Abstract

本发明公开了两株具有高选择性催化cyperenoic acid羟基化的细胞色素P450 BM3突变酶,此类突变酶是由父本细胞色素P450 BM3(F87A/A330W)突变酶出发经过突变得到的。其中细胞色素P450 BM3(F87A/A330W/F331L)变体酶能高效率、高选择性的催化cyperenoic acid得到7β‑hydroxycyperenoic acid,而细胞色素P450 BM3(L75V/F87A/T88F/A330W)变体酶能高效率、高选择性的催化cyperenoic acid得到9α‑hydroxycyperenoic acid。

催化cyperenoic acid羟基化的细胞色素P450 BM3突变酶
技术领域:
本发明涉及生物技术领域,涉及两株具有高选择性催化cyperenoic acid羟基化的细胞色素P450 BM3 突变酶,该突变酶是由野生型的细胞色素P450 BM3酶出发经过突变得到的,具体涉及高效获取这两株突变酶的方法和突变酶的高效表达制备方法以及利用该细胞色素P450 BM3突变酶高选择性催化cyperenoic acid化合物的羟基化,得到具有光学纯度的羟基化产物的方法。
背景技术
细胞色素P450单氧化酶(P450)能够高效、高选择性地催化种类繁多的具有复杂骨架有机化合物的氧化反应,被誉为自然界的万能催化剂,近年来,利用P450单氧化酶催化C-H键活化的研究越来越多。
P450 BM3(CYP102A1)是一种来源于巨大芽胞杆菌(Bacillus megaterium)的脂肪酸羟化酶,其独特性在于P450 BM3是一种天然的血红素铁依赖的单氧化酶和能传递电子的NADPH依赖的黄素还原酶的融合酶,其无需额外的电子传递系统,仅需添加NADPH就能参加反应。P450 BM3单氧化酶的这种优势使其成为近年来的研究热点,特别是针对其对非天然底物的选择性低的问题,通过蛋白质工程对其进行定向进化,以控制其选择性,甚至创造原始P450所不能催化的新的反应类型。例如德国马普研究所的Reetz及其同事利用CAST策略,将P450 BM3进行定向进化,使其同时能以极高的对映选择性和立体选择性氧化睾丸激素和黄体酮。初始的P450 BM3能够氧化氧化睾丸激素中未活化的C-H键,主要得到2β-OH和15β-OH的产物,二者的比例为52:45。将P450 BM3经过CAST突变后,突变KSA-1(A330W/F87A)能够将睾丸激素氧化得到 97%的2β-OH产物,而突变KSA-14(R47Y/T49F/V78L/A82M/F87A)则能将其氧化得到96%的15β-OH产物。美国罗切斯特大学Rudi Fasan领导的研究组最近报道了一种快速的、利用三步进化P450s的策略,利用此策略可将P450 BM3进化为能够在抗疟疾药物青蒿素的未活化位点高产率和高立体选择性地引入羟基的酶。除上述的通过进化P450BM3控制其羟基化选择性的研究外,美国加州理工大学Frances H.Arnold等最近在Science及Nature Chemical Biology上报道了将P450 BM3进化为能够催化环丙烷化反应的酶,而该种类型的反应是不能够被天然的P450 BM3所催化,野生型的P450 BM3能够催化环氧化或者羟基化反应,而当使用重氮酯作为卡宾转移的前体时,通过筛选P450 BM3突变库,得到了能够催化环丙烷化反应的且具有选择性的催化剂。另外,Frances H.Arnold还报道了利用P450 BM3突变体催化叠氮化合物分子内C-H键氨化及C-H 键硫化的研究。近年国内多家单位对P450单氧化酶研究也越加深入,如清华大学、浙江大学、华东理工大学、东南大学、科学院微生物所、科学院天津工业生物技术研究所和科学院南海海洋研究所,其中清华大学林章凛教授课题组通过对P450sca2的定向进化,提高了其把美伐他汀转化为普伐他汀的活性及选择性;中国科学院青岛生物能源与过程研究所李盛英课题组发现了P450单氧化酶与还原蛋白的作用的新机制。
