Abstract

本发明公开了一种多能干细胞形成必需蛋白CREPT的在诱导多能干细胞中的应用。本发明发现降低成纤维细胞中CREPT的表达,成纤维细胞不能被4种Yamanaka因子诱导形成iPS细胞,在不表达CREPT的细胞中表达CREPT后,细胞能被诱导为iPS细胞。表明,CREPT及其编码基因可以用于调控细胞向iPS细胞的转化,可以用于制备不能诱导为多能干细胞的细胞模型,还可以用于制备iPS细胞。

技术领域
本发明涉及分子细胞生物学技术领域中,一种多能干细胞形成必需蛋白CREPT的在诱导多能干细胞中的应用。
背景技术
干细胞是一类能够自我更新和分化形成功能性细胞的多潜能细胞。按照不同的分类标准,干细胞可分为不同的类型,如根据干细胞所处的发育阶段不同可分为胚胎干细胞和成体干细胞;根据干细胞所具有的分化潜能可分为全能干细胞、多能干细胞、专能干细胞和单能干细胞。胚胎干细胞是一种高度未分化细胞,可以分化形成人体各种类型的细胞,具有发育的全能性,为医学移植治疗疾病带来了便利。
但是,伦理问题和移植排斥限制了以胚胎干细胞为基础的医学应用。2006年,Takahashi和Yamanaka利用四种转录因子(Oct4、Sox2、Klf4和c-Myc,Yamanaka因子)成功将鼠的成纤维细胞重编程为未分化的诱导多能干(induced pluripotent stem,iPS)细胞。一年后,James Thomson团队用Oct4、Sox2、Nanog和LIN28四种因子成功将人的体细胞重编程为多能干细胞,即人iPS细胞。iPS细胞的诱导成功具有划时代的意义,它使干细胞的来源不再局限于胚胎干细胞或成体干细胞,人身体上的任何一种细胞都可能被诱导成为iPS细胞供治疗疾病使用。iPS细胞的研究为生命开拓了新的契机,iPS细胞的多潜能性赋予了其非凡的再生能力,在医学领域具有不可估量的价值。
体细胞的重编程成功进一步体现了干细胞多能性的维持是多因子协同控制的结果。其中包括转录因子、染色质重塑酶、衔接蛋白和染色质结构调节蛋白等。许多肿瘤相关基因,如c-Myc、Klf4、STAT3等均在胚胎干细胞多能性维持上发挥重要功能,其中c-Myc和Klf4即属于Yamanaka因子参与iPS细胞的形成过程。CREPT(Cell-cycle Regulated andExpression-elevated Protein in Tumor)是近年申请人克隆并命名的一个新基因(GeneID:58490),它能够促进肿瘤的增殖和转移,是一个新的肿瘤相关基因。
发明内容
本发明所要解决的技术问题是如何制备多能干细胞及不能诱导为多能干细胞的细胞模型。
为解决上述技术问题,本发明首先提供了下述X1)或X2)的方法:
X1)多能干细胞的制备方法,包括:增加名称为动物细胞1的出发动物细胞中CREPT蛋白质的含量和/或活性,得到重组动物细胞,利用所述重组动物细胞制备多能干细胞;所述动物细胞1非干细胞;
X2)不能诱导为多能干细胞的细胞模型的制备方法,包括:降低名称为动物细胞2的出发动物细胞中所述CREPT蛋白质的含量和/或活性,得到不能诱导为多能干细胞的细胞模型;所述动物细胞2非干细胞且表达所述CREPT蛋白质。
所述动物细胞1和所述动物细胞2均为离体的动物细胞。
利用所述重组动物细胞制备多能干细胞可利用Yamanaka因子进行。Yamanaka因子为Oct4、Sox2、Klf4和c-Myc。
上述方法中,所述动物可为a1)或a2):
a1)哺乳动物;
a2)小鼠。
上述方法中,所述动物细胞1和所述动物细胞2均可为b1)或b2):
b1)体细胞;
b2)成纤维细胞。
上述方法中,所述CREPT蛋白质可为如下A1)、A2)或A3):
