背景技术

发明内容

发明内容
最近功能性纳米技术和分子疗法的出现为新型的和更有效的医学方法铺平了道路。本文提供了提供荧光量子点纳米颗粒(QD)的实施方案,其具有与肿瘤靶向部分缀合的高安全性和生物相容性特征,其中缀合物具有治疗和诊断用途。QD是具有极好光学性质的荧光半导体纳米颗粒。它们比任何常规荧光染料(如吲哚菁绿(ICG))亮20倍左右,光稳定性高出许多倍。重要的是,由于其化学性质和纳米尺寸,QD停留时间较长。QD可以吸收和发射更强的光强度,从而提高检测的便利性。QD还提供发射可调性,以便在自发荧光干扰的最小背景下实现空间成像。另一个优点是,与小染料不同,每个QD可以配备多个结合标签,形成多特异性纳米装置,例如但不限于具有最大结合概率并因此具有最高检测效率的双或三特异性纳米装置。
QD的独特性质使得若干医疗应用能够满足体外和体内诊断、临床成像、靶向药物递送和光动力疗法的未满足需求。因此,缀合物是具有用于癌症成像和治疗的多模态特性的“治疗诊断性”纳米装置。在某些实施方案中,所公开的治疗诊断性纳米装置具有成像和治疗能力,可以用于癌症的术前、术中和术后检测和治疗。这些装置特别适用于治疗目前手术切除提供唯一治愈希望的癌症。某些癌症如腺癌生长相对缓慢,因此对靶向快速分裂细胞的化学治疗和放射疗法具有抗性。一种潜在的治愈方法是在腺癌变转移性之前手术切除腺癌。问题在于,某些癌症,例如外分泌胰腺的腺癌,在诊断时通常是转移性的。与此相结合,肿瘤组织在切除期间难以与正常组织区分开,从而使肿瘤组织留在原位。
部分地基于分期困难,许多患者的推测播散性和切除手术的难度,目前仅有8%的英国胰腺癌患者接受手术切除。目前,胰腺癌的10年存活率<1%。在胰腺癌的治疗中急需的是术中识别和去除包括小转移的癌组织的能力。
已经进行了努力以提供足以支持充分切除的肿瘤细胞的体内标记和鉴定,但是小分子染料、有机染料和炭黑墨水缺乏特异性,易于迅速污染所有周围环境。最近,已经引入了使用有机染料(荧光素或吲哚菁绿(ICG))的荧光成像,虽然荧光染料可以提高选择性,但是它们受到它们的快速清除、快速褪色、快速代谢降解和低信号强度的限制。参见,例如Condeelis J和Weissleder R.In Vivo Imagi ng in Cancer.Cold Spri ng HarbPerspect Biol.2010,2:a003848。本发明人认识到,用于支持更完全切除的理想术中诊断工具也将为剩余细胞提供直接的杀肿瘤模式。
鉴于现有技术的缺点,本发明人致力于提供对在癌细胞中过表达的靶分子具有选择性的诊断性纳米装置,其初始关注于胰腺癌细胞。一种这样的靶分子是表皮生长因子受体(EGFR)。EGFR,又名HER1,是一种细胞表面分子,是erbB受体酪氨酸激酶家族的成员。与EGF结合后,EGFR同二聚化(或与其他erbB家族成员异二聚体化),并通过磷酸肌醇3-激酶(PI-3K)/AKT和丝裂原活化蛋白(MAP)激酶途径启动信号传导。通过EGFR/HER1介导的细胞信号传导促进肿瘤增殖、血管生成、转移和细胞凋亡的逃避。EGFR在几种恶性肿瘤(包括头颈癌、肾癌、结肠直肠癌、肺癌、乳腺癌、前列腺癌和胰腺癌)中过表达。因为EGFR是癌症治疗的有希望的靶标,已经开发了结合EGFR并阻断EGF配体的结合的单克隆抗体(mAb)。一种这样的mAb是人-鼠嵌合抗体西妥昔单抗(由Eli Lilly以商品名销售)。西妥昔单抗被FDA批准用于治疗EGFR阳性结肠直肠癌和某些类型的头颈癌以及特定的化学治疗方案。
EGFR也是用于诊断和治疗胰腺癌的鉴定靶标。在约70%的患者中,EGFR被胰腺肿瘤细胞过表达,并且这种过表达与预后不良有关。在此基础上,已经在胰腺癌中研究了通过使用西妥昔单抗靶向EGFR。不幸的是,II期和III期试验未能始终显示西妥昔单抗单独治疗或与细胞毒性剂联合治疗晚期胰腺癌的疗效。参见Luedke,E.等人,Monoclonal AntibodyTherapy of Pancreatic Cancer with Cetuximab:Potential for I mMune Modulation.J I mMunother.35(5)(2012)367-373。除了单独使用西妥昔单抗外,还尝试使用西妥昔单抗缀合物。为此,西妥昔单抗已被用作靶向剂以递送~5nm的与细胞毒性剂吉西他滨共同衍生的金纳米颗粒。参见Patra,CR等人Targeted Delivery of Gemcitabine toPancreatic Adenocarcinoma:Usi ng Cetuximab as a Targeti ng Agent.Cancer Res68(6)(2008)1970-1978。在Patra的研究中,金纳米颗粒被用作易于合成和表征的无毒可衍生载体。然而,金颗粒载体不具有活性诊断性质,并且金纳米颗粒的生物分布必须在死后进行,从而排除了术中使用的可能性。
在本文提供的某些实施例中,靶标癌症是胰腺癌,因为它通常是致命的,因此迫切需要有效的诊断和治疗策略来提高存活率。此外,胰腺肿瘤病变的分期和精确手术由于其柔软和不可切除的性质而具有挑战性。在某些实施方案中,将纳米装置设计为生物相容的、无毒的荧光量子点纳米颗粒(QD)与抗EGFR mAb的缀合物。在其他实施方案中,提供了生物相容的、无毒的荧光QD与抗PD-L1检查点mAb的缀合物。在更进一步的实施方案中,提供具有针对EGFR和PD-L1的组合特异性的多价QD纳米颗粒。使用抗EGFR和PD-L1举例说明的构思适用于通过使用相关的mAb或其衍生物靶向其他类型的癌症。
