Abstract
本发明的目的在于提供一种可再生的铜载体抗癌试剂,即通过膜通透性铜离子载体2,2'‑二硫二吡啶(DPY),将二价铜离子(Cu2+)运载至细胞内,在细胞内释放出Cu2+,同时生成二巯基吡啶(DMe),DMe在细胞内氧化的条件下产生新的DPY,达到可再生、可循环运载铜离子的目的。在此过程中提升细胞内的铜和氧化应激水平,打破癌细胞氧化还原适应的阈值和铜水平差异的治疗窗,实现选择性地诱导癌细胞死亡。

技术领域
本发明涉及一种可再生的铜载体抗癌试剂及其在抗肿瘤中的用途。属于医药领域。
背景技术
癌症是当今社会危害人类健康和生命的重大疾病之一。癌症的治疗不仅是医学的一大挑战,而且也导 致了严重的社会与经济问题。近年来,利用癌细胞与正常细胞不同的生化特征实现恶性肿瘤的化学药物治 疗备受学者关注。
癌细胞与正常细胞相比,表现出增加的活性氧(ROS)水平以维持其恶性表型增长,从而更依赖于“氧 化还原适应”的机制,即将ROS水平保持在允许其逃脱死亡的范围内(NatRev Drug Discov.,2013,12(12): 931-947)。基于此种生化差异,当抗癌药物进一步提升细胞内的ROS水平,超越癌细胞的临界“毒性阈 值”时便可引发其死亡(Antioxid RedoxSignal.,2017,26(6):262-273.),而正常细胞由于基础ROS水平 低,故可以抵抗此范围内的ROS刺激,免受死亡。基于此特点,近年来通过靶向调节细胞内的主要抗氧 化系统硫氧还蛋白(Trx)系统和谷胱甘肽(GSH)系统,从而调控细胞内的ROS水平,诱导癌细胞死亡, 成为抗癌药物发展的策略之一(Trends Pharmacol Sci.,2017,38(9):794-808)。此外,癌细胞中的铜水 平与正常细胞相比显著增高(Cancer Treat Rev.,2009,35(1):32-46),当细胞内的铜浓度被增加时,正常 细胞和癌细胞之间的治疗窗使得铜离子载体抗癌药物发展成为可能(Chem Rev.,2014,114(1):815-62)。 更重要的是,由于临床主流抗癌药物铂类的毒副作用及抗药性等问题,使得用铜代替铂类药物中的铂成为 金属类抗癌药物研发的趋势之一(Metallomics,2015,7(11):1459-1476)。
靶向细胞氧化还原系统的铜类抗癌药虽有报道(Cancer Treat Rev.,2009,35(1):32-46),但利用癌细 胞特殊的氧化还原调控机制,结合铜离子载体将铜循环地运载至细胞内,以氧化应激介导的细胞凋亡方式 选择性诱导癌细胞死亡的研究目前尚未见报道。
发明内容
本发明的第一目的在于提供一种可再生的铜载体抗癌试剂,即膜通透性铜离子载体2,2'-二硫二吡啶 (DPY),将二价铜离子(Cu2+)运载至细胞内,在细胞内释放出Cu2+,同时生成DMe,DMe在细胞内 氧化的条件下产生新的DPY,达到可再生、可循环运载铜离子的目的。在此过程中提升细胞内的铜和氧化 应激水平,打破癌细胞氧化还原适应的阈值和铜水平差异的治疗窗,实现选择性地诱导癌细胞死亡。
本发明的第二目的是DPY与Cu2+协同的抗癌作用的应用。
本发明的第三目的是提供DPY与Cu2+形成的一新配合物。

