Abstract
本发明提供一种用于提高种猪精液品质的中药组合物及其制备方法,涉及中药组合物生产技术领域。所述中药组合物由以下重量份的原料制成:枸杞子18‑22份、菟丝子19‑21份、黄芪13‑17份、覆盆子8‑12份、五味子2‑3份、仙灵脾8‑10份、车前子4‑6份、山茱萸7‑11份、黄精8‑10份、首乌7‑11份、金银花4‑8份、蔓荆子4‑8份、通草4‑6份、紫地丁2‑6份、沉香4‑8份、仙茅4‑8份、薤白2‑6份、墨旱莲2‑6份、糊精2‑4份。本发明克服了现有技术的不足,使制得的中药组合物具有滋阴补气、补肾壮阳、理气健脾、补血行血、清热解毒、抗衰老的功效,不仅能改善、提高精液品质,还能增强种公猪机体的免疫力,提高新陈代谢和生理机能,降低应激和协调内分泌的功能。

技术领域
本发明涉及中药组合物生产技术领域,具体涉及一种用于提高种猪精液品质的中药组合物及其制备方法。
背景技术
种猪是一个养猪场的核心,担负着母猪的供精繁殖任务,其质量的好坏对全场是否正常生产和经济效益是否得以提高具有重要的影响。在生产实践中,猪人工授精技术的应用越来越广泛,极大提高了公猪的利用率。通过人工授精的方式,采用精子密度仪对精液质量进行实时检测,可以提高受胎率,及时淘汰无价值公猪,保证猪场效益。然而保证公猪的精液品质,需要进行科学的饲养管理,采精不宜太早,频率不宜太高,否则会造成精液品质下降。不仅造成母猪不妊娠,而且直接影响后代。
多数养殖场通过在饲粮中添加一些中草药添加剂,用以改善精液品质,提高公猪利用率。中草药主要是调解阴阳、扶正祛邪,激发机体抵抗力从而提高免疫机能和繁殖机能,以现代营养学和饲养学为基础,研制成的复合型纯天然添加剂。为了改善精液品质、提高公猪利用率,中草药添加剂成为研究者们研究的热点。
发明内容
针对现有技术不足,本发明提供一种用于提高种猪精液品质的中药组合物及其制备方法,本发明克服了现有技术的不足,使制得的中药组合物具有滋阴补气、补肾壮阳、理气健脾、补血行血、清热解毒、抗衰老的功效,不仅能改善、提高精液品质,还能增强种公猪机体的免疫力,提高新陈代谢和生理机能,降低应激和协调内分泌的功能。
为实现以上目的,本发明的技术方案通过以下技术方案予以实现:
一种用于提高种猪精液品质的中药组合物,所述中药组合物由以下重量份的原料制成:枸杞子18-22份、菟丝子19-21份、黄芪13-17份、覆盆子8-12份、五味子2-3份、仙灵脾8-10份、车前子4-6份、山茱萸7-11份、黄精8-10份、首乌7-11份、金银花4-8份、蔓荆子4-8份、通草4-6份、紫地丁2-6份、沉香4-8份、仙茅4-8份、薤白2-6份、墨旱莲2-6份、糊精2-4份。

一种用于提高种猪精液品质的中药组合物的制备方法,所述中药组合物的制备方法包括以下步骤:
(1)将枸杞子、菟丝子、黄芪、覆盆子和五味子混合后用去离子水浸泡,后过滤得主原料a,并且将仙灵脾、车前子、山茱萸、黄精和首乌混合后用去离子水浸泡,后过滤得主原料b,将主原料a和主原料b混合后切碎,再将其混合物加入提取罐中,加热至沸腾并保持沸腾一段时间,后自然冷却至室温,重复提取两次,得主原料提取液备用;
(2)将金银花、蔓荆子、通草和紫地丁晒干后进行研磨,再加入1-2倍体积的去离子水进行冲泡,并通过水浴加热的方式进行保温提取,后过滤并静置取上清液得第一辅料提取液备用;
(3)将沉香、仙茅、薤白和墨旱莲用去离子水进行浸泡,浸泡后过滤并切成块状,再加入2-4倍体积的去离子水于超声波震荡仪中,升温并超声震荡后过滤得第二辅料提取液备用;
(4)将制得的主原料提取液、第一辅料提取液和第二辅料提取液混合并搅拌均匀,再将其混合液加入浓缩罐中进行浓缩,后将浓缩液加入发酵罐中,并加入异常汉逊酵母进行发酵,升温并发酵后过滤得组合液粗液备用;
(5)将制得的组合液粗液与3-5倍体积的乙醇混合并搅拌均匀,静置冷藏后通过蒸馏回收乙醇,再次浓缩至稠膏,采取大孔树脂法除去杂质,再加入糊精,搅拌均匀后通过制粒机制粒,再经流化床干燥后整粒得到产品。

