Abstract
本发明涉及一种具有止血抑菌功能的组合物及其制备方法和应用,属于保健品、药品、消毒产品技术领域。本发明所述具有止血抑菌功能的组合物包括三七、蒲公英、金银花、木糖醇、薄荷脑、丁香油。本发明所述三七、蒲公英、金银花和丁香油相互之间药效互补,可达到活血止血的功效,各组分间具有协同作用,可达到抑制细菌繁殖,缓解炎症的功效。

本发明涉及一种具有止血抑菌功能的组合物及其制备方法和应用,属于保健品、药品、消毒产品技术领域。
背景技术
口腔问题包括牙龈出血、牙龈肿痛、口腔异味等齿龈炎症困扰着绝大多数人,牙龈出血常发生在刷牙时或咀嚼硬物时,一些严重的患者在平时说话或者微笑时也会出现牙龈出血的问题,让人尴尬不已。牙龈出血的主要原因是口腔内细菌滋生,导致慢性牙齿炎症,在受到外界刺激的时候会产生血丝,严重者会大量出血。目前的一些牙膏中添加了具有凝血功能的物质,这些成分只能暂时的缓解牙龈出血并不能从根本上抑制口腔内细菌滋生,解决细菌性牙龈炎症、牙龈出血问题。
发明内容
本发明通过三七、蒲公英、金银花、木糖醇、薄荷脑与丁香油的复配,使复配后的混合物具有预防牙龈出血、抑制细菌繁殖的功能,解决了上述问题。
本发明提供了一种具有止血抑菌功能的组合物,所述具有止血抑菌功能的组合物包括三七、蒲公英、木糖醇、薄荷脑。
本发明所述三七有“止血神药”之称,能够散瘀血,止血而不留瘀,对出血兼有瘀滞者更为适宜。
本发明所述蒲公英含有蒲公英醇、蒲公英素、胆碱、有机酸、菊糖等多种健康营养成分。性味甘,微苦,寒。归肝、胃经。治热毒、痈肿、疮疡、目赤肿痛、湿热、黄疸、小便淋沥涩痛、牙痛、咽痛。蒲公英可生吃、炒食、做汤,是药食兼用的植物。
本发明优选为所述具有止血抑菌功能的组合物,按重量份,由下述组份组成:三七1-200份、蒲公英1-200份、木糖醇10-100份、薄荷脑1-20份。
本发明优选为所述具有止血抑菌功能的组合物包括三七、蒲公英、金银花、木糖醇、薄荷脑、丁香油。
本发明所述金银花性寒,味甘,入肺、心、胃经,具有清热解毒、抗炎、补虚疗风的功效,其功效主要是清热解毒,主治温病发热、热毒血痢、痈疽疔毒等。现代研究证明,金银花含有绿原酸、木犀草素苷等药理活性成分,对溶血性链球菌、金黄葡萄球菌等多种致病菌及上呼吸道感染致病病毒等有较强的抑制力,另外还可增强免疫力、护肝、抗肿瘤、消炎、解热、止血凝血。
本发明所述丁香油中的丁香酚具有镇痛抑菌功能,可使充血牙髓得到安抚。丁香酚还具有很强的杀菌力,作为局部镇痛药可用于龋齿,且兼有局部防腐作用。
本发明优选为所述具有止血抑菌功能的组合物,按重量份,由下述组份组成:三七1-200份、蒲公英1-200份、金银花0-200份、木糖醇10-100份、薄荷脑1-20份、丁香油0-50份。
本发明另一目的为提供一种上述组合物的制备方法,所述制备方法包括如下步骤:按预定重量份,将三七、蒲公英和金银花加入到乙醇溶液Ⅰ中浸泡,回流,过滤,浓缩滤液,干燥,得到干粉Ⅰ;按预定重量份,将木糖醇与干粉Ⅰ混匀,得到干粉Ⅱ;按预定重量份,将薄荷脑溶于乙醇溶液Ⅲ中,得到薄荷脑的乙醇溶液;按预定重量份,将丁香油溶于乙醇溶液Ⅳ中,得到丁香油的乙醇溶液;将薄荷脑的乙醇溶液与丁香油的乙醇溶液分别喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,得到具有止血抑菌功能的组合物。。
本发明优选为所述制备方法包括如下步骤:按预定重量份,先将三七、蒲公英和金银花粉碎后加入到乙醇溶液Ⅰ中浸泡2-24h,60-150℃回流1-5h,过滤,得到滤液Ⅰ,三七、蒲公英与金银花粉碎后的粒度为10-80目,三七、蒲公英和金银花的混合物与乙醇溶液Ⅰ的重量比为1:10-100,乙醇溶液Ⅰ的体积分数为60-80%,再将滤渣加入到乙醇溶液Ⅱ中60-150℃回流1-5h,过滤,得到滤液Ⅱ,滤渣与乙醇溶液Ⅱ的重量比为1:10-50,乙醇溶液Ⅱ的体积分数为60-80%,合并滤液Ⅰ和滤液Ⅱ,将合并后的滤液Ⅰ和滤液Ⅱ浓缩至相对密度为1.1-1.25的清膏,喷雾干燥得到干粉Ⅰ;按预定重量份,将木糖醇粉碎后与干粉Ⅰ混匀,木糖醇粉碎后的粒度为60-200目,得到干粉Ⅱ;按预定重量份,将薄荷脑溶于乙醇溶液Ⅲ中,薄荷脑与乙醇溶液Ⅲ的重量比为1:4-49,乙醇溶液Ⅲ的体积分数为70-95%,得到薄荷脑的乙醇溶液;按预定重量份,将丁香油溶于乙醇溶液Ⅳ中,丁香油与乙醇溶液Ⅳ的重量比为1:4-49,乙醇溶液Ⅳ的体积分数为70-95%,得到丁香油的乙醇溶液;将薄荷脑的乙醇溶液与丁香油的乙醇溶液分别喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,得到具有止血抑菌功能的组合物。
本发明又一目的为提供一种上述组合物在食品、保健品和日用化学品中的应用。
本发明优选为将所述组合物加入到辅料中,制成口香糖、片剂、牙膏、漱口水。