在一个实施方案中,无毒水溶性QD与针对人表皮生长因子受体(EGFR)的细胞外结构域的抗体化学连接。西妥昔单抗是一种目前经FDA批准的抗体,用于治疗头颈癌、结肠直肠癌和肺癌的局部或区域性晚期鳞状细胞癌。许多研究也在考虑将西妥昔单抗用于具有高EGFR表达的胰腺癌和许多其他癌症。
因此,在一个实施方案中,无毒的和水溶性的(生物相容的)QD被合成并且表面配备有能够缀合的功能(COOH、OH、NH2、SH、叠氮化物、炔烃)。在一个示例性实施方案中,水溶性无毒QD是羧基官能化的或变成羧基官能化的。通过采用水溶性1-乙基-3-(-3-二甲基氨基丙基)碳二亚胺盐酸盐(EDC)的碳二亚胺连接技术,将COOH-QD与肿瘤靶向抗体(例如西妥昔单抗)的胺末端连接。将羧基官能化的QD与EDC混合以形成活性O-酰基异脲中间体,其随后被来自反应混合物中单克隆抗体上的伯氨基的亲核攻击而置换。如果需要,在与含伯胺抗体的反应过程中加入N-羟基琥珀酰亚胺(磺基-NHS)的磺基衍生物。加入磺基-NHS后,EDC将NHS与羧基偶联,形成比O-酰基异脲中间体更稳定的NHS酯,同时允许在生理pH下与伯胺有效缀合。在任一情况下,结果是在QD和抗体之间形成共价键。也可以备选地使用其他化学过程如Suzuki-Miyaura交叉偶联(SMCC)或基于醛的反应。
如图4A和B所示,该实施方案的基本原理是利用抗EGFR mAb(例如西妥昔单抗)特异性结合和抑制肿瘤上的EGFR的能力的优点,通过缀合荧光QD将其用作特异性标记。形成的(QD-西妥昔单抗)药剂具有四种不同的功能:

·由于西妥昔单抗与EGFR结合导致的EGFR抑制,

据信上述四种机制的组合提供了协同效应,其可以为表达EGFR的癌症提供非常有效的治疗并且具有比单独的西妥昔单抗治疗更低的副作用。

纳米颗粒与介质的相容性以及纳米颗粒对聚集、光氧化和/或猝灭的敏感性主要由纳米颗粒的表面组成介导。关于任何核、核-壳或核/多壳纳米颗粒中的最终无机表面原子的配位可能是不完全的,在表面上具有高反应性“悬空键”,这可导致颗粒聚集。通过用保护性有机基团钝化(封端)“裸”表面原子来克服该问题,所述保护性有机基团在本文中称为封端配体或封端剂。颗粒的封端或钝化防止颗粒发生聚集,但也保护颗粒免受其周围的化学环境的影响,并且在核材料的情况下为颗粒提供电子稳定化(钝化)。封端配体通常是与颗粒的最外层无机层的表面金属原子结合的路易斯碱。封端配体的性质很大程度上决定了纳米颗粒与特定介质的相容性。

包含单一半导体材料(例如,CdS、CdSe、ZnS、ZnSe、InP、GaN等)的纳米颗粒因为在纳米颗粒表面的缺陷或悬浮键处发生的非辐射电子-空穴复合可具有相对低的QY。为了至少部分地解决这些问题,纳米颗粒核可以至少部分地涂覆有不同于核的材料的一个或更多个层(在本文中也称为“壳”),例如不同于“核”的半导体材料。包含在一个或多个壳中的材料可以掺入来自周期表的第2-16族中的任何一个或更多个族的离子。当纳米颗粒具有两个或更多个壳时,每个壳可以由不同的材料形成。在示例性的核/壳材料中,核由上面指定的材料之一形成,壳包括具有比核材料更大的带隙能量和与其类似的晶格尺寸的半导体材料。示例性壳材料包括但不限于ZnS、ZnO、mgS、mgSe、mgTe和GaN。多壳纳米颗粒的一个实例是InP/ZnS/ZnO。核内和远离表面状态的电荷载体的限制提供了更高稳定性和更高QY的纳米颗粒。
然而,尽管期望获得缺乏有毒重金属的QD,但已证明特别难以修饰无镉QD的表面。当使用诸如上述配体交换方法的方法来修饰这种无镉QD的表面时,无镉QD容易降解。例如,已经观察到尝试修饰无镉QD的表面会引起这种纳米颗粒的QY显著降低。对于本文公开的体内目的,需要具有高QY的表面修饰的无镉QD。出于本发明的目的,当提及无镉QD时:<20%的QY被认为是非常低的;<30%的QY被认为是低的;30-40%的QY为被认为是中等;>40%的QY被认为是高的;>50%的QY被认为是非常高的。
本文公开的高QY无镉水分散性QD具有大于约40%的QY。对于某些体内实施方案,优选不含重金属的半导体铟基QD或含有铟和/或磷的QD。
在某些实施方案中,对无毒QD纳米颗粒进行表面修饰以使其是水溶性的并具有允许通过将其暴露于配体相互作用剂以实现配体相互作用剂与QD表面的缔合而衍生的表面部分。配体相互作用剂可包含链部分和对连接/交联剂具有特定亲和力或与其反应的官能团,如下所述。链部分可以是例如烷烃链。官能团的实例包括亲核试剂,例如硫代基、羟基、甲酰胺基、酯基和羧基。配体相互作用剂也可包含或可不包含对QD表面具有亲和力的部分。这些部分的实例包括硫醇、胺、羧基和膦。如果配体相互作用基团不包含这样的部分,则配体相互作用基团可以通过嵌入封端配体与纳米颗粒的表面缔合。配体相互作用剂的实例包括C8-20脂肪酸及其酯,例如肉豆蔻酸异丙酯。
应当注意,配体相互作用剂可以简单地通过用于合成纳米颗粒的方法与QD纳米颗粒结合,从而避免将纳米颗粒暴露于额外量的配体相互作用剂的需要。在这种情况下,可能不需要将其他配体相互作用剂与纳米颗粒缔合。备选地,或另外,在合成和分离纳米颗粒之后,可将QD纳米颗粒暴露于配体相互作用剂。例如,可以将纳米颗粒在含有配体相互作用剂的溶液中温育一段时间。这种温育或一部分温育期可以处于升高的温度,以促进配体相互作用剂与纳米颗粒表面的缔合。在配体相互作用剂与纳米颗粒表面缔合后,QD纳米颗粒暴露于连接/交联剂和表面修饰配体。连接/交联剂包括对配体相互作用剂的基团和表面修饰配体具有特定亲和力的官能团。配体相互作用剂-纳米颗粒缔合复合物可以连续地暴露于连接/交联剂和表面修饰配体。例如,纳米颗粒可以暴露于连接/交联剂一段时间以实现交联,然后暴露于表面修饰配体以将其掺入纳米颗粒的配体壳中。备选地,纳米颗粒可以暴露于连接/交联剂和表面修饰配体的混合物,从而在单个步骤中实现交联并掺入表面修饰配体。
