Abstract
本发明涉及一种农药制剂中非登记成分的气相色谱‑质谱快速测定方法,属于农药质量检测技术领域。所述农药制剂中非登记成分的气相色谱‑质谱快速测定方法包括:标准溶液的配制;样品溶液的配制;空白溶液的配制;GC‑MS检测:将标准工作溶液和空白溶液分别注入GC‑MS,选择离子监测,扣除空白后各非登记成分的定量离子峰面积对其相应浓度进行回归分析,得标准工作曲线;在相同条件下将待测样品溶液注入GC‑MS,得到扣除空白后样品中各非登记成分的定量离子峰面积,代入标准工作曲线,求得样品中各非登记成分的含量。本发明的方法实现了对农药制剂中131种非登记成分进行高通量准确定性筛查和精确定量检测的能力,大大提高了检测种类和检测效率。

技术领域
本发明涉及一种农药制剂中非登记成分的气相色谱-质谱快速测定方法,属于农药质量检测技术领域。
背景技术
农药是重要的农业生产资料和救灾物资,对防治农业有害生物,保障农业生产、农民增收及农产品贮存具有不可或缺的作用。然而,近年来一些农药生产企业为了追求经济效益、降低生产成本、提高农药产品防治效果和逃避农药登记政策监管等,在农药制剂中擅自添加非登记成分,也称隐性成分或未经登记成分,已成为行业内“公开的秘密”和“潜规则”,严重制约了我国农药行业的可持续发展。福建省农产品质量安全检验检测中心尹兰香等随机检测了在福建省销售的23批啶虫脒产品,其中21.7%的农药检出非登记成分。深圳市宝安区无公害农产品监督检验站唐淑军等于2006-2009年抽样检测了深圳市宝安区内的83个农药产品,其中28.9%的农药检出非登记成分。浙江省农药检定管理所黄晓华等随机抽取了在浙江省销售的41批水稻用药,其中46.3%的农药检出非登记成分。农业部农药检定所袁善奎等报道,从2010~2015年农业部发布的16批次农药产品质量监督抽检结果也表明,检出含非登记成分的样品总数仍约占质量不合格样品总数的31%。
总体而言,农药制剂中擅自添加非登记成分呈现出以下三个特点,一是大部分样品中非登记成分的添加量明显高于其登记成分(或一种)的含量;二是擅自添加的非登记成分主要是一些速效性、高活性、广谱性、廉价性以及处于专利保护期或刚过保护期的农药,包括克百威、特丁硫磷等禁限用高剧毒农药;三是生物农药中擅自添加非登记成分的行为比化学农药的更加突出和严重。
农药制剂中的非登记成分除易引起药害事故外,还会加速病虫害产生抗药性,破坏生态环境,导致农产品农药残留超标,危害人畜健康安全,应该引起我国有关管理部门的高度重视,及时出台有关农药制剂中非登记成分限量的国家或行业标准,从限量标准上对违法犯罪分子产生强大的威慑力。
目前,农药制剂检测机构主要按照标签标称的产品标准对产品质量进行检测,再根据仪器上可能显示的异常信号对其他农药成分进行随机检测,检测的种类十分有限,检测的随机性和偶然性较大,缺乏系统全面性。
文献报道有关农药制剂中非登记成分的检测方法主要有气相色谱法、液相色谱法、气相色谱-质谱法、气相色谱-串联质谱法、液相色谱-串联质谱法和液相色谱-四极杆-飞行时间质谱法等,涉及检测参数主要针对某几种或某一类农药非登记成分。现行行业推荐标准NY/T2990-2016《禁限用农药定性定量分析方法》也仅包括了涕灭威、灭多威等24种禁限用农药,而目前我国注册登记的农药有效成分多达670余种,尚有数百种可能成为擅自添加的非登记成分的农药仍缺乏行之有效的检测方法。
事实上,农药中的非登记成分在确证之前是个未知物,具有极强的隐蔽性、随机性、多样性和复杂性,常规的气相色谱法、液相色谱法等色谱方法由于只能依靠保留时间进行定性定量分析,均不能高效且准确的检测所有可能擅自添加的农药成分,应用于农药中非登记成分的检测存在较大缺陷,而气相色谱-质谱法、液相色谱-串联质谱法等色谱-质谱联用方法兼顾色谱的高分离效能和质谱准确鉴定化合物结构的特点,非常适合农药中非登记成分的全面、准确、高效检测。
因此,建立基于质谱法的农药制剂中非登记成分高通量检测方法迫在眉睫,对于推动农药行业可持续发展,确保农业安全生产,降低农产品重大安全隐患,维护消费者身体健康,保护生态环境均具有重要意义。
发明内容
本发明要解决的第一个技术问题是提供一种农药制剂中非登记成分的气相色谱-质谱快速测定方法,该方法能快速测定农药制剂中131种非登记成分。
为解决上述第一个技术问题,本发明的一种农药制剂中非登记成分的气相色谱-质谱快速测定方法包括:
(1)标准溶液的配制:取待测非登记成分的标准品用乙腈溶解配制非登记成分的混合标准溶液;精确吸取适量的混合标准溶液,用乙腈逐级稀释成不同质量浓度的系列标准工作溶液;
(2)样品溶液的配制:取待测农药样品与乙腈混合均匀,再用乙腈稀释得到待测样品溶液;
(3)空白溶液的配制:除不称取样品外,按照步骤(2)进行;
(4)GC-MS检测:将步骤(1)中的系列标准工作溶液和步骤(3)中的空白溶液分别注入GC-MS,选择离子监测,以扣除空白后各非登记成分的定量离子峰面积对其相应浓度进行回归分析,得到标准工作曲线;在相同条件下将步骤(2)中的待测样品溶液注入GC-MS,选择离子监测,得到扣除空白后样品中各非登记成分的定量离子峰面积,代入标准工作曲线,求得样品中各非登记成分的含量。
优选的,步骤(1)所述混合标准溶液的浓度为5.0mg/L;步骤(2)所述混合均匀为涡旋振荡,优选涡旋振荡2min;所述稀释的倍数为2500倍。
优选的,所述步骤(2)为:准确称取1g待测农药样品于50mL容量瓶中,加入20mL乙腈涡旋振荡提取2min,用乙腈定容至刻度,混匀,吸取5mL提取液于8000r/min下离心5min,准确吸取1mL上清液于另一50mL容量瓶中,用乙腈定容至刻度,混匀,过0.22μm尼龙滤膜。
优选的,所述GC-MS的色谱柱为:Agilent VF-1701MS毛细管色谱柱;所述乙腈优选为色谱纯乙腈,乙腈的质量分数w≥99.9%。
优选的,所述GC-MS测定采用的色谱条件为:进样口温度:270℃,升温程序:70℃保持2min,以25℃/min升温至180℃,然后3℃/min升温至250℃,再以5℃/min升温至300℃,保持27min;进样模式:不分流进样;柱流速:1.0mL/min,载气:氦气,纯度≥99.999%;进样量:1μL;
所述质谱的条件为:电离方式:EI;电离能量:70eV;离子源温度:230℃;四极杆温度:150℃,接口温度:300℃,溶剂延迟:7.5min;扫描方式:选择离子监测模式。