本发明有益效果为:
①本发明所述木糖醇本身不能被微生物利用,因此,具有良好的抑菌、防龋齿效果,同时,木糖醇与中药组分合用具有增强药效的功能;
②本发明所述三七、蒲公英、金银花和丁香油相互之间药效互补,可达到活血止血的功效,各组分间具有协同作用,可达到抑制细菌繁殖,缓解炎症的功效;
③本发明采用乙醇溶解后喷涂的方式将薄荷脑与丁香油添加到干粉Ⅱ中,可以达到更好的分散效果,将乙醇烘干后不会残留在产品中,产品的安全性更高。

实施例1
一种具有止血抑菌功能组合物的制备方法,所述制备方法包括如下步骤:
按重量份,先将10份的三七干品、5份的蒲公英干品加入到300份的乙醇溶液Ⅰ中浸泡5h,65℃回流3h,过滤,得到滤液Ⅰ,乙醇溶液Ⅰ的体积分数为75%,再将滤渣加入到300份的乙醇溶液Ⅱ中,65℃回流3h,过滤,得到滤液Ⅱ,乙醇溶液Ⅱ的体积分数为75%,合并滤液Ⅰ和滤液Ⅱ,将合并后的滤液Ⅰ和滤液Ⅱ浓缩至相对密度为1.2的清膏,喷雾干燥得到干粉Ⅰ;
按重量份,将50份的木糖醇与干粉Ⅰ混匀,得到干粉Ⅱ;
按重量份,将3份的薄荷脑溶于15份的乙醇溶液Ⅲ中,乙醇溶液Ⅲ的体积分数为95%,得到薄荷脑的乙醇溶液;
将薄荷脑的乙醇溶液喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,得到具有止血抑菌功能的组合物。
实施例2
一种具有止血抑菌功能组合物的制备方法,所述制备方法包括如下步骤:
按重量份,先将20份的三七干品、6份的蒲公英干品和6份的金银花干品加入到640份的乙醇溶液Ⅰ中浸泡5h,65℃回流3h,过滤,得到滤液Ⅰ,乙醇溶液Ⅰ的体积分数为75%,再将滤渣加入到640份的乙醇溶液Ⅱ中65℃回流3h,过滤,得到滤液Ⅱ,乙醇溶液Ⅱ的体积分数为75%,合并滤液Ⅰ和滤液Ⅱ,将合并后的滤液Ⅰ和滤液Ⅱ浓缩至相对密度为1.2的清膏,喷雾干燥得到干粉Ⅰ;
按重量份,将50份的木糖醇与干粉Ⅰ混匀,得到干粉Ⅱ;
按重量份,将2份的薄荷脑溶于10份的乙醇溶液Ⅲ中,乙醇溶液Ⅲ的体积分数为95%,得到薄荷脑的乙醇溶液;
按重量份,将5份的丁香油溶于20份的乙醇溶液Ⅳ中,乙醇溶液Ⅳ的体积分数为95%,得到丁香油的乙醇溶液;
将薄荷脑的乙醇溶液与丁香油的乙醇溶液分别喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,得到具有止血抑菌功能的组合物。
实施例3
一种具有止血抑菌功能口含片的制备方法,所述制备方法包括如下步骤:
按重量份,先将10份的三七干品、3份的蒲公英干品和6份的金银花干品加入到380份的乙醇溶液Ⅰ中浸泡5h,65℃回流3h,过滤,得到滤液Ⅰ,乙醇溶液Ⅰ的体积分数为75%,再将滤渣加入到380份的乙醇溶液Ⅱ中65℃回流3h,过滤,得到滤液Ⅱ,乙醇溶液Ⅱ的体积分数为75%,合并滤液Ⅰ和滤液Ⅱ,将合并后的滤液Ⅰ和滤液Ⅱ浓缩至相对密度为1.2的清膏,喷雾干燥得到干粉Ⅰ;
按重量份,将50份的木糖醇与干粉Ⅰ混匀,得到干粉Ⅱ;
按重量份,将3份的薄荷脑溶于15份的乙醇溶液Ⅲ中,乙醇溶液Ⅲ的体积分数为95%,得到薄荷脑的乙醇溶液;
按重量份,将3份的丁香油溶于15份的乙醇溶液Ⅳ中,乙醇溶液Ⅳ的体积分数为95%,得到丁香油的乙醇溶液;
按重量份,先将薄荷脑的乙醇溶液与丁香油的乙醇溶液分别喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,再与3份阿拉伯胶制备的40w/v%阿拉伯胶溶液混合造粒,添加1份硬脂酸镁,压片,得到具有止血抑菌功能的口含片。

