【技术领域】
本发明涉及豆类发酵制品的深加工领域,尤其是一种具有溶栓作用的植物蛋白固体饮料及其制备方法。
【背景技术】
黑豆为豆科植物大豆(学名:Glycinemax(L.)merr)的黑色种子。黑豆蛋白质含量36%,易于消化,对满足人体对蛋白质的需要具有重要意义;脂肪含量16%,主要含不饱和脂肪酸,吸收率高达95%,除满足人体对脂肪的需要外,还有降低血液中胆固醇的作用;黑豆含有丰富的维生素、蛋黄素、黑色素及卵磷脂等物质,其中B族维生素和维生素E含量很高,具有营养保健作用,黑豆中还含有丰富的微量元素,对保持机体功能完整,延缓机体衰老、降低血液粘度、满足大脑对微量物质需求都是必不可少的。
山药,人类自古食用,是人类食用最早的植物之一。作为高营养食品含有大量蛋白质、糖类、B族维生素、维生素C、维生素E、葡萄糖、粗蛋白氨基酸及碘、钙、铁、磷等人体不可缺少的无机盐和微量元素等营养成分。现代科学分析,山药的最大特点是含有大量的黏蛋白。黏蛋白是一种多糖蛋白质的混合物,对人体具有特殊的保健作用,能防止脂肪沉积在心血管上,保持血管弹性,阻止动脉粥样硬化过早发生。
茯苓,为多孔菌科真菌茯苓Poria cocos(Schw.)Wolf的干燥菌核,别名:茯菟、茯灵、茯蕶、伏苓、伏菟、松腴、绛晨伏胎、云苓、茯兔、松薯、松木薯、松苓;具有利水渗湿、益脾和胃、宁心安神之功用,能增强机体免疫功能,其中的茯苓多糖还具有明显的抗肿瘤及保肝脏作用。
人参(Panax ginseng C.A.Mey),为多年生草本植物,别称黄参、地精、神草、百草之王;人参适用于调整血压、恢复心脏功能、神经衰弱及身体虚弱等症,也有祛痰、健胃、利尿、兴奋等功效;人参对于高血压病、心肌营养不良、冠状动脉硬化、心绞痛等,都有一定时治疗作用,可以减轻各种症状。特别是人参皂甙Rb2能改善血脂,降低血中胆固醇、甘油三酯、升高血清高密度脂蛋白胆固醇,降低动脉硬化指数,对于高脂血症、血栓症和动脉硬化有治疗价值。
蛹虫草为子囊菌门,肉座目,麦角菌科、虫草属的模式种。学名为Cordycepsmilitaris,又名北蛹虫草,有地区亦称为踊虫草,一般把活体虫蛹培养的北虫草称为蛹虫草,两者实际上是同种真菌。主要有保肺益肾,止血化痰,调节身体机能,主治肺结核咳血,阳痿遗精,腰膝酸痛,美容护肤,神精衰弱,糖尿病,升白血球,防癌、抗癌,对化疗作用也很大。人工栽培的北虫草药用功效与野生虫草的功效相差无几,某些方面的医用表现甚至优于野生虫草。
陈皮,也叫橘皮,贵老,红皮,黄橘皮,广橘皮,新会皮,柑皮,广陈皮。分布于长江以南各地区,10至12月果实成熟时,摘下果实,剥取果皮,阴干或通风干燥;对胃肠道有温和的刺激作用,可促进消化液的分泌,排除肠管内积气。
豆类发酵饮品是以豆浆为主要原料经过乳酸菌发酵而成的类似酸奶产品。相比于酸牛奶,大豆或黑豆酸奶不含胆固醇,热量低,蛋白质含量丰富,钙含量高,是一种很好的酸牛奶替代品。但是由于大豆或黑豆具有豆腥味,味道不佳;现有技术中,大多数大豆或黑豆酸奶通过豆浆与牛奶混合,降低大豆或黑豆比例以降低豆腥味。但是,从本质上讲,牛奶在其中比例较高,从而形成的豆酸奶非纯植物性饮品;其次,由于采用乳制品商用菌种,添加乳糖等发酵基质,引入了乳糖,不适合乳糖不耐症患者。由于大豆或黑豆中本身具有多聚糖,因此,利用自身营养成分即可发酵,也可添加非乳糖类糖类物质促进发酵;此外,现今加工工艺生产的豆类发酵饮品还存在各种添加剂,存在着一定的安全隐患。
【发明内容】
为了解决现有豆类发酵饮品存在的味道不佳、添加剂过多等缺陷,本发明提供一种口感好,具有预防脑梗、心梗的作用,且无任何添加剂、安全可靠的具有溶栓作用的植物蛋白固体饮料及其制备方法。