优选的,所述方法的平均加标回收率为75.4%~109.8%,相对标准偏差均<6%,检出限为5.3~149.1mg/kg,定量限为17.8~497.0mg/kg。
本发明所述检出限是指方法能测出该物质时的最低浓度。本发明所述定量限是指方法能准确定量出该物质时最低的浓度。
有益效果:
(1)本发明的方法采用气相色谱-质谱法,有效解决了现有技术方法(气相色谱法、液相色谱法)只能依靠保留时间定性,易受农药制剂中其它共存成分干扰而造成假阳性或假隐性结果的问题,同时实现了对农药制剂中非登记成分进行高通量准确定性筛查和精确定量检测的能力。
(2)本发明的方法气相色谱-质谱法配合Agilent VF-1701MS中等极性毛细管色谱柱,可以同时满足有机磷类、有机氯类、拟除虫菊酯类等多种类别农药的良好分离,实现131种非登记成分的高通量检测,大大提高了检测种类和检测效率。
(3)本发明的方法采用高灵敏度和强抗干扰能力的选择离子监测模式进行检测,具有操作简单、快速、准确、灵敏度高及重复性好等显著优点,可快速检测农药制剂中擅自添加的非登记成分,为农药制剂的生产和质量监管提供可靠技术支撑,有效推动农药产业可持续发展。
(4)本发明的方法测定131种非登记成分的平均加标回收率为75.4%~109.8%,相对标准偏差均<6%,检出限为5.3~149.1mg/kg,定量限为17.8~497.0mg/kg,远远低于常量添加(质量分数0.1%以上)非登记成分含量,可很好满足农药制剂中非登记成分含量检测和质量监管需要。

