸如美国专利号3,538,23和美国专利号3,862,307中所述的二氧化硅干凝胶。实例为由W.R.Grace&Company,Davison Chemical Division以商品名销售的二氧化硅干凝胶。还优选的是沉淀的二氧化硅物质,诸如由Huber公司以商品名销售的那些,特别是携带标识“Zeodent 119”的二氧化硅。适用于本发明的牙膏中的二氧化硅牙齿研磨剂的类型更详细地描述于美国专利号4,340,583中。二氧化硅研磨剂也描述于Rice的美国专利号5,589,160;5,603,920;5,651,958;5,658,553;以及5,716,601。在本文描述的牙膏组合物中的研磨剂通常以所述组合物的6重量%至70重量%或15重量%至70重量%的水平存在。通常,牙膏含有所述洁齿剂组合物的10重量%至50重量%的研磨剂。
本发明可包含增白剂,例如选自由以下组成的组:过氧化物、金属亚氯酸盐、过硼酸盐、过碳酸盐、过氧酸、次氯酸盐、以及其组合。在一个实施方案中,所述增白剂为过氧化物源。过氧化物源选自由以下组成的组:过氧化氢、过氧化钙、过氧化脲、以及其混合物。优选的过氧化物源为过氧化钙。以下量表示过氧化物原料的量,尽管过氧化物源可含有除过氧化物原料之外的成分。本发明组合物可含有所述洁齿剂组合物的0.01重量%至10重量%、优选0.1重量%至5重量%、更优选0.2重量%至3重量%、以及最优选0.3重量%至0.8重量%的过氧化物源

“高级烷基”例如意指C6-30烷基。在具体实施方案中,阴离子型表面活性剂选自月桂基硫酸钠和月桂基醚硫酸钠。阴离子型表面活性剂可以有效量存在,例如>制剂的0.01重量%,而不是以刺激口腔组织的浓度存在,例如<10%,并且最佳浓度取决于特定制剂和特定表面活性剂。例如,用于漱口水的浓度通常为用于牙膏的浓度的约十分之一。在一个实施方案中,阴离子型表面活性剂以0.3重量%至4.5重量%,例如1.5重量%存在于牙膏中。所述组合物可任选地含有表面活性剂的混合物,例如包含阴离子型表面活性剂和可为阴离子型、阳离子型、两性离子型或非离子型表面活性剂的其他表面活性剂。通常,表面活性剂为在整个宽pH范围内合理稳定的那些表面活性剂。表面活性剂更充分地描述于美国专利号3,959,458;美国专利号3,937,807;以及美国专利号4,051,234。在某些实施方案中,本文中适用的阴离子型表面活性剂包括烷基中具有10至18个碳原子的烷基硫酸盐的水溶性盐和具有10至18个碳原子的脂肪酸的磺化甘油单酯的水溶性盐。月桂基硫酸钠、月桂酰肌氨酸钠和椰子基单酸甘油酯磺酸钠为这种类型的阴离子型表面活性剂的实例。在一个特定实施方案中,所述组合物包含月桂基硫酸钠。
表面活性剂或相容性表面活性剂的混合物可以总体组合物的0.1重量%至5重量%,在一个实施方案中0.3重量%至3重量%并且在另一个实施方案中0.5重量%至2重量%存在于所述组合物中。
二氧化钛也可加入到本发明组合物中。二氧化钛为增加组合物的不透明度的白色粉末。二氧化钛通常构成所述组合物的0.25重量%至5重量%。
所述组合物还可含有干扰或预防细菌粘附的试剂,例如对羟基苯甲酸甲酯或壳聚糖。
所述组合物还可包含钙和磷酸源,其选自(i)钙玻璃络合物,例如磷硅酸钙钠,以及(ii)钙-蛋白质络合物,例如酪蛋白磷酸肽无定形磷酸钙。
所述组合物还可包含可溶性钙盐,例如选自硫酸钙、氯化钙、硝酸钙、乙酸钙、乳酸钙、以及其组合。
着色剂也可加入到本发明组合物中。着色剂可处于水性溶液形式中,优选地水溶液中的1%着色剂。着色溶液通常构成所述组合物的0.01重量%至5重量%。
