Abstract

Description
技术领域
本发明涉及一种用于补充或替换硬组织和/或软组织的生物可吸收植入物,例如软骨、骨骼或任何其他组织。生物可吸收的材料包括至少一种增强纤维。所述至少一种增强纤维优选为增强纤维束/增强纤维结构或纤维结构体的形式。所述至少一种增强纤维由第一材料组分制成,并在与第二材料组分混合后嵌入基质中。第一材料组分的材料包含由丝、壳聚糖、胶原、聚己内酯、聚DL-丙交酯聚、聚乳酸乙醇酸、聚乙交酯、聚氨酯和聚丙烯组成的一组元素中的至少一种元素或至少一种元素组合。
需要再一次强调的是,本文中的纤维可理解为纤维束和纤维结构以及纤维结构体。
同样地,本发明还涉及一种生物可吸收材料的制备方法。
背景技术
一方面,现有技术中生物可吸收的植入物是已知的,其仅由一种原料制成。另一方面,为了改善降解可吸收材料的生物反应,基于多种材料的技术在植入物领域中占据了优势。然而,生物反应改进的同时则面临机械强度的损失,这在某些情况下可能导致植入物在机械载荷下过早失效。例如,这种用于再生缺陷的植入物通常没有足够的机械稳定性,用以在手术插入后直接完全替代待更换的组织。
德国专利申请10 2010 034 471 A1中已知一种通用的植入物。它公开了一种包含细丝的植入物,所述细丝包括细长的,优选为编织的细丝体和至少部分围绕着细丝体的涂层。该专利申请的细丝由聚乙烯和/或聚丙烯制成,而涂层由可吸收材料以及在必要时所需的添加剂组成。
欧洲专利文献1 537 883 B1也可知一种植入物。其针对修复组织损伤和缺陷,并且具有包含增强材料的生物相容性结构。
在欧洲专利申请2 081 020 A2中,公开了一种组织植入物,其由生物可吸收的组分和/或由非生物可吸收的组分制成。作为不可吸收的组分,可考虑例如天然丝或合成丝。
美国专利申请2004/0054372 A1针对生物可降解复合材料,可用作植入物。所述植入物包括生物可降解的纤维增强复合材料、基质以及纤维。
上述现有技术的一个缺点是,当植入物的几何形状适配个体特征时,增加了所用材料的量。除了由于材料使用增加而导致的成本效益降低之外,导致生物过程恶化,这对临床结果产生负面影响。因为,众所周知,浓度相关的负面临床反应可能是由于植入物中需降解的物质的量增加所致。
同样,在现有技术中,待连接的复合材料主要通过所谓的预浸料技术(缩写为“预浸渍纤维”)制备。它是用反应树脂预浸渍的纺织纤维基质半成品,其在温度和压力下硬化以制成植入物。这种生产方法成本高且耗时。
因此,本发明的目标是消除或至少减轻现有技术中的已知缺点,特别是提供一种植入物,该植入物能使用尽可能少的材料,实现快速的生物可吸收特性以及经济的制备。
发明内容
依据本发明,其发明目标的实现是通过以下事实来确保的:第二材料组分在与至少一种增强纤维/纤维束/纤维结构或纤维结构体混合时以颗粒或粉末形式存在。因此,第一材料组分以纤维/纤维束的形式或以纤维结构或纤维结构体的形式存在,且第二材料组分例如以粉末或颗粒的形式存在,其二者被以不同的量和不同的材料组成相互混合。这种方式确保了所要实现的特性的高度灵活性,因为其可以在确保高可靠性的同时,个性化地设计至少一种纤维/纤维束/纤维结构或纤维结构体和粉末或颗粒的混合。
有利的实施方式是从属权利要求的主题,并且将在下面做详细解释说明。
优选地,作为另外一种选择或补充的选择,第二材料组分也可以液体形式存在。至少一种纤维可以以高度灵活的方式置于液体第二材料组分中。此外,液态第二材料组分能够实现不同的密度和强度。
有利的是,当基质由第二材料组分和至少一种其他材料组分制成。通过添加另外的组分可针对性地影响基质的特征,例如关于强度或关于降解动力学或生物适应性的特征。