为解决上述第一个技术问题,本发明的一种农药制剂中非登记成分的气相色谱-质谱快速测定方法如下:
具体步骤如下:
(1)标准溶液的配制:准确称取适量农药标准品,以色谱纯乙腈为溶剂,配制丙烯酰胺、氟嘧磺隆、异丁子香酚、虫螨畏、内吸磷、六氯苯、氟乐灵等131种农药的单一标准储备液,于4℃下保存,有效期6个月。将131种农药按保留时间分为A、B两组,分别移取适量农药单一标准储备液于10mL容量瓶中,乙腈定容至刻度,配制浓度为5.0mg/L的农药混合标准溶液。精确吸取适量的混合标准溶液,用乙腈逐级稀释成不同质量浓度的系列标准工作溶液。
(2)样品溶液的配制:准确称取1g农药样品(精确至0.001g)于50mL容量瓶中,加入20mL乙腈涡旋振荡提取2min,用乙腈定容至刻度,混匀,吸取5mL提取液于8000r/min下离心5min,准确吸取1mL上清液于另一50mL容量瓶中,用乙腈定容至刻度,混匀,过0.22μm尼龙滤膜,待GC-MS分析。根据样品中非登记成分的含量适当调整稀释倍数。
(3)空白溶液的配制:除不称取样品外,按照步骤(2)进行。
(4)GC-MS检测:将步骤(1)中的系列标准工作溶液和步骤(3)中的空白溶液分别注入GC-MS,选择离子监测,以扣除空白后各非登记成分的定量离子峰面积对其相应浓度进行回归分析,得到标准工作曲线;在相同条件下将步骤(2)中的待测样品溶液注入GC-MS,选择离子监测,得到扣除空白后样品中各非登记成分的定量离子峰面积,代入标准工作曲线,求得样品中各非登记成分的含量。
本发明所用提取稀释溶剂色谱纯乙腈的质量分数w≥99.9%。
同时检测的A组67种非登记成分为丙烯酰胺、氟嘧磺隆、异丁子香酚、虫螨畏、内吸磷、六氯苯、氟乐灵、五氯甲氧基苯、扑灭通、特丁硫磷、百治磷、扑灭津、γ-BHC、灭蝇胺、脱乙基另丁津、氟虫脲、五氯苯胺、叠氮津、乙草胺、艾氏剂、甲基嘧啶磷、莠灭净、呋草黄、噻节因、异丙甲草胺、马拉硫磷、仲丁灵、乙氧呋草黄、环氧七氯、啶斑肟、异戊乙净、2-甲-4-氯丁氧乙基酯、炔丙菊酯、1-萘乙酰胺、氯硫磷、烯草胺、丙硫磷、脱叶磷、啶氧菌酯、丙溴磷、噻嗪酮、己唑醇、杀螨酯、4,4-二溴二苯甲酮、丙酯杀螨醇、氧环唑、麦草氟异丙酯、苯虫醚、生物苄呋菊酯、苯霜灵、烯炔菊酯、丙环唑、丰索磷、酯菌胺、倍硫磷砜、噁霜灵、胺菊酯、戊唑醇、联苯肼酯、氟草敏、亚胺硫磷、氟丙菊酯、苯醚氰菊酯、氯菊酯、联苯三唑醇、咪酰胺、氟氰戊菊酯;B组64种非登记成分为4-氨基吡啶、苯磺隆、土菌灵、速灭磷、二硫氰基甲烷、四氯硝基苯、萘草胺、灭线磷、残杀威、氯苯胺灵、氟铃脲、氧乐果、丁基嘧啶磷、溴烯杀、胺丙畏、环萎隆、咯喹酮、烯丙苯噻唑、解草腈、乐果、丁脒酰胺、丙蝇驱、八氯苯乙烯、十二环吗啉、仲草丹、氯酞酸甲酯、倍硫磷、吡咪唑、异丙乐灵、生物烯丙菊酯、哌草丹、异氯磷、毒虫畏、甲苯氟磺胺、alpha-硫丹、灭螨猛、腐霉利、氟咯草酮、抑霉唑、格草净、麦草氟甲酯、噁唑磷、乙硫磷、beta-硫丹、噻螨酮、苯草醚、炔螨特、三氟苯唑、氟草烟-1-甲庚酯、苯腈膦、苯嗪草酮、丙硫菌唑、新燕灵、苯氧威、哌草磷、环戊噁草酮、叶菌唑、氧化萎锈灵、灭菌唑、哒螨灵、醚菊酯、双唑草腈、高效氯氰菊酯、啶酰菌胺。
GC-MS测定时的色谱条件为:色谱柱:AgiLent VF-1701MS毛细管色谱柱,进样口温度:270℃,升温程序:70℃保持2min,以25℃/min升温至180℃,然后3℃/min升温至250℃,再以5℃/min升温至300℃,保持27min;进样模式:不分流进样;柱流速:1.0mL/min,载气:氦气,纯度≥99.999%;进样量:1μL。
采用的质谱条件为:电离方式:EI;电离能量:70eV;离子源温度:230℃;四级杆温度:150℃,接口温度:300℃,溶剂延迟:7.5min;扫描方式:选择离子监测模式。
131种非登记成分的保留时间和选择离子监测参数见表1和表2。