调味系统也可加入到所述组合物中。适合的调味组分包括冬青油、薄荷油、留兰香油、丁香花油、薄荷醇、茴香脑、水杨酸甲酯、桉叶油素、肉桂皮、1-乙酸盖酯、鼠尾草、丁子香酚、欧芹油、羟苯基丁酮、α-紫罗酮、甘牛至草、柠檬、橙、丙烯基乙基愈创木酚(propenylguaethol)、肉桂、香草醛、乙基香草醛、天芥菜精、4-顺式-庚烯醛、联乙酰、甲基-对-叔-丁基苯基乙酸酯、以及其混合物。清凉剂也可为调味系统的一部分。本发明组合物中优选的清凉剂为对薄荷烷羧基酰胺剂,例如N-乙基-对-薄荷烷-3-甲酰胺(在商业上称为“WS-3”)以及其混合物。调味系统通常以所述组合物的0.001重量%至5重量%的水平用于所述组合物中。
甜味剂可加入到所述组合物中。这些甜味剂包括糖精、葡萄糖、蔗糖、乳糖、木糖醇、麦芽糖、左旋糖、阿斯巴甜、环氨酸钠、D-色氨酸、二氢查耳酮、丁磺氨、以及其混合物。不同着色剂也可并入到本发明中。甜味剂和着色剂通常以所述组合物的0.005重量%至5重量%的水平用于牙膏中。
精氨酸当存在时可以例如0.1重量%至20重量%(表示为游离碱基的重量),例如1重量%至10重量%的水平存在。
本发明还可包含其他试剂,诸如抗微生物剂。此类试剂包括水不溶性非阳离子抗微生物剂,诸如卤化二苯醚、酚类化合物(包括酚及其同系物、单烷基和多烷基以及芳族卤代酚、间苯二酚及其衍生物、双酚化合物和卤化N-水杨酰基苯胺)、苯甲酸酯、以及卤化碳酰替苯胺、多酚和草药。水溶性抗微生物剂包括季铵盐和双-二胍盐、以及其他抗微生物剂。三氯生一磷酸盐为另一种优选的水溶性抗微生物剂。季铵试剂包括其中季氮上的一个或两个取代基具有8至20、通常10至18个碳原子的碳链长度(通常为烷基),而其余取代基(通常为烷基或苄基)具有较低数目的碳原子,诸如1至7个碳原子,通常为甲基或乙基。十二烷基三甲基溴化铵、十四烷基氯化吡啶鎓、溴化度米芬、N-十四烷基-4-乙基氯化吡啶鎓、十二烷基二甲基(2-苯氧基乙基)溴化铵、苄基二甲基硬脂基氯化铵、氯化十六烷基铵基吡啶、季铵化5-氨基-1,3-二(2-乙基-己基)-5-甲基六氢嘧啶、苯扎氯铵、苄索氯铵以及甲基苄索氯铵为典型季铵类抗菌剂的实例。其他化合物为双[4-(R-氨基)-1-吡啶嗡]链烷烃,如在1980年6月3日提交的Bailey的美国专利号4,206,215所公开的。
还可包括其他抗微生物剂,诸如双甘氨酸铜、甘氨酸铜、柠檬酸锌、以及乳酸锌。同样适用的是酶,包括内切糖苷酶、木瓜蛋白酶、葡聚糖酶、变异酶(mutanase)、以及其混合物。此类试剂公开于Norris等的1960年7月26日的美国专利号2,946,725和Gieske等的1977年9月27日的美国专利号4,051,234中。特定抗微生物剂包括氯己定、三氯生、三氯生单磷酸盐、以及调味油(诸如百里酚)。三氯生为用于包含在本发明组合物中的优选抗微生物剂。三氯生和这种类型的其他试剂公开于Parran等的1991年5月14日发布的美国专利号5,015,466和Nabi等的1990年1月16日的美国专利号4,894,220中。水不溶性抗微生物剂、水溶性试剂和酶可存在于第一或第二洁齿剂组合物中。季铵试剂、亚锡盐和取代的胍优选存在于第二洁齿剂组合物中。这些试剂可以所述洁齿剂组合物的0.01重量%至1.5重量%的水平存在。
草药试剂包括但不限于黄连提取物、金银花提取物、以及其混合物,所述草药试剂也可以0.01%至0.05%的水平存在于本文中的组合物中。认为此类草药试剂提供抗菌功效。多酚类也可以0.01%至2%的水平包含在内。一种优选的多酚为茶多酚。
有效量的脱敏剂也可并入到本发明组合物中。脱敏剂包括选自碱金属盐与第II族金属或铝或阻塞小管的可聚合单体的氯化物、硝酸盐、硫酸盐或乙酸盐、碱金属硝酸盐或硝酸铵、草酸铵、柠檬酸以及柠檬酸钠的那些。优选的盐为硝酸钾、柠檬酸钾、氯化钾以及其混合物。此类脱敏剂公开于例如美国专利号5,718,885。
