技术领域
本发明涉及植物的新用途,特别涉及金樱子提取物在美白保湿抗衰老护肤品中的应用。
背景技术
随着生活水平的提高,现代人越来越重视皮肤的保养,俗话说,一白遮百丑,美白淡斑、保湿是女性对护肤的基本需求。
对比过去具有安全隐患并逐渐被淘汰的含铅汞、氢醌、曲酸的化学美白保湿剂,现代天然植物美白保湿剂有着更明显的优势。目前,针对临床及市场上使用的美白保湿成分及产品存在的相关问题,国内外企业不断加大在天然植物化妆品领域的研究和投入,从而顺应消费者对天然、健康、绿色产品的需求。
金樱子是蔷薇科植物金樱子(Rasa laevigata Michx.)的干燥成熟果实。其在临床上应用广泛,是卫生部颁布的保健食品之一。其味酸,甘,涩,性平,归肾,膀胱,大肠经,具有固精缩尿,固崩止带,涩肠止泻的功效,富含维生素C,胡萝卜素,氨基酸,无机盐,微量元素及多糖,黄酮,萜类,鞣质等多种物质,具有提高免疫力,降糖降脂,抗氧化,抗菌抗炎,抗肿瘤等作用,主治神经衰弱、肾虚不固、遗精、慢性肾炎、自汗盗汗、脾虚泄泻等,是古汉养生精、壮腰健肾丸、汇仁肾宝等常用中成药的主要原料之一。
随着科技的发展,金樱子还在生物制药、保健品、饮料、果酒等的开发研究中得到广泛应用。可用于生产各种保健饮料、制备果干、酸奶、酿酒、熬糖等。但目前市场上还未出现金樱子的化妆品,关于金樱子美白保湿作用也尚未有人研究。
黑色素是决定人体肤色的关键因素。皮肤受紫外线、遗传、内分泌、炎症介质、饮食不当等因素的影响,黑素代谢异常,皮肤黑素过快增长和分布不均,就会造成局部皮肤过黑及色素沉着,表现为黄褐斑、雀斑和炎症后色素沉着等。美白的核心是抑制皮肤黑色素的生成。减少黑色素的形成可采用抑制酪氨酸酶活性和清除自由基抗氧化的方式来完成。文献报道金樱子中含有VC、胡萝卜素、没食子酸、鞣花酸、儿茶素、槲皮素、芦丁等成分,这些成份是已确定可抑制酪氨酸酶活性,具有美白作用的成份。金樱子提取物、金樱子鞣质、金樱子多糖、金樱子黄酮类成份均具有很强的抗氧化功能。金樱子籽油也富含多种营养成分,不饱和脂肪酸含量高达86%,主要为亚油酸和α-亚麻酸,具有很强的抗氧化活性。从天然产物中提取的多糖具有营养和优异的保湿作用,如透明质酸、玉竹多糖、杏鲍菇多糖、红枣多糖、芦荟多糖、细柱五加叶多糖、白及多糖、茯苓多糖等。而金樱子中亦富含丰富的多糖类成份,也具有优异的保湿作用。
发明内容
本发明的目的之一是提供金樱子提取物在美白保湿抗衰老护肤品中的应用,金樱子提取物为金樱子籽浸膏、金樱子果肉浸膏、金樱子籽油、金樱子精多糖。本发明的另一目的在于提供金樱子提取物的制备方法。
为实现以上目的,本发明采用的技术方案为:金樱子提取物在美白保湿抗衰老护肤品中的应用。
优选的,所述的金樱子是蔷薇科植物金樱子Rasa laevigata Michx的干燥成熟果实。
优选的,所述的金樱子提取物在护肤品中以质量百分数计为:金樱子籽浸膏0.2-2%,金樱子果肉浸膏0.2-2%,金樱子籽油1-4%,金樱子精多糖1-2%。
进一步,所述的金樱子提取物的制备方法:金樱子籽浸膏和金樱子果肉浸膏的制备方法:取金樱子全果干燥,将果肉和籽分开,分别打粉过筛 ,然后在籽和果肉中分别加入10-20倍的60-80%乙醇,采用水浴回流提取,最后真空干燥得到金樱子籽浸膏和金樱子果肉浸膏;金樱子籽油的制备方法:取金樱子籽加入10倍的石油醚,放入索氏提取器中提取,真空抽滤,回收溶剂,真空干燥至恒重得金樱子籽初榨油,最后金樱子籽初榨油经水化脱胶、碱液脱酸、活性炭脱色、真空脱臭后得到金樱子籽油;金樱子精多糖的制备方法:取金樱子果肉加入20倍去离子水,超声,重复操作三次合并滤液,减压浓缩,冷却后加入3倍量的无水乙醇,经醇沉,离心,沉淀干燥得到金樱子精多糖。
