Abstract
本发明公开了一种治疗湿疹的草本酵素药膏及其制备方法和应用,采用包括苦参、白蔹、黄柏、黄莲、绵茵陈、地肤子和薄荷的草本中药和包括博伊丁假丝酵母、布氏乳杆菌和塞内加尔醋酸菌的酵素菌剂提取发酵而成;制备方法包括:煎煮草本中药得中药提取物,加入糖类调配灭菌得中药调配液,接种酵素菌剂发酵,灭活后过滤得药渣和中药发酵液,粉碎药渣,浓缩中药发酵液,混合即得。本发明通过对草本中药的合理配伍,采用酵素菌剂共同发酵,对草本中药进行预消化、分解和转化,将大分子的中间物质分解成为能被直接吸收的有效小分子物质,增加疗效,显著提高吸收速度和透皮吸收率,且能降低草本中药的副作用;制备方法简单,工艺可控,实际推广价值重要。

技术领域
本发明属于中医药技术领域,涉及湿疹的预防和治疗技术领域,更具体的,涉及一种治疗湿疹的草本酵素药膏及其制备方法和应用。
背景技术
湿疹是一种常见的由多种内外因素引起的表皮及真皮浅层的炎症性皮肤病,一般认为与变态反应有一定关系。湿疹是一种容易复发的皮肤病,也是一种过敏性炎症性皮肤病,以皮疹多样性、对称分布、剧烈瘙痒、反复发作和易演变成慢性为特征,可发生于任何年龄、任何部位和任何季节,但常在冬季复发或加剧,有渗出倾向,慢性病程,易反复发作。
具体地,湿疹在临床上按皮损表现可分为急性、亚急性、慢性三期,即:
(1)急性湿疹:皮损初为多数密集的粟粒大小的丘疹、丘疱疹或小水疱,基底潮红,逐渐融合成片,由于搔抓,丘疹、丘疱疹或水疱顶端抓破后呈明显的点状渗出及小糜烂面,边缘不清;如继发感染,炎症更明显,可形成脓疱、脓痂、毛囊炎、疖等。自觉剧烈瘙痒。好发于头面、耳后、四肢远端、阴囊、肛周等,多对称发布。
(2)亚急性湿疹:急性湿疹炎症减轻后,皮损以小丘疹、结痂和鳞屑为主,仅见少量丘疱疹及糜烂,但仍有剧烈瘙痒。
(3)慢性湿疹:常因急性湿疹和亚急性湿疹反复发作不愈而转为慢性湿疹,也可开始即为慢性湿疹。具体表现为患处皮肤增厚、浸润,棕红色或色素沉着,表面粗糙,覆鳞屑,或因抓破而结痂;自觉瘙痒剧烈;常见于小腿、手、足、肘窝、腘窝、外阴、肛门等处。病程不定,易复发,经久不愈。
目前,湿疹的治疗分西药和中药两种方式,其中,西药治疗上通常以抗生素、抗组胺和糖皮质激素等药物治疗,但治标不治本,停药易复发,且长期应用激素毒副作用大,不良反应多,容易出现耐药性,不易为患者家属接受;中药治疗方面,市面上所使用的皮肤湿疹外用制剂产品所使用的原料和制备方法都相对简单,不具有良好的透皮吸收功效,对皮肤湿疹造成的各种瘙痒、感染等得不到较快的缓解,同时中药水提液色素污染衣物、疗效不佳、药液容易霉变等,导致了制备出来的皮肤湿疹外用制剂达不到理想的效果。
发明内容
针对现有技术的不足,本发明提供了一种具有良好的抑菌及透皮吸收功效、能较快缓解皮肤湿疹造成的感染和各种瘙痒、疗效可靠、标本兼治、临床应用安全且无任何毒副作用的治疗湿疹的草本酵素药膏。
本发明的另一目的是提供该治疗湿疹的草本酵素药膏的制备方法。
本发明的又一目的是提供该治疗湿疹的草本酵素药膏的应用。
本发明的目的是通过以下技术方案来实现的:
一种治疗湿疹的草本酵素药膏,采用草本中药和酵素菌剂提取发酵而成,所述草本中药包括:苦参、白蔹、黄柏、黄莲、绵茵陈、地肤子和薄荷,所述酵素菌剂包括:博伊丁假丝酵母、布氏乳杆菌和塞内加尔醋酸菌。
其中,苦参,苦,寒,归心、肝、胃、大肠、膀胱经,清热燥湿,利尿,杀虫,宁心止悸,用于湿热泻痢,便血,黄疸尿赤,淋证涩痛,小便不利,赤白带下,阴肿阴痒,湿疹湿疮,皮肤瘙痒,疥癣麻风,心悸不宁。
