日化产品中化学防腐剂的滥用,对人们皮肤及身体健康造成损害。据报道,长期使用含有MIT、CIT的日化产品,易造成使用者皮肤过敏;三氯生、三氯卡班、尼泊金酯类化学物质防腐剂还会干扰人体内分泌,甚至致癌,对人体健康造成危害;另外,洗涤后的防腐剂废水,不易降解,对水生生物危害很大,不利水环境保护。
天然植物和藻类中含有很多营养成分和抑菌成分,将具有抑菌杀菌作用的天然植物用于日化产品中,效果好,易降解,不仅有利于人体健康,而且有利于环境保护,因此,使用天然植物代替化学防腐剂,是热点研究方向。
樟科新木姜子属植物是一类重要的药用植物,具有杀菌、抗癌、抗肿瘤、抑制血小板凝聚等功效;香菇味道鲜美,香气沁人,营养丰富,素有“植物皇后”美誉,香菇柄具有杀菌、美容、抗氧化等功效;繁枝蜈蚣藻为海膜科、蜈蚣藻属海水藻类植物,藻体直立,紫红色,黏滑,具有养胃功效,可制成藻胶和天然色素。然以繁枝蜈蚣藻、香菇和舟山新木姜子果实提取液作为原料制备的手工皂,目前国内还未见报道。
碧玺粉,又称电气石粉,是一种带有能量的硼硅酸盐结晶体,其表面流动着0.06毫安的微电流,能发射对人体有益的远红外线(波长在4-14um),促进新陈代谢,减小心脏压力。
现有技术如授权公告号为CN 104479923B的中国发明专利,公开了一种栀子香型手工皂及其制作方法,包括基础油、鲜花提取精油和基质;所述基础油包括椰子油、棕榈油和橄榄油;所述鲜花提取精油包括栀子花精油和桂花精油;所述基质包括氢氧化钠和水。但是该手工皂的抗炎抑菌效果不好。
发明内容
本发明的目的在于提供一种抑菌藻菌草保健手工皂制备及提取方法,含有繁枝蜈蚣藻提取物、香菇提取物和新木姜子果实提取物等天然抗菌和营养成分,具有持久抗菌、去污功能,且具有良好保湿作用。
本发明的另一目的是提供一种抑菌藻菌草保健手工皂的原料提取方法。
本发明的再一目的是提供一种抑菌藻菌草保健手工皂的制备方法。
本发明针对上述技术中提到的问题,采取的技术方案为:
一种抑菌藻菌草保健手工皂,包括基础油、活性提取物和基质:所述基础油包括10-30%的椰子油、30-70%的蓖麻油、20-50%的橄榄油;所述活性提取物包括新木姜子果实提取物、繁枝蜈蚣藻提取物和香菇提取物,以基础油的重量为基数,对应添加0.1-20%的活性提取物,其中,新木姜子果实提取物、繁枝蜈蚣藻提取物和香菇提取物的重量配比优选为1:0.5-3:0.1-5;所述基质包括氢氧化钾和无患子果皮提取液,基质以无患子果皮提取液作为水相,基质中有效氢氧化钾用量为相应基础油重量与基础油的皂化价乘积的加和;以无患子果实皂苷提取液作为天然皂基,利用繁枝蜈蚣藻提取物的抗氧化作用、香菇提取物的抗菌抗氧化作用和新木姜子果实提取物的抗炎杀菌作用,以及它们的天然营养成分,产生协同增益效果,制成的液体手工皂不仅具有较强的的抑菌杀菌能力又具有去污能力和优秀的保湿保健功能,实现资源整合及循环再利用。
作为优选,一种抑菌藻菌草保健手工皂,所用新木姜子果实提取物的提取方法包括:新木姜子果实粉碎,按料液重量比1:5-20加入65-85%的乙醇水溶液,搅拌均匀,在温度80-100℃下超声提取1-3h,过滤浓缩,加入3-5倍体积的95%乙醇,置于4℃冰箱24-48h,冷冻离心,得新木姜子果实提取物;新木姜子果实提取物含有异瑟林宁、冬青油烯、黄酮类及挥发油等多种活性成分,具有抗炎杀菌作用,其中含有的挥发精油和脂肪酸,尤其对人体皮肤有积极作用;冷冻低温离心,去除杂质,提高手工皂在低温下的稳定性,避免出现分层现象。
