Abstract
本发明属于功能敷料技术领域,具体涉及一种含糖分的远红外竹原纤维敷料的制备方法,通过在竹原纤维中引入纳米远红外粉,并在加工制备医用纺制品敷料时将糖分与竹原纤维复合,最终形成的功能敷料具有抗菌、抗炎、清创、除臭、止痛、促进组织愈合等多重功效,特别适合于各类急慢性伤口和复杂伤口的护理。

技术领域
本发明属于医疗器械功能性敷料的生产加工技术领域,具体涉及一种含糖分的远红外竹原纤维敷料的制备方法。
背景技术
各种原因引起的伤口,尤其是难愈合的慢性伤口问题已经成为危害公共健康的主要因素之一,慢性伤口的治疗周期长、难度大,且易出现并发症,因此容易给病人带来沉重的经济负担和心理负担。伤口护理的过程不再是简单的伤口消毒和更换敷料,目前的研究认为,清创技术、清洁伤口方法的合理应用、敷料正确选择与合理应用、正确覆盖伤口方法的实施以及局部药物使用都与伤口愈合密切相关。其中,国内外学者关于敷料的研究繁多,传统敷料如纱布、绷带等在使用过程中容易存在创面粘连、保湿性差、易感染、无促进愈合作用等问题。
糖类用于护理伤口的历史可追溯到四五千年前,在抗生素发现之前,糖类疗法一直是伤口治疗的重要方法之一,抗生素发现之后,其应用才逐渐被抗生素所取代。但随着抗生素的滥用及细菌耐药性的增加,临床研究者又开始对治疗感染的古老方法进行重新评价,其中包括糖类及蜂蜜疗法。在国外,糖类及蜂蜜作为一种重要的伤口治疗方法,在远古时代便有记载。1679年,研究者就发现将砂糖撒在感染的伤口上,坏死组织脱落加速,伤口变得清洁。随后的研究表明,糖类在治疗溃疡性伤口方面也具有奇效。现有技术中,关于对糖类护理伤口,尤其是蜂蜜对伤口的护理效果,已有不少的临床报道反馈其良好的效果。然而,由于糖类或质量不佳的蜂蜜其本身结构的固有特性,一方面容易滋生细菌,另一方面其功能的单一性导致其的护理伤口时表现出一定的不足。因此,其难以适应于较复杂的伤口或慢性伤口对敷料的护理要求。
发明内容
本发明的目的在于解决现有技术中的缺陷,克服现有产品的不足,提供一种抗菌、抗炎、清创、除臭、止痛、促进组织愈合的含糖分的远红外竹原纤维敷料的制备方法。
为解决上述技术问题,本发明采用的技术方案是:一种含糖分的远红外竹原纤维敷料的制备方法,包括如下步骤:(1)糖分混悬液的制备:按重量份数计,将1~5份的壳聚糖与50~200份的稀醋酸充分搅拌溶解后形成A液,将10~50份的蜂蜜与0.1~2份的制霉菌素充分搅拌均匀形成B液,然后将A液与B液充分混合即形成糖分混悬液;(2)远红外竹原纤维的制备:将竹原纤维充分浸于去油液和粗化液中进行去油和粗化处理,洗净后浸渍于远红外复合整理液中,浸渍的浴比为1:10~15,浴液温度为25~50℃,浸渍时间为30~60min;(3)功能敷料的加工形成:将上述远红外竹原纤维经拉伸、定型、干燥后,浸渍于糖分混悬液中,或将糖分混悬液喷涂于远红外竹原纤维上,其后再经开松、梳理、铺网、针刺、整烫、收卷,最后经分切、包装、灭菌,得到含糖分的远红外竹原纤维敷料。
优选地,步骤(1)中所述的稀醋酸的体积分数为0.5~1%。
优选地,步骤(1)中所述的蜂蜜为桃金娘科属植物蜂蜜,即经蜜蜂对桃金娘科属植物采集而生成的蜂蜜。
优选地,步骤(2)中所述去油所用的去油液制备方法为:将氢氧化钠5~15重量份、乙酸钠30~60重量份、碳酸钠30~60重量份加入蒸馏水中配制而成。
优选地,步骤(2)中所述粗化所用的粗化液制备方法为:将高锰酸钾1~10重量份、浓度为98%的浓硫酸1~10重量份加入蒸馏水中配制而成。