其中,白蔹,苦,微寒,归心、胃经,清热解毒,消痈散结,敛疮生肌,用于疮疡肿毒,瘰疬,烫伤,湿疮,温疟,惊痫,血痢,肠风,痔漏,白带,跌打损伤,外伤出血。
其中,黄柏,味苦,性寒,归肾、膀胱经,清热燥湿,泻火除蒸,解毒疗疮,用于湿热泻痢,黄疸尿赤,带下阴痒,热淋涩痛,脚气痿蹙,骨蒸劳热,盗汗,遗精,疮疡肿毒,湿疹湿疮,盐黄柏滋阴降火,用于阴虚火旺,盗汗骨蒸。
其中,黄莲,苦,寒,归心、脾、胃、肝、胆、大肠经,清热燥湿,泻火解毒,用于湿热痞满,呕吐吞酸,泻痢,黄疸,高热神昏,心火亢盛,心烦不寐,血热吐衄,目赤,牙痛,消渴,痈肿疔疮;外治湿疹,湿疮,耳道流脓。
其中,绵茵陈,苦、辛、微寒,归脾、胃、肝、胆经,清湿热,退黄疸,用于风湿寒热邪气,热结黄疸,治通身发黄,小便不利,降低头热,去伏瘕,通关节,去滞热,伤寒用之。
其中,地肤子,味辛、苦,性寒,归肾、膀胱经,清热利湿,祛风止痒,用于小便涩痛,阴痒带下,风疹,湿疹,皮肤瘙痒。
其中,薄荷,味辛,性凉,入肺、肝经,疏散风热,清利头目,利咽透疹,疏肝行气,用于外感风热,头痛,咽喉肿痛,食滞气胀,口疮,牙痛,疮疥,瘾疹,温病初起,风疹瘙痒,肝郁气滞,胸闷胁痛。
在上述技术方案中,所述治疗湿疹的草本酵素药膏采用如下重量份的草本中药和酵素菌剂提取发酵而成;
所述草本中药包括:
苦参40-120份、白蔹40-100份、黄柏20-50份、黄莲15-30份、绵茵陈20-50份、地肤子20-50份和薄荷20-50份;
所述酵素菌剂包括:
博伊丁假丝酵母0.1-1份、布氏乳杆菌0.1-1份和塞内加尔醋酸菌 0.1-1份,所述博伊丁假丝酵母、布氏乳杆菌和塞内加尔醋酸菌的活菌数分别为5×107-7.5×107cfu/g、1.0×107-1.25×107cfu/g和1.0×107- 1.25×107cfu/g。
优选地,在上述技术方案中,所述治疗湿疹的草本酵素药膏采用如下重量份的草本中药和酵素菌剂提取发酵而成;
所述草本中药包括:
苦参65-110份、白蔹60-95份、黄柏32-48份、黄莲18-25份、绵茵陈36-42份、地肤子35-46份和薄荷28-35份;
所述酵素菌剂包括:
博伊丁假丝酵母0.4-0.75份、布氏乳杆菌0.36-0.72份和塞内加尔醋酸菌0.35-0.85份。
进一步优选地,在上述技术方案中,
所述博伊丁假丝酵母为由保藏于中国典型培养物保藏中心(武汉) 保藏编号为CCTCC M 2018162 BST003的博伊丁假丝酵母菌株制成;
所述布氏乳杆菌为由保藏于中国典型培养物保藏中心(武汉)保藏编号为CCTCC M2018164 BST005的布氏乳杆菌菌株制成;
所述塞内加尔醋酸菌为由保藏于中国典型培养物保藏中心(武汉) 保藏编号为CCTCC M 2018167 BST008的塞内加尔醋酸菌菌株制成。
进一步优选地,在上述技术方案中,所述治疗湿疹的草本酵素药膏还包括调膏基质,所述调膏基质为甘油、液体石蜡、凡士林、蜂蜡和二甲基硅油中的一种或几种。
再进一步优选地,在上述技术方案中,所述调膏基质为蜂蜡,蜂蜡的加入量为20-50份。
本发明提供了该治疗湿疹的草本酵素药膏的制备方法,包括如下步骤:
S1、按配比称取草本中药,并加入所述草本中药8-12倍重量的水,浸泡45-90min,煮沸75-120min后,滤取一次煎煮液,滤渣再加入所述草本中药8-12倍重量的水煮沸75-120min,并将一次煎煮液加入混合得中药提取物;