鸡骨香为大戟科巴豆属植物,广泛应用于风湿痹痛、腰腿痛、胃痛、咽喉肿痛、黄疽、病痛、蛇咬伤、跌打肿痛等疾病的治疗。Cyperenoic acid是从鸡骨香的干燥根中分离出一种含量高的广藿香烷型(Patchoulane)倍半萜类化合物,其生合成起始于FPP,经过环合形成七环正碳离子中间体,然后经过再次环合,形成Patchoulane型倍半萜,文献报道它对前列腺癌细胞、神经胶质瘤细胞、绿猴肾细胞、黑色素瘤细胞、乳腺癌细胞、肺癌细胞、卵巢癌细胞、结肠癌细胞等均存在增殖抑制作用。有相关文献报道以Cyperenoic acid为基础,通过对其进行化学改造。但由于本身极性小水溶解性差及结构改造单一,但大部分是在羧基和双键位置上发生修饰的,很少在环上进行修饰,抗肿瘤活性开发困难。前期本实验室利用真菌雅致小克银汉霉AS3.2028静息细胞转化cyperenoic acid得到了五个羟基化产物,结构分别为7β-hydroxycyperenoic acid、8α-hydroxycyperenoic acid、9α-hydroxycyperenoic acid、10β-hydroxycyperenoic acid和 13-hydroxycyperenoicacid。其中7β-hydroxycyperenoic acid和9α-hydroxycyperenoic acid产物潜在抗肿瘤活性高于cyperenoic acid(图1)。因此从酶水平提高cyperenoic acid的转化产率,并提高7β-hydroxy-cyperenoic acid 和9α-hydroxy-cyperenoic acid的区域选择性,实现产物的制备具有十分重要的价值。
发明内容:
本发明提供了两株细胞色素P450 BM3变体酶,分别解决了现有突变体酶催化cyperenoic acid生成 7β-hydroxycyperenoic acid和9α-hydroxycyperenoic acid选择性低的问题。
本发明首先提供了一株细胞色素P450 BM3变体酶,其基因序列如SEQ ID NO.15所示,命名为细胞色素P450 BM3(F87A/A330W/F331L)变体酶。即在细胞色素P450 BM3(F87A/A330W)变体酶的基础上引入了一个突变位点:331位点由苯丙氨酸(F)突变为亮氨酸(L)。
该基因通过如下方法获得:根据细胞色素P450 BM3(F87A/A330W)变体酶基因(基因序列:SEQ ID NO.1)的血红素域部分设计定点突变引物,以质粒pET-M11-P450 BM3(F87A/A330W)为模板,进行PCR 扩增,扩增完成后,用Sac I和Nco I内切酶进行双酶切,并与Sac I和Nco I双酶切后的模板质粒相连。将连接产物转入大肠杆菌(E.coli)BL21中,筛选获得突变体。提取质粒,经测序可知,突变酶P450 BM3 (F87A/A330W)基因上的编码第75位氨基酸的碱基序列由“GAT”突变为“CTG”,同时该质粒命名为:质粒 pET-M11-P450 BM3(L75V/F87A/A330W)。
本发明还提供了一株编码所述细胞色素P450 BM3变体酶的基因,其碱基序列如SEQ ID NO.22所示,命名为细胞色素P450 BM3(L75V/F87A/T88F/A330W)变体酶,即75位点由亮氨酸(L)突变为缬氨酸(V), 88位点由苏氨酸(T)突变为苯丙氨酸(F)。
该基因通过如下方法获得:首先,根据细胞色素P450 BM3(F87A/A330W)变体酶基因的血红素域部分设计两对定点突变引物,以质粒pET-M11-P450 BM3(F87A/A330W)为模板,进行PCR扩增,扩增完成后,用Sac I和Nco I内切酶双酶切,并与Sac I和Nco I双酶切后模板质粒相连。将连接后产物转入大肠杆菌(E.coli)BL21中,筛选获得突变体。抽取质粒,经测序可知,突变酶P450 BM3(F87A/A330W)基因上的编码第75位氨基酸的碱基序列由“GAT”突变为“CTG”,同时该质粒命名为:质粒pET-M11-P450 BM3(L75V/F87A/A330W)。接着,同样根据细胞色素P450 BM3(L75V/F87A/A330W)变体酶基因的血红素域部分设计饱和定点突变引物,以质粒pET-M11-P450 BM3(L75V/F87A/A330W)为模板,进行PCR扩增,扩增完成后,用Sac I和Nco I内切酶进行双酶切,并与Sac I和Nco I双酶切后模板质粒相连。将连接后产物转入大肠杆菌(E.coli)BL21中,筛选获得突变体,抽取质粒,经测序可知,突变酶P450 BM3 (L75V/F87A/A330W)基因上的编码第88位氨基酸的碱基序列由“GAA”突变为“AAA”,同时该质粒命名为:质粒pET-M11-P450 BM3(L75V/F87A/T88F/A330W)。