洁齿剂组合物可为糊剂、凝胶或任何构型或其组合。本发明的组令物可在单相中。本发明的另一个实施方案包括双相或多相组合物,其包含本发明低水组合物作为一相并且包含含有另外的洁齿剂组分以进一步增强洁齿剂产品的稳定性、性能和/或美学的至少一个另一单独相。例如,双相组合物可包含含有本发明低水组合物与多磷酸盐的第一相和含有另外的活性剂的单独第二相,所述另外的活性剂诸如亚锡离子源、漂白剂,优选为过氧化物源或牙齿表面调节剂,以提供改进的清洁、增白、抗污和口感益处。
牙齿调节剂的实例为聚硅氧烷和改性聚硅氧烷,包括二有机基聚硅氧烷,诸如聚二甲基硅氧烷(PDMS);烷基-和烷氧基-聚二甲基硅氧烷共聚醇,诸如C12至C20烷基聚二甲基硅氧烷共聚醇;以及氨基烷基硅氧烷。这些硅氧烷聚合物描述于例如美国专利号5,759,523;6,024,891;6,123,950;6,019,962;6,139,823,这些专利全部转让给Procter&Gamble公司。

在实施方案的实践中,使用者仅需要将本文中的洁齿剂组合物应用于人类或动物的牙齿表面希望的区域,以便获得希望的作用,例如增白、呼吸清新、预防龋病、减轻疼痛、维持牙龈健康、控制牙垢、控制侵蚀等等。洁齿剂控制牙齿表面侵蚀或预防矿质脱去的用途为已知的并且描述于例如美国专利号6,685,920中,所述专利的公开内容以引用的方式整体并入本文。所述组合物也可应用于其他口腔表面,诸如齿龈和粘膜组织,尽管认为当洁齿剂应用于牙齿时获得最佳益处。洁齿剂组合物可直接或间接接触牙齿和/或口腔表面,然而,优选的是洁齿剂组合物直接应用。洁齿剂组合物可通过任何方式应用,但优选地使用牙刷或通过用洁齿剂浆液冲洗来应用。
本发明的口腔组合物的制造可通过用于产生此类组合物的任何各种标准技术来实现。为了制备洁齿剂,可制备含有保湿剂(如甘油、丙三醇、山梨醇、以及丙二醇中的一种或多种)、增稠剂和抗菌剂(例如,三氯生)的媒介物,并且加入所述媒介物以及阴离子表面活性剂和两性表面活性剂的混合物,接着将抛光剂以及氟盐与预混合物共混。最后,掺混调味剂并将pH调节至6.8至7.0之间。
在第一实施方案中,本发明提供了一种包含以下的口腔护理组合物(组合物1):
(a)亚锡离子源;
(b)锌离子源;
(c)多磷酸盐;以及
(d)增稠剂,其包含:
(i)1重量%至3.5重量%聚乙烯吡咯烷酮,
(ii)0.2重量%至0.45重量%多糖胶,以及
(iii)0.05重量%至0.3重量%羧甲基纤维素;例如:
1.1.组合物1,其中亚锡离子源的量按所述组合物的重量计为0.01%至10%,例如0.1%至3%、0.3%至0.7%、0.4%至0.6%;
1.2.任一项前述组合物,其中锌离子源的量按所述组合物的重量计为0.5%至5%或1%至4%;
1.3.任一项前述组合物,其中多磷酸盐的量按所述组合物的重量计为0.1%至30%、或2%至20%、或1%至10%或3%至7%;
1.4.任一项前述组合物,其中所述亚锡离子源包括但不限于,氟化亚锡、其他亚锡卤化物诸如氯化亚锡二水合物、焦磷酸亚锡、有机羧酸亚锡盐诸如甲酸亚锡、乙酸亚锡、葡糖酸亚锡、乳酸亚锡、酒石酸亚锡、草酸亚锡、丙二酸亚锡和柠檬酸亚锡、亚乙基果绿定亚锡、或其混合物;
1.5.任一项前述组合物,其中所述锌离子源为氟化锌、氯化锌、氯氟化锌、乙酸锌、六氟锆酸锌、硫酸锌、酒石酸锌、葡萄糖酸锌、柠檬酸锌、乳酸锌、苹果酸锌、甘氨酸锌、焦磷酸锌、偏磷酸锌、草酸锌、磷酸锌、碳酸锌、氧化锌、或其混合物;
1.6.任一项前述组合物,其中所述多磷酸盐为焦磷酸四钠(TSPP)、三聚磷酸钠(STPP)、酸式焦磷酸钠(SAPP)、或其混合物;