S2、往所述中药提取物中加入所述草本中药0.015-0.025倍重量的糖类,混合调配,再加热至121℃,保温灭菌15-20min,随后冷却至 28-32℃,得到中药调配液;
S3、往所述中药调配液中接种布氏乳杆菌,发酵36-72h,随后再接种博伊丁假丝酵母和塞内加尔醋酸菌,继续发酵18-30h,加热至 115℃,保温10-20min,将所述中药调配液中的活菌灭活,随后过滤得药渣和中药发酵液;
S4、将所述药渣粉碎成膏状药渣,并将所述中药发酵液浓缩至草本中药1.8-2.2倍重量的中药浓缩液,向其中加入所述膏状药渣混合均匀,即得草本酵素药膏。
在上述技术方案中,步骤S1中,所述煮沸为采用文火慢煎。
在上述技术方案中,步骤S3中,所述布氏乳杆菌的发酵时间大于 48h,所述博伊丁假丝酵母和塞内加尔醋酸菌的发酵时间大于24h。
优选地,在上述技术方案中,步骤S4中,还包括,在加入膏状药渣前,加入调膏基质搅拌融化。
本发明还提供了该草本酵素药膏或该制备方法制备得到的草本酵素药膏在治疗湿疹中的应用。
本发明的优点:
(1)本发明所提供的治疗湿疹的草本酵素药膏通过对苦参、白蔹、黄柏、黄莲、绵茵陈、地肤子和薄荷的合理配伍,并采用博伊丁假丝酵母、布氏乳杆菌和塞内加尔醋酸菌的共同发酵,对草本中药进行预消化、分解和转化,将大分子的中间物质分解转化成为能够被直接吸收的有效小分子物质,并产生强烈的抑菌效果,能加快草本中药的吸收,增加疗效,显著提高吸收速度和透皮吸收率;
(2)本发明所提供的治疗湿疹的草本酵素药膏通过采用多种酵素菌剂混合发酵草本中药,不仅能显著提高草本中药的疗效,使其药效提高至传统草本中药的4-28倍,而且能通过发酵分解草本中药的毒性,有效降低草本中药的副作用,此外,还能去除了普通草本中药煎煮后所具有的难闻的草药味道,使制备得到的草本酵素药膏具有特异性酵素香味,患者使用效果良好;
(3)本发明所提供的治疗湿疹的草本酵素药膏采用加热煎煮、混合发酵和调制成膏相结合的方式制备,能有效提取各草本中药中的活性成分,所需条件和设备简单,工艺简单可控,具有十分重要的实际推广价值;
(4)本发明所提供的治疗湿疹的草本酵素药膏通过选取容易接受的给药途径,利用透皮吸收直接进入血液,结合中医经络、脏腑学说,结合穴位刺激有助于药物的有效吸收,具有十分重大的临床意义。

所述博伊丁假丝酵母采用来自百士泰(武汉)生物科技有限公司保藏于中国典型培养物保藏中心(武汉)保藏编号为CCTCC M 2018162 BST003的博伊丁假丝酵母(分类命名:Candida biodinin BST003,保藏地址:中国典型培养物保藏中心(武汉),中国武汉-武汉大学,保藏日期:2018年03月29日)菌株制成,具体方法如下,在无菌条件下,将博伊丁假丝酵母菌株接入LB琼脂试管斜面培养基上,在30-35℃下恒温培养66-78h,直至长出明显光滑的菌落,即得博伊丁假丝酵母试管菌种,置于2-5℃冰箱中备用,随后将LB液体培养基装入已烘干洗净的1000ml三角瓶中,每瓶装量600~700ml,塞好棉塞,用牛皮纸包扎瓶口,在121℃下恒温灭菌30min,冷却至30-35℃,在无菌条件下接入培养成熟的博伊丁假丝酵母试管菌种,在135-170rpm、28-32℃下恒温培养36-72h,直至其活菌数大于5×107cfu/g,并置于2-5℃冰箱中待用;