实施例
为了公开的完整性,并说明制备本发明组合物和复合物的方法以及呈现组合物的某些特征。这些实施例决不是要限制本公开的范围或教导,包括了以下实施例。
实施例1
无毒量子点的合成
分子接种过程用于产生无毒QD。简而言之,如下进行在500-700nm范围内发射的非官能化铟基QD的制备:将二丁基酯(约100ml)和肉豆蔻酸(MA)(10.06g)置于三颈烧瓶中并在-70℃和真空下脱气1小时。此后,引入氮气并将温度升至-90℃。加入约4.7g的ZnS分子簇[Et3NH]4[Zn10S4(SPh)16],并将混合物搅拌约45分钟。然后将温度升至~100℃,然后逐滴加入In(MA)3(1M,15ml),接着加入三甲基甲硅烷基膦(TMS)3P(1M,15ml)。搅拌反应混合物,同时将温度升至~140℃。在140℃下,进一步滴加溶于癸二酸二正丁酯的肉豆蔻酸铟(In(MA)3)(1M,35ml)(搅拌5分钟)和溶于癸二酸二正丁酯的(TMS)3P(1M,35ml)。然后将温度缓慢升至180℃,进一步滴加In(MA)3(1M,55ml),然后滴加(TMS)3P(1M,40ml)。通过以这种方式添加前体,形成发射最大值从500nm逐渐增加到720nm的铟基颗粒。当获得所需的发射最大值时停止反应并在反应温度下搅拌半小时。此后,将混合物退火多至约4天(在比反应温度低-20-40℃的温度下)。在该阶段还使用UV灯来辅助退火。
通过套管技术添加干燥的脱气甲醇(约200ml)来分离颗粒。使沉淀物沉降,然后借助滤棒通过套管除去甲醇。加入干燥的脱气氯仿(约10ml)以洗涤固体。将固体在真空下干燥1天。该过程导致在ZnS分子簇上形成铟基纳米颗粒。在进一步的处理中,通过在稀释的氢氟酸(HF)中洗涤,进一步提高了所得铟基纳米颗粒的量子产率。铟基核材料的量子产率范围为约25%-50%。
ZnS壳的生长:将20ml的部分HF蚀刻的铟基核颗粒在三颈烧瓶中干燥。加入1.3g肉豆蔻酸和20ml癸二酸二正丁酯并脱气30分钟。将溶液加热至200℃,逐滴加入2ml的1M(TMS)2S(速率为7.93ml/h)。该添加完成后,将溶液静置2分钟,然后加入1.2g无水乙酸锌。将溶液在200℃保持1小时,然后冷却至室温。通过加入40ml无水脱气甲醇并离心分离所得颗粒。弃去上清液,向剩余的固体中加入30ml无水脱气己烷。使溶液静置5小时,然后再次离心。收集上清液,弃去剩余的固体。最终的非官能化铟基QD纳米颗粒材料的量子产率在有机溶剂中为约60%-90%。
实施例2
水溶性表面修饰QD
本文提供了用于产生和使用三聚氰胺交联剂六甲氧基甲基三聚氰胺(HMMM)修饰的荧光纳米颗粒作为纳米探针以在体外和体内检测和/或调节生物靶标的方法的一个实施方案。独特的三聚氰胺基涂层表现出出色的生物相容性、低毒性和非常低的非特异性结合。这些独特的功能允许在体外和体内广泛的生物医学应用。
制备合适的水溶性QD的一个实例如下:将在608nm处发红光的200mg无镉QD纳米颗粒(如实施例1所述,其具有包含铟和磷的合金作为核材料,具有含Zn的壳)用肉豆蔻酸异丙酯(100微升)分散在甲苯(1ml)中。包含肉豆蔻酸异丙酯作为配体相互作用剂。将混合物在50℃加热约1-2分钟,然后在室温下缓慢振荡15小时。将六甲氧基甲基三聚氰胺(HMMM)(CYMEL 303,购自Cytec Industries,Inc.,West Paterson,NJ)(400mg)的甲苯溶液(4ml)、单甲氧基聚环氧乙烷(CH3O-PEG2000-OH)(400mg)和水杨酸(50mg)加入到纳米颗粒分散体中。包含在官能化反应中的水杨酸起到三种作用,作为催化剂、交联剂和COOH源。部分由于HMMM对OH基团的偏好,水杨酸提供的许多COOH基团在交联后仍可在QD上获得。
HMMM是基于三聚氰胺的连接/交联剂,具有以下结构:
HMMM可以在酸催化的反应中反应以交联各种官能团,例如酰胺、羧基、羟基和硫醇。
将混合物脱气并在130℃下回流持续第一个小时,然后在140℃下回流3小时,同时用磁力搅拌器以300rpm搅拌。在第一个小时期间,使氮气流通过烧瓶以确保除去由HMMM与亲核试剂反应产生的挥发性副产物。将混合物冷却至室温并在惰性气体下储存。与未修饰的纳米颗粒相比,表面修饰的纳米颗粒在荧光量子产率方面显示出很少的损失或没有损失,并且在发射峰或半峰全宽(FWHM)值方面没有变化。将表面修饰的纳米颗粒的等分试样在真空下干燥,并将去离子水加入到残余物中。表面修饰的纳米颗粒很好地分散在水性介质中并且保持永久分散。相反,未修饰的纳米颗粒不能悬浮在水性介质中。根据上述步骤的表面修饰的纳米颗粒的荧光QY约为47。
在另一个实施方案中,将具有608nm红光的无镉量子点纳米颗粒(200mg)用胆固醇(71.5mg)分散在甲苯(1ml)中。将混合物在50℃加热约1-2分钟,然后在室温下缓慢振荡15小时。将HMMM(Cymel 303)(400mg)、单甲氧基聚环氧乙烷(CH3O-PEG2000-OH)(400mg)、愈创木酚甘油醚(100mg)、二氯甲烷(DCM)(2ml)和水杨酸(50mg)的甲苯溶液(4ml)加入到纳米颗粒分散体中。将混合物脱气并在140℃下回流4小时,同时用磁力搅拌器以300rpm搅拌。与先前的方法一样,在第一个小时期间,使氮气流通过烧瓶以确保除去由HMMM与亲核试剂反应产生的挥发性副产物。将混合物冷却至室温并在惰性气体下储存。将表面修饰的纳米颗粒的等分试样在真空下干燥,并将去离子水加入到残余物中。使用100mM KOH溶液将溶液的pH调节至6.5,并使用Amicon过滤器(30kD)通过三次超滤循环除去过量的未反应物质。将最终的水溶液冷藏至使用。