所述的水浴回流提取,温度为75-79℃,时间为1-2h,过滤,重复操作三次,合并提取液减压浓缩至相对密度1.3-1.5。
所述放入索氏提取器中提取的温度为70-80℃,时间为3-4h。
所述超声的温度为70-80℃,时间为90-100min;减压浓缩至体积的1/10;醇沉时间为24-28h,离心时间为3-8min,转速为6000-7000r/min。
本发明还包括一种美白保湿抗衰老护肤品,其特征在于,其有效成分包括金樱子提取物,其中金樱子提取物为金樱子籽浸膏、金樱子果肉浸膏、金樱子籽油、金樱子精多糖。
优选的,所述的美白保湿抗衰老护肤品,其组分及以质量百分数计为:金樱子籽油3.5%、凡士林2%、荷荷巴油1%、乳木果油5%、蜂蜡2%;聚山梨醇酯801.5%、甘油2%、1,3-丁二醇3%、金樱子籽浸膏0.5%、金樱子精多糖1%、纯净水77.5%;防腐剂0.5%、香精0.5%。
所述制备的美白保湿抗衰老护肤品包括但不限于爽肤水、乳液、精华素、面霜。
采用上述技术方案,本发明所述的金樱子提取物在美白保湿抗衰老护肤品中的应用,金樱子提取物经试验表明具有很强的美白保湿抗氧化防衰老的作用;可将之与其他化妆品用辅料与添加剂混合制成各种皮肤护理的化妆品组合物及各种剂型,具有实际应用价值:是一种天然无污染的植物美白保湿抗衰老提取物,对人体无副作用,在美白保湿抗衰老化妆品领域中有广泛应用前景。
本发明的有益效果:(1)金樱子籽浸膏和金樱子果肉浸膏可有效抑制酪氨酸酶的活性及黑色素的合成,可以用于制备皮肤美白的外用化妆品组合物;(2)金樱子籽油不饱和脂肪酸含量高达86%,主要为亚油酸和α-亚麻酸,具有很强的抗氧化防衰老美白保湿的作用,可作为油相制备外用化妆品组合物;(3)金樱子精多糖具有优良的保湿作用,可以用于制备皮肤保湿的外用化妆品组合物;(4)在制备金樱子籽浸膏和金樱子果肉浸膏的过程中采用的水浴回流法,提取效率高,原料利用率高;(5)在制备金樱子籽油的过程中采用的索氏提取法,完成一次溶剂提取后,溶剂被加热,挥发的溶剂经过冷凝管冷凝,重新进入提取的环境,开始新的一次提取,这样能节省溶剂;且提取的完成度高,原料的利用率也高。

具体实施方式
下面结合附图和具体实施方式,对本发明做进一步说明。为了使本领域技术人员更好地理解本发明的技术方案,下面对本发明进行详细描述,本部分的描述仅是示范性和解释性,不应对本发明的保护范围有任何的限制作用。
1、金樱子提取物抑制酪氨酸酶活性研究
取金樱子全果干燥,将果肉和籽分开,分别打粉过40目筛。
籽分别加20倍纯水、20%乙醇、40%乙醇、60%乙醇、80%乙醇、纯乙醇水浴回流提取2h,过滤,重复操作三次,合并提取液减压浓缩,真空干燥得6种金樱子籽提取物。
果肉分别加20倍纯水、20%乙醇、40%乙醇、60%乙醇、80%乙醇、纯乙醇水浴回流提取2h,过滤,重复操作三次,合并提取液减压浓缩,真空干燥得6种金樱子果肉提取物。
分别称取金樱子提取物各0.1000g,分别溶于20ml二甲基亚砜,得到5mg/ml母液,再对倍稀释到2.500、1.250、0.625、0.312、和0.156mg/ml。按下表配制总反应体系为5ml。

依次加入Ph=6.8的磷酸盐缓冲液,不同浓度梯度的样品液和酪氨酸酶,在37℃水浴10min,加入底物L-酪氨酸,计时,20min时测定在475nm波长下的吸光值。
测定时以相应的阴性液做参照,酪氨酸酶抑制率公式如下:
抑制率=(A-B)/A*100
其中A为标准对照的吸光值,B为样品液的吸光值。
由图1可知,金樱子籽提取物抑制酪氨酸酶的活性高于金樱子果肉提取物,醇提物抑制酪氨酸酶活性高于水提物,60%醇提金樱子籽提取物体抑制酪氨酸酶活性最高。
2、金樱子提取物保湿活性研究