进一步优选,一种抑菌藻菌草保健手工皂,所用繁枝蜈蚣藻提取物的提取方法包括:将繁枝蜈蚣藻干燥、粉碎、过筛,加入4-6倍蒸馏水,微波预处理90-150s,加入无水乙醇,使料液比达到1:10-25,超声波提取15-30min,过滤、浓缩、使用95%乙醇醇析、离心过滤、复溶脱蛋白、二次醇析,得繁枝蜈蚣藻提取物;繁枝蜈蚣藻富含蜈蚣藻多糖,具有优异的抗氧化作用,使用微波预处理的热作用和超声波提取技术的空化作用,产生协同增益效果,有效加快繁枝蜈蚣藻多糖的提取效率和增高多糖提取率,醇析步骤减少杂质,提高提取多糖的纯度。
更进一步优选,一种抑菌藻菌草保健手工皂,所用香菇提取物的提取方法包括:取香菇干粉,加入7-12倍蒸馏水溶解,微波预处理90-150s,功率600W的超声波破碎15-25min,加入纤维素酶200U/g,调pH4.6,45℃水解50-70min,调pH为10,在10℃提取30min,离心15min,上清液减压浓缩,80%醇沉,4℃静置、离心、弃去上清液,真空冷冻干燥,得香菇蛋白;然后,调节pH6.0,以香菇蛋白重量计,加入0.2%的蛋白酶,50℃水解2-4h,高温灭酶,离心取上清液,得香菇提取物;香菇中含有大分子香菇多糖和小分子香菇肽,具有抗菌消炎和抗氧化的作用,而且小分子肽易于皮肤吸收,使用微波热作用和超声波技术协同作用,利用酶解法,筛选适宜的酶解条件,一步高效提取香菇中的小分子香菇肽,所提取的香菇肽的分子质量多为5-10ku的肽段,此分子区间的香菇肽具有最大的抗菌和抗氧化活性。
再进一步优选,一种抑菌藻菌草保健手工皂,所用无患子果皮提取液的提取方法包括:取无患子果实,去核粉碎,按料液比1:10-30,加入水,100℃煎煮30min,用纱布过滤3次,得澄清的无患子果皮提取液,备用;无患子果皮中含有大量的皂苷,具有杀虫作用,更是优异的天然清洁剂,可以有效去除油污,使用无患子果皮提取液作为水相,有效增加了手工皂的清洁能力。
更进一步优选,一种抑菌藻菌草保健手工皂,手工皂原料还包括碧玺粉0.1-3%,其中,碧玺粉优选为晶体碧玺粉,细度为200目;碧玺粉,含多种多人体有益的矿物质,表面流动着0.06毫安的微电流,能发射对人体有益的4-14um波长的远红外线,促进新陈代谢;使用200目细度,洗涤时的颗粒摩擦,具有保健、去污和轻微的去角质作用,增强洗手液的有益效果。
一种抑菌藻菌草保健手工皂的制备方法,按比例称取氢氧化钾,加入3-5倍体积的无患子果皮提取液,搅拌溶解,保持温度70-90℃,加入基础油,搅拌1.5-2h,加入活性提取物和碧玺粉,继续搅拌0.5-1.5h,完成皂化反应,此时反应体系由液体糊状变成软固体状;密封,30-40℃下保温静置15天以上;以0.8-2倍的水稀释皂体,调节pH至5.5-6.8,静置,得抑菌藻菌草保健手工皂;静置低于15天,反应不完全,不利经济效益。
椰子油具有清洁能力,蓖麻油是软油,保护皮肤,且能使皂体透明透亮,橄榄油含有不饱和脂肪酸,有益人体健康,具有温和保湿作用,无患子果皮提取液含有天然皂基,具优异的清洁能力,以椰子油、蓖麻油和橄榄油为基础油,以无患子果皮提取液作为水相,使用氢氧化钾做碱,配合新木姜子、繁枝蜈蚣藻和香菇活性提取物和碧玺粉,制备得到清洁能力优异的液体手工皂。
本发明制得的抑菌藻菌草保健手工皂为液体手工皂,真皂率在25-45%之间;pH值稳定,呈弱酸性;保湿性不低于15%;对大肠埃希菌与金黄色葡萄球菌抑菌率大于98%,对白色念珠菌和乙型副伤寒杆菌的抑菌率大于95%;人体皮肤呈弱酸性,常规的洗涤产品都呈弱碱性,有的pH值甚至达到10,对皮肤具有很强的刺激作用,易导致过敏,而本发明洗手液将pH值为弱酸性,完美贴合人体皮肤的酸碱性,温和不刺激。
与现有技术相比,本发明的优点在于:
1)本发明抑菌藻菌草保健液体手工皂不含化学物质防腐剂,减少皮肤刺激性,完全避免了化学物质防腐剂带来的致敏、干扰内分泌甚至致癌的风险。
2)本发明抑菌藻菌草保健液体手工皂使用天然植物新木姜子提取物、繁枝蜈蚣藻提取物和香菇提取物等达到杀菌抑菌和防腐的目的,且保留了植物提取物中的营养成分,协同晶体碧玺粉微电流和红外线特性,使产品具清洁作用的同时具有良好的抗菌保湿和保健功效,而且易降解,无残留,对人体健康和水环境无害,实现资源整合及循环再利用。
3)本发明利用超声波技术提取新木姜子,微波和超声波技术配伍提取繁殖蜈蚣藻,利用微波、超声波和酶解技术配伍提取香菇肽,分别提高了相应活性物质的提取率。
具体实施方式
下面通过实施例对本发明方案作进一步说明:
实施例1:
一种抑菌藻菌草保健手工皂的配方成分包括:
基础油包括:20%椰子油、50%的蓖麻油、40%的橄榄油;
活性提取物包括:新木姜子果实提取物、繁枝蜈蚣藻提取物和香菇提取物,其重量配比为1:2:2;以基础油的重量为基数,对应添加10%的活性提取物;
基质包括:氢氧化钾和无患子果皮提取液,基质以无患子果皮提取液作为水相,基质中有效氢氧化钾用量为相应基础油重量与基础油的皂化价乘积的加和;
晶体碧玺粉2%,细度为200目,余量为水。
一种抑菌藻菌草保健手工皂的制备方法包括:
1)新木姜子果实提取物的提取方法:新木姜子果实粉碎,按料液重量比1:15加入75%的乙醇水溶液,搅拌均匀,在温度90℃下超声提取2h,过滤浓缩,加入4倍体积的95%乙醇,置于4℃冰箱36h,冷冻离心,得新木姜子果实提取物;
2)繁枝蜈蚣藻提取物的提取方法:将繁枝蜈蚣藻干燥、粉碎、过筛,加5倍蒸馏水,微波预处理120s,加入无水乙醇,使料液比达到1:20,超声波提取20min,过滤、浓缩、使用95%乙醇醇析、离心过滤、复溶脱蛋白、二次醇析,得繁枝蜈蚣藻提取物;
3)香菇提取物的提取方法:取香菇干粉,加入9倍蒸馏水溶解,微波预处理120s,功率600W的超声波破碎20min,加入纤维素酶200U/g,调pH4.6,45℃水解60min,调pH10,在10℃提取30min,离心15min,上清液减压浓缩,80%醇沉,4℃静置、离心、弃去上清液,真空冷冻干燥,得香菇蛋白;然后,调节pH6.0,以香菇蛋白重量计,加入0.2%的蛋白酶,50℃水解3h,高温灭酶,离心取上清液,得香菇提取物;
4)无患子果皮提取液的提取方法:取无患子果实,去核粉碎,按料液比1:20,加入水,100℃煎煮30min,用纱布过滤3次,得澄清的无患子果皮提取液,备用;
5)抑菌藻菌草保健手工皂的制备:按比例称取氢氧化钾,加入4倍体积的无患子果皮提取液,搅拌溶解,保持温度80℃,加入基础油,搅拌2h,加入活性提取物和碧玺粉,继续搅拌1h,完成皂化反应,此时反应体系由液体糊状变成软固体状;密封,35℃下保温静置15天以上;以1.5倍的水稀释皂体,调节pH至6.3,静置,得抑菌藻菌草保健手工皂。
本实施例制得的抑菌藻菌草保健手工皂为液体手工皂,真皂率为34%;pH为6.3;保湿性为17.8%;对大肠埃希菌与金黄色葡萄球菌抑菌率为99.90%,对白色念珠菌的抑菌率为99.76%,对乙型副伤寒杆菌的抑菌率为97.83%。
实施例2:
一种抑菌藻菌草保健手工皂的配方成分包括:
基础油包括:15%椰子油、40%的蓖麻油、45%的橄榄油;
活性提取物包括:新木姜子果实提取物、繁枝蜈蚣藻提取物和香菇提取物,其重量配比为1:2:1;以基础油的重量为基数,对应添加5%的活性提取物;
基质包括:氢氧化钾和无患子果皮提取液,基质以无患子果皮提取液作为水相,基质中有效氢氧化钾用量为相应基础油重量与基础油的皂化价乘积的加和;