为达到上述明目的,采用的技术方案如下:
一种具有溶栓作用的植物蛋白固体饮料,包括黑豆粉、大豆粉、谷物粉、益生元、黑纳豆粉和复合益生菌;
所述黑豆粉使用量占固体饮料总重量的40%~60%;
所述大豆粉使用量占固体饮料总重量的5%~10%;
所述谷物粉使用量占固体饮料总重量的20%~40%;
所述益生元使用量占固体饮料总重量的10%~20%;
所述黑纳豆粉在固体饮料中的活性单位100-400FU/g;
所述复合益生菌在固体饮料中的含量为5~20×108个/g。
优选地,该植物蛋白固体饮料还包括山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物中的一种或几种,其中,山药提取物使用量占固体饮料总重量的2%~6%,茯苓提取物使用量占固体饮料总重量的2%~6%,人参提取物使用量占固体饮料总重量的0.2%~1%,蛹虫草提取物使用量占固体饮料总重量的0.2%~1%,陈皮提取物使用量占固体饮料总重量的0.02%~1%。
优选地,所述人参提取物中选取的人参为5年以下、人工种植的用于食品中的人参。
优选地,所述益生元是低聚果糖、低聚木糖或低聚麦芽糖中的一种或几种。
优选地,所述复合益生菌为嗜酸乳杆菌、干酪乳杆菌、罗伊氏乳杆菌、副干酪乳杆菌、植物乳杆菌、鼠李糖乳杆菌、两歧双歧杆菌、乳双歧杆菌或短双歧杆菌中的一种或几种,复合益生菌的发酵终含量为5~20×108个/g
一种具有溶栓作用的植物蛋白固体饮料的制备方法,包括以下步骤:
1)、黑纳豆粉制备:
1.1)、菌种培养:
1.1.1)、菌种活化及斜面培养:
菌种活化培养基为马铃薯200g/L,胡萝卜200重量份,琼脂20重量份,蛋白胨1.2重量份,葡萄糖21重量份,磷酸二氢钾0.4重量份,硫酸镁0.18重量份,维生素B1少许,接种后菌种活化时间为7-8天;
1.1.2)、种子菌培养:种子菌培养基成分的重量份为:葡萄糖2重量份,黑豆蛋白胨1重量份,Na2HPO4 0.6重量份,NaH2PO4 0.1重量份,MgSO4 0.05重量份,CaCl2 0.02重量份,吐温1重量份;培养基pH调整范围为pH 7.5~8.0;
1.2)、黑纳豆制备:
黑豆经浸泡、去种皮、蒸煮、沥水,然后接种种子菌和发酵,制成黑纳豆;
1.3)、黑纳豆粉制取:
将发酵后黑纳豆物料匀浆,经离心过滤、浓缩、冷冻干燥,粉碎,制得黑纳豆粉;
2)、山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物的制备:
选取10Kg的山药,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得山药提取物;
选取10Kg的茯苓,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得茯苓提取物;
选取10Kg的人参,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得人参提取物;
选取10Kg的蛹虫草,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得蛹虫草提取物;
选取10Kg的陈皮,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得陈皮提取物;
3)、物料混合:
将黑豆粉、大豆粉、谷物粉、黑纳豆粉、山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物、益生元和复合益生菌按一定比例混合,最终在固体饮料中,纳豆激酶的含量活性单位100~400FU/g,复合益生菌的含量为5~20×108个/g。
