背景技术
光门控、内向整流阳离子通道,通道视紫红质-1(ChR1)和通道视紫红质2(ChR2)已成为体外和体内神经元靶向光激活的优选工具1-5。尽管野生型(WT)ChR2可用于光诱导去极化,但目前正在寻找具有更快动力学和增加光敏性的ChR2突变体,以用于未来潜在的临床应用(WO 03/084994和6-8)。

动力学是一个主要问题,因为光敏感度是通过通道的开放时间来调节的。这是由于其他通道参数(如单通道电导,开放概率,量子效率)的不变性。换句话说,具有长开放时间的通道在低光强度下达到最大活动,而短活性通道需要更多光以相对于光饱和度达到饱和。虽然“快速”通道需要更多的光来激活,但由于不同神经元细胞的高频率放电,高速对于神经生物学中的许多应用是必不可少的。这是有根据的,例如对于大脑中间神经元的听觉系统中的神经节细胞,其达到高达1000Hz的放电速率。因此,仍然需要结合稳健表达和更快响应动力学的突变型光诱导离子通道。
发明内容
发明人通过修饰七个跨膜螺旋基序的螺旋6对Chrimson进行了系统研究。光激活期间的螺旋6运动是微生物型视紫红质的常见特征(15,17).因此,修改螺旋6以改变通道的关闭时间。可以证明,Chrimson中螺旋6中位置261,267和268的突变加速了通道的关闭时间(关闭动力学),并且突变的组合导致了关闭动力学(off-kinetics)的进一步加速。
本公开描述了通过螺旋6中的具体定点突变修饰Chrimson的速度的一般方法。这些新变体的使用将提供对神经元的光刺激,直至其达到800至1000Hz的极限。
在NG108-15细胞(神经母细胞瘤细胞),HEK293细胞和来自小鼠脑的海马细胞中测试了针对增加的速度的实验验证。

优选实施方案的详细描述

发明人的目的是鉴定Chrimson的第六跨膜结构域内的残基,其突变能够加速关闭动力学。
实施例1-在NG108-15细胞中的Chrimson突变体的光电流
以全细胞模式通过膜片钳测量在-60mV的钳夹电位研究了瞬时表达Chrimson-YFP和Chrimson-YFP突变体的NGl08-15细胞。浴溶液含有140mM NaCl,2mM CaCl2,2mM MgCl2,10mM HEPES,pH 7.4,移液管溶液含有110mM NaCl,2mM MgCl2,10mM EGTA,10mM HEPES,pH7.4。响应于3ms光脉冲(强度23mW/mm2,波长594nm)测量光电流。τ关闭值是由照明停止后的电流与单指数函数的拟合确定的。电流密度(J-60mV)是通过将响应于500ms光脉冲(强度23mW/mm2,波长594nm)的稳定电流除以细胞电容来确定的。
结果显示在图1和2中,并总结在下表1中。