优选地,步骤(2)中所述的远红外复合整理液由以下成分构成:30~50g/L的纳米远红外粉体,15~30g/L的纳米远红外分散液SK-814,15~20g/L的交链剂AF6600,20~40g/L的护肤加工剂SEW3260。
优选地,所述的纳米远红外粉体的组分按重量份数计,由10~20份的SiO2、15~30份的A2O3、1~5份的TiO2、10~20份的MgO、5~15份的Fe2O3、30~60份的ZrO2、0.5~2份的AgCl组成。
蜂蜜是一种成分复杂、营养丰富的过饱和糖溶液,具有极高的渗透压和较低的pH值,含有过氧化氢(hydrogen peroxide,H2O2)及丙酮醛类(Methylglyoxal,MGO)等杀菌成分,具有广谱的抗菌活性,且其抗菌活性没有产生耐药性的危险。这些良好特点为蜂蜜用于伤口治疗奠定了重要基础。桃金娘科属植物蜂蜜是一种抗氧化剂的天然生物材料,是一种非常好的治疗烧伤、烫伤、溃疡、褥疮等各类伤口的防感染、促进愈合作用的敷料,桃金娘科属植物蜂蜜的粘稠性可为创伤、烧伤、烫伤及溃疡感染提供保护层;桃金娘科属植物蜂蜜能刺激组织再生,刺激新的毛细血管及纤维的形成,代替表皮深层的结缔组织,并能产生胶原纤维增强修复组织;桃金娘科属植物蜂蜜不会粘附在创伤组织上,在更换桃金娘科属植物蜂蜜敷料时,不会撕去新形成的肉芽组织,更不至于引起疼痛,同时其具有抗炎作用,能减轻创作的肿胀,从而改善血液循环,加速伤口愈合,并减轻疼痛,还能减少创伤流出液。
竹原纤维就是从自然生长的竹子中提取出的一种纤维素纤维,是继棉、麻、毛、丝之后的第五大天然纤维。竹纤维具有良好的透气性、瞬间吸水性和较强的耐磨性,同时又具有天然抗菌、抑菌、除螨、防臭和抗紫外线功能。
远红外线是红外线中波长最长的一段红外线其波长范围在5-100um之间。它是电磁波的一种。远红外线微量照射治疗对微循环的改善效果显著。尤以微血流状态改善明显,表现为照射后毛细血管血流速度加快,红细胞聚集现象减少,乳头下静脉淤血现象减轻或消失。从而对改善机体组织及重要脏器的营养、代谢、修复功能等有非常积极作用。
本发明通过在竹原纤维中引入纳米远红外粉,并在加工制备医用纺制品敷料时将糖分与竹原纤维复合,最终形成的功能敷料可产生如下的有益效果:
1)利用制霉菌素对微生物的抑制作用以及壳聚糖本身的广谱抗菌性,防止蜂蜜被微生物污染,从而可充分发挥桃金娘科属植物蜂蜜对伤口表现出的良好的护理作用;
2)由于竹原纤维本身的天然抑菌性,结合壳聚糖固有的抗菌特性,二者产生协同的抗菌作用,有效遏制伤口滋生细菌,保护创面,避免感染;
3)在蜂蜜与壳聚糖的联合作用下,可刺激刺激淋巴细胞和吞噬细胞活性增强,激活人体对感染的免疫反应;蜂蜜中的抗氧化成分有助于自由基清除,可有效减轻炎症反应。
4)远红外线能够深入人体的皮下组织,所以利用远红外线反应,使皮下深层皮肤温度上升,扩张微血管,促进血液循环,复活酵素,强化血液及细胞组织代谢,缓解伤口疼痛,更加有利于伤口的愈合;
5)远红外线深透力可达肌肉关节深处,使身体内部温暖,放松肌肉,在其与蜂蜜的共同作用下,可极大地带动微血管网的氧气及养分交换,并排除积存体内的疲劳物质和乳酸等老化废物对消除内肿,缓解伤口疼痛的功效更加显著。
具体实施方式
下面结合实施例对本发明的技术方案做进一步的说明。
实施例1
一种含糖分的远红外竹原纤维敷料的制备方法,包括如下步骤:
(1)糖分混悬液的制备:按重量份数计,将1份的壳聚糖与200份体积分数为1%的稀醋酸充分搅拌溶解后形成A液,将40份的桃金娘科属植物蜂蜜与0.5份的制霉菌素充分搅拌均匀形成B液,然后将A液与B液充分混合即形成糖分混悬液;