进一步地,所述步骤1.2)中还包括:黑豆浸泡6小时,去种皮、蒸煮、沥水,然后接种种子菌;在温度为26~28℃、相对湿度为≤75%的条件下,发酵培养13~15天,制成黑纳豆。
进一步地,所述步骤2)中山药提取物、茯苓提取物、人参提取物、蛹虫草提取物或陈皮提取物的制备中,所有提取物蒸煮温度为90~100℃。
进一步地,所述步骤2)中山药提取物、茯苓提取物、人参提取物、蛹虫草提取物或陈皮提取物的制备中,所有提取物加热时的温度为170℃~190℃,受热时间5~7秒。
进一步地,所述步骤2)中山药提取物、茯苓提取物、人参提取物、蛹虫草提取物或陈皮提取物的制备中,所有提取物喷雾干燥时,喷雾压力为0.15pa,并控制出口温度为80~90℃。
本发明的优点:
(1)、制备的具有溶栓作用的植物蛋白固体饮料,在食品饮料行业,为天然纯植物性蛋白饮品,口感好、没有新鲜纳豆的不愉快味道,且无任何添加剂,既保留了大豆的营养成分,又增加了一定的功能成分,特别适合中老年人饮用,具有预防脑梗、心梗的作用,具有很好的经济效益和市场前景。
(2)、制备的具有溶栓作用的植物蛋白固体饮料,因使用了人参,所以不适用于孕妇、哺乳期妇女及14周岁以下儿童。
(3)、制备过程中,使用的发酵设备简单,易操作,通过添加纳豆粉、山药、茯苓、人参、蛹虫草、陈皮等药食同源提取物及活性益生菌,相比于单纯的豆类固体饮料,具备了溶栓、清肠功效,具有预防脑梗、心梗的作用,是一种很好的植物性活性乳酸菌固体饮料。

菌种活化培养基为马铃薯200g/L,胡萝卜200重量份,琼脂20重量份,蛋白胨1.2重量份,葡萄糖21重量份,磷酸二氢钾0.4重量份,硫酸镁0.18重量份,维生素B1少许,接种后菌种活化时间为7-8天;
1.1.2)、种子菌培养:种子菌培养基成分的重量份为:葡萄糖2重量份,黑豆蛋白胨1重量份,Na2HPO4 0.6重量份,NaH2PO4 0.1重量份,MgSO4 0.05重量份,CaCl2 0.02重量份,吐温1重量份;培养基pH调整范围为pH 7.5~8.0;
1.2)、黑纳豆制备:
黑豆浸泡6小时左右,去种皮、蒸煮、沥水,然后接种种子菌;在温度为26~28℃、相对湿度为≤75%的条件下,发酵培养13~15天,制成黑纳豆。
1.3)、黑纳豆粉制取:
将发酵后黑纳豆物料匀浆,经离心过滤、浓缩、冷冻干燥,粉碎,制得黑纳豆粉;
2)、山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物的制备:
选取10Kg的山药,用90Kg水蒸煮1-2小时,经打浆、过滤、加热、浓缩和喷雾干燥,制得山药提取物,其中,蒸煮时的温度为90℃~100℃;加热时的温度为170℃~190℃,受热时间5~7秒;喷雾压力为0.15pa,并控制出口温度为80~90℃;
选取10Kg的茯苓,用90Kg水蒸煮1-2小时,经打浆、过滤、加热、浓缩和喷雾干燥,制得山药提取物,其中,蒸煮时的温度为90℃~100℃;加热时的温度为170℃~190℃,受热时间5~7秒;喷雾压力为0.15pa,并控制出口温度为80~90℃;
选取10Kg的人参,用90Kg水蒸煮1-2小时,经打浆、过滤、加热、浓缩和喷雾干燥,制得山药提取物,其中,蒸煮时的温度为90℃~100℃;加热时的温度为170℃~190℃,受热时间5~7秒;喷雾压力为0.15pa,并控制出口温度为80~90℃;

经选取10Kg的陈皮,用90Kg水蒸煮1-2小时,经打浆、过滤、加热、浓缩和喷雾干燥,制得山药提取物,其中,蒸煮时的温度为90℃~100℃;加热时的温度为170℃~190℃,受热时间5~7秒;喷雾压力为0.15pa,并控制出口温度为80~90℃;
