1.本公开提供了一种巴比妥酸衍生物,该巴比妥酸衍生物对于检测硝基芳香族爆炸物具有高灵敏度、良好的选择性、低检测限等优点。
2.通过以本公开的巴比妥酸衍生物作为传感材料的传感器检测硝基芳香族爆炸物时,具有明显的荧光淬灭现象,从而使检测硝基芳香族爆炸物的方法更为简便。

本公开的另一种实施方式,提供了一种上述巴比妥酸衍生物的制备方法,4-二苯胺基苯甲醛与巴比妥酸类化合物进行羟醛缩合反应后获得巴比妥酸衍生物;

本公开中4-二苯胺基苯甲醛的结构式为可以采用市售产品,该实施方式的一种或多种实施例中,提供了4-二苯胺基苯甲醛的制备方法为:三苯胺与二甲基甲酰胺(DMF)在三氯氧磷(POCl3)的催化作用下进行反应。
该系列实施例中,4-二苯胺基苯甲醛的制备步骤为:在-0.5~0.5℃下将DMF与POCl3混合,再加入三苯胺加热至44~46℃,进行反应。
当巴比妥酸类化合物中的R为苯基时,为1,3-二苯基巴比妥酸,可以采用市售产品,也可以自行合成。为了降低成本,该实施方式的一种或多种实施例中,1,3-二苯基巴比妥酸的制备方法为:将丙二酰卤与N,N-二苯基脲进行取代反应获得。丙二酰卤可以为丙二酰氯、丙二酰溴、丙二酰碘等。
该系列实施例中,1,3-二苯基巴比妥酸的制备过程中,将丙二酰卤加入至N,N-二苯基脲的溶液中,加热至75~85℃,进行反应。
本公开的第三种实施方式,提供了一种上述巴比妥酸衍生物在检测硝基芳香族爆炸物中的应用。本公开所述的硝基芳香族爆炸物为硝基芳烃,例如2,4-二硝基甲苯(DNT)、2,4,6-三硝基甲苯(TNT)、2,4,6-三硝基苯酚(PA)等。该巴比妥酸衍生物在检测硝基芳香族爆炸物具有选择性高、检测灵敏等优点,而且可以用于检测水介质中的硝基芳烃。
本公开的第四种实施方式,提供了一种检测硝基芳香族爆炸物的方法,将待测样品置于含有上述巴比妥酸衍生物的溶液,然后紫外-可见光光谱或荧光光谱进行检测。
该实施方式的一种或多种实施例中,采用荧光光谱时,检测波长为620~650nm。
该实施方式的一种或多种实施例中,含有巴比妥酸衍生物的溶液中溶剂为四氢呋喃与水的混合物,四氢呋喃与水的体积比为1:8.9~9.1。实验证明,该体系下对硝基芳香族爆炸物的检测更为灵敏。
本公开的第五种实施方式,提供了一种检测传感器,以上述巴比妥酸衍生物作为传感材料。经过实验证明,采用本公开的固相纸质传感器检测硝基芳烃时,具有明显的荧光淬灭现象。
该实施方式的一种或多种实施例中,包括基体,所述基体为滤纸。
该系列实施例中,制备方法为将滤纸浸泡在含有巴比妥酸衍生物的溶液中,然后进行干燥。
本公开的第六种实施方式,提供了一种检测硝基芳香族爆炸物的方法,将上述传感器置于待测气体或待测液体中,检测传感器的荧光。
为了使得本领域技术人员能够更加清楚地了解本公开的技术方案,以下将结合具体的实施例详细说明本公开的技术方案。
实施例1
4-(二苯胺基)-苯甲醛的合成,路线如下:
在0℃下,将2mL无水(N,N-二甲基甲酰胺)DMF(25.7mmol)加入到5ml三氯氧磷(POCl3)(54mmol)中,并保持15分钟,然后在搅拌下加入三苯胺(1g,4mmol)。将混合物加热至45℃,保持2.5小时。冷却后,将澄清的红色溶液滴入冰水中。将得到的混合物过滤,洗涤并重新溶解在二氯甲烷中。将该溶液用水(150mL)洗涤并用硫酸镁干燥,在溶剂蒸发后,柱色谱层析得到纯净的产物,即4-(二苯胺基)-苯甲醛。产量:0.78g(80%)。1H NMR(400MHz,CDCI3):δ9.82(s,1H),7.74–7.61(m,2H),7.35(t,J=7.7Hz,4H),7.22–7.13(m,6H),7.07–6.96(m,2H).