(2)远红外竹原纤维的制备:将竹原纤维充分浸于去油液和粗化液中进行去油和粗化处理,洗净后浸渍于远红外复合整理液中,浸渍的浴比为1:12,,浴液温度为30℃,浸渍时间为30min;
(3)功能敷料的加工形成:将上述远红外竹原纤维经拉伸、定型、干燥后,浸渍于糖分混悬液中,或将糖分混悬液喷涂于远红外竹原纤维上,其后再经开松、梳理、铺网、针刺、整烫、收卷,最后经分切、包装、灭菌,得到含糖分的远红外竹原纤维敷料。
上述工艺步骤(2)中去油液由以下成分构成:将氢氧化钠15重量份、乙酸钠30重量份、碳酸钠60重量份加入蒸馏水中配制而成。步骤(2)中粗化液的构成:将高锰酸钾1重量份、浓度为98%的浓硫酸10重量份加入蒸馏水中配制而成。步骤(2)中远红外复合整理液由以下成分构成:30g/L的纳米远红外粉体,30g/L的纳米远红外分散液SK-814,18g/L的交链剂AF6600,30g/L的护肤加工剂SEW3260。所述纳米远红外粉体的组分按重量份数计,由20份的SiO2、30份的A2O3、4份的TiO2、15份的MgO、5份的Fe2O3、30份的ZrO2、2份的AgCl组成。
实施例2
一种含糖分的远红外竹原纤维敷料的制备方法,包括如下步骤:
(1)糖分混悬液的制备:按重量份数计,将2份的壳聚糖与50份体积分数为0.8%的稀醋酸充分搅拌溶解后形成A液,将30份的桃金娘科属植物蜂蜜与2份的制霉菌素充分搅拌均匀形成B液,然后将A液与B液充分混合即形成糖分混悬液;
(2)远红外竹原纤维的制备:将竹原纤维充分浸于去油液和粗化液中进行去油和粗化处理,洗净后浸渍于远红外复合整理液中,浸渍的浴比为1:10,浴液温度为40℃,浸渍时间为40min;
(3)功能敷料的加工形成:将上述远红外竹原纤维经拉伸、定型、干燥后,浸渍于糖分混悬液中,或将糖分混悬液喷涂于远红外竹原纤维上,其后再经开松、梳理、铺网、针刺、整烫、收卷,最后经分切、包装、灭菌,得到含糖分的远红外竹原纤维敷料。
上述工艺步骤(2)中去油液由以下成分构成:将氢氧化钠10重量份、乙酸钠40重量份、碳酸钠40重量份加入蒸馏水中配制而成。步骤(2)中粗化液的构成:将高锰酸钾10重量份、浓度为98%的浓硫酸1重量份加入蒸馏水中配制而成。步骤(2)中远红外复合整理液由以下成分构成:40g/L的纳米远红外粉体,20g/L的纳米远红外分散液SK-814,16g/L的交链剂AF6600, 40g/L的护肤加工剂SEW3260。所述纳米远红外粉体的组分按重量份数计,由10份的SiO2、15份的A2O3、3份的TiO2、10份的MgO、15份的Fe2O3、40份的ZrO2、0.5份的AgCl组成。
实施例3
一种含糖分的远红外竹原纤维敷料的制备方法,包括如下步骤:
(1)糖分混悬液的制备:按重量份数计,将5份的壳聚糖与150份体积分数为0.6%的稀醋酸充分搅拌溶解后形成A液,将10份的桃金娘科属植物蜂蜜与0.1份的制霉菌素充分搅拌均匀形成B液,然后将A液与B液充分混合即形成糖分混悬液;
(2)远红外竹原纤维的制备:将竹原纤维充分浸于去油液和粗化液中进行去油和粗化处理,洗净后浸渍于远红外复合整理液中,浸渍的浴比为1:15,浴液温度为50℃,浸渍时间为50min;
(3)功能敷料的加工形成:将上述远红外竹原纤维经拉伸、定型、干燥后,浸渍于糖分混悬液中,或将糖分混悬液喷涂于远红外竹原纤维上,其后再经开松、梳理、铺网、针刺、整烫、收卷,最后经分切、包装、灭菌,得到含糖分的远红外竹原纤维敷料。