金樱子果肉提取物制备:果肉加20倍去离子水70℃超声90min,重复操作三次合并滤液,减压浓缩,干燥即得。
金樱子全果提取物制备:金樱子全果加20倍去离子水70℃超声90min,重复操作三次合并滤液,减压浓缩,干燥即得。
金樱子精多糖制备:果肉加20倍去离子水70℃超声90min,重复操作三次合并滤液,减压浓缩至体积的1/10,冷却后加入3倍量的无水乙醇,醇沉24h,于6000r/min离心3min,沉淀干燥得金樱子多糖。
在20℃恒温条件下,分别测定金樱子果肉和金樱子全果提取物、金樱子精多糖、甘油在RH42%和RH80%的环境中48h的吸湿率,吸收率的公式如下:
吸湿率(%)=(A-B)/A*100
其中A为样品实始质量,B为放置n小时后的样品质量。
从图2,3中可以看出,金樱子精多糖的吸湿效果优于金樱子果肉和全果提取物,且在低湿度环境下,吸湿性能更好。
将吸湿达到饱和的金樱子果肉和金樱子全果提取物、金樱子精多糖在温度为20℃,在放置了干硅胶的恒温恒湿箱内继续放置,24h后精确称取各试样质量,由试样前后的质量差求保湿率,保湿率的公式如下:
保湿率=Hn/Ho×100%
式中Ho为试样含水量,Hn为放置24h后试样含水量。
48h后保湿率见下表:

从上表可知,金樱子精多糖保湿率在第24h时是最高的。
3、安全性实验
参照化妆品卫生规范(2007版)中人体封闭式斑贴实验方法测试产品的安全性。
1)实验材料与受试者
受试物是金樱子护肤品终产品。阴性对照为空白对照。受试者共30人,男0人,女30人,年龄19-51岁,符合受试者志愿者人选标准。
2)实验方法
选用合格的斑试器,以封闭式斑贴试验方法,将受试物0.03g涂于斑器内,外用胶带贴敷于受试者背部,24h去除受试者,分别于斑贴试验0.5、24、48h观察皮肤反应,根据《化妆品卫生规范》中皮肤反应分级标准记录其结果:

人体斑贴试验检验受试物引起人体皮肤不良反应的可能性。30例受试者中出现2级皮肤不良反应的人数多于2例或出现任何1例3级或3级以上皮肤不良反应时,判定受试物对人体有不良反应,从表3中可以看出,本实验结果显示30人中1例出现2级反应,根据《化妆品卫生规范》中规范,该受试物对人体皮肤未引起不良反应。通过本试验结果可以得出金樱子护肤品的人体安全性很高,对人体皮肤未引起不良反应。
实施例1:
一种金樱子美白保湿抗衰老乳液,其组分及以质量百分数计为:A相:金樱子籽油3.5%、凡士林2%、荷荷巴油1%、乳木果油5%、蜂蜡2%;B相:聚山梨醇酯801.5%、甘油2%、1,3-丁二醇3%、金樱子籽浸膏0. 5%、金樱子精多糖1%、纯净水77.5%;C相:防腐剂0.5%、香精0.5%。
金樱子提取物的制备方法:取金樱子籽干燥,打粉过筛,然后在籽中分别加入20倍的60%乙醇,采用75℃水浴回流提取1h,过滤,重复操作三次,合并提取液减压浓缩至相对密度1.5,最后真空干燥得金樱子籽浸膏;取金樱子籽加入10倍石油醚,放入索氏提取器中提取,在80℃温度下提取3h,真空抽滤,回收溶剂,真空干燥至恒重得金樱子籽初榨油,最后金樱子初榨油经水化脱胶、碱液脱酸、活性炭脱色、真空脱臭后得到金樱子籽油;取金樱子果肉加20倍去离子水,在70℃温度下超声90min,重复操作三次合并滤液,减压浓缩至体积的1/10,冷却后加入3倍量的无水乙醇,经醇沉24h,离心3min,转速为6000r/min,沉淀干燥得金樱子精多糖。
一种金樱子美白保湿抗衰老乳液的制备方法,其工艺过程是:将A相混合并加热至85℃,搅拌均匀,再将B相混合并加热至75℃,搅拌均匀;将A相加入B相并搅拌至乳化完全,降温至45℃,最后加入C相并搅拌均匀,得到了金樱子美白保湿抗衰老乳液。
通过实施例1得到的金樱子美白保湿抗衰老乳液是一种无污染纯天然,具有美白保湿抗衰老功能,对人体无副作用的乳液护肤品。