值得注意的是,用于修饰纳米颗粒以增加其水溶性的传统方法(例如,与巯基官能化的水溶性配体的配体交换)在温和条件下使纳米颗粒具有水溶性是无效的。在更加严苛的条件下,例如加热和超声处理,变成水溶性的部分具有非常低的QY(<20%)。相反,本方法提供具有大于40%的高的或非常高的QY的水溶性纳米颗粒。如本实施例中制备的表面修饰的纳米颗粒在其他极性溶剂(包括乙醇、丙醇、丙酮、甲乙酮、丁醇、甲基丙烯酸三丙基甲酯或甲基丙烯酸甲酯)中也很好地分散并保持永久分散。
实施例3
QD治疗诊断装置
对于体内目的,如果没有要求,具有低毒性特征的QD是理想的。在一个实施方案中,提供缺乏重金属(如镉、铅和砷)的低毒性QD用于癌症(包括胰腺癌、结肠直肠癌、头颈癌、胃癌、脑癌、前列腺癌和食道癌)的图像引导手术。
QD的独特性质实现了几种潜在的医学应用,包括未满足的体外和体内诊断、临床成像、靶向药物递送和光动力疗法。关于QD的医学应用的主要关注点之一已经是大多数研究都已经集中在含有有毒重金属(如镉、铅或砷)的QD上。本文所述的生物相容且水溶性的无重金属QD可安全地用于体外和体内的医学应用。在某些实施方案中,提供体内相容的水分散性无镉QD,其具有10-20nm的流体动力学尺寸(在完整IgG2抗体的空间尺寸范围内)。在一个实施方案中,体内相容的水分散性无镉QD根据本文实施例1和2中所述的步骤制备。在某些实施方案中,体内相容的水分散性的无镉QD被羧基官能化并且用一个或多个配体结合部分进一步衍生。
用本文公开的体内相容的水分散性无镉QD的体外和体内毒理学研究表明它们比市售的基于镉的QD的细胞毒性低至少20倍,并且在比有用剂量高出许多倍的剂量下,在动物模型上没有观察到毒性迹象。此外,本文提供的体内相容的水分散性无镉QD纳米颗粒显示没有溶血作用并且没有补体C3活化,这表明有利的临床相容性特征。
许多癌症通过淋巴系统转移,因此淋巴结转移的存在是许多癌症(包括黑素瘤、乳腺癌、结肠癌、肺癌和卵巢癌)中的重要预后标志物。前哨淋巴结(SLN)被定义为从肿瘤部位接受淋巴引流的第一个淋巴结。乳腺癌细胞最有可能扩散到位于腋窝的淋巴结(LN),因此准确评估腋窝淋巴结(ALN)是(早期)乳腺癌患者生存和复发的最具有预后作用的指标。
当作为SLN定位的标记物评估时,将30μg剂量的非官能化的体内相容的水分散性无镉QD颗粒(钝化颗粒)皮下注射到在麻醉状态下的裸鼠的爪中并且显示出能够使用简单的成像系统检测腋窝淋巴结的位置。如图5所示,不含重金属/无镉和生物相容性的QD纳米颗粒可在皮下注射后通过区域淋巴结的离体成像用于淋巴结定位。通过电感耦合等离子体质谱使用基于元素分析的光致发光成像和化学提取测量,显示QD在腋窝和胸部区域淋巴结中快速且选择性地累积。此外,QD光致发光的寿命成像显微术表明在生物系统中对其光致发光性质的微扰。
还已经证明,本文提供的体内相容的水分散性的无镉QD颗粒可以使用碳二亚胺偶联与诸如链霉抗生物素蛋白和抗HER2单克隆抗体曲妥珠单抗的蛋白质缀合,并且QD缀合物显示靶向底物(例如,携带HER2受体的细胞)的特异性标记。
体内相容的水分散性无镉QD与链霉抗生物素蛋白的共价缀合:利用碳二亚胺化学过程使实施例2中引用的水溶性和官能化QD与链霉抗生物素蛋白(SAV)共价连接,以将表面COOH基团与SAV分子的胺末端连接。例如,以下方案用于将633nm发射性水溶性QD与SAV结合。
在一个实施方案中,在Eppendorf管中,将0.5mg(50μl)水溶性纳米颗粒与100μlMES活化缓冲液混合。将MES缓冲液制备成去DI水中的25mM溶液(2-(N-吗啉)乙磺酸半钠盐(MES),Sigma Aldrich),pH为4.0。然后加入100μl 10mg/ml的新鲜EDC(1-乙基-3-(3-二甲基氨基丙基)碳二亚胺盐酸盐,Fisher Scientific)溶液,充分混合并在室温下静置5分钟。将混合物转移至预润湿的NanoSep 300K过滤器中,加入200μl MES活化缓冲液,并使用台式微量离心机(~2000g)以5000rpm/20min旋转过滤单元。将保留的点重新分散在50μl活化缓冲液中,并转移到含有0.5mg 1x PBS中的SAV的Eppendorf管中。将它们充分混合并在室温下温育10分钟,然后在冰箱(4-6℃)中放置过夜。使用Nanosep 300K过滤器和PBS缓冲液通过三次超滤循环除去过量的SAV。每个离心循环在5000rpm下进行20分钟,并将最终残余物用~400μl PBS再分散。纯化的QD-SAV缀合物的产生描绘于图3中。