A1)氨基酸序列是序列3的蛋白质;
A2)将序列表中序列3所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的蛋白质;
A3)在A1)或A2)的N端或/和C端连接标签得到的融合蛋白质。
为了使A1)中的蛋白质便于纯化,可在由序列表中序列1所示的氨基酸序列组成的蛋白质的氨基末端或羧基末端连接上如下表所示的标签。

上述A2)中的CREPT蛋白质,为与序列3所示蛋白质的氨基酸序列具有75%或75%以上同一性且具有相同功能的蛋白质。所述具有75%或75%以上同一性为具有75%、具有80%、具有85%、具有90%、具有95%、具有96%、具有97%、具有98%或具有99%的同一性。
上述A2)中的CREPT蛋白质的编码基因可通过将序列2所示的DNA序列中缺失一个或几个氨基酸残基的密码子,和/或进行一个或几个碱基对的错义突变,和/或在其5′端和/或3′端连上上表所示的标签的编码序列得到。其中,序列2所示的DNA分子编码序列3所示的CREPT蛋白质。
上述方法中,所述增加名称为动物细胞1的出发动物细胞中CREPT蛋白质的含量和/或活性可通过向所述动物细胞1中导入所述CREPT蛋白质的编码基因实现;
所述降低名称为动物细胞2的出发动物细胞中所述CREPT蛋白质的含量和/或活性可通过降低或抑制所述动物细胞2中所述CREPT蛋白质的编码基因的表达实现。
上述方法中,所述降低或抑制所述动物细胞2中所述CREPT蛋白质的编码基因的表达可通过敲除所述动物细胞2中所述CREPT蛋白质的编码基因或其任一片段或向所述动物细胞2中导入抑制所述CREPT蛋白质的编码基因的物质实现。
所述降低或抑制所述动物细胞2中所述CREPT蛋白质的编码基因的表达具体可敲除所述动物细胞2中所述CREPT蛋白质的编码基因的外显子。所述CREPT蛋白质的编码基因的外显子可为所述CREPT蛋白质的编码基因的第三个外显子。
所述CREPT蛋白质的编码基因可为序列表中序列2所示的DNA分子,所述CREPT蛋白质的编码基因的基因组序列为序列表中序列1所示的DNA分子。序列1的第1-459位为第一个外显子的序列,第7500-7692位为第二个外显子的序列,第14910-15043位为第三个外显子的序列,第19423-19532位为第四个外显子的序列,第21591-21717位为第五个外显子的序列,第30156-30331位为第六个外显子的序列,第46434-49711位为第七个外显子的序列。
上述方法中,所述敲除所述CREPT蛋白质的编码基因或其任一片可段利用Cre-loxp方法实现。
所述Cre-loxp方法为利用Cre重组酶与LoxP位点抑制目的基因表达的方法。
所述抑制所述CREPT蛋白质的编码基因的物质可为降低所述CREPT蛋白质表达量的核酸分子。
本发明还提供了利用所述不能诱导为多能干细胞的细胞模型的制备方法制备的不能诱导为多能干细胞的细胞模型。
本发明还提供了所述CREPT蛋白质或与所述CREPT蛋白质相关的生物材料在调控动物细胞转化多能干细胞中的应用;所述动物细胞非干细胞;所述生物材料为下述B1)至B9)中的任一种:
B1)编码所述CREPT蛋白质的核酸分子;
B2)含有B1)所述核酸分子的表达盒;
B3)含有B1)所述核酸分子的重组载体、或含有B2)所述表达盒的重组载体;
B4)含有B1)所述核酸分子的重组微生物、或含有B2)所述表达盒的重组微生物、或含有B3)所述重组载体的重组微生物;
B5)含有B1)所述核酸分子的转基因动物细胞系、或含有B2)所述表达盒的转基因动物细胞系;