本发明提供了所述细胞色素P450 BM3变体酶在选择性催化cyperenoic acid分别生成7β-hydroxy cyperenoic acid和9α-hydroxycyperenoic acid中的应用。与父本细胞色素P450 BM3(F87A/A330W)变体酶相比,突变体P450 BM3(L75V/F87A/A330W)催化生成7β-hydroxycyperenoic acid的选择性由73%提高至 94%,突变体P450 BM3(L75V/F87A/T88F/A330W)催化生成9α-hydroxycyperenoic acid的选择性由12%提高至90%。
本发明所获得的变体酶的酶学参数较父本细胞色素P450 BM3(F87A/A330W)变体酶有显著改善,其中细胞色素P450 BM3(L75V/F87A/A330W)突变酶催化效率(Kcat/KM)是父本的5.5倍,细胞色素P450 BM3 (L75V/F87A/T88F/A330W)突变酶是父本的3.5倍。说明本发明所涉及的变体酶对底物cyperenoic acid具有更高的催化活力。
进一方面,本发明的一些实施方案中,含有突变的细胞色素P450 BM3突变体针对cyperenoic acid也展现出较高的活性及选择性,这些突变包含下列一项或者多项突变。其中氨基酸序列74残基为Val(SEQ ID NO.2),氨基酸序列78残基为Ala(SEQ ID NO.5),氨基酸序列87残基为Ala(SEQ ID NO.7),氨基酸序列 268残基为Leu(SEQ ID NO.12),氨基酸序列328残基为Val(SEQ ID NO.13),氨基酸序列330残基为Phe (SEQ ID NO.14),氨基酸序列331残基为Trp(SEQ ID NO.16),基酸序列437残基为Phe(SEQ ID NO.18) 能高选择性催化cyperenoic acid得到7β-hydroxycyperenoic acid。

下面通过具体的实施方案描述本发明的细胞色素P450 BM3突变酶选择性催化cyperenoic acid,生成 7β-hydroxy-cyperenoic acid和9α-hydroxy-cyperenoic acid。除特别指明,本发明中所用的实验方案均为本领域技术人员所公知的方法。此外,实施例应理解为说明性的,而不是限制本发明。
某些术语的定义。
区域选择性(regioselectivity)是指在一定的反应条件下,优先选择与分子内不同位置的某一相同功能基团起化学反应。与区域选择性相对应的是立体选择性,立体选择性就是反应物对某一个碳原子进行不同方向的攻击碰撞,从而产生的产物具有不同的手性。

实施例涉及培养基配方
a.LB培养基:称取10gNaCl,10g胰蛋白胨(Tryptone),5g酵母提取物(YeastExtract)溶于900mL 蒸馏水,定溶至1000mL后用NaOH调pH至7.5,(如配置LB平板,可在灭菌前加入15g琼脂),分装后, 121℃高压灭菌20min。
b.LB转化平板:LB培养基+1.5%琼脂(g/l)。
c.TB培养基:称取12g胰蛋白胨(Tryptone),24g酵母提取物(Yeast Extract),4mL甘油(Glycerol) 溶于800mL蒸馏水充分搅拌溶解,加蒸馏水定容至1000mL后,分装,121℃高压灭菌20min。。
d.磷酸盐缓冲溶液(PH7.4):称取12.54g K2HPO4·3H2O与2.31g KH2PO4于90mL的去蒸馏水中,搅拌溶解后,定容至100mL,121℃高压灭菌20min。
e.1M KPi母液(PH7.4):称取114.11g K2HPO4-3H2O(0.5mol),27.2g KH2PO4(0.5mol)溶于蒸馏水后中,定容至700mL。稀释到0.1M KPi待后续用。
f.KPi洗液:量取90mL的0.1MKPi母液,10mL甘油(Glycerol)。
实施例1 P450 BM3父本突变体P450 BM3(F87A/A330W)质粒提取
本发明所使用的父本细胞色素P450 BM3(F87A/A330W)突变酶的菌种由德国马克斯-普朗克研究所的 Manfred.T.Reetz教授惠赠,它是将细胞色素P450 BM3(F87A/A330W)变体酶基因插入原始载体 pET-M11的启动子下游制得,其中突变基因的序列和重组质粒的结构已记载在文献(“Regio-and stereoselectivity of P450-catalysed hydroxylationof steroids controlled by laboratory evolution[J].Nature chemistry,2011,3(9):738-743.”)中。接种细胞色素P450 BM3(F87A/A330W)突变酶菌种,用LB液体培养过夜后,发酵液用1.5mL离心管12000r/min离心1min,弃上清收集菌体,可重复几次以得到足够量的细胞,随后参照TIAN prep Mini Plasmid Kit提取含细胞色素P450 BM3(F87A/A330W)变体酶基因的质粒。