1.7.任一项前述组合物,其中增稠剂的总量为所述组合物的1重量%至5重量%;
1.8.任一项前述组合物,其中所述多糖胶为黄原酸胶、阿拉伯树胶、瓜尔豆胶、刺槐豆胶、黄蓍胶、结冷胶、他拉胶或其混合物;
1.9.任一项前述组合物,其中聚乙烯吡咯烷酮的量按所述组合物的重量计为1%至3.5%、或2.75%至3.25%、或1%至1.5%,多糖胶的量按所述组合物的重量计为0.2%至0.45%、或0.35%至0.45%、或0.2%至0.3%,并且羧甲基纤维素的量按所述组合物的重量计为0.05%至0.3%、或0.05%至0.15%、或0.2%至0.3%;
1.10.任一项前述组合物,所述组合物的屈服应力为15Pa至70Pa或20Pa至50Pa,并且粘度为150,000cP至1,000,000cP或200,000cP至700,000cP,如在25℃下通过布鲁克费尔德粘度计测量的;
1.11.任一项前述组合物,其中所述口腔护理组合物在所述组合物制备后七天在25℃温度下表现出相对于初始粘度小于70%的粘度增加;或者在所述组合物制备后七天在25℃温度下表现出相对于初始粘度小于60%的粘度增加;或者在所述组合物制备后七天在25℃温度下表现出相对于初始粘度40%-60%的粘度增加。
1.12.任一项前述组合物,其还包含有效量的氟化物,例如其中所述氟化物为选自以下的盐:氟化亚锡、氟化钠、氟化钾、氟化铟、氟化锌、单氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺(例如,N′-十八烷基三亚甲基二胺-N,N,N′-三(2-乙醇)-二氢氟化物)、氟化铵、氟化钛、六氟硫酸盐、以及其组合;
1.13.任一项前述组合物,其包含游离或口腔可接受盐形式的L-精氨酸;
1.14.任一项前述组合物,其包含缓冲剂,例如柠檬酸缓冲液或磷酸钠缓冲液(例如,磷酸二氢钠和磷酸二钠);
1.15.任一项前述组合物,其包含保湿剂,例如选自甘油、山梨醇、丙二醇、聚乙二醇、木糖醇、以及其混合物;
1.16.任一项前述组合物,其还包含研磨剂或微粒;
1.17.前一项所述组合物,其中所述研磨剂或微粒选自碳酸氢钠、磷酸钙(例如,二水合磷酸二钙)、硫酸钙、沉淀的碳酸钙、焦磷酸钙、二氧化硅(例如,水合二氧化硅)、氧化铁、氧化铝、珠光体、塑性粒子(例如,聚乙烯)、以及其组合;
1.18.任一项前述组合物,其包含所述总组合物重量的6%至70%、或15%至70%、或10%至50%的量的研磨剂;
1.19.任一项前述组合物,其包含一种或多种表面活性剂,例如选自阴离子型、阳离子型、两性离子型、以及非离子型表面活性剂、以及其混合物,例如包含阴离子型表面活性剂,例如选自月桂基硫酸钠、月桂基醚硫酸钠、以及其混合物的表面活性剂,例如量为0.3重量%至4.5重量%;
1.20.任一项前述组合物,其为牙膏或牙齿凝胶的形式;
1.21.任一项前述组合物,其还包含调味剂、芳香剂和/或着色剂;
1.22.任一项前述组合物,其还包含小于15%水;
1.23.任何前述组合物,其还包含一种或多种抗菌剂,例如包含选自以下的抗菌剂:卤化二苯基醚(例如三氯生)、草药提取物和香精油(例如,迷迭香提取物、茶提取物、木兰提取物、百里酚、薄荷醇、桉油精、香叶醇、香芹酚、柠檬醛、扁柏酚、儿茶酚、水杨酸甲酯、表没食子儿茶素没食子酸酯、表没食子儿茶素、没食子酸、米斯瓦克(miswak)提取物、沙棘提取物)、双胍防腐剂(例如,氯己定、阿立西定或奥替尼啶)、季铵化合物(例如,十六烷基氯化吡啶鎓(CPC)、苯扎氯铵、十四烷基氯化吡啶鎓(TPC)、N-十四烷基-4-乙基氯化吡啶鎓(TDEPC))、酚类防腐剂、海克替啶、奥替尼啶、血根碱、聚乙烯吡咯烷酮碘、地莫匹醇、5-正辛酰-3′-三氟甲基苯基水杨酰胺(salifluor)、金属离子(例如,锌盐(例如柠檬酸锌)、亚锡盐、铜盐、铁盐)、血根碱、蜂胶和氧合剂(例如,过氧化氢、缓冲的过硼酸钠或过氧碳酸钠)、邻苯二甲酸及其盐、单过氧邻苯二甲酸(monoperthalic