在另一个实施方案中,在Eppendorf管中,将1.2mg羧基官能化的水溶性量子点与pH为4.5的100μl MES活化缓冲液(即将25μl50mg/ml储存液稀释至100μl MES)混合。向其中加入33μl新鲜EDC溶液(在去离子水中的30mg/ml储存液,1-乙基-3-(3-二甲基氨基丙基)碳二亚胺盐酸盐(EDC),Fisher Scientific)并将溶液混合。向其中加入4ul新鲜磺基-NHS(100mg/ml储存液,ThermoFisher Scientific,在去离子水中)并将溶液混合。将NanoSep300K过滤器(PALL NanoSep 300K Omega超滤器)在100μl MES中预润湿。将MES/EDC/磺基-NHS/QD溶液加入NanoSep 300K过滤器中并用MES加满至500μl。将过滤器以5000rpm离心15分钟。将保留的点重新分散在50μl活化缓冲液中,并转移至含有5ul链霉抗生物素蛋白溶液(SAV,Sigma Aldrich)(在HEPES中的1mg/100ul SAV储存液,在去离子水中25mM,pH为7.4)+20μl HEPES的Eppendorf管。将溶液充分混合并在室温下温育过夜(约16-18小时)。
用16μl的6-氨基己酸溶液(19.7mg/100mM)淬灭溶液。将溶液转移至预润湿的Nanosep 300K过滤器(100μl 1xPBS),并用1x PBS加满至500μl线。使用Nanosep 300K过滤器和1x PBS缓冲液通过三次超滤循环除去过量的SAV。每个离心循环在5000rpm下持续20分钟,每次循环后用~400μl的1x PBS再分散。将最终浓缩物再分散在100μl PBS中。通过将10μl生物素化的球体(Spherotech 10微米)与5μlSAV-QD缀合物混合来进行活性验证。向其中加入85μl 1xPBS,将混合物摇动放置1小时,然后加入1ml 1x PBS并以1000rpm/5min离心。
为了证明检测系统的活性,将制备的QD-SAV缀合物用于检测聚苯乙烯球表面上的生物素分子。在Eppendorf管中,将10μl生物素球(Spherotech1%,~7微米)与10μl如上所述制备的缀合物混合,并在室温下温育30分钟,然后用1ml PBS洗涤并使用微量离心机(2000rpm/5min))沉淀。使用具有紫色单带通激发滤光器的荧光显微镜(Olympus BX51)观察最终的球体。如图7所示,SAV-QD缀合物特异性结合至生物素球的表面。在这种情况下,红光和绿光体内相容的水分散性无镉QD纳米颗粒与链霉抗生物素蛋白缀合并用于染色生物素化的聚合物球体。当以相同方式处理时,普通的非生物素化球体未显示任何染色,这表明QD的结合是特异性的并且归因于QD纳米颗粒上的SAV功能性。
体内相容的水分散性无镉QD与曲妥珠单抗的共价缀合:在Eppendorf管中,将1.2mg羧基官能化的水溶性QD与100μl MES活化缓冲液(即将25μl50mg/ml储存液稀释至100μl MES)混合。将MES缓冲液制备成在DI水中的25mM溶液(2-(N-吗啉)乙磺酸半钠盐(MES),Sigma Aldrich)pH为4.5。向其中加入33μl新鲜EDC溶液(在去离子水中的30mg/ml储存液,1-乙基-3-(3-二甲基氨基丙基)碳二亚胺盐酸盐(EDC),Fisher Scientific)并将溶液混合。向其中加入4μl新鲜磺基-NHS(100mg/ml储存液,ThermoFisher Scientific,在DI水中)并将溶液混合。将NanoSep 300K过滤器(PALLNanoSep 300K Omega超滤器)在100μl MES中预润湿。将MES/EDC/磺基-NHS/QD溶液加入NanoSep 300K过滤器中并用MES加满至500μl。将过滤器以5000rpm/15min离心。将保留的点再分散在50μl活化缓冲液中,并转移至含有10μl曲妥珠单抗(25mM HEPES缓冲液中的100mg/ml储存液,pH为8.5)+40μlHEPES的Eppendorf管中,pH为8.5。将溶液充分混合并在室温下温育过夜(约16-18小时)。溶液用16μl的6-氨基己酸(6AC)(19.7mg/100mM)淬灭。注意,淬灭可以替代地用其它具有伯胺的化合物进行,但是对于该实施方案选择6AC,因为它具有COOH并且可以保持产物的胶体稳定性。将溶液转移至预润湿的Nanosep300K过滤器(100μl 1xPBS),并用1x PBS加满至500μl线。使用Nanosep300K过滤器和1x PBS缓冲液通过三次超滤循环除去过量的SAV。每个离心循环在5000rpm下持续20分钟,每次循环后用~400μl的1x PBS再分散。将最终浓缩物重新分散在100μlPBS中。图6A和B显示与抗HER2mAb(曲妥珠单抗,)缀合的红色纳米颗粒结合至表达HER2的细胞并鉴定表达HER2的细胞。在图6A中,与抗HER2mAb缀合的红色纳米颗粒用于治疗HER2阳性4T1细胞。图6B显示了对照HER2阴性4T1细胞的相同处理,几乎没有显示出荧光。
进行实验以确保体内相容的水分散性的无镉QD不非特异性结合。对于具有图8A和8B中所示结果的实验,进行斑点印迹。首先将浓度为0.15mg/ml的纯化HER2的等分试样(1μl)垂直点在两个条带的每一个上的三个点上。图8A显示了将不同浓度的缀合物添加到先前点有纯化HER2的条带的结果:i.纯的(neat)缀合物,ii.1/10稀释的缀合物,iii.1/100稀释的缀合物。图8B显示了将不同浓度的未官能化的体内相容的水分散性无镉QD添加到先前点有纯化HER2的条带上的结果。斑点印迹实验的结果显示,与Herceptin缀合的QD结合至HER2,并且当通过将条带暴露于激发光诱导发荧光时,在相对高的稀释度下容易检测到。先前没有点有HER2的条带不会引起未缀合的QD的结合。图9显示点在斑点印迹条上的HER2稀释系列的结果。结果显示,与缀合物温育后,少至7.5ng的在斑点印迹条上点的HER2可以很容易地检测到。