B6)含有B1)所述核酸分子的转基因动物组织、或含有B2)所述表达盒的转基因动物组织;
B7)含有B1)所述核酸分子的转基因动物器官、或含有B2)所述表达盒的转基因动物器官;
B8)降低所述CREPT蛋白质表达量的核酸分子;
B9)含有B8)所述核酸分子的表达盒、重组载体、重组微生物、转基因动物细胞系、转基因动物组织或转基因动物器官。
上述应用中,B1)所述核酸分子可为如下b11)或b12)或b13)或b14):
b11)编码序列是序列表中序列2的cDNA分子或DNA分子;
b12)序列表中序列1所示的DNA分子;
b13)与b11)或b12)限定的核苷酸序列具有75%或75%以上同一性,且编码所述CREPT蛋白质的cDNA分子或DNA分子;
b14)在严格条件下与b11)或b12)或b13)限定的核苷酸序列杂交,且编码所述CREPT蛋白质的cDNA分子或DNA分子。
其中,所述核酸分子可以是DNA,如cDNA、基因组DNA或重组DNA;所述核酸分子也可以是RNA,如mRNA或hnRNA等。
本领域普通技术人员可以很容易地采用已知的方法,例如定向进化和点突变的方法,对本发明的编码CREPT蛋白质的核苷酸序列进行突变。那些经过人工修饰的,具有与本发明分离得到的CREPT蛋白质的核苷酸序列75%或者更高同一性的核苷酸,只要编码CREPT蛋白质且具有CREPT蛋白质功能,均是衍生于本发明的核苷酸序列并且等同于本发明的序列。
这里使用的术语“同一性”指与天然核酸序列的序列相似性。“同一性”包括与本发明的编码序列3所示的氨基酸序列组成的蛋白质的核苷酸序列具有75%或更高,或85%或更高,或90%或更高,或95%或更高同一性的核苷酸序列。同一性可以用肉眼或计算机软件进行评价。使用计算机软件,两个或多个序列之间的同一性可以用百分比(%)表示,其可以用来评价相关序列之间的同一性。
上述应用中,所述严格条件可为如下:50℃,在7%十二烷基硫酸钠(SDS)、0.5MNaPO4和1mM EDTA的混合溶液中杂交,在50℃,2×SSC,0.1%SDS中漂洗;还可为:50℃,在7%SDS、0.5M NaPO4和1mM EDTA的混合溶液中杂交,在50℃,1×SSC,0.1%SDS中漂洗;还可为:50℃,在7%SDS、0.5M NaPO4和1mM EDTA的混合溶液中杂交,在50℃,0.5×SSC,0.1%SDS中漂洗;还可为:50℃,在7%SDS、0.5M NaPO4和1mM EDTA的混合溶液中杂交,在50℃,0.1×SSC,0.1%SDS中漂洗;还可为:50℃,在7%SDS、0.5M NaPO4和1mM EDTA的混合溶液中杂交,在65℃,0.1×SSC,0.1%SDS中漂洗;也可为:在6×SSC,0.5%SDS的溶液中,在65℃下杂交,然后用2×SSC,0.1%SDS和1×SSC,0.1%SDS各洗膜一次;也可为:2×SSC,0.1%SDS的溶液中,在68℃下杂交并洗膜2次,每次5min,又于0.5×SSC,0.1%SDS的溶液中,在68℃下杂交并洗膜2次,每次15min;也可为:0.1×SSPE(或0.1×SSC)、0.1%SDS的溶液中,65℃条件下杂交并洗膜。
上述75%或75%以上同一性,可为80%、85%、90%或95%以上的同一性。
上述应用中,B2)所述的含有编码CREPT蛋白质的核酸分子的表达盒(CREPT基因表达盒),是指能够在宿主细胞中表达CREPT蛋白质的DNA,该DNA不但可包括启动CREPT基因转录的启动子,还可包括终止CREPT基因转录的终止子。进一步,所述表达盒还可包括增强子序列。
可用现有的表达载体构建含有所述CREPT基因表达盒的重组载体。
上述应用中,所述载体可为质粒、黏粒、噬菌体或病毒载体。