acid)及其盐和酯、硬脂酸抗坏血酸酯、油酰肌氨酸、烷基硫酸酯、琥珀酸二辛酯磺酸盐、N-水杨酰基苯胺、溴化度米芬、地莫匹醇、辛哌醇和其他哌啶子基衍生物、烟酸类制备物、亚氯酸盐;任何以上各项的混合物;例如,包含三氯生或十六烷基氯化吡啶鎓;

1.29.任一项前述组合物,其还包含有效于减小牙本质敏感性的量的生理上或口腔上可接受的钾盐,例如硝酸钾或氯化钾;
1.30.任一项前述组合物,其还包含呼吸清新剂、芳香剂和/或调味剂;
1.31.任一项前述组合物,其例如通过刷牙应用于口腔后有效于(i)减小牙齿超敏反应、(ii)减少菌斑累积、(iii)减少或抑制脱矿质并促进牙齿补充矿质、(iv)抑制口腔内的微生物生物膜形成、(v)减少或抑制牙龈炎、(vi)促进口腔内疮疡或切口的愈合、(vii)减少产酸细菌的水平、(viii)增加非生龋齿和/或非菌斑形成细菌的相对水平、(ix)减少或抑制龋齿形成、(x)减少、修复或抑制牙釉质龋前损伤(例如,通过定量光激发荧光技术(QLF)或电龋齿测量(ECM)检测的)、(xi)治疗、减轻或减少口干、(xii)清洁牙齿和口腔、(xiii)减少侵蚀、(xiv)增白牙齿;和/或(xv)促进全身性健康,包括心血管健康,例如通过经由口腔组织减少全身性感染的可能性;
1.32.一种通过将任一项前述组合物中陈述的成分组合来获得或可获得的组合物;
1.33.任一项前述组合物,其中所述组合物为牙膏,其任选地还包含以下一种或多种:水、研磨剂、表面活性剂、发泡剂、维生素、聚合物、酶类、保湿剂、增稠剂、抗微生物剂、防腐剂、调味剂、着色剂和/或其组合。

粘度和屈服应力
程序
针对含有结果部分指定的不同量的CMC7(羧基取代度为每个单体0.7,羧甲基纤维素钠和聚阴离子型纤维素来自Quimica Amtex,Mexico)、PVP(Polyplasdone XL-10,其为一种平均粒度为110-140um且容积密度为0.3g/cm3的N-乙烯基-2-吡咯烷酮的交联共聚物)和黄原酸胶的配制品A的组合物测量粘度和屈服应力。在25℃下使用布鲁克费尔德Model RVT粘度计Spindle V74以1RPM测量粘度;粘度以厘帕(cP)计。在25℃下使用布鲁克费尔德Model RVT粘度计Spindle V74测量屈服应力(Ysup为以Pa计的屈服应力)。

Claims (25)
1.一种口腔护理组合物,其包含:
(a)亚锡离子源;
(b)锌离子源;
(c)多磷酸盐;以及
(d)增稠剂,其包含:
(i)1重量%至3.5重量%聚乙烯吡咯烷酮,
(ii)0.2重量%至0.45重量%多糖胶,以及
(iii)0.05重量%至0.3重量%羧甲基纤维素
其中所述重量%按所述组合物的重量计。
2.如权利要求1所述的组合物,其中亚锡离子源的量按所述组合物的重量计为0.01%至10%,并且所述亚锡离子源选自氟化亚锡、其他亚锡卤化物诸如氯化亚锡二水合物、焦磷酸亚锡、有机羧酸亚锡盐诸如甲酸亚锡、乙酸亚锡、葡糖酸亚锡、乳酸亚锡、酒石酸亚锡、草酸亚锡、丙二酸亚锡和柠檬酸亚锡、亚乙基果绿定亚锡、以及其混合物。
3.如任一项前述权利要求所述的组合物,其中亚锡离子源的量按所述组合物的重量计为0.1%至3%。
4.如任一项前述权利要求所述的组合物,其中亚锡离子源的量按所述组合物的重量计为0.3%至0.7%。