本发明提供一种用于提高种猪精液品质的中药组合物及其制备方法,与现有技术相比优点在于:
(1)本发明采用枸杞子、菟丝子、黄芪、覆盆子、五味子、仙灵脾、车前子、山茱萸、黄精和首乌作为主原料,使制得的中药组合物具有滋阴补气、补肾壮阳、理气健脾和补血行血的功效,解决公猪高频度采精造成精液品质下降、性欲降低的问题,其中覆盆子、仙灵脾、山茱萸、黄精和首乌相互配合,能温肾助阳、促进精液分泌,并且抗衰老,从而有效改善精液品质,提高其生理机能,通过浸泡、切碎、沸腾提取的方式有效提高了主原料中有效成分的提取率,提取效果好,利用率高;
(2)本发明采用金银花、蔓荆子、通草和紫地丁等辅料,使制得的中药组合物具有清热解毒、提高新陈代谢并且协调内分泌的功能,通过晒干、研磨、冲泡、保温提取的方式有效保留了各组分中的精华,从而有效提高了种猪的舒适度和精液品质;
(3)本发明采用沉香、仙茅、薤白和墨旱莲等辅料,有效提高了种猪机体的免疫力,并且降低应激,该辅料与主原料相互配合,使制得的中药组合物温肾助阳的功效更为温和,防止药效过大不利于种猪的吸收,吸收效果好;
(4)本发明在制备的后期,通过将各提取液混合浓缩后进行发酵,能够在有效保留其中有益成分的同时,降低所得中药组合物中苦涩的口感,并且对其他大分子物质进行有效分解,从而便于种猪的吸收,有助消化,所得中药组合物效果显著,适宜推广。

实施例4:
以实施例二中药组合物的配方制备后作为以下公猪使用,下面是统计某猪场种公猪90头使用本发明中药组合物配方的临床统计资料:
一、一般资料
试验将某猪场90头日龄、体况相近的大约克种公猪,体重约80kg。每场随机分为3组,对照组饲喂基础日粮,试验1组饲喂基础日粮+0.2%中草药组合物(本品),试验2组饲喂基础日粮+0.3%中草药组合物,测定公猪精液量、精液pH值、精子畸形率、精液密度、精子活率、促卵泡生成素(FSH)、促黄体生成素(LH)和睾酮(T)含量等相关数据。公猪群体间差异均无统计学意义(P﹥0.05),具有可比性。
二、诊断评价标准
精液品质检查用手握法采集精液,用纱布过滤。
1、颜色和气味:正常精液为乳白色或灰白色,略有腥味。如有呈黄色(尿)、淡红色(血)、黄棕色(脓)及臭味者中任一种情况的判为不合格。
2、精液pH值:可用pH值试纸进行测定。正常精液呈弱碱性或中性,pH值在7.0-7.8。pH值>7.8者为不合格精液。
3、精子活率:用悬滴标本检查法,在恒温载物台将精液加热至35-37℃,在100-400倍显微镜下观察精子,直线前进运动的精子所占的百分率即表示精子的活率。采用十级制评分。新鲜采集的精液和新鲜稀释的精液活率要求不得低于0.7,保存24h以上的精液使用前检查活率不得低于0.6,否则为不合格精液。
4、采精量:将采集的精液,立即用4-6层消毒纱布滤除胶状物质,观察射精量。
5、在试验结束后,每头公猪采血4-5mL,静置后离心,分离血清,用化学发光免疫法测定种公猪促卵泡生成素(FSH)、促黄体生成素(LH)和睾酮(T)含量。
6、精子密度:指每毫升精液中所含的精子数,是确定稀释倍数的重要指标。用血细胞计数板进行计数或用精液密度仪测定。
7、精子畸形率:畸形率指异常精子的百分率。异常精子包括巨型精子、短小精子、双头或双尾精子、顶体膨胀或脱落、头部残缺或尾部分离、尾部变曲精子。采用显微镜观察法确定:精液涂片、染色(伊红或姬姆萨)后,在400-600倍显微镜下观察,计算畸形精子占所数精子总数的百分率。通常检查3个视野,计算500个以上精子。公猪精子畸形率超过20%不宜使用。要求每头公猪每2周检查1次精子畸形率。
三、结果分析

