Description

Compound essential oil composition, application thereof and product for improving sleep
Technical Field
The invention relates to the technical field of traditional Chinese medicine health care, in particular to a compound essential oil composition, application thereof and a product for improving sleep.
Background
Insomnia is a common sleep disturbance phenomenon, usually referred to as a subjective experience of the patient that does not meet the quality of sleep time (and/or) and affects day-to-day social functioning. The clinical manifestations of insomnia patients mainly include the following aspects:
1. disorders of the sleep process: difficulty falling asleep, reduced sleep quality and decreased sleep time.
2. Daytime cognitive dysfunction: the memory function is decreased, the attention function is decreased, and the planning function is decreased, thereby causing drowsiness in the daytime, the working ability is decreased, and the daytime sleepiness is easy to occur when the work is stopped.
3. Vegetative nerve functional disturbance of the limbic system of the brain and its surroundings: the cardiovascular system is manifested by chest distress, palpitation, unstable blood pressure and peripheral vasoconstriction and expansion disorder; the digestive system is manifested by constipation or diarrhea, and gastric distress and distention; the locomotor system is characterized by muscular tension in the neck and shoulders, headache and lumbago. The emotional control ability is reduced, and the feeling of anger is easy to generate or is not happy; impotence is likely to occur in men, and sexual dysfunction is often seen in women.
4. Other systemic symptoms: short-term endosomal weight loss, immune dysfunction and endocrine dysfunction are likely to occur.
With the development of social economy, the pressure of life of people is increased, the incidence rate of insomnia shows a rising trend, and according to the investigation of six cities in China, the method has the following advantages: insomnia is a problem in 57% of adults. The normal life and work of the patient can be affected by long-term insomnia, and the physical and mental health of the patient can be affected by serious insomnia. The current commonly used medicines for treating sleep disorder include barbiturates and benzodiazepine

In order to further prove the advantages of the present invention, the present invention provides the effect verification data of examples 1 to 6 and comparative examples 1 to 6 to prove the beneficial effects of the present invention.
Verification of nerve-soothing and sleep-assisting effects of examples 1-6 and comparative examples 1-6
Animal experiments
1) Experimental animals: SD mice, clean grade, half male and female, weight 18 g-20 g. After the mice were bred adaptively for 1 week, the mice were randomly divided into a blank control group, examples 1-6 groups and comparative examples 1-6 groups, each group containing 10 mice.
2) The experimental method comprises the following steps: distilled water is inhaled in an atomizing mode in a blank control group, the compound essential oil described in examples 1-6 in an atomizing mode, and the compound essential oil described in comparative examples 1-6 in an atomizing mode. The dosage is 100 μ L for 30min for 7 days. On day 7, after all groups of mice were separately inhaled with the corresponding substances by nebulization, pentobarbital solution (50mg/kg) was intraperitoneally injected, the tail of the mouse was lifted up, and the mice were placed flat on a pad plate and then the nebulization was continued. The sleep latency period is from the time of intraperitoneal injection to the disappearance of mouse righting reflex, and the duration of righting reflex is the sleep time. Sleep latency time and sleep time were recorded.
3) Data processing: the measured data is averaged + -SD

And (3) representing that a one-factor analysis of variance method is adopted for data processing. P<0.05 means statistically significant.
4) The results of the experiment are shown in the following table:
influence of different groups of compound essential oil on sleep time of mice
Figure BDA0001662508520000122
Figure BDA0001662508520000131
Note: the shoulder marks indicate that each group of the examples and the comparative examples has significant difference compared with a blank control group, and P is less than 0.05; the letters (a, b, c, d, e, f, g, h, i, j and k) in the same row indicate that the comparison is carried out between different groups of the examples and the comparative examples, and the letters indicate that the comparison shows that the letters have significant difference, and P is less than 0.05
According to the tests, compared with the blank control group, the sleep time of the mice is improved, the sleep latency is reduced, and the significant difference (P is less than 0.05) is achieved in the groups of examples 1-6 and the groups of comparative examples 1-6, so that the essential oils of the examples 1-6 and the comparative examples 1-6 have certain nerve calming and sleep aiding effects. The comparison result between the groups of examples 1-6 and the groups of comparative examples 1-6 shows that the sleep time of the groups of examples 1-6 is far longer than that of the groups of comparative examples 1-6, the sleep latency of the groups of examples 1-6 is far shorter than that of the groups of comparative examples 1-6, and the optimal effect is the group of example 3.