实施例2
1,3-二苯基巴比妥酸的合成,路线如下:
将丙二酰氯(33.3mg,2mmol)加入到含有6ml CHCl3的N,N-二苯基脲(424.5mg,2mmol)中,并将混合物加热至80℃,保持4小时。然后用二氯甲烷萃取反应混合物,在Na2SO4上干燥。过滤后,将滤液减压浓缩,并通过柱色谱进一步纯化,获得1,3-二苯基巴比妥酸。产量:343.1mg(75%)。1H NMR(400MHz,DMSO-d6):δ7.45(d,J=7.5Hz,3H),7.41(d,J=7.5Hz,1H),7.28(d,J=7.4Hz,3H),4.01(s,1H).
实施例3
5-(4-二苯基氨基苯乙烯)-巴比妥酸(PB-1)的合成,路线如下:
将4-(二苯胺基)-苯甲醛(0.447g,3mmol)和巴比妥酸(0.384g,3mmol)在(10ml)乙醇中的混合物回流4小时。过滤,过滤后,将滤液减压浓缩,并通过柱色谱(乙酸乙酯:石油醚=2:1)进一步纯化。产量:(0.75g)90%。1H NMR(400MHz,CDCl3)δ(ppm):8.42(s,1H),8.31(d,J=8.9Hz,2H),7.97(s,1H),7.80(s,1H),7.40(t,J=7.7Hz,4H),7.24(t,J=6.3Hz,6H),6.94(d,J=8.9Hz,2H).FT-IR(KBr,cm-1):3217(N-H),3061(=C-H),1672(C=O).
实施例4
5-(4-二苯基氨基苯乙烯)-1,3-二甲基-巴比妥酸(PB-2),路线如下:
将4-(二苯胺基)-苯甲醛(0.447g,3mmol)和1,3-二甲基-巴比妥酸(0.489g,3mmol)在(10ml)乙醇中的混合物回流4小时。过滤,过滤后,将滤液减压浓缩,并通过柱色谱(乙酸乙酯:石油醚=2:1)进一步纯化。产率:(0.84g)90%1H NMR(400MHz,CDCl3)δ(ppm):8.44(s,1H),8.24(d,2H),7.38(t,4H),7.23(m,6H),6.95(d,2H),3.40(d,6H).FT-IR(KBr,cm-1):3052(=C-H),1667(C=O).
实施例5
5-(4-二苯基氨基苯乙烯)-1,3-二苯基-巴比妥酸(PB-3),路线如下:
将4-(二苯胺基)-苯甲醛(0.447g,3mmol)和1,3-二苯基巴比妥酸(0.840g,3mmol)在(10mL)乙醇中的混合物回流4小时。过滤,过滤后,将滤液减压浓缩,并通过柱色谱(乙酸乙酯:石油醚=2:1)进一步纯化。产量:(1.158g)90%。1H NMR(400MHz,DMSO-d6)δ(ppm):8.30(s,1H),8.23(d,J=8.4Hz,2H),7.49–7.38(m,12H),7.34(d,J=7.5Hz,6H),7.24(d,J=8.4Hz,8H),6.74(d,J=8.5Hz,2H).FT-IR(KBr,cm-1):3065(=C-H),1667(C=O).
