本发明属于生物医药或化妆品技术领域,具体涉及一种提高皮肤耐受性的组合物及其制备方法。
背景技术
耐受性(Tolerance)是指人体对药物反应性降低的一种状态,按其性质有先天性和后天获得性之分。耐受性是一种生物学现象,是药物应用的自然结果。
皮肤耐受性就是指皮肤对抵御外界的能力,主要是指抵御外界刺激物的能力。耐受性差的皮肤可以分为三大类:遗传过敏性皮肤:如异位性皮炎或具有酒渣鼻倾向的皮肤。遗传过敏性皮肤具有角质层通透性异常和易于发生瘙痒的特点,而具有酒渣鼻倾向的皮肤在受到刺激时会产生血管充血反应。被诱发的皮肤敏感:使皮肤敏感性增加的因素包括:气候,环境,皮肤磨损、激光等机械因素以及维甲酸、皮肤磨削等治疗因素。具有遗传过敏素质的皮肤在接触某些化妆品成分如防腐剂和香料时其敏感性会进一步上升。特发性皮肤敏感:没有病原学基础,患者常声称他们不能耐受任何类型的化妆品
皮肤常见的刺激物:表面活性剂,这类成分可以提高皮肤的渗透性,尽管表活成分本身不一定有问题,但对于皮肤耐受性差的皮肤,有可能会促进有刺激性的成分对皮肤的渗透;香精,本身香精是一些小分子的萜烯、芳香烃类化合物,本身对皮肤没有明确的功能,但渗透性很强,而且目前临床上统计因香精出现的致敏率明显比其他成分高,所以对耐受性差的皮肤,尽量选清淡或不含香的成分。防腐剂,第三类成分非常有挑战,因为多数人会想当然的认为这类成分很安全。这两组数据表明,1组数据是来自2000-2005瑞士接触性皮炎研究所的调查,发现对当时常用的季铵盐-15,咪唑烷基脲及尼泊金酯类成分进行斑贴测试,43.4%的人会呈现阳性反应。另一组测试是Fisher博士的调查,发现CIT的皮肤阳性反应率是8.5%。而含有尼泊金酯的地塞米松膏剂对在湿疹治疗中,比不含尼泊金酯类成分的治愈率低,复发率高。植物类成分,除此之外,还要注意一些植物类成分,比如芦荟中的大黄素,橙类中的芸香甙,当归等伞形科的香豆素。如果在提取过程中分离的不充分,遇到太阳,容易产生光毒反应引起皮肤刺激。
现有技术中,增加皮肤耐受性就是增加皮肤抵御外界的能力,增厚角质层,使皮肤达到更加健康的状态。现今化妆品行业增加皮肤耐受性的方法大多是通过补充外源性神经酰胺、胆甾醇等物质去增强皮肤中的"灰浆"结构,效果不明显。
因此,研究新的可以提高皮肤耐受性的产品具有重要意义。
发明内容
基于上述原因,申请人经过多年研究,得到一种能够提高皮肤耐受性的组合物,该组合物中原料包括:交替单胞菌发酵产物提取物、β-葡聚糖,进一步,组合物原料包括:交替单胞菌发酵产物提取物、β-葡聚糖、野大豆(GLYCINE SOJA)籽提取物、糖鞘脂类,制备成液体制剂,能够显著降低SDS(十二烷基硫酸钠)所致溶血率;将本发明的液体制剂加入到化妆品中,具有很好的提高皮肤耐受性的作用。
本发明通过下述技术方案实现的。
一种提高皮肤耐受性的组合物,该组合物原料包括:交替单胞菌发酵产物提取物、β-葡聚糖。
优选的组合物原料包括:交替单胞菌发酵产物提取物、β-葡聚糖、野大豆(GLYCINESOJA)籽提取物。
优选的组合物原料包括:交替单胞菌发酵产物提取物、β-葡聚糖、野大豆(GLYCINESOJA)籽提取物、糖鞘脂类。
所述的一种提高皮肤耐受性的组合物,其中交替单胞菌发酵产物提取物8-12重量份、β-葡聚糖0.5-1.5重量份。
所述的一种提高皮肤耐受性的组合物,其中交替单胞菌发酵产物提取物8-12重量份、β-葡聚糖0.5-1.5重量份、野大豆(GLYCINE SOJA)籽提取物0.8-1.2重量份。
所述的一种提高皮肤耐受性的组合物,其中交替单胞菌发酵产物提取物8-12重量份、β-葡聚糖0.5-1.5重量份、野大豆(GLYCINE SOJA)籽提取物0.8-1.2重量份、糖鞘脂类0.5-1.0重量份。
所述的一种提高皮肤耐受性的组合物,其中交替单胞菌发酵产物提取物10重量份、β-葡聚糖1重量份、野大豆(GLYCINE SOJA)籽提取物1重量份、糖鞘脂类0.8重量份。