所述布氏乳杆菌采用来自百士泰(武汉)生物科技有限公司保藏于中国典型培养物保藏中心(武汉)保藏编号为CCTCC M 2018164 BST005的布氏乳杆菌(分类命名:Lactobacillus buchneri BST005,保藏地址:中国典型培养物保藏中心(武汉),中国武汉-武汉大学,保藏日期:2018年03月29日)菌株制成,具体方法如下,在无菌条件下,将布氏乳杆菌菌株接入已灭菌的MRS琼脂试管斜面培养基上,在 30-35℃下恒温培养66-78h,即得布氏乳杆菌菌种,置于2-5℃冰箱中备用,随后将MRS液体培养基装入已烘干洗净的1000ml三角瓶中,每瓶装量600~700ml,塞好棉塞,用牛皮纸包扎瓶口,在121℃下恒温灭菌30min,冷却至30-35℃,在无菌条件下接入培养成熟的布氏乳杆菌菌种,在28-32℃下静置恒温培养36-72h,,直至其活菌数大于1 ×107cfu/g,并置于2-5℃冰箱中待用;
所述塞内加尔醋酸菌采用来自百士泰(武汉)生物科技有限公司保藏于中国典型培养物保藏中心(武汉)保藏编号为CCTCC M 2018167 BST008的塞内加尔醋酸菌(分类命名:Acetobacter senegalensis BST008,保藏地址:中国典型培养物保藏中心(武汉),中国武汉- 武汉大学,保藏日期:2018年03月29日)菌株制成,所采用液体培养基配方(质量比)为:酵母浸粉1%、葡萄糖1%和无水乙醇3%,所采用固体培养基配方(质量比)为:酵母浸粉1%、葡萄糖1%、无水乙醇3%和琼脂粉2%,具体方法如下,在无菌条件下,将塞内加尔醋酸菌菌株接入已灭菌的塞内加尔醋酸菌固体试管斜面培养基上,在30-35℃下恒温培养66-78h,即得布氏乳杆菌菌种,置于2-5℃冰箱中备用,将塞内加尔醋酸菌液体培养基中的酵母浸粉和葡萄糖装入已烘干洗净的1000ml三角瓶中,每瓶装量600~700ml,塞好棉塞,用牛皮纸包扎瓶口,在121℃下恒温灭菌30min,冷却至30-35℃,在无菌条件下加入3%的无水乙醇,摇匀后接入培养成熟的布氏乳杆菌菌种,在135-170rpm、28-32℃下恒温培养36-72h,直至其活菌数大于1×107 cfu/g,并置于2-5℃冰箱中待用。

实施例1一种治疗湿疹的草本酵素药膏
1、原料配比:
草本中药包括:苦参90份、白蔹85份、黄柏42份、黄莲24份、绵茵陈42份、地肤子38份和薄荷32份;
所述酵素菌剂:博伊丁假丝酵母0.48份、布氏乳杆菌0.42份和塞内加尔醋酸菌0.38份;
2、制备方法:
S1、按上述配比称取草本中药,并加入草本中药12倍重量的水,浸泡60min,文火慢煎90min后,滤取一次煎煮液,滤渣再加入草本中药12倍重量的水文火慢煎90min,并将一次煎煮液加入混合得中药提取物;
S2、往中药提取物中加入草本中药0.02倍重量的糖类,混合调配,再加热至121℃,保温灭菌15min,随后冷却至30℃,得到中药调配液;
S3、往中药调配液中接种布氏乳杆菌,发酵40h,随后再接种博伊丁假丝酵母和塞内加尔醋酸菌,继续发酵24h,加热至115℃,保温15min,将中药调配液中的活菌灭活,随后过滤得药渣和中药发酵液;
S4、将药渣粉碎成膏状药渣,并将中药发酵液浓缩至草本中药2.0 倍重量的中药浓缩液,加入膏状药渣混合均匀,即得草本酵素药膏。
实施例2一种治疗湿疹的草本酵素药膏
1、原料配比:
草本中药包括:苦参75份、白蔹60份、黄柏48份、黄莲28份、绵茵陈24份、地肤子32份和薄荷42份;
所述酵素菌剂:博伊丁假丝酵母0.36份、布氏乳杆菌0.75份和塞内加尔醋酸菌0.28份;
2、制备方法:
