Abstract

离子液体溶解骨骼的一种方法工艺,以鸡骨为原料,选择氯化胆碱‑甘油低共融体系离子液体,采用研磨法对骨骼进行溶解,设计单因素、正交实验确定离子液体中骨骼的最佳溶解工艺。其工艺条件为:鸡骨与氯化胆碱‑甘油质量比(固液比)为1∶20,反应溶解时间4h,溶解温度115℃。离子液体回收率为95.17%,所得溶解产物为胶原与羟基磷灰石。

离子液体溶解骨骼的一种方法工艺
技术领域 本发明涉及一种溶解天然骨的新型方法与工艺,属于精细化学中天然物提取制备新型材料的技术领域。
背景技术 据统计我国每年产生的动物骨骼量高达1500万吨,其中有100万吨未在骨骼加工领域得到有效利用,造成了资源浪费。骨骼主要是由有机成分胶原和无机成分羟基磷灰石组成。其中,骨骼中胶原主要用于生物医学、食品加工、化妆品制备等领域;骨骼中羟基磷灰石具有良好的生物相容性和组织活性,被广泛用做骨损伤修复材料。一方面,离子液体具有无污染、较低蒸汽压、不易燃、易与产物分离、可设计性、易回收、可循环使用等优点,选用新型绿色溶剂溶解骨骼符合可持续发展技术要求,具有经济效益;另一方面,动物骨骼中含有丰富的骨胶原和相容性良好的生物材料羟基磷灰石,探究离子液体溶解的工艺条件,提取有机成分骨胶原,并分离出无机成分羟基磷灰石,为新型生物材料的制备提供了研究方法,避免了资源浪费。
发明内容 本发明的目的是提供一种新的溶解骨骼的方法及其溶解工艺条件,其设备简单,原料易得又经济节能。本发明利用氯化胆碱-甘油离子液体溶解鸡骨,确定最佳溶解工艺,提取制备生物材料。离子液体溶解骨骼的原理是根据氢键破坏理论,利用离子液体破坏骨骼中胶原的三股螺旋结构、胶原多肽之间的肽键、胶原与羟基磷灰石分子间氢键,从而溶解骨骼中有机成分胶原,提取分离出羟基磷灰石,达到溶解骨骼的目的。与常规制备羟基磷灰石的方法相比,此工艺设备简单,原料易得,变废为宝,成本较低,采用生物提取法所得产物相容性更好,方法更为新颖。其特征在于所述的研磨法溶解骨骼过程界面更新更为均匀,所述原料鸡骨的粒度为60~80目;所述离子液体的合成原料氯化胆碱与丙三醇摩尔比为1∶2;所述离子液体溶解鸡骨的反应时,鸡骨与离子液体的质量比(固液比)可为1∶10~1∶50;溶解时间可为2h~6h;溶解温度可为80℃~160℃。
具体实施方式 称取1g鸡骨(60目~80目)置于盛有15g离子液体的研钵中,在加热装置中加热研磨搅拌。在115℃下溶解;4h后结束反应,趁热迅速用铜网过滤,去除未反应鸡骨颗粒,将滤渣洗净后干燥称重,得到鸡骨在离子液体中的溶解率为47.3%;待离子液体温度降至室温时加入甲醇进行稀释,充分混合后迅速进行真空抽滤,得到提取产物羟基磷灰石,蒸馏水少量多次洗涤,干燥后称重,提取率为22.3%;滤液用旋转蒸发仪旋蒸回收再利用,回收次数为3次,平均回收率为95.17%。

Claims (4)

1.一种溶解天然骨的新型方法与工艺,其特征在于所述的研磨法溶解骨骼过程界面更新更为均匀,选用离子液体溶解骨骼中胶原,从而提取分离出骨骼中羟基磷灰石,并应用于骨损伤修复材料。选用的原料鸡骨粒度为60~80目。
2.按权利要求1所述的离子液体溶解骨骼的方法工艺,所述离子液体的合成原料氯化胆碱与丙三醇摩尔比为1∶2。
3.按权利要求1所述离子液体溶解骨骼的方法时,鸡骨与离子液体的质量比(固液比)可为1∶10~1∶50;溶解时间可为2h~6h;溶解温度可为80℃~160℃。
4.按权利要求3所述离子液体溶解骨骼的工艺,溶骨率可达47.3%,羟基磷灰石提取率可达22.3%。