从上述数据可以看出,对应于位置267,268和261的位置的突变能够加速Chrimson的关闭动力学(τ关闭)。这些突变的组合导致了Chrimson的关闭动力学(τ关闭)的进一步加速。突变可以有利地彼此组合或与已知的K176R突变组合。
实施例2-Chrimson突变体在NG108-15细胞中的作用光谱
以全细胞模式通过膜片钳测量在-60mV的钳夹电位研究了Chrimson和Chrimson突变体的作用光谱。浴溶液含有140mM NaCl,2mM CaCl2,2mM MgCl2,10mM HEPES,pH 7.4,移液管溶液含有110mM NaCl,2mM MgCl2,10mM EGTA,10mM HEPES,pH 7.4。使用Opolette 355可调谐激光系统(OPTOPRIM)生成脉冲长度为7ns的光脉冲。对于作用光谱的记录,将不同波长的脉冲能量设定为对应于Chrimson wt的1018光子/m2和Chrimson突变体的1019光子/m2的相等光子计数的值。
结果如图3所示。Y268F突变产生了作用光谱的蓝移(γ最大=580nm)。值得注意的是,与野生型Chrimson的作用光谱(γ最大=590nm)相比,未携带Y268F突变的测试Chrimson突变体的作用光谱没有显著转移。
实施例3-Chrimson突变体在大鼠海马神经元中显示出增加的尖峰频率
为了测试Chrimson突变体对神经元应用的适合性,构建体在培养的大鼠海马神经元中表达。
海马神经元培养。从出生后的P1 Sprague-Dawley大鼠()分离出海马,并用木瓜蛋白酶(20U ml-1)在37℃下处理20分钟。用补充有10%胎牛血清的DMEM(Invitrogen/Gibco,高葡萄糖)洗涤海马,并在少量该溶液中研磨。将约75,000个细胞接种在24孔板中的聚-D-赖氨酸/层粘连蛋白涂覆的玻璃盖玻片上。3小时后,用培养基(含有2%B-27补充剂,2mMGlutamaxI的Neurobasal A)替换平板培养基。
以全细胞模式通过膜片钳实验在-70mV的钳夹电位研究了异源表达Chrimson-YFP,Chrimson-YFP Y261F/S267M和Chrimson-YFP K176R/Y261F/S267M的大鼠海马神经元。野生型Chrimson或相应的Chrimson突变体的异源表达通过在测量前14至21天用腺相关病毒转导完成。
简言之,在接种后4-9天,向每个孔中加入1×109基因组拷贝/ml(GC/ml)病毒。转导后5天表达变得可见。在培养物的寿命期间(约5周)未观察到神经毒性。对于本文所述的任何实验,没有将全反式视黄醛添加到培养基或记录培养基中。
腺相关病毒(AAV2/1)。rAAV2/1病毒由Botond Roska博士(FMI,Basel)实验室使用含有Chrimson-YFP野生型或Chrimson-YFP突变体的具有人突触蛋白启动子(16)的pAAV2载体制备。病毒滴度名义上是1×1012-1×1013GC/ml
来自海马神经元的电生理记录。对于培养的海马神经元中的全细胞记录,将电阻为3-8Mω的钳移液管填充129mM葡萄糖酸钾,10mM HEPES,10mM Kcl,4mM MgATP和0.3mMNa3GTP,滴定至pH 7.2。使用Tyrodes溶液作为细胞外溶液(125mM NaCl,2mM Kcl,2mMCaCl2,1mM MgCl2,30mM葡萄糖和25mM HEPES,滴定至pH7.4)。记录在兴奋性突触传递阻断剂1,2,3,4-四氢-6-硝基-2,3-二氧代-苯并[f]喹喔啉-7-磺酰胺(NBQX,10μM,Sigma)和D(-)-2-氨基-5-膦酰戊酸(AP-5,50μM,Sigma)存在下进行。为了测定τ关闭和J-70mV,还在1μM TTX存在下进行测量。
使用Axopatch 200B放大器(Axon Instruments,Union City,CA)扩大电生理信号,在10kHz过滤,用Axon Digidata 1322A(50Hz)数字化,并使用pClamp9软件(AxonInstruments)获取和分析。
动作电位由指定频率的40个光脉冲触发。光脉冲的脉冲宽度为3ms,波长λ=594nm,饱和强度11-30mW/mm2。
τ关闭值是由照明停止后的电流与单指数函数的拟合确定的。电流密度(J-70mV)是通过将响应于500ms光脉冲(饱和强度20-40mW/mm2波长594nm)的稳定电流除以细胞电容来确定的。为了确定以100%概率(J100)诱发动作电位所需的最低光强度,应用不同光强度的40个脉冲(λ=594nm,脉冲宽度=3ms,v=10Hz)。通过将光触发尖峰的数量除以光脉冲的总数来计算尖峰概率。
结果如图4所示。虽然表达野生型Chrimson的大鼠海马神经元不能在10Hz以上的频率下以100%的概率诱导动作电位(参见图4A),但是Chrimson突变体Y261F/S267M(未显示)和K176R/Y261F/S267M(参见图4B)能够在20Hz和40Hz下适当地诱导动作电位。此外,Chrimson突变体K176R/Y261F/S267M(未显示)和Y261F/S267M(参见图4C)甚至能够在高达80-100Hz的频率下适当地诱导动作电位。这些测量清楚地证明本公开的快速Chrimson突变体能够实现快速神经光刺激。
与野生型Chrimson相比,在大鼠海马神经元中异源表达的Chrimson突变体K176R/Y261F/S267M和Chrimson突变体Y261F/S267M的测量特征显示在下表2中。
表2:关闭动力学(τ关闭),电流密度(J-70mV)和以100%概率(J100)诱导动作电位所需的最低光强度。显示的是平均τ关闭(n=3-11),平均J-70mV(n=10-14),平均J100(n=7)和相应的标准偏差。
如表2所示,与野生型相比,Chrimson突变体K176R/Y261F/S267M和Chrimson突变体Y261F/S267M的关闭动力学(τ关闭)显著加速。因此,与野生型Chrimson相比,ChrimsonY261F/S267M和Chrimson K176R/Y261F/S267M的电流密度(J-70mV)仅略微降低。大鼠海马神经元中的强烈表达使得能够在低光强度下进行神经光刺激。这些数据证实了在NG108-15细胞中观察到的更快的动力学(表1)并解释了在高频下精确诱导动作电位的能力,如本文图4所示。
实施例4-听觉途径的光遗传学刺激
听觉神经的光遗传学刺激有望在通过诸如人工耳蜗和听觉脑干植入物之类的听觉假体进行声音编码方面具有较大优势。由于光可以方便地聚焦,因此光学听觉假体可以大大提高编码的频率分辨率。目前可用的光遗传学工具的一个局限性是相对于听觉信号处理速度的慢动力学。在此,应用先前在Hernandez等.J Clin Invest.2014;124(3):1114-29中描述的系统,使用出生后AAV介导的小鼠SGN转导测试了快速门控Chrimson-YFP Y261F/S267M用于耳蜗光遗传学的潜力。通过p3小鼠中的圆窗将AAV2/6-hSyn-Chrimson-YFPY261F/S267M注射到鼓阶中,并在注射后4-8周分析表达和功能。注射的小鼠在群体中没有显示任何明显的表型。使用注射和未注射的耳朵的冷冻切片免疫组织化学和YFP共焦成像和钙视网膜蛋白免疫荧光分析Chrimson-YFP Y261F/S267M表达和SGN密度(图5A,5B)。每只注射的耳朵显示Chrimson-YFP Y261F/S267M,在注射的耳中在所有耳蜗转弯中,接近90%的SGN被转导(相比之下未注射的耳朵中小于5%)并且SGN密度美国有显著改变(图1B)。转导率远远高于我们之前使用AAV2/6-hSyn-CatCh-YFP的经子宫注射(Hernandez等人,2014)进行的研究所达到的转导率,并且与此不同,独立于频率排布位置。进行后鼓室切开术,并通过圆窗插入50μm光纤,以将595nm连续波激光的光投射到基底转弯的SGN上。我们可以很容易地引出光学听觉脑干响应(oABR,图6A),其振幅和波形与声学ABR(aABR)相似。随着光强度的增加,oABR振幅增加,oABR潜伏期缩短(图6B,6E)。刺激低至0.5mW(图6C,刺激持续时间:1ms,刺激频率:10s-1)并且短至80μs(图6D,刺激强度:11mW,刺激频率:10s-1)足以驱动oABR。oABR的振幅和波形在不同动物之间变化(图6A),但通常随着超过一个数量级的光强度变化而变化(图6C,输出动态范围>20dB)。当减少刺激持续时间(图6D)或提高刺激频率时,oABR振幅减小,但发现高达200Hz的oABR(图6F)。值得注意的是,oABR反映在5-7毫秒内延伸的群体响应,因此假设响应可能已被后续刺激之间的相互作用减弱。结论是Chrimson-YFP Y261F/S267M介导的耳蜗光遗传学可以在175Hz或更高驱动SGN,这在生理声音驱动的SGN激发频率率范围内(Liberman,M.C.,J.Acoust.Soc.Am.63,442-455(1978);Winter,等.,Hear.Res.45,191-202(1990))。总之,已经有效地建立了SGN的出生后转导,并且在低光强度和短暂刺激下实现了SGN激发。预计Chrimson-YFP Y261F/S267M(可提供足够的时间保真度,并且在更深入的光线穿透,减少组织中的散射,并降低光毒性风险方面优于蓝色通道视紫红质)将成为光遗传学听觉恢复和听觉研究的宝贵工具。