5.如任一项前述权利要求所述的组合物,其中亚锡离子源的量按所述组合物的重量计为0.4%至0.6%。
6.如任一项前述权利要求所述的组合物,其中所述亚锡源为氟化亚锡。
7.如任一项前述权利要求所述的组合物,其中锌离子源的量按所述组合物的重量计为0.5%至5.0%并且所述锌离子源选自氟化锌、氯化锌、氯氟化锌、乙酸锌、六氟锆酸锌、硫酸锌、酒石酸锌、葡萄糖酸锌、柠檬酸锌、乳酸锌、苹果酸锌、甘氨酸锌、焦磷酸锌、偏磷酸锌、草酸锌、磷酸锌、碳酸锌、氧化锌、以及其混合物。
8.如任一项前述权利要求所述的组合物,其中锌离子源的量按所述组合物的重量计为1%至4%。
9.如任一项前述权利要求所述的组合物,其中所述锌源为氧化锌。
10.如任一项前述权利要求所述的组合物,其中多磷酸盐的量按所述组合物的重量计为0.1%至30%、或2%至20%、或1%至10%或3%至7%,并且所述多磷酸盐选自焦磷酸四钠、三聚磷酸钠、酸式焦磷酸钠、以及其混合物。
11.如任一项前述权利要求所述的组合物,其中多磷酸盐的量按所述组合物的重量计为2%至20%。
12.如任一项前述权利要求所述的组合物,其中多磷酸盐的量按所述组合物的重量计为1%至10%。
13.如任一项前述权利要求所述的组合物,其中多磷酸盐的量按所述组合物的重量计为3%至7%。
14.如任一项前述权利要求所述的组合物,其中聚乙烯吡咯烷酮的量按所述组合物的重量计为2.75%至3.25%,多糖胶的量按所述组合物的重量计为0.35%至0.45%,并且羧甲基纤维素的量按所述组合物的重量计为0.05%至0.15%。
15.如任一项前述权利要求所述的组合物,其中聚乙烯吡咯烷酮的量按所述组合物的重量计为1%-1.5%,多糖胶的量按所述组合物的重量计为0.2%至0.3%,并且羧甲基纤维素的量按所述组合物的重量计为0.2%至0.3%。
16.如任一项前述权利要求所述的组合物,其中多糖胶为黄原酸胶、阿拉伯树胶、瓜尔豆胶、刺槐豆胶、黄蓍胶、结冷胶、他拉胶或其混合物。
17.如任一项前述权利要求所述的组合物,其中多糖胶为黄原酸胶。
18.如任一项前述权利要求所述的组合物,所述组合物在25℃下的布鲁克费尔德屈服应力为15Pa至70Pa,并且在7天后在25℃下的最大布鲁克费尔德粘度为150,000cP至1,000,000cP。
19.如任一项前述权利要求所述的组合物,所述组合物在25℃下的布鲁克费尔德屈服应力为20Pa至50Pa,并且在7天后在25℃下的最大布鲁克费尔德粘度为200,000cP至700,000cP。
20.如任一项前述权利要求所述的组合物,其中所述口腔护理组合物在所述组合物制备后七天在25℃温度下表现出相对于初始粘度小于70%的粘度增加。
21.如任一项前述权利要求所述的组合物,其中所述口腔护理组合物在所述组合物制备后七天在25℃温度下表现出相对于初始粘度小于60%的粘度增加。
22.如任一项前述权利要求所述的组合物,其中所述口腔护理组合物在所述组合物制备后七天在25℃温度下表现出相对于初始粘度40%-60%之间的粘度增加。
23.如任一项前述权利要求所述的组合物,其还包含选自以下的氟盐:氟化亚锡、氟化钠、氟化钾、氟化铟、氟化锌、单氟磷酸钠、氟硅酸钠、氟硅酸铵、氟化胺(例如,N′-十八烷基三亚甲基二胺-N,N,N′-三(2-乙醇)-二氢氟化物)、氟化铵、氟化钛、六氟硫酸盐、以及其组合。
24.如任一项前述权利要求所述的组合物,其还包含所述总组合物重量的6%至70%、或15%至70%、或10%至50%的量的研磨剂。
25.如任一项前述权利要求所述的组合物,其还包含保湿剂、增白剂、表面活性剂、抗菌剂、着色剂、调味剂、或这些两种或更多种的任何组合。