上述应用中,所述微生物可为酵母、细菌、藻或真菌。
上述应用中,所述转基因动物细胞系、转基因动物组织和转基因动物器官均不包括繁殖材料。
所述动物可为a1)或a2):
a1)哺乳动物;
a2)小鼠。
所述动物细胞可为b1)或b2):
b1)体细胞;
b2)成纤维细胞。
在本发明的一个实施例中,所述成纤维细胞可为胚胎成纤维细胞。在本发明的另一个实施例中,所述成纤维细胞为鼠尾成纤维细胞。
本发明发现,降低成纤维细胞中CREPT的表达,成纤维细胞不能被4种Yamanaka因子诱导形成iPS细胞,在不表达CREPT的细胞中表达CREPT后,细胞能被诱导为iPS细胞。表明,CREPT及其编码基因可以用于调控细胞向iPS细胞的转化,可以用于制备不能诱导为多能干细胞的细胞模型,还可以用于制备iPS细胞。

具体实施方式
下面结合具体实施方式对本发明进行进一步的详细描述,给出的实施例仅为了阐明本发明,而不是为了限制本发明的范围。下述实施例中的实验方法,如无特殊说明,均为常规方法。下述实施例中所用的材料、试剂、仪器等,如无特殊说明,均可从商业途径得到。以下实施例中的定量试验,均设置三次重复实验,结果取平均值。下述实施例中,如无特殊说明,序列表中各核苷酸序列的第1位均为相应DNA的5′末端核苷酸,末位均为相应DNA的3′末端核苷酸。
实施例1、CREPT基因的敲除抑制MEF细胞转化为iPS细胞
本实施例发现CREPT基因(Gene ID:70470,NCBI(美国国立生物技术信息中心))在维持MEF细胞向iPS细胞的转化中起着重要的作用。CREPT基因的序列为序列表中序列1,序列1的第1-459位为第一个外显子的序列,第550-7499位为第一个内含子的序列,第7500-7692位为第二个外显子的序列,第7693-14909位为第二个内含子的序列,第14910-15043位为第三个外显子的序列,第15044-19422位为第三个内含子的序列,第19423-19532位为第四个外显子的序列,第19533-21590位为第四个内含子的序列,第21591-21717位为第五个外显子的序列,第21718-30155位为第五个内含子的序列,第30156-30331位为第六个外显子的序列,第30332-46433位为第六个内含子的序列,第46434-49711位为第七个外显子的序列,CREPT基因的CDS序列为序列表中序列2,编码序列3所示的CREPT蛋白质。
一、材料的准备
1、MEF细胞的制备
MEF细胞(Mouse Embryonic Fibroblast)均取自怀孕13.5天小鼠(C57BL/6N,北京维通利华实验动物技术有限公司)胚胎,经扩大培养至第3代后得到MEF细胞。
2、包装CREPT shRNA病毒的制备
根据addgene的说明书将CREPT的shRNA序列构建至慢病毒载体,shRNA靶序列为5’-GGACCTGAGTTCACAAGAGA-3’,得到重组慢病毒载体;将该重组病毒载体与包装质粒(addgene,慢病毒载体为pLL3.7(http://www.addgene.org/11795/);包装质粒为psPAX2(http://www.addgene.org/12260/)和pMD2.G(http://www.addgene.org/12259/))一起转染293T细胞,48小时后收集培养上清,即为包装CREPT shRNA的病毒上清。利用慢病毒载体与包装质粒一起转染293T细胞得到的上清液作为对照上清。
3、CREPTFlox/Flox小鼠的制备
所用CREPTFlox/Flox小鼠来源于英国MRC Harwell Institute,该小鼠通过基因编辑技术将LoxP位点整合到C57BL/6N小鼠的CREPT基因第3个外显子的两侧而得到。