一旦第二材料组分包含基于陶瓷磷酸盐的组分,就可以确保承担支架功能的基质具有高强度。这增加了植入物的可靠性及其生物相容性。
在一个优选的实施方式中,纤维的整体包含由丝,壳聚糖,胶原,聚己内酯,聚DL-丙交酯聚,聚乳酸乙醇酸,聚乙交酯,聚氨酯和聚丙烯组成的一组元素中的至少两种元素。这里可能的是,一种纤维由多种材料制成且单个纤维包含相同的材料,但彼此之间包含不同的材料。来自上述一组元素的每个元素提供其从材料科学中已知的各自的优点。因此,选择要选择哪个元素取决于各自的通用条件。在这种情况下,强度、生物相容性、成本效益以及纤维组分与基质的体积和质量比均被列为影响因素。
另一个优点是,当纤维具有至少一个凸起和/或凹陷时,其能够增加基质和纤维之间相互作用的表面,这使得纤维可以坚固地固定在基质内。以这种方式,即使在受到出乎意料高的外力和内力冲击的情况下,植入物也能承受负荷。
在一个有利的实施方式中,多个固体颗粒排列在基质中,使其可控制吸收时间,其固体颗粒占测量的生物可吸收材料/植入物质量的质量百分比为5%至25%。基质中固体颗粒的质量百分比占比越高,其对吸收时间的影响越大。在这种情况下,必须在符合强度通用条件并考虑生物学特性的条件下对可以使用多少质量百分比进行评估。由于第二种材料组分在混合时呈粉末和/或颗粒形式,因此在生产植入物时无需引入很高的额外开销,即可实现固体颗粒的混入。
即使通过创成式、减法和3D成形制造方法可以实现各个材料组分的混合,但是精确可靠地制备生物可吸收的材料更有利。粉末/颗粒形式和线形有利于创成式的分层处理过程。这种创成式制造省去了其他快速原型制作工艺所必需的支撑结构,其对所使用的材料的量具有积极影响。
类似地,这也适用于进一步的3D成形方法,例如,压缩成型。作为混合各个组分的另外的一种选择,还有LCM(“基于平版印刷的陶瓷制造”)或静电纺丝法。
此外,当进行混合时第二材料组分颗粒状或粉末状的的微粒在预定时间窗之后形成一个整体的、合并的形状,其优点是显而易见的。以这种方式,省去了在相应的后处理时对每个第二材料组分的粉末形式进行处理,这对植入物内的动力磨损具有有利影响,因为不再存在材料内部的相界了。
在另一个优选的实施方式中,在进行混合时第二材料组分颗粒状或粉末状的的微粒含有由镁、钙、羟基磷灰石、α-和/或β-磷酸三钙和石灰组成的一组元素中的至少一种元素,以便有针对性地影响生物可吸收植入物的降解行为。以这种方式,植入物的生物吸收是根据患者的健康状况可变的,以确保快速且没有任何并发症的愈合。

根据本发明,纤维比例占植入物的质量百分比的为5%至95%。特别优选的是质量百分比为5%、15%、20%、30%至55%或60%至95%的实施例。

优选地,全部纤维整体组成一个纤维结构体,该纤维结构体例如是呈网状且彼此相向的。所述结构体可以在平面中设计或者也可以在三维上设计。纤维结构体从各个部分的纤维分别得到一个方向取向,各个部分的纤维的取向以网状方式相互交织。
优选地,纤维具有一个结构化的表面拓扑以增大单个纤维和基质之间的相互作用表面,从而增强相互作用,这增加了本发明的植入物的机械稳定性/坚固性/强度。
上述作为纤维材料的壳聚糖除强化功能外,还表现出优异的抗菌效果。
通过将例如镁、镁-钙-锌(MgCaZn)、铁、钡、锶、钙、羟基磷灰石、α-和/或β-磷酸三钙和石灰的微粒与相应的的纤维混合,以这种方式实现了对一个时间域上/时间上的发展进度的控制,使得可吸收材料的降解行为可以加速,或也可以根据需要减速。