实施例2:
一种金樱子美白保湿抗衰老爽肤水,其组分及以质量百分数计为:黄瓜提取液4%、芦荟汁25%、金樱子籽浸膏2%、金樱子精多糖2%、矿泉水67%。
金樱子提取物的制备方法:取金樱子籽干燥,打粉过筛,然后在籽中分别加入10倍的80%乙醇,采用79℃水浴回流提取2h,过滤,重复操作三次,合并提取液减压浓缩至相对密度1.3,最后真空干燥得金樱子籽浸膏;取金樱子果肉加20倍去离子水,在80℃温度下超声100min,重复操作三次合并滤液,减压浓缩至体积的1/10,冷却后加入3倍量的无水乙醇,醇沉28h,离心8min,转速为7000r/min,沉淀干燥得金樱子精多糖。
一种金樱子美白保湿抗衰老爽肤水的制备方法,其工艺过程是:先将矿泉水煮沸,放至温度为35℃,再将黄瓜提取液、芦荟汁、金樱子籽浸膏、金樱子精多糖混合,搅拌均匀,过滤,最后加入矿泉水得到金樱子美白保湿抗衰老爽肤水。
通过实施例2得到的金樱子美白保湿抗衰老爽肤水是一种无污染纯天然,具有美白保湿功能,对人体无副作用的爽肤水护肤品。
实施例3:
一种金樱子美白保湿抗衰老精华素,其组分及以质量百分数计为:肌肽1%、乙酰基六肽-8液体1%、葡聚糖0.5%、透明质酸钠0.5%、增稠剂1%、精氨酸0.5%、金樱子籽浸膏0.2%、金樱子果肉浸膏0.2%、金樱子精多糖1.5%、金樱子籽油1%、防腐剂1%、香精0.05%、去离子水91.55%。
金樱子提取物的制备方法:取金樱子全果干燥,将果肉和籽分开,分别打粉过筛,然后在籽和果肉中分别加入15倍的70%乙醇,采用77℃水浴回流提取1.5h,过滤,重复操作三次,合并提取液减压浓缩至相对密度1.4,最后真空干燥得金樱子籽浸膏和金樱子果肉浸膏;取金樱子籽加入10倍石油醚,放入索氏提取器中提取,在75℃温度下提取3.5h,真空抽滤,回收溶剂,真空干燥至恒重得金樱子籽初榨油,最后金樱子初榨油经水化脱胶、碱液脱酸、活性炭脱色、真空脱臭后得到金樱子籽油;取金樱子果肉加20倍去离子水,在75℃温度下超声95min,重复操作三次合并滤液,减压浓缩至体积的1/10,冷却后加入3倍量的无水乙醇,经醇沉26h,离心5min,转速为6500r/min,沉淀干燥得金樱子精多糖。
一种金樱子美白保湿抗衰老精华素的制备方法,其工艺过程是:将葡聚糖、透明质酸钠增稠剂溶于去离子水中,升温至75℃,分散后作为A相备用;将肌肽、乙酰基六肽、精氨酸、金樱子籽浸膏0.2%、金樱子果肉浸膏0.2%、金樱子精多糖1.5%、金樱子籽油4%、防腐剂1%、香精0.05%作为B相备用;开启搅拌器,搅拌速度250r/min,温度控制在75℃,将B相加入A相中,均质2000r/min,保持3-5min,之后再持续搅拌15min,开始冷却,冷却速度控制在1℃/min,冷却至40℃时,停止搅拌,即可得到金樱子美白保湿抗衰老精华素。
通过实施例3得到的金樱子美白保湿抗衰老精华素是一种无污染纯天然,具有美白保湿抗衰老功能,对人体无副作用的精华素护肤品。
实施例4:
一种金樱子美白保湿抗衰老面霜,其组分及以质量百分数计为:金樱子籽油4%,金樱子精多糖2%,简易乳化剂1%,甘油10%,化妆品抗菌剂0.5%,去离子水82.5%。
金樱子提取物的制备方法:取金樱子籽加入10倍石油醚,放入索氏提取器中提取,在76℃温度下提取3h,真空抽滤,回收溶剂,真空干燥至恒重得金樱子籽初榨油,最后金樱子初榨油经水化脱胶、碱液脱酸、活性炭脱色、真空脱臭后得到金樱子籽油;取金樱子果肉加20倍去离子水,在76℃温度下超声98min,重复操作三次合并滤液,减压浓缩至体积的1/10,冷却后加入3倍量的无水乙醇,经醇沉25h,离心3min,转速为6000r/min,沉淀干燥得金樱子精多糖。