图10A和10B显示了体内相容的水分散性无镉QD与具有和不具有靶向部分的SK-BR-3细胞系结合的结果。SK-BR-3系是过表达HER2的人乳腺癌细胞系。将SK-BR-3细胞在含有10%FCS的McCoy′s改良培养基中培养,并用50μg/ml钝化的或曲妥珠单抗官能化的无镉QD处理过夜,其在630nm发荧光。用于使细胞可视化的激发源是汞弧灯(Osram HBO50W/ACLI Short Arc,2000流明)和具有宽发射带通的DAPI滤光器立方体(激发带为380-450nm)。染色后的细胞在Olympus BX51荧光显微镜上观察。细胞核用Hoechst 33342复染。图10A显示用未官能化的体内相容的水分散性无镉QD处理SK-BR-3细胞的结果。图10B显示用曲妥珠单抗官能化的体内相容的水分散性无镉QD处理SK-BR-3细胞的结果,并且显示当用抗HER2官能化时大大增强了QD摄取。
实施例4
针对胰腺癌的多配体治疗诊断纳米装置
选择多个配体的基本原理是能够靶向与肿瘤细胞相关的广谱受体。在某些实施方案中,多个配体的特异性包括特异性结合选自由以下各项组成的组中的靶标的至少一种配体:EGFR、PD-L1、PD-L2、HER2、CEA、CA19-9、CA125、端粒酶蛋白和亚基、CD20、CD25、CD30、CD33、CD52、CD73、CD109、VEGF-A、CTLA-4以及RANK配体。
在一个特定实施方案中,肿瘤是胰腺癌。如前所述,约70%的胰腺肿瘤表达EGFR,并且这与约80%表达PD-L1重叠。
活化的细胞毒性T细胞、B细胞和骨髓细胞表达称为程序性死亡-1(PD-1)受体的细胞表面受体。PD-1是免疫检查点受体,其调节这些细胞对细胞活性的激活或抑制。PD-1受体具有两个配体,程序性死亡配体-1(PD-L1)和程序性死亡配体-2(PD-L2)。PD-L1也称为分化簇274(CD274)或B7同源物1(B7-H1)。靶细胞上的PD-L1和PD-L2与活化的T细胞上PD-1受体的接合向T细胞递送灭活信号,包括通过抑制TCR介导的IL-2产生的激活和T细胞增殖。通常,细胞毒性T细胞上的PD-1与靶细胞上的PD-L1和PD-L2之间的相互作用可用于在妊娠期间抑制免疫系统并预防自身免疫疾病。然而,肿瘤细胞通过过表达PD-L1来利用该途径,从而抑制T细胞介导的抗肿瘤反应。肿瘤浸润性免疫细胞也表达PD-L1,潜在地导致抑制肿瘤微环境中活化的T细胞。
在表达PD-L1的那些胰腺肿瘤中,20%具有PD-L1表达的高度上调,并且这些肿瘤倾向于高侵入性和复发性。因此,在一个实施方案中,提供了双特异性纳米装置,用于检测和治疗基于QD表达EGFR和PD-L1的癌症。该纳米装置将水溶性荧光QD与两种免疫治疗性单克隆抗体(mAb)-抗EGFR mAb和抗PD-L1mAb组合。
在一个实施方案中,抗EGFR mAb是西妥昔单抗,抗PD-L1mAb是阿特珠单抗。图4A显示了多配体纳米装置的示意图和对癌细胞的作用机制。在这种多配体装置的生成中,将所需量的所选抗体如西妥昔单抗和阿特珠单抗混合,然后使用标准EDC化学过程与羧基官能化QD反应。也可以使用其他化学过程如Suzuki-Miyaura交叉偶联(SMCC)或基于醛的反应。
西妥昔单抗和阿特珠单抗的使用将确保纳米装置针对所有胰腺癌病变的靶向通用性并克服癌性扩散内的异质性。在一个实施方案中,多配体纳米装置用于利用微创内窥镜检查的术前诊断。在其他实施方案中,多配体装置用作术中和术后检测和跟进剂(followup agent)。在该方法中使用无毒水溶性QD是有用的不仅由于它们的亮度和光稳定性,而且还由于其能够连接至少两种不同的靶向分子,这是使用常规标记染料无法实现的特征。
在某些实施方案中,提供了多配体纳米装置,其支持微创分子成像和低风险和高风险胰腺癌的检测,导致早期诊断和治疗,同时减少不必要的手术切除。
在这样的实施方案中,将多配体纳米装置注射到循环中或腹部中并使其浓缩在假定的肿瘤中。腹腔镜装置被引入腹部,其包括用于诱导QD的荧光的光源和成像相机。临床检测和前驱病变的适当管理是降低胰腺癌死亡率的重要策略。本文公开的纳米装置的附加益处在于,与小染料不同,可以应用光动力疗法。激发体内的QD可以导致由官能化的QD结合的细胞诱导的凋亡。
该装置和方法可用于其他类型的恶性肿瘤,以增加用于癌症治疗的可用选择的库(arsenal)。每个QD可以配备有一个以上的特异性结合标签,形成多配体纳米装置,包括但不限于具有最大结合概率并因此具有最高检测效率的双特异性或三特异性纳米装置。针对EGFR和PD-L1过表达的胰腺癌和其他癌症所描述的纳米装置的相同概念可以通过使用相关的mAb或其衍生物用于靶向其他类型的癌症。
在其他的实施方案中,至少一种所述把特异性配体对选自由以下各项组成的组中的靶标具有特异性:EGFR、PD-L1、PD-L2、HER2、CEA、CA19-9、CA125、端粒酶蛋白和亚基、CD20、CD25、CD30、CD33、CD52、CD73、CD109、VEGF-A、CTLA-4以及RANK配体。
人表皮生长因子受体2(HER2)也称为CD340、原癌基因Neu或ERBB2,是表皮生长因子受体家族的成员。ERBB2基因在许多癌症中过表达,包括乳腺癌、子宫癌、胃癌、胃癌和唾液腺癌。单克隆抗体曲妥珠单抗目前用于治疗HER2阳性乳腺癌。
癌胚抗原(CEA)是糖基磷脂酰肌醇(GPI)细胞表面锚定的糖蛋白,其免疫学上表征为CD66a-f的成员。CEA分子参与细胞粘附,通常在出生后不产生,因此在健康成人中以非常低的水平存在。CEA水平在结肠直肠癌中特别高,但在胃癌、胰腺癌、肺癌、乳腺癌和甲状腺髓样癌中也可能高。
