Abstract

本发明公开了一种鉴别江枳壳的特征图谱法,包含(1)特征图谱建立(2)供试液的制备(3)高效液相色谱法检测(4)特征峰鉴别;通过筛选出了具有共性的8个特征峰作为标准HPLC特征峰,结合以3号色谱峰为参照,计算各峰的保留时间和峰面积比值建立成标准图谱;将供试样的HPLC特征图与标准图谱进行比较,即可快速鉴别江枳壳;当八个特征色谱峰均出现,而且其保留时间和峰面积比值均符合标准图谱规定,方可认为其为道地江枳壳,否则,缺任一,均为非江枳壳;鉴定方法简单,快速准确。

一种鉴别江枳壳的特征图谱法
技术领域
本发明属于中药技术领域,涉及一种江西道地药材‒江枳壳的鉴别方法。
背景技术
枳壳为芸香科植物酸橙(Citrus aurantium L.)及其栽培变种的干燥未成熟果实,味苦、辛、酸,性寒,入肺、脾、大肠经。枳壳为临床上常用的理气药,具有理气宽中、行滞消胀的功效,能破气、行痰、消积,主要用于治疗胸胁气滞、胀满疼痛、食积不化、痰饮内停、胃下垂、脱肛、子宫脱垂、经期延长、高血脂症、椎间盘退变类疾病等。枳壳主要分布在江西、四川、浙江以及湖南等地。根据产地的不同,枳壳分为江枳壳、川枳壳、湘枳壳等,而传统中药理论认为江枳壳为道地药材。江西道地枳壳被称为“江枳壳”,皮青、肉厚、色白、质坚、香味纯正、菊花芯为其主要特色,是近1700年长期栽培地理环境、立地环境、品种及种植技术的综合体现。
江枳壳中又以商州、清江地区的枳壳最为著名,即江西省新干县、樟树一带的枳壳。
随着枳壳人工种植技术的进步,四川、浙江以及湖南等地枳壳种植面积逐年增加,尤其湖南省枳壳种植面积增长迅速。由于地域及品种、栽培技术的综合差别,枳壳的品质差异极大,对江枳壳的市场冲击尤其显著。
2015版《中国药典》,对枳壳中的柚皮苷和新橙皮苷含量进行了限量检查。近年来,研究发现江枳壳相对于非道地产区的川枳壳、湘枳壳、浙枳壳等,在柚皮苷和新橙皮苷含量上并未体现优势,对于柚皮苷含量甚至低于湘枳壳。现行标准下,导致药材市场上以假乱真,充当江枳壳的药材频现。
基于此,开发一种能够鉴别道地江枳壳的方法具有必要性。
发明内容
针对中国药典标准,不能准确识别道地江枳壳的现状,本发明开发了一种准确鉴定江枳壳的方法。
为实现上述发明目的,本发明通过下述技术方案来实现:
1、供试液制备
将药材经粉碎后过60目筛,取0.1g,精密称定,置于具塞锥形瓶中,分别精密加入 pH为3-5的50-80%甲醇水溶液5-15 mL,加热至60-90℃,搅拌提取3-5小时,连续提取1-3次,过滤,转移至25mL容量瓶中,用提取溶剂定容至刻度,摇匀,取提取液2mL过0.22μm微孔滤膜,即得供试液;
2、高效液相色谱法(HPLC)法检测
将供试液注入高效液相色谱仪中,进行测定;测定条件为C18色谱柱为(4.6 mm×250mm,5μm)或(4.6 mm×150 mm,5μm);检测波长为254 nm‒285nm;流速为0.5 mL/min‒1.0 mL/min;流动相中有机相A为乙腈或甲醇,水相B为0.1%-1%磷酸水溶液,含乙腈或甲醇5%‒15%;
洗脱程序如下:
3、特征峰鉴别
以3号色谱峰为参照,其峰面积的相对比值关系,如表2所示;
在高效液相色谱图中,要求至少出现8个特征峰,而且其保留时间和峰面积比值均符合表2,方可认为其为道地江枳壳,否则,缺任一,均为非江枳壳。
本发明具有如下优点:
1.本发明供试液制备工艺简单,便于江枳壳的快速鉴别;
2.本发明通过筛选出具有共性的8个特征峰,结合各峰出现的时间及峰面积的相对比值关系建立成了江枳壳的标准图谱,使得江枳壳的鉴别准确度高,满足市场对江枳壳鉴别的需要;
3.本发明根据江枳壳中黄酮苷类药效成分,设计供试液制备工艺,建立HPLC特征图谱,专属性强,准确性高。