将序列4所示的重组DNA分子电转入C57BL6/N小鼠的ES细胞,通过G418筛选获得重组正确的ES细胞克隆(即在C57BL6/N小鼠两条染色体中序列1所示CREPT基因的第8948-19283位间的DNA片段替换为重组DNA分子(其序列为序列表中序列4)),重组方式如图1,图1中,1-6分别代表CREPT基因的第一至第六个外显子。将重组正确的ES细胞克隆注射C57BL6/N小鼠囊胚,得到嵌合鼠,然后,将嵌合鼠与C57BL6/N小鼠交配,选取子代中含重组片段的小鼠,即为能够稳定遗传重组片段的小鼠,经过自交后获得纯合重组片段的小鼠,最后,将该小鼠与FRT工具鼠(即B6/JNju-H11em1Cin(CAG-FLPo)/Nju,清华大学动物房保存,来源于南京大学模式动物研究所,详细信息见http://www.nrcmm.cn/strain/2063)交配得到CREPTFlox /Flox小鼠。
其中,序列4的第1-5472位和第13323-17292位位同源臂序列,第5537-5584位和第12441-12488位为FRT位点,第10584-10617位、第12495-12528位和第13266-13299位为LoxP位点,第12945-13078位为CREPT基因第3个外显子。
最终,CREPTFlox/Flox小鼠为将C57BL6/N小鼠两条染色体中序列1所示CREPT基因的第8948-19283位间的DNA片段替换为序列表中序列5所示的重组DNA分子得到的小鼠。序列5的第1-5472位和第6419-10388位为同源臂序列,第5537-5584位为FRT位点,第5591-5624位和第6362-6395位为LoxP位点,第6041-6174位为CREPT基因第3个外显子。
4、CREPTFlox/Flox;ERTII-Cre+/+基因型的小鼠的制备
通过将CREPTFlox/Flox小鼠与EIIa-Cre工具鼠(即B6.FVB-TgN(EIIa-Cre)C5379Lmgd,E2a-Cre,清华大学动物房保存,来源于The Jackson Laboratory,详细信息见https://www.jax.org/strain/003724)交配繁殖鉴定选出CREPTFlox/Flox;ERTII-Cre+/+基因型的小鼠。
二、CREPT基因的敲除对iPS细胞形成的影响
1、利用C57BL/6J小鼠的MEF细胞诱导iPS细胞
1)细胞计数,将MEF细胞计数传代到明胶预处理过底部的6孔板中,每孔加入约4000个细胞,加入培养基(即向DMEM(Gibco,C11965500BT)中添加如下物质得到的培养基:10%(体积百分比)血清(Gibco,10091-148)、100μM巯基乙醇(SIGMA,M7522),各物质的浓度均为在培养基中的浓度)在37℃、5%CO2的细胞培养箱(SANYO)中进行培养,每孔加入培养基的体积为2mL,每组3个平行孔。
2)步骤1)完成并培养16小时后,将等体积的4种Yamanaka因子病毒上清与步骤一的包装CREPT shRNA的病毒上清液一起加入到六孔板中继续培养,即实验组。每孔中Oct4、Sox2、Klf4和c-Myc病毒上清的添加量为50μL,包装CREPT shRNA的病毒上清液的添加量为100μL。利用步骤一的对照上清同步实验作为对照组。
其中,4种Yamanaka因子病毒上清的制备方法如下:根据addgene的说明书将Yamanaka因子的编码基因构建至慢病毒载体pLL3.7,得到重组慢病毒载体;将该重组病毒载体与包装质粒psPAX2和pMD2.G一起转染293T细胞,48小时后收集培养上清,即为Yamanaka因子病毒上清。