上述工艺步骤(2)中去油液由以下成分构成:将氢氧化钠5重量份、乙酸钠50重量份、碳酸钠50重量份加入蒸馏水中配制而成。步骤(2)中粗化液的构成:将高锰酸钾8重量份、浓度为98%的浓硫酸5重量份加入蒸馏水中配制而成。步骤(2)中远红外复合整理液由以下成分构成:50g/L的纳米远红外粉体,15g/L的纳米远红外分散液SK-814,15g/L的交链剂AF6600,20g/L的护肤加工剂SEW3260。所述纳米远红外粉体的组分按重量份数计,由15份的SiO2、20份的A2O3、1份的TiO2、18份的MgO、10份的Fe2O3、50份的ZrO2、1份的AgCl组成。
实施例4
一种含糖分的远红外竹原纤维敷料的制备方法,包括如下步骤:
(1)糖分混悬液的制备:按重量份数计,将4份的壳聚糖与100份体积分数为0.9%的稀醋酸充分搅拌溶解后形成A液,将50份的桃金娘科属植物蜂蜜与1份的制霉菌素充分搅拌均匀形成B液,然后将A液与B液充分混合即形成糖分混悬液;
(2)远红外竹原纤维的制备:将竹原纤维充分浸于去油液和粗化液中进行去油和粗化处理,洗净后浸渍于远红外复合整理液中,浸渍的浴比为1:14,浴液温度为25℃,浸渍时间为60min;
(3)功能敷料的加工形成:将上述远红外竹原纤维经拉伸、定型、干燥后,浸渍于糖分混悬液中,或将糖分混悬液喷涂于远红外竹原纤维上,其后再经开松、梳理、铺网、针刺、整烫、收卷,最后经分切、包装、灭菌,得到含糖分的远红外竹原纤维敷料。
上述工艺步骤(2)中去油液由以下成分构成:将氢氧化钠8重量份、乙酸钠60重量份、碳酸钠30重量份加入蒸馏水中配制而成。步骤(2)中粗化液的构成:将高锰酸钾8重量份、浓度为98%的浓硫酸8重量份加入蒸馏水中配制而成。步骤(2)中远红外复合整理液由以下成分构成:45g/L的纳米远红外粉体,18g/L的纳米远红外分散液SK-814,20g/L的交链剂AF6600,25g/L的护肤加工剂SEW3260。所述纳米远红外粉体的组分按重量份数计,由18份的SiO2、25份的A2O3、5份的TiO2、20份的MgO、12份的Fe2O3、60份的ZrO2、1.5份的AgCl组成。
实施例5 动物试验
实验组D~G分别对应为实施例1~4制备的含糖分的远红外竹原纤维敷料产品;对照组为两组,一组为现有的藻酸盐功能性敷料(对照品1),另一组为不加壳聚糖组分,其余与实施例1工艺相同而加工得到的产品(对照品2);
取雄性新西兰兔,84只,每只2~3kg,随机分为七组,每组12只。各兔子均按照临床动物实验烧伤标准模型制备方法均造成30%TBSA浅II度烧伤,其中A组兔子不使用任何敷料,B组兔子敷用对照品1,C组兔子敷用对照品2,D、E、F、G组敷用本发明实施例1~4的复合液体敷料,每天更换两次,直到D、E、F、G组的创面愈合便结束本次动物实验。
从动物实验的结果上看,D~G组兔子创面敷用各自的敷料后,创面均无感染现象、无红肿发生。而从愈合时间上看,D~G组兔子创面的愈合时间明显少于B、C组,时间差异具有显著意义,见表1:

从六组实验的整体结果上看,对照品和本发明制备的敷料对创面的愈合能起到加速的作用,而D~G组敷料因为含抑菌、消炎及促进血液循环等功能成分,有利于促进创面愈合并缩短愈合时间。且从表格中可以看出C组与D~G组试验结果的差别,不难发现,蜜蜂与壳聚糖两种糖分在敷料护理伤口的过程中联合起到护创的作用,其联合作用的结果是显著的。
实施例6 抗菌试验
对照组为两组,一组为现有的壳聚糖功能性敷料(对照品1),另一组为不加壳聚糖和制霉菌素组分,其余与实施例1工艺相同而加工得到的产品(对照品2);