一种金樱子美白保湿抗衰老面霜的制备方法,其工艺工程是:将金樱子籽油、金樱子精多糖、简易乳化剂、甘油和去离子水混合在一起,在35℃温度下搅拌,时间为30min,搅拌完成,最后加入化妆品抗菌剂,再搅拌10min,即可得到金樱子美白保湿抗衰老面霜。
通过实施例4得到的金樱子美白保湿抗衰老面霜是一种无污染纯天然,具有美白保湿抗衰老功能,对人体无副作用的面霜护肤品。

1.金樱子提取物在制备护肤品中的应用,其特征在于,所述金樱子提取物为金樱子籽浸膏、金樱子果肉浸膏、金樱子籽油、金樱子精多糖,金樱子提取物在护肤方面具有美白保湿抗衰老的作用;所述的金樱子提取物在护肤品中以质量百分数计为:金樱子籽浸膏0.2-2%,金樱子果肉浸膏0.2-2%,金樱子籽油1-4%,金樱子精多糖1-2%;所述金樱子籽浸膏和金樱子果肉浸膏的制备方法:取金樱子全果干燥,将果肉和籽分开,分别打粉过筛,然后在籽和果肉中分别加入10-20倍的60-80%乙醇,采用水浴回流提取,最后真空干燥得到金樱子籽浸膏和金樱子果肉浸膏;所述金樱子籽油的制备方法:取金樱子籽加入10倍的石油醚,放入索氏提取器中提取,真空抽滤,回收溶剂,真空干燥至恒重得金樱子籽初榨油,最后金樱子籽初榨油经水化脱胶、碱液脱酸、活性炭脱色、真空脱臭后得到金樱子籽油;所述金樱子精多糖的制备方法:取金樱子果肉加入20倍去离子水,超声,重复操作三次合并滤液,减压浓缩,冷却后加入3倍量的无水乙醇,经醇沉,离心,沉淀干燥得到金樱子精多糖。
2.根据权利要求1所述的金樱子提取物在制备护肤品中的应用,其特征在于,所述的水浴回流提取,温度为75-79℃,时间为1-2h,过滤,重复操作三次,合并提取液减压浓缩至相对密度1.3-1.5。
3.根据权利要求1所述的金樱子提取物在制备护肤品中的应用,其特征在于,所述放入索氏提取器中提取的温度为70-80℃,时间为3-4h。
4.根据权利要求1所述的金樱子提取物在制备护肤品中的应用,其特征在于,所述超声的温度为70-80℃,时间为90-100min;减压浓缩至体积的1/10;醇沉时间为24-28h,离心时间为3-8min,转速为6000-7000r/min。
5.一种美白保湿抗衰老护肤品,其特征在于,其有效成分包括金樱子提取物,其中金樱子提取物为金樱子籽浸膏、金樱子果肉浸膏、金樱子籽油、金樱子精多糖;所述的金樱子提取物在护肤品中以质量百分数计为:金樱子籽浸膏0.2-2%,金樱子果肉浸膏0.2-2%,金樱子籽油1-4%,金樱子精多糖1-2%;所述金樱子籽浸膏和金樱子果肉浸膏的制备方法:取金樱子全果干燥,将果肉和籽分开,分别打粉过筛,然后在籽和果肉中分别加入10-20倍的60-80%乙醇,采用水浴回流提取,最后真空干燥得到金樱子籽浸膏和金樱子果肉浸膏;所述金樱子籽油的制备方法:取金樱子籽加入10倍的石油醚,放入索氏提取器中提取,真空抽滤,回收溶剂,真空干燥至恒重得金樱子籽初榨油,最后金樱子籽初榨油经水化脱胶、碱液脱酸、活性炭脱色、真空脱臭后得到金樱子籽油;所述金樱子精多糖的制备方法:取金樱子果肉加入20倍去离子水,超声,重复操作三次合并滤液,减压浓缩,冷却后加入3倍量的无水乙醇,经醇沉,离心,沉淀干燥得到金樱子精多糖;所述金樱子精多糖的制备方法:取金樱子果肉加入20倍去离子水,超声,重复操作三次合并滤液,减压浓缩,冷却后加入3倍量的无水乙醇,经醇沉,离心,沉淀干燥得到金樱子精多糖。
6.根据权利要求5所述的一种美白保湿抗衰老护肤品,其特征在于,其组分及以质量百分数计为:金樱子籽油3.5%、凡士林2%、荷荷巴油1%、乳木果油5%、蜂蜡2%;聚山梨醇酯801.5%、甘油2%、1,3-丁二醇3%、金樱子籽浸膏0.25%、金樱子果肉浸膏0.25%、金樱子精多糖1%、纯净水77.5%;防腐剂0.5%、香精0.5%。