碳水化合物抗原19-9(CA19-9),又名癌抗原19-9或唾液酸化的Lewis(a)抗原,是用CA19-9抗体检测的唾液酸-LewisA碳水化合物抗原,其参与细胞间识别(cell-to-cellrecognition)并与晚期胃肠癌(包括结肠直肠癌和胰腺癌)相关。
癌抗原-125(CA125)也称为粘蛋白16(MUC16),其参与能够转移的细胞间相互作用(cell-to-cell interaction),并且是卵巢癌等的生物标志物。
CD20,分化簇20,是由MS4A1基因编码的活化糖基化磷蛋白,且在B细胞表面上表达,但不在早期pro-B细胞或浆细胞的表面上表达。CD20在B细胞淋巴瘤、毛细胞白血病、B细胞慢性淋巴细胞白血病中表达并由黑素瘤干细胞等表达。用于治疗B细胞淋巴瘤和白血病的许多单克隆抗体针对CD20,包括利妥昔单抗、奥奴珠单抗(obinutuzumab)、替伊莫单抗(ibritumomab)和托西莫单抗(tositumomab)。
CD25,分化簇25,是存在于活化的T和B细胞和某些其他细胞上的白细胞介素-2受体α链(IL2RA)蛋白,并且在大多数B细胞肿瘤、一些急性非淋巴细胞白血病、神经母细胞瘤和肥大细胞增多症中表达并由肿瘤浸润淋巴细胞表达。人源化单克隆抗体,达克珠单抗(daclizumab),针对CD25,但目前FDA仅批准用于多发性硬化症。
CD30,分化簇30,有名TNFRSF8,是由活化的T细胞和B细胞表达的肿瘤坏死因子受体家族蛋白。CD30在间变性大细胞淋巴瘤和胚胎癌中表达。人源化单克隆抗体,brentuximab,被FDA批准用于治疗霍奇金淋巴瘤和全身性间变性大细胞淋巴瘤。
CD33,分化簇33,有名唾液酸结合Ig样凝集素3(SIGLEC-3),是髓单核细胞衍生细胞的粘附分子,其介导对细胞的唾液酸依赖性结合并优先结合α-2,6-连接的唾液酸。人源化单克隆抗体吉妥单抗(gemtuzumab)与CD33结合。吉妥单抗和卡奇霉素(calicheamicin)细胞毒剂奥佐米星(ozogamicin)的组合用于治疗急性淋巴细胞白血病。
CD52,分化簇52,有名剑桥病理学抗原-1(Campath-1抗原),是小细胞表面糖蛋白。CD52与某些类型的淋巴瘤有关,并且被单克隆抗体阿仑单抗(alemtuzumab)靶向。
CD73,分化簇73,有名胞外-5′-核苷酸酶(NT5E),是在大多数组织中发现的70kDa细胞表面酶,但似乎在肿瘤细胞中上调,包括在结肠癌、肺癌、胰腺癌和卵巢癌,其作用是削弱抗肿瘤T细胞应答。
CD109,分化簇109,是糖基磷脂酰肌醇(GPI)-连接的糖蛋白,其参与转化生长因子β结合。CD109通常在血小板、活化的T细胞和内皮细胞的细胞表面上发现,但是由CD34+急性髓细胞白血病细胞表达,并且由胰腺导管癌细胞过表达。
血管内皮生长因子A(VEGFA)是血液生长的主要诱导物,并且在成人中在器官重塑和伤口愈合期间表达。然而,VEGF在肿瘤血管生成、糖尿病性视网膜病和年龄相关性黄斑变性中也是致病性的。人源化单克隆抗体贝伐单抗(bevacizumab)用于治疗结肠癌、肺癌、胶质母细胞瘤和肾细胞癌以及年龄相关性黄斑变性。
细胞毒性T淋巴细胞相关蛋白4(CTLA-4),也叫作CD152,是下调免疫应答的免疫检查点蛋白受体。单克隆抗体易普利姆玛(ipilimumab)通过与CTLA-4结合激活免疫系统,并且被FDA批准用于黑素瘤的再治疗和治疗其他癌症的临床试验中。
核因子Kappa-B的受体活化剂(RANK)配体(或RANKL)也称为肿瘤坏死因子配体超家族成员11(TNFSF11)、TNF相关激活诱导细胞因子(TRANCE)、骨保护素配体(OPGL)和破骨细胞分化因子(ODF)。
包括但不限于包括EGFR和PD-L1特异性的双特异性纳米装置的多配体纳米装置的优点可总结如下:
·由于QD的荧光特性导致的最大肿瘤标记以及由于使用两种或更多种不同的结合标签进行官能化而导致的高标记概率,
·西妥昔单抗与EGFR结合导致的EGFR抑制和PD-L1介导的活化T细胞下调的抑制,
·由于缀合物的纳米尺寸导致的EPR(增强的渗透性和保留)效应引起的协同肿瘤摄取,
·由于QD产生能够引起细胞自杀级联(细胞凋亡)的单线态氧或热量形式的凝聚能量的能力而导致的肿瘤光疗效应
据信上述四种机制的组合提供了协同效应,这可以导致表达EGFR和PD-L1的癌症的非常有效的治疗,以及由于能够递送较低剂量的结合QD的西妥昔单抗和阿特珠单抗的较低的副作用。

4.权利要求2所述的纳米装置,其中至少一种靶特异性配体是针对PD-L1的单克隆抗体。
5.权利要求2所述的纳米装置,其中所述表面修饰的水溶性QB纳米颗粒与至少两种配体化学缀合,其中一种配体特异性结合EGRF且其中一种配体特异性结合PD-L1。
6.权利要求5所述的纳米装置,其中所述表面修饰的水溶性QB纳米颗粒与包括西妥昔单抗和阿特珠单抗的至少两种配体化学缀合。
7.权利要求1-6中任一项所述的纳米装置,其中所述表面修饰的无毒水溶性QD纳米颗粒是无镉的并且包含选自由以下各项材料组成的组的半导体材料:ZnS、ZnSe、ZnTe、InP、InAs、InSb、AlP、AlS、AlAs、AlSb、GaN、GaP、GaAs、GaSb、PbS、PbSe、AgInS2、AgInS2/ZnS、Si、Ge及其合金和掺杂衍生物。
8.权利要求7所述的纳米装置,其中所述无镉表面修饰的无毒水溶性QD纳米颗粒包含由一种所述材料形成的核和另一种所述材料形成的一个或更多个壳。
9.权利要求8所述的纳米装置,其中所述无镉表面修饰的无毒水溶性QD纳米颗粒是核/多壳QD。