GC-MS测定时,若扣除空白后样品中非登记成分色谱峰保留时间与标准溶液中相应非登记成分保留时间相一致,并且在扣除背景后的样品质谱图中,所选择的离子均出现,而且定量离子与定性离子的丰度比与标准溶液的离子丰度比相一致,即:相对丰度>50%,允许±10%偏差;相对丰度在20%~50%之间,允许±15%偏差;相对丰度在10%~20%之间,允许±20%偏差;相对丰度≤10%,允许±50%偏差,则可判断样品中存在这种非登记成分;若上述两个条件不能同时满足,则判断不含有该种非登记成分。

下面结合实施例对本发明的具体实施方式做进一步的描述,并不因此将本发明限制在所述的实施例范围之中。
实施例1
1.仪器与设备
Agilent 7890B-5977B型气相色谱-质谱联用仪;Eppendorf 5810R型高速冷冻离心机;Sartorius BP 211D型电子天平;XW-80A型漩涡混合器。
2.材料与试剂
农药标准品:丙烯酰胺、氟嘧磺隆、虫螨畏、内吸磷、六氯苯、氟乐灵、扑灭通、特丁硫磷、百治磷等131种农药标准品,购自于德国Dr.Ehrenstorfer公司。
试剂:色谱纯乙腈,购自于美国Sigma-Aldrich公司。
样品:市售农药样品A——桉油精(5%可溶液剂)。
3.标准溶液的配制:准确称取适量农药标准品,以色谱纯乙腈为溶剂,配制丙烯酰胺、氟嘧磺隆、异丁子香酚、虫螨畏、内吸磷、六氯苯、氟乐灵等131种农药的单一标准储备液,于4℃下保存,有效期6个月。将131种农药按保留时间分为A、B两组,分别移取适量农药单一标准储备液于10mL容量瓶中,乙腈定容至刻度,配制浓度为5.0mg/L的农药混合标准溶液。精确吸取适量的混合标准溶液,用乙腈逐级稀释成不同质量浓度的系列标准工作溶液。
4.样品前处理:
准确称取1g农药样品(精确至0.001g)于50mL容量瓶中,加入20mL乙腈涡旋振荡提取2min,用乙腈定容至刻度,混匀,吸取5mL提取液于8000r/min下离心5min,准确吸取1mL上清液于另一50mL容量瓶中,用乙腈定容至刻度,混匀,过0.22μm尼龙滤膜,待GC-MS分析。
5.空白试验:
除不称取样品外,按照上述步骤4进行。
6.测定方法:
将配制好的系列标准工作溶液和空白溶液分别注入GC-MS,选择离子监测,以扣除空白后各非登记成分的定量离子峰面积对其相应浓度进行回归分析,得到标准工作曲线;在相同条件下将待测样品溶液注入GC-MS,选择离子监测,得到扣除空白后样品中各非登记成分的定量离子峰面积,代入标准工作曲线,求得样品中各非登记成分的含量见表5。