按重量份,先将薄荷脑的乙醇溶液喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,再与3份阿拉伯胶制备的40w/v%阿拉伯胶溶液混合造粒,添加1份硬脂酸镁,压片,得到口含片。
对比例2
一种口含片的制备方法,所述制备方法包括如下步骤:
按重量份,先将6份的蒲公英干品加入到120份的乙醇溶液Ⅰ中浸泡5h,65℃回流3h,过滤,得到滤液Ⅰ,乙醇溶液Ⅰ的体积分数为75%,再将滤渣加入到120份的乙醇溶液Ⅱ中65℃回流3h,过滤,得到滤液Ⅱ,乙醇溶液Ⅱ的体积分数为75%,合并滤液Ⅰ和滤液Ⅱ,将合并后的滤液Ⅰ和滤液Ⅱ浓缩至相对密度为1.2的清膏,喷雾干燥得到干粉Ⅰ;
按重量份,将50份的木糖醇与干粉Ⅰ混匀,得到干粉Ⅱ;
按重量份,将3份的薄荷脑溶于15份的乙醇溶液Ⅲ中,乙醇溶液Ⅲ的体积分数为95%,得到薄荷脑的乙醇溶液;

按重量份,先将3份的金银花干品加入到60份的乙醇溶液Ⅰ中浸泡5h,65℃回流3h,过滤,得到滤液Ⅰ,乙醇溶液Ⅰ的体积分数为75%,再将滤渣加入到60份的乙醇溶液Ⅱ中65℃回流3h,过滤,得到滤液Ⅱ,乙醇溶液Ⅱ的体积分数为75%,合并滤液Ⅰ和滤液Ⅱ,将合并后的滤液Ⅰ和滤液Ⅱ浓缩至相对密度为1.2的清膏,喷雾干燥得到干粉Ⅰ;
按重量份,将50份的木糖醇与干粉Ⅰ混匀,得到干粉Ⅱ;
按重量份,将3份的薄荷脑溶于15份的乙醇溶液Ⅲ中,乙醇溶液Ⅲ的体积分数为95%,得到薄荷脑的乙醇溶液;