实施例2 PCR扩增构建突变库A
突变库A的构建是利用突变引物和侧翼引物,重叠延伸PCR进行的。(设计的突变引物如下:)
以提取的P450 BM3(F87A/A330W)质粒为模板进行PCR扩增,反应体系如下。
PCR扩增条件:95℃:10min,(95℃:30s,46.5℃:30s,72℃:120s)30个循环,72℃,10min。
所得基因片段经过胶回收,按如下体系进行PCR扩增,而PCR扩增条件如上述条件。
PCR扩增得到的DNA片段用胶回收试剂盒纯化。所得到目的基因与质粒pET-M11-P450 BM3 (F87A/A330W)同时进行双酶切,酶切体系如下:
经37℃反应2h,对双酶切反应液使用胶回收试剂盒纯化,所得的目的基因与质粒pET-M11-P450 BM3 (F87A/A330W)双酶切片段进行酶连,酶连体系如下:
酶连反应液16℃反应6h后待用。
实施例3 E.coli BL21感受态细胞的制备及转化
(a)从种子培养基中取0.4mL接种于20mL LB液体培养基中培养3h;(b)3000r/min,5min分两次富集 2mL菌体于1.5mL EP管中,弃上清;(c)加入100弃上冰冷CaCl2溶液,重新悬浮菌体,冰浴30min;(d) 加入20μL连接液,轻轻旋转混匀,冰浴30min;(e)42℃热休克90s,冰浴3min,加入600μL LB液体培养基。37℃培养,150r/min振荡培养1h;(f)分别取150μL涂布于LB(Kana)抗性平板。
实施例4突变库A中突变体诱导表达
(a)菌体活化:从4℃冰箱保藏的平板培养物上挑取单菌落接种于4mL含4μL卡纳霉素(50mg/ml)抗性的LB液体培养基中,37℃,200r/min摇床过夜培养;(b)TB表达培养基诱导表达:取种子液1mL接种至表达培养基中(100mL TB,10mL K2HPO4-KH2PO4,1mL Power-mix,1mL TES,100μL MgCl2(203mg/ml), 100μL kan+),放置200rpm,37℃摇床2.5h,冷却到室温后,30℃培养20h;(c)菌体预处理:离心富集菌体,4000rpm离心15min,弃上清,用15mL Kpi洗液洗涤菌体,4000rpm离心15min,弃上清,将富集得到的菌体按照200mg湿菌体/mL的0.1MKpi缓冲液重悬,-20℃,24h。将菌体冻融后取1mL加入100μL 溶菌酶(174mg/mL),37℃破碎40min,4℃,4000rpm,离心10min,收集上清粗酶液,立即使用或者存于 -20℃冰箱备用。
实施例5突变库A对cyperenoic acid的筛选
将上述所得粗酶液按如下体系反应:
25℃,200rpm反应20h,所得反应液进行HPLC检测,检测条件为:色谱柱:DiamonsilTM C18 (250×4.6mm);流动相:甲醇-水(含0.1%的三氟乙酸,0-18min 60%-73.5%;18-20min 73.5%-100%;20-27min 100%);检测器:JASCO UV-2075型UV检测器;检测波长:240nm;流速:0.8mL/min;进样量:10μL。
检测结果如下表所示:
实施例6细胞色素P450 BM3(F87A/A330W/F331L)突变酶的纯化。
经过筛选得到细胞色素P450 BM3(F87A/A330W/F331L)突变酶能高选择性的催化cyperenoic acid得到产物7β-hydroxy-cyperenoic acid。因此对其按实施例4中粗酶液用金属螯合亲和层析(IMAC)纯化,其中金属螯合介质为Ni-NTA介质,透过的纯酶在去除咪唑之后,存于-4℃冰箱备用。父本细胞色素P450 BM3(F87A/A330W)变体酶的制备方法同上。
实施例7细胞色素P450 BM3(F87A/A330W/F331L)突变酶的酶学参数的测定。
酶学参数通过对酶动力学曲线的测定获得,反应条件为于1ml的100mM,pH 7.4的Kpi反应体系中加入lnmol的纯酶、终浓度1~1000μM的cyperenoic acid,置于25℃保温10min,加入0.4mg NADPH 启动反应,反应在温度为25℃水浴,转速为150rpm避光条件下进行,反应10min后立即取出,迅速加入6M KOH终止反应,于分光光度计上测定波长为240nm处的光吸收值,即为cyperenoic acid的光吸收情况.