在GC-MS测定时,采用的色谱条件为:色谱柱:Agilent VF-1701MS毛细管色谱柱,进样口温度:270℃,升温程序:70℃保持2min,以25℃/min升温至180℃,然后3℃/min升温至250℃,再以5℃/min升温至300℃,保持27min;进样模式:不分流进样;柱流速:1.0mL/min,载气:氦气,纯度≥99.999%;进样量:1μL。
采用的质谱条件为:电离方式:EI;电离能量:70eV;离子源温度:230℃;四级杆温度:150℃,接口温度:300℃,溶剂延迟:7.5min;扫描方式:选择离子监测模式。
131种非登记成分的保留时间和选择离子监测参数见表1和表2。
GC-MS测定时,若扣除空白后样品中非登记成分色谱峰保留时间与标准溶液中相应非登记成分保留时间相一致,并且在扣除背景后的样品质谱图中,所选择的离子均出现,而且定量离子与定性离子的丰度比与标准溶液的离子丰度比相一致(相对丰度>50%,允许±10%偏差;相对丰度在20%~50%之间,允许±15%偏差;相对丰度在10%~20%之间,允许±20%偏差;相对丰度≤10%,允许±50%偏差),则可判断样品中存在这种非登记成分;若上述两个条件不能同时满足,则判断不含有该种非登记成分。
实施例2
如实施例1所述,选择另一农药制剂样品B,测得样品中各非登记成分的含量见表6。

与实施例1相似,唯一不同的是:GC-MS测定采用的色谱条件为:进样口温度:270℃,升温程序:80℃保持0min,以40℃/min升温至200℃,保持2min,然后8℃/min升温至280℃,保持45min;进样模式:不分流进样;柱流速:1mL/min,载气:氦气,纯度≥99.999%;进样量:1μL;
所述质谱条件为:电离方式:EI;电离能量:70eV;离子源温度:230℃;四极杆温度:150℃,接口温度:280℃,溶剂延迟:4min;扫描方式:全扫描,扫描范围:35~450u。
对比例2
与实施例1相似,唯一不同的是:GC-MS测定采用的色谱条件为:进样口温度:270℃,升温程序:70℃保持1min,以20℃/min升温至200℃,保持5min,然后8℃/min升温至280℃,保持40min;进样模式:不分流进样;柱流速:1mL/min,载气:氦气,纯度≥99.999%;进样量:1μL;
所述质谱条件为:电离方式:EI;电离能量:70eV;离子源温度:230℃;四级杆温度:150℃,接口温度:280℃,溶剂延迟:4min;扫描方式:全扫描,扫描范围:40~450u。
对比例3
与对比例2相似,唯一不同的是:标准溶液的配制:131种农药不分组,分别移取适量农药单一标准储备液于10mL容量瓶中,乙腈定容至刻度,配制浓度为5.0mg/L的131种非登记成分混合标准溶液。
对比例1~3检测结果如表8、表9和图7~图11所示,可以看出:对比例1、对比例2等其他色谱与质谱条件下,A组67种非登记成分、B组64种非登记成分以及对比例3未分组131种非登记成分均不能进行良好的分离。

Claims (4)
1.农药制剂中非登记成分的气相色谱-质谱快速测定方法,其特征在于,所述方法包括:
(1)标准溶液的配制:取待测非登记成分的标准品用乙腈溶解配制非登记成分的混合标准溶液;精确吸取适量的混合标准溶液,用乙腈逐级稀释成不同质量浓度的系列标准工作溶液;所述混合标准溶液的浓度为5.0 mg/L;
(2)样品溶液的配制:取待测农药样品与乙腈混合均匀,再用乙腈稀释得到待测样品溶液;准确称取1g待测农药样品于50 mL容量瓶中,加入20 mL乙腈涡旋振荡提取2 min,用乙腈定容至刻度,混匀,吸取5mL提取液于8000 r/min下离心5min,准确吸取1mL上清液于另一50 mL容量瓶中,用乙腈定容至刻度,混匀,过0.22 μm尼龙滤膜;
(3)空白溶液的配制:除不称取样品外,按照步骤(2)进行;
(4)GC-MS检测:将步骤(1)中的系列标准工作溶液和步骤(3)中的空白溶液分别注入GC-MS,选择离子监测,以扣除空白后各非登记成分的定量离子峰面积对其相应浓度进行回归分析,得到标准工作曲线;在相同条件下将步骤(2)中的待测样品溶液注入GC-MS,选择离子监测,得到扣除空白后样品中各非登记成分的定量离子峰面积,代入标准工作曲线,求得样品中各非登记成分的含量;

3.根据权利要求1所述的农药制剂中非登记成分的气相色谱-质谱快速测定方法,其特征在于,将所述131种非登记成分分为A、B两组;分别进行步骤(1)~(4):

4.根据权利要求1~3任一项所述的农药制剂中非登记成分的气相色谱-质谱快速测定方法,其特征在于,所述方法的平均加标回收率为75.4%~109.8%,相对标准偏差均<6%,检出限为5.3~149.1mg/kg,定量限为17.8~497.0 mg/kg。