3)、物料混合:
将黑豆粉、大豆粉、谷物粉、黑纳豆粉、山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物、益生元和复合益生菌按一定比例混合,最终在固体饮料中,纳豆激酶的含量活性单位为100~400FU/g,复合益生菌的含量为5~20×108个/g。
所制备的植物蛋白固体饮料口感独特、酸甜可口,豆浆种富含大豆蛋白质、大豆肽、大豆异黄酮、大豆皂甙、大豆磷脂、大豆膳食纤维、大豆亚油酸、大豆亚麻酸、大豆卵磷脂和大豆维生素等活性物质,具有降低胆固醇、降血压、降血脂、抑制肿瘤、延缓衰老等作用;豆浆富含的大豆低聚糖不被人体消化吸收,进入人体肠道后促进双歧杆菌增殖,增强肠道功能和解毒作用。
而且,本饮品无任何添加剂,保留了大豆的营养成分,又增加了一定的功能成分,特别适合中老年人饮用,具有预防脑梗、心梗的作用。其中,添加纳豆粉、山药、茯苓、人参、蛹虫草、陈皮等药食同源提取物及活性益生菌,相比于单纯的豆类固体饮料,具备了溶栓、清肠功效,具有预防脑梗、心梗的作用,是一种很好的植物性活性乳酸菌固体饮料。
以下为固体饮料混合实施案例,详见下表:

以上为采用制备方法制取具有溶栓作用的植物蛋白固体饮料的三个实施例,在三个配方中,经试验和试用,有以下不同效果:
(1)、消费者试用:寻找150名有便秘问题的自愿试用人员,分成3组,每组50人,且每组人员年龄、性别基本一致。
A组,试用配方1,每天早餐时饮用1包;
B组,试用配方2,每天早餐时饮用1包;
C组,试用配方3,每天早餐时饮用1包;
不同时间见效的人数见下表

从结果看,三个配方均对改善便秘有显著效果,其中配方3的效果最快。
(2)、溶栓实验:取3块形状、大小基本一致的新鲜的鸡血,鸡血重量0.8g;将上述3个配方产品各取10g,放入烧杯中,用100ml纯净水溶解,将鸡血块放入烧杯,在不同时间鸡血块的变化情况,见下表:

从结果看,三个配方中,溶栓效果依次为配方1>配方2>配方3。
以上所述实施例只是为本发明的较佳实施例,并非以此限制本发明的实施范围,除了具体实施例中列举的情况外;凡依本发明之原理所作的等效变化,均应涵盖于本发明的保护范围内。
Claims (10)
1.一种具有溶栓作用的植物蛋白固体饮料,其特征在于,包括黑豆粉、大豆粉、谷物粉、益生元、黑纳豆粉和复合益生菌;
所述黑豆粉使用量占固体饮料总重量的40%~60%;
所述大豆粉使用量占固体饮料总重量的5%~10%;
所述谷物粉使用量占固体饮料总重量的20%~40%;
所述益生元使用量占固体饮料总重量的10%~20%;
所述黑纳豆粉在固体饮料中的活性单位100-400FU/g;
所述复合益生菌在固体饮料中的含量为5~20×108个/g。
2.根据权利要求1所述的一种具有溶栓作用的植物蛋白固体饮料,其特征在于,该植物蛋白固体饮料还包括山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物中的一种或几种,其中,山药提取物使用量占固体饮料总重量的2%~6%,茯苓提取物使用量占固体饮料总重量的2%~6%,人参提取物使用量占固体饮料总重量的0.2%~1%,蛹虫草提取物使用量占固体饮料总重量的0.2%~1%,陈皮提取物使用量占固体饮料总重量的0.02%~1%。
3.根据权利要求2所述的一种具有溶栓作用的植物蛋白固体饮料,其特征在于,所述人参提取物中选取的人参为5年以下、人工种植的用于食品中的人参。
4.根据权利要求1所述的一种具有溶栓作用的植物蛋白固体饮料,其特征在于,所述益生元是低聚果糖、低聚木糖或低聚麦芽糖中的一种或几种。