3)步骤2)完成并培养24小时后更换新鲜培养基,以后每隔24小时更换一次培养基,所更换的新鲜培养基均为iPS细胞诱导培养基,iPS细胞诱导培养基通过向Knock outDMEM(Gibco,10829-018)中加入如下物质得到:20%(体积百分比)血清(Gibco,10091-148)、2mM GlutMAX(L-丙氨酰-L-谷氨酰胺二肽,Gibco,35050-061)、0.1mM NEAA(七种氨基酸,Gibco,11140-050,每种氨基酸在培养基中的浓度均为0.1mM)、100μM巯基乙醇(SIGMA,M7522)、10ng/mL LIF(Millipore,ESG1107),各物质的浓度均为在ES培养基中的浓度。
4)一周后即可观察到对照组中有iPS细胞开始形成,两周后iPS细胞克隆即清晰可见,而实验组中则只有很少iPS细胞形成。
5)碱性磷酸酶(AP)染色,碱性磷酸酶能够特异性将iPS细胞染色而无法染色成纤维细胞。将已经诱导形成的iPS细胞克隆用AP试剂盒染色。然后,将经AP染色的六孔板放置在倒置显微镜下,观察细胞克隆数,拍照。
结果(图2)显示,实验组中iPS细胞克隆占所有细胞克隆的百分比为9±2%,对照组中iPS细胞克隆占所有细胞克隆的百分比为100±9%,对照组中iPS细胞克隆占所有细胞克隆的百分比显著高于实验组。表明CREPT基因表达的降低可以抑制iPS细胞的形成。
2、利用CREPTFlox/Flox;ERTII-Cre+/+基因型小鼠的鼠尾成纤维细胞(TTF)细胞诱导iPS细胞
1)待CREPTFlox/Flox;ERTII-Cre+/+基因型小鼠出生10周后,至鼠尾端剪取0.5cm鼠尾提取TTF;
细胞计数,将CREPTFlox/Flox;ERTII-Cre+/+基因型小鼠的TTF计数传代到明胶预处理过底部的6孔板中,每孔加入约4000个细胞,加入培养基(即向DMEM(Gibco,C11965500BT)中添加如下物质得到的培养基:10%(体积百分比)血清(Gibco,10091-148)、100μM巯基乙醇(SIGMA,M7522),各物质的浓度均为在培养基中的浓度)在37℃、5%CO2的细胞培养箱(SANYO)中进行培养,每孔加入培养基的体积为2mL,每组3个平行孔。
2)步骤1)完成并培养16小时后,将等体积的4种Yamanaka因子病毒上清与TAM溶液(该溶液为将TAM(他莫昔芬,SIGMA公司,货号为T5648)溶解到无水乙醇中得到的TAM的浓度为5mM的溶液)一起加入到六孔板中,即CREPT表达抑制组。每孔中Oct4、Sox2、Klf4和c-Myc病毒上清的添加量为50μL,TAM溶液的添加量为2μL。
按照上述方法,将步骤1)完成并培养16小时后,将等体积的4种Yamanaka因子病毒上清与TAM溶液(该溶液为将TAM溶解到无水乙醇中得到的TAM的浓度为5mM的溶液)以及CREPT表达病毒上清一起加入到六孔板中,即恢复CREPT表达组。每孔中Oct4、Sox2、Klf4和c-Myc病毒上清的添加量为50μL,TAM溶液的添加量为2μL,CREPT表达病毒上清的添加量为100μL。
利用等体积无水乙醇替换溶TAM溶液同步进行的实验作为对照组。
3)步骤2)完成并培养24小时后更换新鲜培养基,以后每隔24小时更换一次培养基,所更换的新鲜培养基均为iPS细胞诱导培养基。
4)一周后即可观察到对照组中有iPS细胞开始形成,两周后iPS细胞克隆即清晰可见,CREPT表达抑制组则很少有iPS细胞形成,恢复CREPT表达组有iPS细胞开始形成。