S1、按上述配比称取草本中药,并加入草本中药10倍重量的水,浸泡60min,文火慢煎90min后,滤取一次煎煮液,滤渣再加入草本中药10倍重量的水文火慢煎90min,并将一次煎煮液加入混合得中药提取物;
S2、往中药提取物中加入草本中药0.02倍重量的糖类,混合调配,再加热至121℃,保温灭菌15min,随后冷却至30℃,得到中药调配液;
S3、往中药调配液中接种布氏乳杆菌,发酵48h,随后再接种博伊丁假丝酵母和塞内加尔醋酸菌,继续发酵24h,加热至115℃,保温15min,将中药调配液中的活菌灭活,随后过滤得药渣和中药发酵液;
S4、将药渣粉碎成膏状药渣,并将中药发酵液浓缩至草本中药2.0 倍重量的中药浓缩液,加入35份甘油搅拌融化,最后加入膏状药渣混合均匀,即得草本酵素药膏。
实施例3一种治疗湿疹的草本酵素药膏
1、原料配比:
草本中药包括:苦参100份、白蔹82份、黄柏38份、黄莲20份、绵茵陈42份、地肤子40份和薄荷32份;
所述酵素菌剂:博伊丁假丝酵母0.55份、布氏乳杆菌0.48份和塞内加尔醋酸菌0.47份;
2、制备方法:

S4、将药渣粉碎成膏状药渣,并将中药发酵液浓缩至草本中药2.0 倍重量的中药浓缩液,加入36份蜂蜡搅拌融化,最后加入膏状药渣混合均匀,即得草本酵素药膏。
对比例1一种治疗湿疹的草本药膏
1、原料配比:
草本中药包括:苦参100份、白蔹82份、黄柏38份、黄莲20份、绵茵陈42份、地肤子40份和薄荷32份;
2、制备方法:
S1、按上述配比称取草本中药,并加入草本中药10倍重量的水,浸泡60min,文火慢煎120min后,滤取一次煎煮液,滤渣再加入草本中药10倍重量的水文火慢煎120min,并将一次煎煮液加入混合得中药提取物;
S2、将中药提取物过滤得药渣和中药药液,将药渣粉碎成膏状药渣,并将中药药液浓缩至草本中药2.0倍重量的浓缩中药药液,加入 36份蜂蜡搅拌融化,最后加入膏状药渣混合均匀,即得草本药膏。
对比例2一种治疗湿疹的草本酵素药膏
1、原料配比:
草本中药包括:苦参80份、白蔹75份、黄柏68份、黄莲32份、绵茵陈15份、地肤子36份和薄荷14份;
所述酵素菌剂:博伊丁假丝酵母1.2份、布氏乳杆菌0.35份和塞内加尔醋酸菌1.5份;
2、制备方法:
S1、按上述配比称取草本中药,并加入草本中药10倍重量的水,浸泡60min,文火慢煎90min后,滤取一次煎煮液,滤渣再加入草本中药10倍重量的水文火慢煎90min,并将一次煎煮液加入混合得中药提取物;
S2、往中药提取物中加入草本中药0.02倍重量的糖类,混合调配,再加热至121℃,保温灭菌15min,随后冷却至30℃,得到中药调配液;
S3、往中药调配液中接种布氏乳杆菌,发酵36h,随后再接种博伊丁假丝酵母和塞内加尔醋酸菌,继续发酵24h,加热至115℃,保温15min,将中药调配液中的活菌灭活,随后过滤得药渣和中药发酵液;

对比例3一种治疗湿疹的草本酵素药膏
1、原料配比:
草本中药包括:苦参100份、白蔹82份、黄柏38份、黄莲20份、绵茵陈42份、地肤子40份和薄荷32份;
所述酵素菌剂:博伊丁假丝酵母0.55份、布氏乳杆菌0.48份和塞内加尔醋酸菌0.47份;
2、制备方法:
S1、按上述配比称取草本中药,并加入草本中药10倍重量的水,浸泡60min,文火慢煎120min后,滤取一次煎煮液,滤渣再加入草本中药10倍重量的水文火慢煎120min,并将一次煎煮液加入混合得中药提取物;