对比例4
一种口含片的制备方法,所述制备方法包括如下步骤:
按重量份,将3份的薄荷脑溶于15份的乙醇溶液Ⅰ中,乙醇溶液Ⅰ的体积分数为95%,得到薄荷脑的乙醇溶液;
按重量份,将3份的丁香油溶于15份的乙醇溶液Ⅱ中,乙醇溶液Ⅱ的体积分数为95%,得到丁香油的乙醇溶液;
按重量份,将薄荷脑的乙醇溶液与丁香油的乙醇溶液分别喷涂在50份的木糖醇上,混匀,烘干至含水量低于5%,得到干粉;
按重量份,将干粉与3份阿拉伯胶制备的40w/v%阿拉伯胶溶液混合造粒,添加1份硬脂酸镁,压片,得到止血抑菌口含片。
测试例1
抑菌试验
试验菌种:金黄色葡萄球菌(ATCC 6538)、大肠杆菌(8099)。
分别将实施例3、对比例1、对比例2、对比例3和对比例4制备的口含片制成直径为5mm、厚度为4mm圆片,每4片为一组。
阴性对照样片的制备:取无菌干燥滤纸片,每片滴加无菌蒸馏水20μL,干燥后备用。
试验菌的接种:用无菌棉拭子蘸取浓度为5×105cfu/mL-5×106cfu/mL试验菌悬液,在营养琼脂培养基平板表面均匀涂抹3次,每涂抹1次,平板转动60°,最后将无菌棉拭子绕平板边缘涂抹一周,盖好平皿,置室温干燥5min。
抑菌剂样片贴放:每次试验贴放1个染菌平板,每个染菌平板贴放4片试验样片和1片阴性对照样片,用无菌镊子取样片贴放于平板表面,各样片中心之间相距25mm,与平板的周缘相距15mm,贴放好后,用无菌镊子轻压样片,使其紧贴于平板表面,盖好平皿,置37℃温箱,培养18h观察结果,用游标卡尺测量抑菌环的直径(包括贴片)并记录。
测量抑菌环时,应选均匀而完全无菌生长的抑菌环进行,测量其直径应以抑菌环外沿为界。
评价规定
a、抑菌作用的判断:
抑菌环直径大于7mm者,判为有抑菌作用;
抑菌环直径小于或等于7mm者,判为无抑菌作用。
b、3次重复试验均有抑菌作用结果者,判为合格。
c、阴性对照组应无抑菌圈产生,否则试验无效。
实验结果见表1。

结论:由表1得,单独添加三七、金银花的片剂对大肠杆菌和金黄色葡萄球菌均无抑菌性,添加蒲公英的片剂对金黄色葡萄球菌有一定的抑菌性,单独添加丁香油的片剂对金黄色葡萄球菌抑菌性较好,通过比较实施例3与对比例1、2、3、4得,各组分间具有协同增效的抑菌效果。
测试例2
止血试验
小鼠适应性喂养3d后,按性别、体重分为6组,小鼠腹部用脱毛剂硫化钠脱毛,分别将实施例3制备的干粉Ⅱ、对比例1制备的干粉Ⅱ、对比例2制备的干粉Ⅱ、对比例3制备的干粉Ⅱ及对比例4制备的干粉涂于剃毛区,用药面积2cm×1cm,组合物用量为0.5g/cm2,采用玻璃棒局部涂用,每日1次,连续给药7d,空白对照组涂抹去离子水,末次给药1h后,用手术剪将小鼠尾部剪断,待血液自行溢出时开始计时,每隔30s用滤纸吸去血液一次,直到血液自然停止(滤纸吸时无血为止),即为出血时间,用秒测定出血时间,实验结果见表2。

结论:由表2得,实施例3制备的干粉Ⅱ对小鼠具有明显的止血效果,实施例3中多种组分的作用效果明显好于单独使用的效果,证明各组分间具有协同作用。
应用例1
张某,32岁,拔牙后,牙龈出血严重,含服本发明实施例3制备的片剂后,牙龈不再出血,且疼痛减轻。
应用例2
王某,12岁,喜食甜食,经常牙疼,用本发明实施例3制备的片剂代替糖果,自述口感较好,食用1个月后,牙痛次数明显减少。
应用例3
郭某,25岁,患有龋齿、慢性牙龈炎,经常牙龈出血,牙龈肿痛,坚持每天含服本发明实施例3制备的片剂2个月后,自述刷牙时牙龈出血症状明显改善,再未发生过牙龈肿痛。

