Toxicity test on C8166 cells

The test sample was mixed with 4X 105/ml C8166 cell suspension, cultured at 37 ℃ for 3 days in 3 duplicate wells with 5% CO2, tested for cytotoxicity by MTT occlusion and the CC50 value was calculated, and the results of the test were calculated by repeating two experiments as shown in Table 1.

Inhibition experiment for HIV-1 IIIB induced C8166 cytopathy

The test sample and 8X 105/ml C8166 cells 50 ul/well were inoculated into 96-well cell culture plates containing 100 ul/well gradient ratio diluted drug, 50ul HIV-1 IIIB diluted supernatant, 1300TCID 50/well was added, 3 duplicate wells were set, cultured at 37 ℃ for 3 days with 5% CO2, and EC50 values were calculated, and the results of the calculation were shown in Table 2 by repeating the two experiments.

Inhibition experiment of HIV-1 IIIB acute infection C8166 cells

4X 105/ml C8166 cells were infected with HIV-1 III BMOI ((multiplicity of infection) ═ 0.04) at 37 ℃ for 2 hours, washed 3 times by centrifugation with PBS, freed virus ions were removed, 100ul of the cells were inoculated into 96-well plates containing 100ul of test compounds at various dilutions, and cultured at 37 ℃ in 5% CO2 for 3 days. As can be seen from a combination of the data in tables 1, 2 and 3, the CC50 value of the lyophilized powder prepared in example 1 against C8166 cells was 371.31 ± 7.19 (ug/ml); an EC50 value of 13.87 ± 3.08(ug/ml) for HIV-1 iiib-induced C8166 cytopathic, a therapeutic index T1 value of 26.78; the EC50 value for HIV-1 IIIB acutely infected C8166 cells was 11.93. + -. 4.14(ug/ml) and the therapeutic index T1 was 31.12.