实施例5-用于恢复视力的光遗传学方法
Macé等.Mol Ther.2015;23(1):7-16是由发明人中一些撰写的早期出版物,其描述了由腺相关病毒(AAV)基因疗法介导的视网膜神经元的光遗传学再激活。大多数遗传性视网膜营养不良显示出进行性感光细胞变性,导致严重的视力损害。腺相关病毒(AAV)基因疗法介导的视网膜神经元的光遗传学再激活具有恢复视力的潜力,无论患者特异性突变如何。临床可译性的挑战是尽可能恢复视力接近自然视力,同时针对脆弱的变性视网膜使用手术安全的递送途径。为了保持内部视网膜的视觉处理,靶向ON双极细胞,其于疾病的晚期阶段仍然存在。对于安全的基因递送,使用最近工程化的AAV变体,其可以在注射到眼睛的容易接近的玻璃体液中之后转导双极细胞。结果表明,在ON双极细胞特异性启动子下的编码通道视紫红质的AAV介导玻璃体内施用后限于ON-双极细胞的长期基因递送。ON双极细胞中的通道视紫红质表达导致视网膜和皮质水平的ON和OFF响应的恢复。此外,在治疗的盲小鼠中光诱导的运动行为恢复。
例如由Macé等描述将本文鉴定的螺旋6基序引入构建体中代表常规实践。或者,可以通过本公开的光诱导离子通道的编码序列简单地替换结构中的通道视紫红质的编码序列。本发明的新的光诱导离子通道插入用于激活ON双极细胞的盒以及用于视网膜中的神经节细胞的盒中。