具体实施方式
实施例1:
(1)取江枳壳药材粉末(过60目筛)0.1g,精密称定,置于具塞锥形瓶中,分别精密加入pH 为5的70%甲醇水溶液8 mL,加热至60℃,搅拌提取3小时,连续提取3次,过滤,转移至25mL容量瓶中,用提取溶剂定容至刻度,摇匀,取提取液2mL过0.22μm微孔滤膜,即得供试液;
(2)高效液相色谱法(HPLC)法检测
C18色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈(A)-0.1%磷酸水(含10%乙腈)(B);检测波长为280 nm;流速为1.0 mL/min;洗脱程序如下:
(3)结果分析
图2为HPLC检测图谱,从图2可以得知,具有标准图谱中的8个特征色谱峰,依次将其列举如表4;
根据表4,可以见得特征峰出峰时间能够与标准图谱对齐,另外,特征峰的峰面积比值在标准图谱的规定范围之内,该结果提示该鉴定药材为道地江枳壳。
实施例2:
(1)取浙枳壳药材粉末(过60目筛)0.1g,精密称定,置于具塞锥形瓶中,分别精密加入pH 为5的70%甲醇水溶液8 mL,加热至60℃,搅拌提取3小时,连续提取3次,过滤,转移至25mL容量瓶中,用提取溶剂定容至刻度,摇匀,取提取液2mL过0.22μm微孔滤膜,即得供试液;
(2)高效液相色谱法(HPLC)法检测
C18色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈(A)-0.1%磷酸水(含10%乙腈)(B);检测波长为280 nm;流速为1.0 mL/min。洗脱程序如下:
(3)结果分析
图3为HPLC检测图谱,从图3可以得知,具有标准图谱中的8个特征色谱峰,依次将其列举如表6;
根据表6,可以见得特征峰出峰时间基本能够与标准图谱对齐。但是,特征峰的峰面积比值除 “峰2”和“峰4”在标准图谱的规定范围之内;其他特征峰均在标准图谱的规定范围之外,而且“峰6”未检测出;因此,该结果提示该鉴定药材不是道地江枳壳。

Claims (5)

1.一种鉴别江枳壳的特征图谱法,通过以特征图谱中关键色谱峰及其保留时间和峰面积相对比值为依据进行鉴别,其特征在于:包含(1)标准图谱建立;(2)供试液的制备;(3)高效液相色谱法检测;(4)特征峰鉴别。
2.根据权利要求1所述的一种鉴别江枳壳的特征图谱法,其特征在于:
步骤(1)中所述的标准图谱建立为:在高效液相色谱图中,至少出现8个特征峰,且以3号色谱峰为参照,其峰面积的相对比值关系如下:

步骤(3)高效液相色谱法检测:将供试液注入高效液相色谱仪中,进行测定;测定条件为C18色谱柱为(4.6 mm×250 mm,5μm)或(4.6 mm×150 mm,5μm);检测波长为254 nm‒285nm;流速为0.5 mL/min‒1.0mL/min;流动相中有机相A为乙腈或甲醇,水相B为0.1%-1%磷酸水溶液,含乙腈或甲醇5%‒15%;

步骤(4)特征峰鉴别:要求八个特征色谱峰均出现,而且其保留时间和峰面积比值均符合标准图谱的规定,方可认为其为道地江枳壳,否则,缺任一,均为非江枳壳。