按统计分析采用SPSS15.0软件分析,结果以x±s组间采取t检验,计算资料用%表示,采用χ2分析进行统计学检验,以P﹤0.05及P﹤0.01具有统计学意义。
1、治疗方法
按照本发明的方法,对照组试验公猪每天饲喂基础日粮,试验1组公猪每天饲喂2%中草药添加剂(本品)+基础日粮,试验2组公猪每天饲喂0.3%的中草药组合物(本品)+基础日粮,一天一副,连用5-7天,一般10个月为一个疗程,随后定期检测公猪精液量、精液pH值、精子畸形率、精液密度、精子活率、促卵泡生成素(FSH)、促黄体生成素(LH)和睾酮(T)含量等相关数据。
2、结果统计
治疗效果分析:公猪精液质量指标测定结果如表1,公猪血清激素指标测定结果如表2,可知试验1组和2组较对照组提高精液质量的效果更佳。

4、典型案例
(1)荣昌县某猪场随机选择种公猪20头,随机分为试验组和对照组,每组各10头公猪。使用本发明中草药添加剂(本品)。预饲7天,然后进入正式试验阶段。试验组种公猪每天每头饲喂0.2%中草药添加剂(本品),连用10天,对照组不喂中草药添加剂。其它饲养管理条件和日粮组成两组完全相同,停止饲喂0.2%中草药添加剂(本品)后10天内,继续采精检查。试验组种公猪饲喂中草药添加剂后10-18天、19-30天射精量、精子密度有显著增加,精子活率明显提高,停止使用中草药添加剂后10天内,试验组公猪的精液量、精子密度、精子活率仍保持在一个较高的水平。
(2)蚌埠某猪场公猪阳痿30头,采用本发明中草药配方治疗(0.2%饲料中添加),共治愈28例,有效2例;治疗后公猪的精液量、精子密度、精子活率仍保持在一个较高的水平。通过本发明中药配方对治疗种公猪无精死精症有明显的效果。
(3)在福建省某猪场,将体重、体况、繁殖性能相近的大约克公猪20头随机分为2组,分别为对照组、试验组、每组各10头公猪。所有试验公猪均在同一栋猪舍中,为期2个月。试验期间猪舍内平均温度为20-24℃。对照组试验公猪饲喂基础日粮,试验组饲喂0.2%中草药添加剂(本品)+基础日粮。与对照组相比,试验组精液量增加了5.97%。试验组精子畸形率均低于对照组,降低了11.00%,试验组精子密度比对照组增加了20.90%,试验组精子活力增加了11.70%。
(4)河南省某猪场,从该猪场公猪群中随机选取60头健康公猪,体重约80kg。实验猪只在封闭式猪舍内饲养,保持猪舍内通风良好。试验期间自由采食和饮水,按猪场常规程序进行饲养管理。随机分为2组,分别为对照组、试验组、每组各30头公猪。对照组试验公猪饲喂基础日粮,试验组饲喂中草药添加剂(0.2%饲料中添加)+基础日粮,基础日粮中添加一定量的中草药添加剂能够显著降低公猪精子畸形率,试验组和与对照组相比精子畸形率降低25%,显著提高精子活力,试验组与对照组相比精子活力提高14.3%,显著提高精子密度,试验组和与对照组相比精子密度提高6.6%,显著提高精液量,试验组和对照组相比精液量提高7.28mL。