对实施例3~5制备的PB-1、PB-2、PB-3进行性能表征。
PB-1、PB-2和PB-3在不同THF/H2O百分比(fw分别为0%、30%、50%、70%,其中,fw=H2O的体积÷(THF的体积+H2O的体积)×100%)下的吸收光谱,如图1所示,表明PB-1、PB-2和PB-3具有相似的吸收光谱。随着水分的增加,吸光度降低,原因可能是分子的扭转程度随着水分的增加而增加,导致弱的分子内电荷转移。在454nm、455nm和464nm处的吸收峰(分别是PB-1、PB-2和PB-3),这是由于从三苯胺基团到巴比妥酸基团的分子内电荷转移(ICT)。PB-1、PB-2和PB-3的荧光强度在纯THF中较弱(通过肉眼观察),这是由于溶液中的分子间相互作用相对较弱,PB-1、PB-2和PB-3在溶液状态下自由旋转,所以以非辐射跃迁的方式消耗能量,从而显示出微弱的荧光。
PB-1的吸收光谱在350nm附近具有宽且相对较弱的峰,这与n-π*跃迁的特征一致。该机制可以如图2所示。在PB-1上的巴比妥酸基团的N原子上有一对孤对电子。一个参与与羰基的P-π共轭,因此,不能产生n-π*跃迁,然而,350nm附近的吸收峰是由另一个孤对电子的n-π*跃迁引起的。对于PB-2和PB-3,氮原子上的氢原子分别被甲基和苯基取代,并且不产生负离子。
通过荧光检测,PB-1、PB-2和PB-3在纯THF中在619.0nm、628.0nm和634.0nm处表现出弱发射,如表1所示。

IA是聚集态的荧光强度,IP是纯THF中的荧光强度。
对PB-1、PB-2和PB-3的AIE性质进行表征,结果如图3所示,与PB-2和PB-3相比,PB-1具有更强的AIE效应,在聚集溶液中,PB-1的发光强度比纯THF溶液高7.2倍,荧光量子产率(ΦF)从1.5%增加到9.0%,PB-2的最大荧光增强倍数(fw=90%/fw=0%)仅为3.6倍(ΦF:0.8%至3.3%)。同时,PB-3的最大荧光增强倍数(fw=90%/fw=0%)仅为2.1倍(ΦF:1.7%至4%)。PB-1更好的AIE性能是由于氢键的相互作用,这有助于进一步固化聚合态的分子构象,阻断非辐射性途径。
本公开发现PB-1、PB-2和PB-3的AIE效应与巴比妥酸衍生物的分子结构有关,随着巴比妥酸衍生物体积的增加,AIE指数(IA/IP)下降(表1)。PB-1、PB-2和PB-3的聚集诱导荧光指数分别为7.2、3.6和2.1。PB-3的荧光强度在聚集状态下比PB-1和PB-2弱,但在溶液中强于PB-1和PB-2,PB-2的荧光在聚集态中弱于PB-1,但在纯溶液中强于PB-1。原因是体积较大取代基有较大的空间位阻使得分子难以在纯溶液中自由旋转。另一方面,在具有较大分子间距的基团在聚集状态下,分子间相互作用减弱。
如图3所示,PB-2,PB-3的化合物的发射光谱具有相似的行为。当PB-2的fw≤40%且PB-3的fw≤20%时,分子仍然完全溶于混合物溶液中,并且fw的增加增强了混合物的极性,荧光强度随着ICT的发生而降低,而发射波长伴随着红移。随着含水量的增加(40%<fw(PB-2)<80%)(20%<fw(PB-3)≤60%),在溶液中形成松散的聚集体,并且发光材料的微环境由于自缠绕极性减弱,从而产生发射蓝移,同时,分子的自由旋转导致荧光强度降低。当PB-2的80%<fw≤90%和PB-3的60%<fw≤90%时,强的分子间作用力导致纳米聚集体的形成,因此发射波长发生红移并且荧光强度增加。
对于PB-1,当fw从0%增加到60%时,发射波长从619.0变为644.4nm,这种现象的解释为当fw低于60%时,分子仍然完全可溶于混合物中,并且fw的增加增强了混合物的极性,由于ICT效应导致红移。当60%<fw≤70%时,在溶液中形成松散的聚集体,也导致荧光强度降低,然而,PB-1不同于PB-2和PB-3,分子间氢键(N-H-N,N-H···O)的形成增强了分子间的作用力,并伴随着发射波长的红移。当70%<fw≤90%时,形成紧密聚集的颗粒,随着荧光强度的增强,同时,在疏水条件下,氢键合力减弱并且发射波长伴随着蓝移。