晶体碧玺粉1%,细度为200目,余量为水。
一种抑菌藻菌草保健手工皂的制备方法包括:
1)新木姜子果实提取物的提取方法:新木姜子果实粉碎,按料液重量比1:20加入80%的乙醇水溶液,搅拌均匀,在温度80℃下超声提取1.5h,过滤浓缩,加入3倍体积的95%乙醇,置于4℃冰箱48h,冷冻离心,得新木姜子果实提取物;
2)繁枝蜈蚣藻提取物的提取方法:将繁枝蜈蚣藻干燥、粉碎、过筛,加入6倍蒸馏水,微波预处理150s,加入无水乙醇,使料液比达到1:25,超声波提取30min,过滤、浓缩、使用95%乙醇醇析、离心过滤、复溶脱蛋白、二次醇析,得繁枝蜈蚣藻提取物;
3)香菇提取物的提取方法:取香菇干粉,加入10倍蒸馏水溶解,微波预处理90s,功率600W的超声波破碎15min,加入纤维素酶200U/g,调pH4.6,45℃水解50min,调pH为10,在10℃提取30min,离心15min,上清液减压浓缩,80%醇沉,4℃静置、离心、弃去上清液,真空冷冻干燥,得香菇蛋白;然后,调节pH6.0,以香菇蛋白重量计,加入0.2%的蛋白酶,50℃水解4h,高温灭酶,离心取上清液,得香菇提取物;
4)无患子果皮提取液的提取方法:取无患子果实,去核粉碎,按料液比1:20,加入水,100℃煎煮30min,用纱布过滤3次,得澄清的无患子果皮提取液,备用;
5)抑菌藻菌草保健手工皂的制备:按比例称取氢氧化钾,加入5倍体积的无患子果皮提取液,搅拌溶解,保持温度90℃,加入基础油,搅拌2h,加入活性提取物和碧玺粉,继续搅拌0.5h,完成皂化反应,此时反应体系由液体糊状变成软固体状;密封,40℃下保温静置15天以上;以2倍的水稀释皂体,调节pH至6.0,静置,得抑菌藻菌草保健手工皂。
本实施例制得的抑菌藻菌草保健手工皂为液体手工皂,真皂率为28%;pH为6.0;保湿性为16.5%;对大肠埃希菌与金黄色葡萄球菌抑菌率为99.90%,对白色念珠菌的抑菌率为97.32%,对乙型副伤寒杆菌的抑菌率为96.43%。
对比例1:
不添加新木姜子提取物、繁枝蜈蚣藻提取物、香菇提取物,其余与实施例1完全相同。
对比例2:
市场在售白液体皂。
实施例3:
以实施例1制备的抑菌藻菌草保健液体手工皂为试验组,以实施例2、对比例1、对比例2的液体皂为对照组1、对照组2、对照组3,进行保湿性、发泡能力、发泡稳定性测试,结果见表1。
保湿性测定:称取试验组、对照组1、对照组2的测试样品,以甘油作为阳性对照,放入配制的相对湿度为80%的饱和硫酸铵和相对湿度为40%的饱和乙酸钾溶液中,放置在25℃恒温器中,不同时间进行称量,A0表示初始液体皂质量,分别称重Ax(6、12、24、36、48h)后的液体皂质量,计算公式为保湿性(%)=Ax/A0×100%。
发泡能力测定:采用震荡法:1)取新制备的各组液体皂样品0.5mL加入塑料量筒,再加入20mL蒸馏水,用大小合适的橡胶塞塞紧塑料量筒,充分震摇20次,震摇方向、快慢、力度、气温环境须保持一致;2)震摇完毕后,立即取下比色管管塞,加入20mL的水,沿管壁将比色管中泡沫冲下,记录泡沫震摇后的高度及加水后泡沫破坏到原有高度一半时所需时间;3)重复以上操作,记录数据。
以泡沫高度作为洗手液起泡性能的量度,以泡沫破坏到原有高度一半时所需时间表征泡沫的稳定性。

由表1可知,试验组和对照组1的液体皂,其发泡能力,发泡稳定性都很好,保湿性分别为17.8%和16.5%,都大于15%;而对照组2和对照组3的发泡能力,发泡稳定性,保湿性都比不上试验组,对照组2和对照组3的保湿性均低于15%,说明新木姜子果实提取物、繁枝蜈蚣藻提取物和香菇提取物对保湿性、发泡能力和发泡稳定性都有积极的影响。