Claims (8)
1.一种用于提高种猪精液品质的中药组合物,其特征在于,所述中药组合物由以下重量份的原料制成:枸杞子18-22份、菟丝子19-21份、黄芪13-17份、覆盆子8-12份、五味子2-3份、仙灵脾8-10份、车前子4-6份、山茱萸7-11份、黄精8-10份、首乌7-11份、金银花4-8份、蔓荆子4-8份、通草4-6份、紫地丁2-6份、沉香4-8份、仙茅4-8份、薤白2-6份、墨旱莲2-6份、糊精2-4份。
2.根据权利要求1所述的一种用于提高种猪精液品质的中药组合物,其特征在于,所述中药组合物由以下重量份的原料制成:枸杞子20份、菟丝子20份、黄芪15份、覆盆子10份、五味子2.5份、仙灵脾9份、车前子5份、山茱萸9份、黄精9份、首乌9份、金银花6份、蔓荆子6份、通草5份、紫地丁4份、沉香6份、仙茅6份、薤白4份、墨旱莲4份、糊精3份。
3.一种用于提高种猪精液品质的中药组合物的制备方法,其特征在于,所述中药组合物的制备方法包括以下步骤:
(1)将枸杞子、菟丝子、黄芪、覆盆子和五味子混合后用去离子水浸泡,后过滤得主原料a,并且将仙灵脾、车前子、山茱萸、黄精和首乌混合后用去离子水浸泡,后过滤得主原料b,将主原料a和主原料b混合后切碎,再将其混合物加入提取罐中,加热至沸腾并保持沸腾一段时间,后自然冷却至室温,重复提取两次,得主原料提取液备用;
(2)将金银花、蔓荆子、通草和紫地丁晒干后进行研磨,再加入1-2倍体积的去离子水进行冲泡,并通过水浴加热的方式进行保温提取,后过滤并静置取上清液得第一辅料提取液备用;
(3)将沉香、仙茅、薤白和墨旱莲用去离子水进行浸泡,浸泡后过滤并切成块状,再加入2-4倍体积的去离子水于超声波震荡仪中,升温并超声震荡后过滤得第二辅料提取液备用;
(4)将制得的主原料提取液、第一辅料提取液和第二辅料提取液混合并搅拌均匀,再将其混合液加入浓缩罐中进行浓缩,后将浓缩液加入发酵罐中,并加入异常汉逊酵母进行发酵,升温并发酵后过滤得组合液粗液备用;
(5)将制得的组合液粗液与3-5倍体积的乙醇混合并搅拌均匀,静置冷藏后通过蒸馏回收乙醇,再次浓缩至稠膏,采取大孔树脂法除去杂质,再加入糊精,搅拌均匀后通过制粒机制粒,再经流化床干燥后整粒得到产品。
4.根据权利要求3所述的一种用于提高种猪精液品质的中药组合物的制备方法,其特征在于:步骤(1)中主原料a和主原料b的浸泡时间分别为8-12h和6-10h,沸腾时间为55-65min。
5.根据权利要求3所述的一种用于提高种猪精液品质的中药组合物的制备方法,其特征在于:步骤(2)中研磨至粒度为2-3mm,保温提取时间为1-2h,静置时间为2-4h。
6.根据权利要求3所述的一种用于提高种猪精液品质的中药组合物的制备方法,其特征在于:步骤(3)中浸泡时间为4-6h,升温至60-80℃,超声震荡仪的频率为20-30kHz,超声声强为1-2W/cm2,超声时间为20-30min。
7.根据权利要求3所述的一种用于提高种猪精液品质的中药组合物的制备方法,其特征在于:步骤(4)中浓缩至原体积的1/5,升温至80-100℃,发酵时间为6-10h。
8.根据权利要求3所述的一种用于提高种猪精液品质的中药组合物的制备方法,其特征在于:步骤(5)中乙醇的浓度为95%,冷藏温度为-15℃,冷藏时间为10-14h,浓缩至相对密度为1.3-1.4,干燥温度为60-80℃。