本发明考察了CuCl2自身对肿瘤细胞的毒性以及它与DPY共同作用的最佳浓度。如图2A所示,CuCl2在浓度0-50μM之间作用HepG2细胞24h或者48h后,均对细胞几乎无毒性,但是当CuCl2浓度增加到 100μM时,作用24h或48h后在显微镜下可以明显地观察到细胞死亡,MTT结果与本发明的观察一致, 100μM CuCl2对HepG2细胞作用24h后有一定的毒性。为了进一步验证在其它肿瘤细胞中也存在此现象, 如图2B所示,CuCl2在浓度0-50μM之间作用HeLa细胞48h后,同样地,浓度在0-50μM之间几乎对细 胞无毒性,但是当100μM的CuCl2作用HeLa细胞48h后存在明显的毒性。所以本发明选取了CuCl2的作 用浓度在0-50μM之间。为了寻求最佳的CuCl2浓度,如图2C所示,本发明分别用0、2.5、5、10、20 和40μM的DPY与10、20和50μM的CuCl2协同作用HepG2细胞24h后,发现都能表现出良好的协同 作用,20μM的CuCl2与DPY共同作用细胞毒性梯度更明显,且单独的20μM的CuCl2对肿瘤细胞几乎无 毒性(图2A和B),所以本发明选取20μM的CuCl2作为本发明的最佳浓度。
实施例2:铜离子依赖的细胞毒性的确证
本发明比较了20μM的CuCl2分别与DPY、DPH和DMe共同作用对不同肿瘤细胞的毒性。如图2A-F 所示,20μM的CuCl2分别与DPY、DPH和DMe共同作用HepG2细胞24h(图3A-C)或48h(图3D-F) 后,对细胞生长的抑制情况,本发明发现20μM的CuCl2与不同浓度的DPY共同作用,能很好地抑制HepG2 细胞的增殖,而且具有时间依赖性,当20μM的CuCl2与20μM的DPY作用HepG2细胞48h后,细胞几 乎完全死亡(图3D),而单独的20μM的DPY作用48h对细胞的生长却几乎没有影响(图3D),说明DPY 可能作为铜离子载体把铜带进了细胞内;而作为对照分子,DPH与20μM的CuCl2共同作用HepG2细胞 却不能杀死细胞(图3B和E),因为DPH不能络合二价铜,故不能将Cu2+带进细胞。DMe作为巯基小分 子,其单独作用对细胞的生长几乎无影响,但是与20μM的CuCl2共同作用后,大浓度能明显地抑制细胞 生长(图3C和F),说明DMe作为具有还原性的小分子巯基化合物可能与CuCl2在细胞培养环境下发生氧 化还原反应,生成对细胞具有毒副作用的Cu+,进而抑制细胞生长。为了进一步确认图3A-F的结果。
本发明利用HeLa细胞对以上结果进行了证实,20μM的CuCl2分别与DPY、DPH和DMe共同作用 HeLa细胞48h后,如图2G-I所示,单独20μM的DPY对HeLa细胞生长无影响,但当与20μM的CuCl2共同作用后,可以明显地抑制细胞生长(图3G);而DPH与铜不能产生此作用,进一步证实了DPY作为 铜离子载体能把Cu2+带进了肿瘤细胞内而发挥作用。同样在肺癌细胞A549和肝癌细胞SMMC-7721上, 不同浓度的DPY也能与20μM的Cu2+产生协同毒性(图3J)。比较有意思的是,DPY与Cu2+协同对正常 细胞L02、BEAS-2B以及293T的细胞毒性却比肿瘤细胞较小(图3K),说明二者共同作用可以选择性地 作用于肿瘤细胞。
本发明对MTT结果进行了验证,采用台盼蓝染色法对不同浓度的DPY与20μM的CuCl2共同作用 HepG2细胞48h后的结果进行了确认,当DPY与20μM的CuCl2共同作用细胞后,细胞毒性明显增强, 与前面MTT结果一致。
总体来说,单独的DPY和Cu2+几乎对细胞无毒性,但当二者共同作用后,会选择性地杀死肿瘤细胞, 而DPH却不能,说明DPY可能作为铜离子载体将Cu2+载入细胞内而发挥作用。
实施例1和2的具体操作方法如下:
MTT比色法,是一种检测细胞存活和生长的方法[7]。将每孔5×103至1×104个细胞(根据细胞种类 及作用时间选取不同细胞数)与试验药物种于96孔板中,每孔总体积为100μl,每个药物浓度平行4-6份, 使用0.1%的溶媒(一般是DMSO)作为对照,在5%CO2、95%湿度和37℃的培养条件下培养指定时间。 接着用多道移液枪向每孔加入10μl MTT(终浓度:5mg/ml)继续孵育4小时,MTT与活细胞线粒体中 的琥珀酸脱氢酶完全反应,生成蓝紫色结晶甲瓒,用多道移液枪向每孔加入100μl提取缓冲液(即三联溶 液含10%SDS、0.1%HCl及5%异丁醇),继续过夜培养(一般在8-12小时之间),使蓝紫色的甲瓒结晶 完全溶解,在570nm处测定吸光度值,对照组吸光度最好在0.8-1之间,按照:细胞存活率(%)=(OD药物组-OD空白)/(OD对照-OD空白)×100,计算细胞存活率,最后根据药物作用浓度与细胞存活率作图,计 算出药物对细胞增殖抑制的IC50值。
Trypan blue染色法,将每孔5×104细胞种于24孔板中或者1×105细胞种于12孔板中,待细胞过夜 贴壁,加药物或其它试剂,0.1%的DMSO作为对照,在37℃继续孵育指定的时间[9,10]。若是贴壁细胞需 要用胰酶消化细胞,制备单细胞悬浮液,并根据细胞数的多少进行适当稀释;若是悬浮细胞(HL-60细胞), 则可以直接稀释后用。将细胞用0.4%(重量/体积)的Trypan blue染色(具体比例为细胞悬液:0.4%Trypan blue溶液=9:1,混合混匀),细胞存活时,细胞胞膜结构完整,Trypan blue无法进入,不染色,而当细胞 失活时,细胞膜的通透性增强,Trypan blue易进入染成蓝色,在显微镜下计数活细胞(非染色,圆形透明 状)和死细胞(染色,蓝色)的数目。
实施例3:DPY与Cu2+新配合物的形成
为了确认DPY能作为铜离子载体,可以将Cu2+载入细胞。
本发明在细胞外环境对DPY和CuCl2进行单晶培养,以证实DPY和CuCl2确实能络合。我们将DPY 与CuCl2·2H2O以1:1的比例均匀地分散在CH3Cl3溶液中,制得暗绿色针状单晶,反应方程如图4A,通 过X单晶衍射仪分析结构如图4B,发现Cu2+与DPY以1:1的比例很好地络合。
实例3的操作方法:
配合物的生成,将DPY与CuCl2·2H2O以1:1的比例均匀地分散在CH3Cl3溶液中,发现溶液的颜色 由亮蓝色变成暗绿色,说明有化学反应发生,接着常温搅拌2h,过滤得饱和溶液,室温养单晶,一周后 发现暗绿色针状单晶,通过X单晶衍射仪分析结构。
晶体结构的测定,DPY与CuCl2的晶体数据是通过X-Ray晶体衍射仪(SuperNova(Dual))在低温(100 K)条件下由Cu/Mo Kα射线照射收集所得。晶胞参数通过观察到的设定角度和所选强反 射的探测器位置所优化,然后利用单晶结构解析软件Siemens SHELXTL PLUS program中的直接法确认晶 体结构[28]。使用非等热向热参数优化所有非氢原子,所有氢原子都使用固定等向的热参数并考虑空间几何 结构及结构因子等加以修饰。晶体参数如表1所示,CCDC 1484851包含本发明中DPY与CuCl2晶体的补 充数据。

实施例4:细胞内Cu2+浓度的测定
为了确认DPY能作为铜离子载体,可以将Cu2+载入细胞。
本发明考察了DPY和CuCl2共同作用肿瘤细胞后细胞内的铜离子摄取量(Uptake),假如DPY作为铜 离子载体,将Cu2+载入细胞,那么细胞内的Cu2+含量将会升高。我们分别设立了四组试验:空白对照组; 20μM CuCl2组;20μM CuCl2与20μM DPY共同作用组;20μMCuCl2与20μM DPH共同作用组。