实施例4:
液体皂抑菌试验:以实施例1制备的抑菌藻菌草保健液体手工皂为测试对象,试验菌为金黄色葡萄球菌、大肠埃希菌、白色念珠菌和乙型副伤寒杆菌。
测试方法:抑菌试验在25℃水浴条件下进行,所用器材均经过高温高压灭菌,试剂均经过0.22μm滤膜过滤,在灭菌试管内加入5.0ml的0.5倍实验浓度的液体皂与对照样溶液(PBS)5.0ml,一式3管。分别在每管加0.1ml菌悬液,然后混匀,作用至规定的不同时间,取0.5ml菌药混合液加入到装4.5ml磷酸盐缓冲液试管内,充分混匀。取0.5ml样液接种培养,作活菌计数,计算抑菌率。试验重复3次,测试结果见表2。

由表2可知,在常温25℃条件下,在2min内,试验样品液体手工皂对大肠埃希菌的抑菌率大于99.90%,对金黄色葡萄球菌的抑菌率大于99.90%,对白色念珠菌的抑菌率大于98.13%,对乙型副伤寒杆菌的抑菌率大于95.35%,说明本发明液体手工皂具有优异的抑菌效果。
实施例5:
皮肤急性毒性试验:选取Wistar大鼠10只,雌雄各半,体重为180-220g,按5000mg/kg体重的剂量取样品,涂抹或粘贴在动物背部脱毛区(范围30-40cm2),用两层硫酸纸覆盖,用无刺激性胶布封闭固定24h。24h之后除受试动物,观察动物中毒体征,观察期为7d。第7d称重1次,试验结束处死动物并观察大体病理改变。结果显示涂抹本发明抑菌藻菌草保健液体手工皂后的大鼠无死亡,大体病理解剖学检查未见异常,LD50均>5000mg/kg体重。
本发明的操作步骤中的常规操作为本领域技术人员所熟知,在此不进行赘述。

1.一种手工皂,其特征在于,
所述手工皂由基础油、活性提取物、基质和碧玺粉制成:
所述基础油由10-30%的椰子油、30-70%的蓖麻油、20-50%的橄榄油组成;
所述活性提取物由新木姜子果实提取物、繁枝蜈蚣藻提取物和香菇提取物组成,以基础油的重量为基数,对应添加0.1-20%的活性提取物,其中,新木姜子果实提取物、繁枝蜈蚣藻提取物和香菇提取物的重量配比为1:0.5-3:0.1-5;
所述基质由氢氧化钾和无患子果皮提取液组成,基质以无患子果皮提取液作为水相,基质中有效氢氧化钾用量为相应基础油重量与基础油的皂化价乘积的加和;
所述新木姜子果实提取物的提取方法包括:新木姜子果实粉碎,按料液重量比1:5-20加入65-85%的乙醇水溶液,搅拌均匀,在温度80-100℃下超声提取1-3h,过滤浓缩,加入3-5倍体积的95%乙醇,置于4℃冰箱24-48h,冷冻离心,得新木姜子果实提取物;

所述手工皂的制备方法包括:按比例称取氢氧化钾,加入3-5倍体积的无患子果皮提取液,搅拌溶解,保持温度70-90℃,加入基础油,搅拌1.5-2h,加入活性提取物和碧玺粉,继续搅拌0.5-1.5h,完成皂化反应,此时反应体系由液体糊状变成软固体状;密封,30-40℃下保温静置15天以上;以0.8-2倍的水稀释皂体,调节pH至5.5-6.8,静置,得手工皂,其中含碧玺粉为0.1-3%,其是晶体碧玺粉,细度为200目,所述无患子果皮提取液的提取方法包括:取无患子果实,去核粉碎,按料液比1:10-30,加入水,100℃煎煮30min,用纱布过滤3次,得澄清的无患子果皮提取液,备用。
2.根据权利要求1所述的一种手工皂,其特征在于为液体手工皂,真皂率在25-45%之间;pH值稳定,呈弱酸性;保湿性不低于15%;对大肠埃希菌与金黄色葡萄球菌抑菌率大于98%,对白色念珠菌和乙型副伤寒杆菌的抑菌率大于95%。