1.一种具有止血抑菌功能的组合物,其特征在于:所述具有止血抑菌功能的组合物包括三七、蒲公英、木糖醇、薄荷脑。

按预定重量份,将木糖醇与干粉Ⅰ混匀,得到干粉Ⅱ;
按预定重量份,将薄荷脑溶于乙醇溶液Ⅲ中,得到薄荷脑的乙醇溶液;
按预定重量份,将丁香油溶于乙醇溶液Ⅳ中,得到丁香油的乙醇溶液;
将薄荷脑的乙醇溶液与丁香油的乙醇溶液分别喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,得到具有止血抑菌功能的组合物。
6.根据权利要求5所述的制备方法,其特征在于:所述制备方法包括如下步骤:
按预定重量份,先将三七、蒲公英和金银花粉碎后加入到乙醇溶液Ⅰ中浸泡2-24h,60-150℃回流1-5h,过滤,得到滤液Ⅰ,三七、蒲公英与金银花粉碎后的粒度为10-80目,三七、蒲公英和金银花的混合物与乙醇溶液Ⅰ的重量比为1:10-100,乙醇溶液Ⅰ的体积分数为60-80%,再将滤渣加入到乙醇溶液Ⅱ中60-150℃回流1-5h,过滤,得到滤液Ⅱ,滤渣与乙醇溶液Ⅱ的重量比为1:10-50,乙醇溶液Ⅱ的体积分数为60-80%,合并滤液Ⅰ和滤液Ⅱ,将合并后的滤液Ⅰ和滤液Ⅱ浓缩至相对密度为1.1-1.25的清膏,喷雾干燥得到干粉Ⅰ;
按预定重量份,将木糖醇粉碎后与干粉Ⅰ混匀,木糖醇粉碎后的粒度为60-200目,得到干粉Ⅱ;
按预定重量份,将薄荷脑溶于乙醇溶液Ⅲ中,薄荷脑与乙醇溶液Ⅲ的重量比为1:4-49,乙醇溶液Ⅲ的体积分数为70-95%,得到薄荷脑的乙醇溶液;
按预定重量份,将丁香油溶于乙醇溶液Ⅳ中,丁香油与乙醇溶液Ⅳ的重量比为1:4-49,乙醇溶液Ⅳ的体积分数为70-95%,得到丁香油的乙醇溶液;
将薄荷脑的乙醇溶液与丁香油的乙醇溶液分别喷涂在干粉Ⅱ上,混匀,烘干至含水量低于5%,得到具有止血抑菌功能的组合物。
7.权利要求1、2、3或4所述组合物在食品、保健品和日用化学品中的应用。
8.根据权利要求7所述的应用,其特征在于:将所述组合物加入到辅料中,制成口香糖、片剂、牙膏、漱口水。
9.根据权利要求8所述的应用,其特征在于:所述辅料为甜味剂、酸度调节剂、增稠剂、乳化剂、香精、色素、防腐剂、保湿剂和起泡剂中的至少一种;
所述甜味剂为蔗糖、果糖、山梨糖醇、麦芽糖醇、异麦芽酮糖醇、赤藓糖醇、安赛蜜、阿斯巴甜、甜菊糖苷、三氯蔗糖和淀粉中的至少一种;
所述酸度调节剂为柠檬酸及其盐、苹果酸及其盐、酒石酸及其盐、冰乙酸、乳酸及其盐、富马酸及其盐、碳酸盐和碳酸氢盐中的至少一种;
所述增稠剂为瓜尔胶、果胶、海萝胶、海藻酸丙二醇酯、海藻酸钠、槐豆胶、环状糊精、黄原胶、聚葡萄糖、决明胶、卡拉胶、可得然胶、可溶性大豆多糖、磷酸化二淀粉磷酸酯、氯化钙、罗望子多糖胶、麦芽糖醇、普鲁兰多糖、乳酸钙、双乙酰酒石酸单双甘油酯、羧甲基淀粉钠、羧甲基纤维素钠和阿拉伯胶中的至少一种;
所述乳化剂为磷脂、酪蛋白酸钠、羟丙基淀粉、乳酸脂肪酸甘油酯、辛烯基琥珀酸淀粉钠、乙酰化单双甘油脂肪酸酯、吐温20、吐温40、吐温60、吐温80和硬脂酸及其镁盐中的至少一种;
所述防腐剂为山梨酸及其钾盐、苯甲酸及其钠盐、溶菌酶、乳酸链球菌素、双乙酸钠、丙酸钠、丙酸钙和羟苯甲酯中的至少一种;
所述保湿剂为甘油、丙二醇和多聚磷酸盐中的至少一种;
所述起泡剂为十二烷基硫酸钠、月桂酰肌氨酸钠和椰油酰甘氨酸钠中的至少一种。