结果发现:细胞色素P450 BM3(F87A/A330W/F331L)变体酶对cyperenoic acid的KM值为69.5μM,显著低于父本酶P450BM-3(F87V/A74G/L188Q)的163.7μM,表明突变酶对底物具有更高的亲和力。转化数(kcat)为15.3min-1,与父本(11.5min-1)有轻微提高。变体酶的催化效率(kcat/KM)为0.22 μM-1·min-1,是父本(0.04μM-1·min-1)的5.5倍。

实施例8PCR扩增构建突变库B
突变库B的构建是利用突变引物和侧翼引物,重叠延伸PCR进行的。(设计的突变引物如下:)

Claims (9)

1.一种细胞色素P450 BM3变体酶,其特征在于,氨基酸序列如SEQ ID NO.1所示,该突变酶能催化cyperenoic acid分别生成7β-hydroxycyperenoic acid、8α-hydroxycyperenoic acid和9α-hydroxycyperenoic acid,对应于野生型的P450 BM3,其87位残基为Ala,330位残基为Trp。
2.如权利要求1所述细胞色素P450 BM3变体酶,特征在于,75位、78位、88位、268位、330位、331位或437位氨基酸残基范德华体积减小,氨基酸序列分别如SEQ ID NO.3、SEQ IDNO.5、SEQ ID NO.8、SEQ ID NO.11、SEQ ID NO.14、SEQ ID NO.15、SEQ ID NO.17所示。
3.如权利要求1所述细胞色素P450 BM3变体酶,特征在于,74位、75位、78位、87位、88位、264位、268位、328位、331位或437位氨基酸残基范德华体积增大,氨基酸序列分别如SEQID NO.2、SEQ ID NO.4、SEQ ID NO.6、SEQ ID NO.7、SEQ ID NO.9、SEQ ID NO.10、SEQ IDNO.12、SEQ ID NO.13、SEQ ID NO.16、SEQ ID NO.18所示。
4.如权利要求1所述细胞色素P450BM3变体酶,特征在于,75位氨基酸残基为Val,78位或88位氨基酸残基范德华体积增大,氨基酸序列分别如SEQ ID NO.19、SEQ ID NO.20或SEQID NO.21、SEQ ID NO.22所示。
5.一种表达载体pET-M11,该载体能表达权利要求1-4任何一项所述的P450BM3变体酶。
6.一种宿主细胞,其含有权利要求5所述的表达载体。
7.根据权利要求6所述的宿主细胞,其特征在于,宿主细胞为大肠杆菌E.coli BL21。
8.如权利要求1-4任何一项所述的细胞色素P450 BM3变体酶在催化cyperenoic acid中的应用。
9.如权利要求8所述的应用,其特征在于,SEQ ID NO.1-2、SEQ ID NO.5、SEQ ID NO.7-8、SEQ ID NO.12、SEQ ID NO.14-16或SEQ ID NO.18能高选择的催化cyperenoic acid获得产物7β-hydroxycyperenoic acid,SEQ ID NO.3、SEQ ID NO.19-22能高选择的催化cyperenoic acid获得产物9α-hydroxycyperenoic acid。