根据本发明,同样可以进行表面改性以改善植入物的抗菌效果并优化向内生长行为。因此,优选地,改变组分镁、聚乙烯、聚丙烯、聚醚醚酮、羟基磷灰石、α-和/或β-磷酸三钙和石灰,以使其与纤维相互作用,以产生所需的植入物的行为。

具体实施方式
图1示出了包括至少一种增强纤维2的用于补充或替换硬组织的生物可吸收植入物。根据本发明,所述纤维2又包括由丝、壳聚糖、胶原、聚己内酯、聚DL-丙交酯聚、聚乳酸乙醇酸、聚乙交酯、聚氨酯和聚丙烯组成的一组元素中的至少一种元素。将这种纤维2嵌入基质材料3中,用以与后者基质材料3一起构成这样一种植入物1,其沿纵向和沿横向均具有高强度值。基质材料3在与纤维2混合时是颗粒状或粉末状或液体的。这在图1中的相界4处可以看出。
在图2中示出了一种状态,在这种状态下,基质材料3原始的颗粒状组合物被完全去除,使得植入物1仅包含一个均匀的基质3,增强纤维2排列在该基质中。在这种状态下,植入物1优选地适于插入患者体内。
在这里,形状的设计可以是灵活的。图1和图2仅示出了材料成分,而母体植入物1将被设计成使其与待支撑硬组织互补。
植入物1的生物吸收由于在图2所示的状态下不再出现相界4而提高。
Claims (10)
1.一种用于补充或替换硬组织和/或软组织的生物可吸收植入物(1),包括至少一种增强纤维(2),所述增强纤维(2)由第一材料组分制成并且在与第二材料组分混合后嵌入基质(3)中,其中第一材料组分的材料包含由丝、壳聚糖、胶原、聚己内酯、聚DL-丙交酯聚、聚乳酸乙醇酸、聚乙交酯、聚氨酯和聚丙烯组成的一组元素中的至少一种元素,其特征在于,第二材料组分在与至少一种增强纤维(2)混合的时间点以颗粒状或粉末形式存在。
2.根据权利要求1所述的生物可吸收植入物(1),其特征在于,所述基质(3)由所述第二材料组分和至少一种其他材料组分制成。
3.根据权利要求1或2中任一项所述的生物可吸收植入物(1),其特征在于,所述第二材料组分包含陶瓷磷酸盐基组分。
4.根据权利要求1至3中任一项所述的生物可吸收植入物(1),其特征在于,所述纤维(2)的整体包含由丝、壳聚糖、胶原、聚己内酯、聚DL-丙交酯聚、聚乳酸乙醇酸、聚乙交酯、聚氨酯和聚丙烯组成的一组元素中的的至少两种元素。
5.根据权利要求1至4中任一项所述的生物可吸收植入物(1),其特征在于,所述纤维(2)包括至少一个凸起和/或凹陷,以增加所述基质(3)与所述纤维(2)之间的相互作用表面。
6.根据权利要求1至5中任一项所述的生物可吸收植入物(1),其特征在于,多个固体颗粒排列在所述基质(3)中,使其可控制吸收时间,其固体颗粒占测量的生物可吸收材植入物质量的质量百分比为5%至25%。
7.根据权利要求1至6中任一项所述的生物可吸收植入物(1),其特征在于,通过创成式、减法和3D成形制造方法实现各个材料组分的混合。
8.根据权利要求1至7中任一项所述的生物可吸收植入物(1),其特征在于,在混合时颗粒状或粉末状的第二材料组分的微粒在预定时间窗之后形成一个整体的、合并的形状。
9.根据权利要求1至8中任一项所述的生物可吸收植入物(1),其特征在于,在混合时颗粒状或粉末状的第二材料组分的微粒包含由镁、钙、羟基磷灰石、α-和/或β-磷酸三钙和石灰组成的一组元素中的至少一种元素,以便有针对性地影响生物可吸收植入物(1)的降解行为。
10.根据前述权利要求中任一项所述的用于制备生物可吸收植入物(1)的方法,包括以下步骤:
-提供由第一材料组分制成的至少一种纤维(2);
-提供颗粒状或粉末状的第二材料组分微粒或第二材料组分的液态;
-将第一材料组分与第二材料组分混合以获得生物可吸收的植入物(1);
-对生物可吸收植入物(1)的形状和/或表面纹理进行后处理,以使其被设计成与待替换的硬组织和/或软组织互补。
