Abstract

本发明提供了一种前白蛋白检测试剂盒,包括试剂R1和试剂R2,所述试剂R1和试剂R2组成如下:R1三羟甲基氨基甲烷、PEG 6000、TX‑100 20、异构十醇聚氧乙烯醚E1005、叠氮钠;R2三羟甲基氨基甲烷、羊抗人前白蛋白抗血清。本发明通过加入E1005后,溶液很快变为清亮,透明状态。由此我们可看出,E1005能有效的溶解血清中的乳糜,故能有效避免乳糜样本对测定带来的干扰。

Description

前白蛋白测定试剂盒
技术领域
本发明属于临床体外诊断试剂技术领域,涉及前白蛋白测定试剂盒。
背景技术
血清前白蛋白能敏感反映肝脏蛋白质代谢情况,它比白蛋白更能早期反应肝细胞损害,它的血清浓度明显受营养状况及肝功能改变的影响,有30%患者白蛋白正常而前白蛋白降低,多数肝病患者前白蛋白均降低50%以上,所以前白蛋白被认为是判断早期肝脏损害的灵敏指标。此外,在预测晚期癌症患者生存时间方面,前白蛋白也是一项有价值的指标。前白蛋白测定试剂盒产品适用范围该产品用于体外测定血清中前白蛋白的浓度。
血清PA是一种负急性时相反应蛋白,在炎症和恶性疾病时其血清水平下降。PA在肝脏合成,各类肝炎、肝硬化致肝功能损伤时,血清PA水平降低,是肝功能障碍的一个敏感指标。PA可作为蛋白质营养状况的指征,蛋白质消耗性疾病、肾病、妊娠、高雌激素血症时,PA浓度下降;PA浓度升高可见于Hodgkin′s病。
测定原理:利用抗原Pre-Aib和特异性抗体(羊抗人Pre-Aib抗血清)相结合形成不溶性免疫复合物,使反应液产生浊度,其浊度的高低与样本中的PA浓度呈正比。
现有的试剂为双试剂,试剂R1为缓冲液,R2为抗血清。加入R2后,抗原抗体发生方应,产生浊度,波长为340.而乳糜样本与R1混合后,也会在340nm处有较高吸光度,由于乳糜样本的水不溶性,样本产生的吸光度在R1中随着孵育时间的变长而上升,加入R2后则显著降低(原因不清楚),故而使测定结果偏低。
发明内容
本发明提供了一种改善前白蛋白乳糜样本的稳定性的试剂盒。
本发明的技术方案是:
一种前白蛋白检测试剂盒,包括试剂R1和试剂R2,所述试剂R1和试剂R2组成如下:

三羟甲基氨基甲烷
羊抗人前白蛋白抗血清。
进一步地,上述第一试剂和第二试剂组成为:

羊抗人前白蛋白抗血清 100ml/L。
本发明通过加入E1005后,溶液很快变为清亮,透明状态。由此我们可看出,E1005能有效的溶解血清中的乳糜,故能有效避免乳糜样本对测定带来的干扰。

根据测定原理,该试剂盒是通过抗原抗体反应产生的浊度来反应血清中PA的含量。而对于高乳糜样本,其与试剂组分R1混合后也能产生较高的浊度,故对测定产生较大的干扰。我们通过实验发现,在试剂组分R1中加入异构十醇聚氧乙烯醚E1005,能有效避免高乳糜样本对测定带来的干扰。通过手工实验,我们观察发现,没加E1005,高乳糜样本与R1混合后溶液呈乳白色,浑浊状态。而加入E1005后,溶液很快变为清亮,透明状态。由此我们可看出,E1005能有效的溶解血清中的乳糜,故能有效避免乳糜样本对测定带来的干扰。

对于高乳糜样本,配方1结果异常偏低,而改进后的配方测定结果正常。说明改进后配方能有效消除乳糜对测定带来的个干扰。
结论:改进后配方能有效消除乳糜对测定带来的干扰,满足临床需求。