10.权利要求1至6中任一项所述的纳米装置,其中所述表面修饰的水溶性量子点(QD)纳米颗粒包含配体相互作用剂和表面修饰配体。
11.权利要求10所述的纳米装置,其中所述表面修饰的QD纳米颗粒通过在包含六甲氧基甲基三聚氰胺的溶液中将所述配体相互作用剂和所述表面修饰配体化学加成到所述QD而形成。
12.权利要求10所述的纳米装置,其中所述配体相互作用剂是C8-20脂肪酸及其酯。
13.权利要求10所述的纳米装置,其中所述表面修饰配体是单甲氧基聚环氧乙烷。
14.纳米颗粒缀合物(纳米装置)的混合物,其包含至少两个纳米装置群,包括第一靶特异性群和第二靶特异性群,所述第一靶特异性群包含用第一靶特异性抗体衍生的表面修饰的水溶性量子点(QD)纳米颗粒,所述第二靶特异性群包含用第二靶特异性抗体衍生的表面修饰的水溶性量子点(QD)纳米颗粒。
15.权利要求14所述的混合物,其中所述第一靶特异性抗体对EGFR具有特异性,所述第二靶特异性抗体对PD-L1具有特异性。
16.通过施用包含无毒水溶性量子点(QD)纳米颗粒的纳米颗粒缀合物同时诱导细胞死亡、肿瘤收缩和肿瘤细胞的荧光标记的方法,所述无毒水溶性量子点(QD)与针对由肿瘤细胞过表达的细胞表面部分的抗体化学缀合。
17.权利要求16所述的方法,其中所述细胞表面部分是表皮生长因子受体(EGFR)。
18.权利要求16所述的方法,其中所述肿瘤是胰腺肿瘤。
19.纳米颗粒缀合物(纳米装置),包括附着于量子点(QD)纳米颗粒的两种或更多种不同治疗性抗体,其中所述不同治疗性抗体具有不同的靶特异性。
20.权利要求19所述的纳米装置,其中所述QD纳米颗粒与针对EGFR的抗体和针对PD-L1的抗体化学缀合。
21.检测或治疗性调节生物部分的方法,包括:使包含靶特异性衍生的无镉水溶性量子点(QD)的组合物与生物靶标进行物理相互作用,其中所述无镉水溶性量子点已使用三聚氰胺交联剂六甲氧基甲基三聚氰胺(HMMM)表面修饰以包括选自由以下各项组成的组中的一个或更多个官能团:COOH、NH2、SH、OH、磺酸盐、磷酸盐、叠氮化物、烯丙基、甲硅烷基和PEG链,并通过具有多个不同的靶特异性配体的一个或更多个官能团进一步衍生;
通过施加在所述QD的激发波长下的光来刺激所述靶特异性衍生的无镉水溶性QD的发射;和
对所述组合物与所述生物靶标之间相互作用的光谱发射进行记录和/或成像。
22.权利要求21所述的方法,其中所述配体选自由以下各项组成的组中的一个或更多个:抗体、链霉抗生物素蛋白、核酸、脂质、糖类、药物分子、蛋白质、肽和氨基酸。
23.权利要求21所述的方法,其中所述检测用于对血管生成、肿瘤分界、肿瘤转移、体内诊断和淋巴结进展中的一种或更多种进行成像和检测。
24.权利要求21所述的方法,其中所述检测用于免疫化学、免疫荧光、DNA序列分析、荧光共振能量转移、流式细胞术、荧光激活细胞分选和高通量筛选中的一种或更多种。
25.权利要求21-24中任一项所述的方法,其中所述多种不同的靶特异性配体是对选自由以下各项组成的组中的靶标具有特异性的配体:EGFR、PD-L1、PD-L2、HER2、CEA、CA19-9、CA125、端粒酶蛋白和亚基、CD20、CD25、CD30、CD33、CD52、CD73、CD109、VEGF-A、CTLA-4以及RANK配体。
26.多配体纳米装置,其包含用多个配体群衍生的无镉水溶性量子点,每个群具有不同的靶特异性。
27.权利要求26所述的多配体纳米装置,其包含至少一种靶特异性抗体配体和选自链霉抗生物素蛋白、核酸、脂质、糖、药物分子、蛋白质、肽和氨基酸的其他配体的多配体纳米装置,其中所述抗体配体充当靶向配体以递送所述其他配体。
28.权利要求26所述的多配体纳米装置,其包含至少两种靶特异性抗体配体,其中至少一种是对选自由以下各项组成的组的靶标具有特异性的配体:EGFR、PD-L1、PD-L2、HER2、CEA、CA19-9、CA125、端粒酶蛋白和亚基、CD20、CD25、CD30、CD33、CD52、CD73、CD109、VEGF-A、CTLA-4以及RANK配体。
29.多配体纳米装置,其被制备用作检测和治疗癌症的药物,其中所述多配体纳米装置包含用多个配体群衍生的无镉水溶性量子点(QD),每个配体群对在癌细胞上过表达的不同靶标具有特异性。
30.权利要求29所述的多配体纳米装置,其中所述装置用于用微创内窥镜检查或腹腔镜检查的术前或术中诊断。
31.权利要求29或30所述的多配体纳米装置,其中所述多配体纳米装置适于注射到循环中或腹部中以及在相对于正常细胞的肿瘤细胞中的浓缩。
32.权利要求31所述的多配体纳米装置,其中所述多配体纳米装置通过用单甲氧基聚环氧乙烷进行表面修饰而适于注射到循环中或腹部中以及在相对于正常细胞的肿瘤细胞中的浓缩。
33.权利要求29或30所述的多配体纳米装置,其中浓缩的多配体纳米装置可以通过暴露于内窥镜或腹腔镜施加的QD激发光源诱导发荧光,所述荧光可通过成像相机检测。
34.权利要求29或30所述的多配体纳米装置,其中所述在癌细胞上过表达的不同靶标选自EGFR、PD-L1、PD-L2、HER2、CEA、CA19-9、CA125、端粒酶蛋白和亚单位、CD20、CD25、CD30、CD33、CD52、CD73、CD109、VEGF-A、CTLA-4以及RANK配体。
35.权利要求34所述的多配体纳米装置,其中所述在癌细胞上过表达的不同靶标是EGFR和PD-L1。
36.权利要求29或30所述的多配体纳米装置,其中所述无镉水溶性量子点具有至少50%的量子产率。