在紫外灯照射(365nm)下不同含水量(fw分别为0、10%、20%、30%、40%、50%、60%、70%、80%、90%)的PB-1、PB-2和PB-3的荧光表征中,溶液的荧光强度随着含水量的增加先降低后增加,并且发光颜色发生显着的变化,从黄绿色发射到橙色或红色发射。
PB-1、PB-2和PB-3的光稳定性如图4所示,在连续UV照射下,PB-3荧光在30分钟内没有变化,而PB-1和PB-2的荧光逐渐消失,因此PB-3(fw=90%)具有优异的稳定性并可用于爆炸物检测。PB-1和PB-2快速光化学不稳定的原因可能是在聚集状态下,PB-1和PB-2的溶解度随时间降低,并且化合物开始沉淀并漂浮在比色皿上方的溶液中。因此(THF:H2O(体积比1:9))溶液中的发光分子总数减少,最终导致发光强度降低。
PB-1、PB-2和PB-3对硝基芳香族炸药的检测表征。
在2,4-二硝基甲苯(DNT)、2,4,6-三硝基甲苯(TNT)和2,4,6-三硝基苯酚(PA)存在下对PB-3纳米聚集体进行荧光猝灭的表征如图5所示。在三种硝基芳香炸药中,PA显示出90%的荧光猝灭效率((1-I/I0)×100%)。图5中,PA,TNT或DNT逐渐加入到PB-3(fw=90%)溶液中,荧光逐渐减弱,但可以看到PA的滴落导致最快的荧光减少。记录了不同浓度(0-1.6g/L)猝灭的荧光强度,得到Stern-Volmer曲线。发现低浓度的硝基芳族化合物(0-0.8g/L)的猝灭曲线是线性的。如图6所示,通过拟合数据获得各种硝基芳族化合物的猝灭常数(KSV)。从图6中可以看出,当硝基芳烃的浓度范围(0~0.8g/L),PA、TNT和DNT的KSV值分别为4.1×104,1.4×104,2.0×103M-1。可以看出PA的KSV远高于TNT和DNT。对于PA,THF-H2O混合物(1:9v/v)中PB-3的检出限(LOD)为2.4μM(通过等式LOD=3S/Ksv获得,S为空白样本15个测量值的标准差)。
室温下硝基芳族化合物对(0.8×10-2g/L)PB-3在THF-H2O混合物(1:9v/v)中各种干扰物的猝灭效率如图7所示,可以看出,室温下PB-3对PA的检测效果最好。
为检测不同水体(黄河水、自来水、加尖的池塘水)对荧光猝灭是否有影响,因此,在含等量(0.8×10-2g/L)的PA存在下,检测PB-3(fw=90%)聚集体的荧光猝灭,结果如图8所示,可以看出,不同水环境下的荧光猝灭果是相同的,因此可以得出不同来源的水对荧光猝灭行为没有影响的结论。
不同的金属离子存在下对PA进行荧光检测,如图9所示,金属离子的存在并不影响PA的检测。
为了测试固相传感器设备是否可以用于实际爆炸物检测。本公开将Whatman滤纸浸泡在PB-3(fw=90%)(10-4M)溶液中,然后在空气气流中干燥,制备了试纸。首先是蒸汽模式测试。将荧光试纸放在含有固体PA的玻璃小瓶上5分钟。将测试条的圆形区域暴露于PA蒸汽。可以看出,聚集体的荧光明显在暴露区域中被猝灭,如图10A所示。然后,对溶液状态进行测试,将测试纸浸入纯THF(参考)和含有PA的THF(10-4M)溶液中。如图10B所示,试样的荧光在浸入PA/THF中时完全淬灭(图10B.e),相反,在浸入纯THF的参比纸中没有明显的荧光猝灭现象(图10B.d),实验结果表明,PB-3可用于制备检测硝基芳香族炸药的敏感传感器。
与PB-1和-2相比,PB-3在高水分THF/H2O=1:9(fw=90%)时显示出比较好的稳定性,因此进一步用于高灵敏度检测作为硝基芳族分析物的原型PA。基于PB-3的固相纸试验在蒸汽和溶液中显示出对PA的很好敏感性。

7.如权利要求2所述的应用,其特征是,1,3-二苯基巴比妥酸的制备方法为:将丙二酰卤与N,N-二苯基脲进行取代反应获得。

9.一种检测硝基芳香族爆炸物的方法,其特征是,将待测样品置于含有权利要求1所述的巴比妥酸衍生物的溶液,然后紫外-可见光光谱或荧光光谱进行检测。
10.如权利要求9所述的方法,其特征是,采用荧光光谱时,检测波长为620~650nm。
11.如权利要求9所述的方法,其特征是,含有巴比妥酸衍生物的溶液中溶剂为四氢呋喃与水的混合物,四氢呋喃与水的体积比为1:8.9~9.1。
12.一种检测传感器,其特征是,以权利要求1所述的巴比妥酸衍生物作为传感材料。