S3、往中药调配液中同时接种布氏乳杆菌、博伊丁假丝酵母和塞内加尔醋酸菌,发酵60h,加热至115℃,保温15min,将中药调配液中的活菌灭活,随后过滤得药渣和中药发酵液;
S4、将药渣粉碎成膏状药渣,并将中药发酵液浓缩至草本中药2.0 倍重量的中药浓缩液,加入36份蜂蜡搅拌融化,最后加入膏状药渣混合均匀,即得草本酵素药膏。
对实施例1-3所制备的治疗湿疹的草本酵素药膏的皮肤过敏性进行测试,具体过程如下:
取2-2.5kg健康白兔(市售),9-12月龄,每组6只(3雌3雄),共3组,将白兔背部脊柱两侧毛剪掉,不可损伤表皮,去毛范围左、右各约3cm×3cm,并以此为涂敷点。
实验组1-3以实施例1-3所制备的治疗湿疹的草本酵素药膏(0.5g) 进行涂敷,直接涂敷于白兔一侧皮肤上,然后用两层纱布(2.5cm× 2.5cm)覆盖,并将再用无刺激性胶布或绷带加以固定,另一侧皮肤作为对照。
按照上述方法,连续测试14天,且从第2天开始,每次涂敷前一小时应剪毛并用清水或无刺激性溶剂将残留产品清除,并观察记录结果。
测试结果表明,实施例1-3所制备的治疗湿疹的草本酵素药膏对健康白兔脱毛皮肤未产生红斑、发炎、溃烂或水肿等刺激性作用,说明本发明产品对皮肤无刺激性。
测试实施例1-3和对比例1-3所制备的药膏的临床试验效果,具体过程如下:
选取280名湿疹患者,湿疹分布于额头、手臂、双腿,所患湿疹轻重不等。将患者随机分为7组,每组40名,实验例1-3分别涂覆实施例1-3所制备的治疗湿疹的草本酵素药膏,实验例4-6分别涂覆对比例1-3所制备的药膏,实验例7涂覆市售治疗湿疹的外用药膏产品(皮炎平)。
使用方法:
取产品适量均匀涂抹于患处,每日两次,早晚各一次。
判断标准:
效果显著:皮损消退70%以上,自觉症状明显减轻或消失;
效果一般:皮损消退30%以上70%以下,自觉症状减轻;
无效:皮损消退不足30%,自觉无明显症状变化。
分别统计涂覆7天和14天后的临床效果。
临床效果对比表如下表1所示。

从上表1的结果可以看出,整体来看,实施例1-3所制得的草本酵素药膏的临床效果明显优于对比例1-3所制得药膏,且市售药膏的临床效果最差。
对比分析实施例1-3所制得的草本酵素药膏,可以发现,实施例1 和实施例3的草本酵素药膏的临床效果差别较小,实施例3的草本酵素药膏不管是7天后还是14天后的临床效果略好于实施例1,究其原因,是由草本中药的配比和所加蜂蜡所造成的,蜂蜡具有良好的成膜性以及防氧化变质等特性,同时具有收涩、敛疮和生肌等中药功效。
在对比例1-3中,对比例1的药膏的临床效果最差,其草本中药配比虽然合适,但由于没经过发酵步骤,造成药膏的吸收效果较差,同时未发酵药膏的抑菌效果也较差;在对比例2和对比例3中,由于草本中药的配合和发酵工艺进行了调整,较实施例3来说效果明显下降,但对比例1-3的临床效果在整体上均优于市售药膏。
检测实施例3和对比例1所制备的药膏的抑菌效果,结果如下表2 所示。

从上表2的结果可以看出,本发明中所述产品在未经发酵之前,仅对金黄色葡萄球菌有抑制作用;在经过三支菌种发酵之后,不仅增强了对金黄色葡萄球菌的抑制作用,同时对大肠杆菌、绿脓杆菌、白色念珠菌均产生了较强的抑制作用。相对于传统的中药湿疹外用药剂需额外添加抑菌物质来说,更加安全有效。
最后,以上仅为本发明的较佳实施方案,并非用于限定本发明的保护范围。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (9)
1.一种治疗湿疹的的草本酵素药膏,其特征在于,采用如下重量份的草本中药和酵素菌剂提取发酵而成;