5)碱性磷酸酶(AP)染色,将已经诱导形成iPS细胞克隆用AP试剂盒染色。然后,将经AP染色的六孔板放置在倒置显微镜下,观察细胞克隆数,拍照。
结果(图3)显示,对照组中iPS细胞克隆占所有细胞克隆的百分比为47±4%,CREPT表达抑制组中iPS细胞克隆占所有细胞克隆的百分比为4±3%,恢复CREPT表达组中iPS细胞克隆占所有细胞克隆的百分比为41±7%,对照组和恢复CREPT表达组中iPS细胞克隆占所有细胞克隆的百分比均显著高于CREPT表达抑制组。表明,抑制CREPT基因的表达导致iPS细胞不能形成,重新表达CREPT基因后能够恢复iPS细胞的形成。说明,CREPT基因可以调控iPS细胞的形成。

4.根据权利要求1-3中任一所述的方法,其特征在于:所述CREPT蛋白质为如下A1)、A2)或A3):
A1)氨基酸序列是序列3的蛋白质;
A2)将序列表中序列3所示的氨基酸序列经过一个或几个氨基酸残基的取代和/或缺失和/或添加且具有相同功能的蛋白质;
A3)在A1)或A2)的N端或/和C端连接标签得到的融合蛋白质。
5.根据权利要求1-4中任一所述的方法,其特征在于:
所述增加名称为动物细胞1的出发动物细胞中CREPT蛋白质的含量和/或活性通过向所述动物细胞1中导入所述CREPT蛋白质的编码基因实现;
所述降低名称为动物细胞2的出发动物细胞中所述CREPT蛋白质的含量和/或活性通过降低或抑制所述动物细胞2中所述CREPT蛋白质的编码基因的表达实现。
6.根据权利要求5所述的方法,其特征在于:所述降低或抑制所述动物细胞2中所述CREPT蛋白质的编码基因的表达通过敲除所述动物细胞2中所述CREPT蛋白质的编码基因或其任一片段或向所述动物细胞2中导入抑制所述CREPT蛋白质的编码基因的物质实现。
7.根据权利要求6所述的方法,其特征在于:所述敲除所述CREPT蛋白质的编码基因或其任一片段利用Cre-loxp方法实现。
8.利用权利要求1-7中任一所述方法制备的不能诱导为多能干细胞的细胞模型。
9.权利要求1-4中任一所述CREPT蛋白质或与所述CREPT蛋白质相关的生物材料在调控动物细胞转化多能干细胞中的应用;所述动物细胞非干细胞;所述生物材料为下述B1)至B9)中的任一种:
B1)编码所述CREPT蛋白质的核酸分子;
B2)含有B1)所述核酸分子的表达盒;
B3)含有B1)所述核酸分子的重组载体、或含有B2)所述表达盒的重组载体;
B4)含有B1)所述核酸分子的重组微生物、或含有B2)所述表达盒的重组微生物、或含有B3)所述重组载体的重组微生物;
B5)含有B1)所述核酸分子的转基因动物细胞系、或含有B2)所述表达盒的转基因动物细胞系;
B6)含有B1)所述核酸分子的转基因动物组织、或含有B2)所述表达盒的转基因动物组织;
B7)含有B1)所述核酸分子的转基因动物器官、或含有B2)所述表达盒的转基因动物器官;
B8)降低所述CREPT蛋白质表达量的核酸分子;
B9)含有B8)所述核酸分子的表达盒、重组载体、重组微生物、转基因动物细胞系、转基因动物组织或转基因动物器官。
10.根据权利要求9所述的应用,其特征在于:B1)所述核酸分子为如下b11)或b12)或b13)或b14):
b11)编码序列是序列表中序列2的cDNA分子或DNA分子;
b12)序列表中序列1所示的DNA分子;
b13)与b11)或b12)限定的核苷酸序列具有75%或75%以上同一性,且编码所述CREPT蛋白质的cDNA分子或DNA分子;
b14)在严格条件下与b11)或b12)或b13)限定的核苷酸序列杂交,且编码所述CREPT蛋白质的cDNA分子或DNA分子。