将以 上分组药物分别加入HepG2细胞,作用0、0.5、1、3、6、12、24和48h后,收集细胞定量,用电感耦合 等离子体原子发射光谱法(ICP-AES)检测细胞内的铜离子含量变化,如图5A所示,与空白对照组、DPH 与Cu共同作用组、单独Cu组比较,DPY与Cu共同作用组随着作用时间的增加,细胞内的铜含量明显增 加,存在时间依赖性,24h几乎达到饱和,进一步说明了DPY可以把Cu2+带进细胞内,而且存在时间依 赖性。图4B和C分别是以上指定药物浓度作用HepG2和HeLa细胞24h后细胞内的铜离子含量变化图, 如图5B所示,对于HepG2而言,空白组(Control组)细胞内的铜含量为0.26±0.10,单独Cu组细胞内 的铜含量为0.92±0.31,DPH与Cu共同作用组细胞内的铜含量为1.18±0.14,而DPY与Cu共同作用组 细胞内的铜含量为13.59±0.93,结果明显比前面三组的数值高,同样的结果在HeLa细胞内也发生,如图5C所示,以上药物组对HeLa细胞作用24h后,结果显示DPY与Cu共同作用组细胞内的铜含量明显升 高,充分地说明了DPY作为Cu2+载体,可以将Cu2+载入细胞,从而使得细胞内的铜含量明显升高。
实例4的操作方法:
HepG2或HeLa细胞(1-3×106)一式三份种于100mm培养皿中过夜贴壁,加指定浓度的药物,作用 指定时间,收集细胞。一份用于蛋白定量;另外两份用于铜离子浓度检测:样品用浓硝酸溶解,用电感耦 合等离子体原子发射光谱法(ICP-AES)检测铜含量。
实施例5:细胞外DPY浓度增加,说明有新生成的DPY,能循环运载铜离子
本发明通过液相串联质谱(LC-MS/MS)法检测DPY与CuCl2共同作用HepG2细胞后培养基中DPY 浓度的变化,以考察释放Cu2+后新生成的DPY是否可以从细胞内转到细胞外液中去。用100μM的DPY 与20μM CuCl2共同作用HepG2细胞0、1、2、4、8、12和24h,检测培养基中的DPY浓度,如表2所 示,DPY在培养基中的浓度先降低后升高,说明DPY将Cu2+运载至细胞内,在细胞内巯基分子的作用下 释放出Cu2+,并发生一系列氧化还原反应,新生成的DPY又可以运转至细胞外液,通过循环的方式不断 地运载Cu2+。
该发明试剂巧妙地利用了DPY载体将铜离子载入细胞,释放铜离子的同时产生DMe分子,DMe在细 胞内的氧化条件下可以生产新的DPY,从而达到循环运载铜离子的目的。

实例5的操作方法:
LC-MS/MS检测DPY浓度,配置0、0.5、1、2.5、5、10和25μg/ml的标准DPY系列作标准曲线, 得Y=1.71x-2.16,R2=0.9939。
将1×106个HepG2细胞种在60mm培养皿中,过夜贴壁(12h后),加100μM的CuCl2与20μM的 DPY,分别于0、1、2、4、8、12、24、36和48h收集60μl培养基,每个点再补加60μl新培养基,用 LC-MS/MS检测不同时间点的DPY浓度,根据标准曲线定量。
Claims (5)
1.一种可再生的铜载体抗癌试剂,其特征在于可以将二价铜离子(Cu2+)运载至细胞内,在细胞内释放Cu2+,同时产生新的载体分子,继续循环地运载铜离子。
2.根据权利要求1所述,具体载体为DPY,能运载Cu2+进入细胞,释放Cu2+,同时产生DMe,DMe在细胞内氧化的条件下生成DPY,达到可再生、可循环运载铜离子的目的。
3.根据权利要求2所述,DPY与Cu2+在体外模拟产生的新配合物。
4.根据权利要求2所述的结构类似且能运载Cu2+的其它载体,若能在细胞内再生,继续运载Cu2+,所产生的应用。
5.根据权利要求1所述的一种可再生的铜载体抗癌试剂对四种癌细胞:人肝细胞癌细胞系HepG2和SMMC-7721,人宫颈癌细胞系HeLa细胞和人肺腺癌细胞A549的细胞毒性作用的应用。