抗菌性能试验:根据GB/T 20944.3-2008纺织品抗菌性能的评价第3部分:振荡瓶法,测定本发明产品的抗菌性能。所用菌种为白色念珠菌、大肠杆菌和金黄色葡萄球菌,菌液浓度为105~107cfu/mL。首先在三角瓶中加入缓冲生理盐水及敷料产品,在0.1MPa蒸汽及121℃高温中灭菌20min,冷却至室温,用1mL无菌刻度吸管移取1mL菌液,加入上述三角瓶中。为使试样和细菌强行接触,在37℃摇床振荡数小时。取振荡前及振荡后溶液0.1mL涂于营养琼脂培养基上,经37℃、24h恒温培养后比较,其结果如下表所示。

由抑菌试验的结果我们可以看出,本发明的敷料产品对格兰氏阳性和阴性菌及真菌等均具有良好的效果,对各类细菌和真菌的抑菌率达90%以上,且高于壳聚糖敷料的抗菌效果,这是因为由于竹原纤维本身的天然抑菌性,结合壳聚糖固有的抗菌特性,二者产生协同的抗菌作用,有效遏制伤口滋生细菌,保护创面,避免感染。与此同时,对照品2相对其他组,其抗菌性最低,这可能是蜂蜜中本身容易滋生细菌的原因。因此,该敷料能有效防止伤口遭受感染。
应当指出的是,具体实施方式只是本发明比较有代表性的例子,显然本发明的技术方案不限于上述实施例,还可以有很多变形。本领域的普通技术人员,以本发明所明确公开的或根据文件的书面描述毫无异议的得到的,均应认为是本专利所要保护的范围。
Claims (7)
1.一种含糖分的远红外竹原纤维敷料的制备方法,其特征在于,包括如下步骤:
(1)糖分混悬液的制备:按重量份数计,将1~5份的壳聚糖与50~200份的稀醋酸充分搅拌溶解后形成A液,将10~50份的蜂蜜与0.1~2份的制霉菌素充分搅拌均匀形成B液,然后将A液与B液充分混合即形成糖分混悬液;
(2)远红外竹原纤维的制备:将竹原纤维充分浸于去油液和粗化液中进行去油和粗化处理,洗净后浸渍于远红外复合整理液中,浸渍的浴比为1:10~15,浴液温度为25~50℃,浸渍时间为30~60min;
(3)功能敷料的加工形成:将步骤(2)所得远红外竹原纤维经拉伸、定型、干燥后,浸渍于步骤(1)所得糖分混悬液中,或将步骤(1)所得糖分混悬液喷涂于步骤(2)所得远红外竹原纤维上,其后再经开松、梳理、铺网、针刺、整烫、收卷,最后经分切、包装、灭菌,得到含糖分的远红外竹原纤维敷料。
2.根据权利要求1所述的一种含糖分的远红外竹原纤维敷料的制备方法,其特征在于,步骤(1)中所述的稀醋酸的体积分数为0.5~1%。
3.根据权利要求1所述的一种含糖分的远红外竹原纤维敷料的制备方法,其特征在于,步骤(1)中所述的蜂蜜为桃金娘科属植物蜂蜜。
4.根据权利要求1所述的一种含糖分的远红外竹原纤维敷料的制备方法,其特征在于,步骤(2)中所述的去油液的制备方法为:将氢氧化钠5~15重量份、乙酸钠30~60重量份、碳酸钠30~60重量份加入蒸馏水中配制而成。
5.根据权利要求1所述的一种含糖分的远红外竹原纤维敷料的制备方法,其特征在于,步骤(2)中所述的粗化液的制备方法为:将高锰酸钾1~10重量份、浓度为98%的浓硫酸1~10重量份加入蒸馏水中配制而成。
6.根据权利要求1所述的一种含糖分的远红外竹原纤维敷料的制备方法,其特征在于,步骤(2)中所述的远红外复合整理液由以下成分构成:30~50g/L的纳米远红外粉体,15~30g/L的纳米远红外分散液SK-814,15~20g/L的交链剂AF6600,20~40g/L的护肤加工剂SEW3260。
7.根据权利要求6所述的一种含糖分的远红外竹原纤维敷料的制备方法,其特征在于,所述的纳米远红外粉体的组分按重量份数计,由10~20份的SiO2、15~30份的A2O3、1~5份的TiO2、10~20份的MgO、5~15份的Fe2O3、30~60份的ZrO2、0.5~2份的AgCl组成。