所述草本中药组成为:
苦参40-120份、白蔹40-100份、黄柏20-50份、黄莲15-30份、绵茵陈20-50份、地肤子20-50份和薄荷20-50份;
所述酵素菌剂组成为:
博伊丁假丝酵母0.1-1份、布氏乳杆菌0.1-1份和塞内加尔醋酸菌0.1-1份,所述博伊丁假丝酵母、布氏乳杆菌和塞内加尔醋酸菌的活菌数分别为5×107-7.5×107cfu/g、1.0×107-1.25×107cfu/g和1.0×107-1.25×107cfu/g。
2.根据权利要求1所述的草本酵素药膏,其特征在于,采用如下重量份的草本中药和酵素菌剂提取发酵而成;
所述草本中药组成为:
苦参65-110份、白蔹60-95份、黄柏32-48份、黄莲18-25份、绵茵陈36-42份、地肤子35-46份和薄荷28-35份;
所述酵素菌剂组成为:
博伊丁假丝酵母0.4-0.75份、布氏乳杆菌0.36-0.72份和塞内加尔醋酸菌0.35-0.85份。
3.根据权利要求1-2任一项所述的草本酵素药膏,其特征在于,
所述博伊丁假丝酵母为由保藏于中国典型培养物保藏中心(武汉) 保藏编号为CCTCCM 2018162 BST003的博伊丁假丝酵母菌株制成;
所述布氏乳杆菌为由保藏于中国典型培养物保藏中心(武汉)保藏编号为CCTCC M2018164 BST005的布氏乳杆菌菌株制成;
所述塞内加尔醋酸菌为由保藏于中国典型培养物保藏中心(武汉)保藏编号为CCTCC M2018167 BST008的塞内加尔醋酸菌菌株制成。
4.根据权利要求1-2任一项所述的草本酵素药膏,其特征在于,还包括调膏基质,所述调膏基质为甘油、液体石蜡、凡士林、蜂蜡和二甲基硅油中的一种或几种。
5.根据权利要求4所述的草本酵素药膏,其特征在于,所述调膏基质为蜂蜡,蜂蜡的加入量为20-50份。
6.一种如权利要求1-5任一项所述的草本酵素药膏的制备方法,其特征在于,包括如下步骤:
S1、按配比称取草本中药,并加入所述草本中药8-12倍重量的水,浸泡45-90min,煮沸75-120min后,滤取一次煎煮液,滤渣再加入所述草本中药8-12倍重量的水煮沸75-120min,并将一次煎煮液加入混合得中药提取物;
S2、往所述中药提取物中加入所述草本中药0.015-0.025倍重量的糖类,混合调配,再加热至121℃,保温灭菌15-20min,随后冷却至28-32℃,得到中药调配液;
S3、往所述中药调配液中接种布氏乳杆菌,发酵36-72h,随后再接种博伊丁假丝酵母和塞内加尔醋酸菌,继续发酵18-30h,加热至115℃,保温10-20min,将所述中药调配液中的活菌灭活,随后过滤得药渣和中药发酵液;
S4、将所述药渣粉碎成膏状药渣,并将所述中药发酵液浓缩至草本中药1.8-2.2倍重量的中药浓缩液,向其中加入所述膏状药渣混合均匀,即得草本酵素药膏。
7.根据权利要求6所述的草本酵素药膏的制备方法,其特征在于:步骤S1中,所述煮沸为采用文火慢煎。
8.根据权利要求6所述的草本酵素药膏的制备方法,其特征在于:
步骤S3中,所述布氏乳杆菌的发酵时间大于48h,所述博伊丁假丝酵母和塞内加尔醋酸菌的发酵时间大于24h;
和/或,步骤S4中,还包括,在加入膏状药渣前,加入调膏基质搅拌融化。
9.权利要求1-5任一项所述的草本酵素药膏或权利要求7-8任一项所述的制备方法制备得到的草本酵素药膏在制备治疗湿疹药物中的应用,其特征在于,所述草本酵素药膏对皮肤无刺激,在治疗湿疹同时兼具抑菌功效。