5.根据权利要求1所述的一种具有溶栓作用的植物蛋白固体饮料,其特征在于,所述复合益生菌为嗜酸乳杆菌、干酪乳杆菌、罗伊氏乳杆菌、副干酪乳杆菌、植物乳杆菌、鼠李糖乳杆菌、两歧双歧杆菌、乳双歧杆菌或短双歧杆菌中的一种或几种,复合益生菌的发酵终含量为5~20×108个/g。
6.一种具有溶栓作用的植物蛋白固体饮料的制备方法,其特征在于,包括以下步骤:
1)、黑纳豆粉制备:
1.1)、菌种培养:
1.1.1)、菌种活化及斜面培养:
菌种活化培养基为马铃薯200g/L,胡萝卜200重量份,琼脂20重量份,蛋白胨1.2重量份,葡萄糖21重量份,磷酸二氢钾0.4重量份,硫酸镁0.18重量份,维生素B1少许,接种后菌种活化时间为7-8天;
1.1.2)、种子菌培养:种子菌培养基成分的重量份为:葡萄糖2重量份,黑豆蛋白胨1重量份,Na2HPO4 0.6重量份,NaH2PO4 0.1重量份,MgSO4 0.05重量份,CaCl2 0.02重量份,吐温1重量份;培养基pH调整范围为pH 7.5~8.0;
1.2)、黑纳豆制备:
黑豆经浸泡、去种皮、蒸煮、沥水,然后接种种子菌和发酵,制成黑纳豆;
1.3)、黑纳豆粉制取:
将发酵后黑纳豆物料匀浆,经离心过滤、浓缩、冷冻干燥,粉碎,制得黑纳豆粉;
2)、山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物的制备:
选取10Kg的山药,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得山药提取物;
选取10Kg的茯苓,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得茯苓提取物;
选取10Kg的人参,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得人参提取物;
选取10Kg的蛹虫草,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得蛹虫草提取物;
选取10Kg的陈皮,用90Kg水蒸煮1~2小时,打浆、过滤、加热、浓缩、喷雾干燥,制得陈皮提取物;
3)、物料混合:
将黑豆粉、大豆粉、谷物粉、黑纳豆粉、山药提取物、茯苓提取物、人参提取物、蛹虫草提取物、陈皮提取物、益生元和复合益生菌按一定比例混合,最终在固体饮料中,纳豆激酶的含量活性单位100~400FU/g,复合益生菌的含量为5~20×108个/g。
7.根据权利要求6所述的一种具有溶栓作用的植物蛋白固体饮料的制备方法,其特征在于,所述步骤1.2)中还包括:黑豆浸泡6小时,去种皮、蒸煮、沥水,然后接种种子菌;在温度为26~28℃、相对湿度为≤75%的条件下,发酵培养13~15天,制成黑纳豆。
8.根据权利要求6所述的一种具有溶栓作用的植物蛋白固体饮料的制备方法,其特征在于,所述步骤2)中山药提取物、茯苓提取物、人参提取物、蛹虫草提取物或陈皮提取物的制备中,所有提取物蒸煮温度为90~100℃。
9.根据权利要求6所述的一种具有溶栓作用的植物蛋白固体饮料的制备方法,其特征在于,所述步骤2)中山药提取物、茯苓提取物、人参提取物、蛹虫草提取物或陈皮提取物的制备中,所有提取物加热时的温度为170℃~190℃,受热时间5~7秒。
10.根据权利要求6所述的一种具有溶栓作用的植物蛋白固体饮料的制备方法,其特征在于,所述步骤2)中山药提取物、茯苓提取物、人参提取物、蛹虫草提取物或陈皮提取物的制备中,所有提取物喷雾干燥时,喷雾压力为0.15pa,并控制出口温度为80~90℃。