该组合物制备成液体制剂。
其中液体制剂的原料包括:去离子水15-25重量份、甘油0.8-1.2重量份、1,3-丁二醇1.5-2.5重量份。
其中液体制剂的制备方法为:
(1)取β-葡聚糖、交替单胞菌发酵产物提取物,加入到野大豆(GLYCINE SOJA)籽提取物,搅拌至完全均匀,然后用均质器均质1.5-2.5分钟,备用原料Ⅰ;
(2)将水、甘油、1,3-丁二醇、糖鞘脂类混合均匀,注入到高压高剪切纳米均质机中,升温至60-65℃,均质3-6分钟,循环3-6次,得到备用原料Ⅱ;
(3)将备用原料Ⅰ和备用原料Ⅱ混合均匀,得到液体制剂。
本发明所述β-葡聚糖购自德国Symrise公司,批号为201701011。
本发明所述交替单胞菌发酵产物提取物LucasMeyer公司,批号为20170103。
本发明上述所述的野大豆(GLYCINE SOJA)籽提取物购自美国Kemin公司,批号为20171202。
本发明上述所述的甘油购自意慕利(EMERY)公司,批号为20171202。
本发明所述1,3-丁二醇购自OXEA公司,批号为20171116。
本发明所述糖鞘脂类购自Kikkoman公司,批号为20171111。
具体实施方式
以下以具体实施例来说明本发明的技术方案,但本发明的保护范围不限于此。
本说明书实施例所述的内容仅仅是对发明构思的实现形式的列举,本发明的保护范围不应当被视为仅限于实施例所陈述的具体形式,本发明的保护范围也及于本领域技术人员根据本发明构思所能够想到的等同技术手段。尽管以下本发明的实施方案进行了描述,但本发明并不局限于上述的具体实施方案和应用领域,下述的具体实施方案仅仅是示意性的、指导性的,而不是限制性的。本领域的普通技术人员在本说明书的启示下和在不脱离本发明权利要求所保护的范围的情况下,还可以做出很多种的形式,这些均属于本发明保护之列。
本发明下述试验,是在多次创造性试验的基础上,以本发明所要保护的技术方案为基础,总结的研发人员的结论性试验。
试验1
对SDS致DNA刺激损伤的抑制
试验1组:交替单胞菌发酵产物提取物10mg、β-葡聚糖1mg;
试验2组:交替单胞菌发酵产物提取物10mg、β-葡聚糖1mg,野大豆(GLYCINE SOJA)籽提取物1mg;
试验3组:交替单胞菌发酵产物提取物10mg、β-葡聚糖1mg,野大豆(GLYCINE SOJA)籽提取物1mg、糖鞘脂类0.8g
试验方法:昆明种小鼠,体质量约25g。常规饲养1周后健康者颈椎脱臼处死,取小块肝放于滤纸上除去被膜及血后置于小烧杯内,加入少量预冷的PBS液清洗,用眼科剪剪碎,110目不锈钢筛网过滤,收集细胞悬液,用PBS液调细胞密度至107~108个/m L,将制得的细胞按105~106个分装于5mL刻度离心管,加DMEM培养液至3mL。向培养体系加入各试验组加入原料0.3mg,孵育1h,再加入40μL质量浓度为1.0mg/mL的SDS,染毒25min。阴性对照组:培养液中加入40μL的PBS;阳性对照组:培养液中仅加入40μL质量浓度为1.0mg/m L的SDS;以100μL Hanks液重悬细胞,用于彗星电泳测定溶血率(委托北京工商大学生物化学系检测)。
试验结果见表1。

注:与阳性对照组比较*P<0.05,**P<0.01;与试验1组比较##P<0.01。
试验结论:上述试验表明,交替单胞菌发酵产物提取物与β-葡聚糖组合后,能够降低SDS所致溶血率,更进一步交替单胞菌发酵产物提取物与β-葡聚糖、野大豆(GLYCINESOJA)籽提取物组合后(试验2组),交替单胞菌发酵产物提取物与β-葡聚糖、野大豆(GLYCINE SOJA)籽提取物、糖鞘脂类组合后(试验3组),能够显著降低SDS所致溶血率,与试验1组比较,具有极显著差异,充分说明试验2组和试验3组,具有极好的抗刺激性的作用。(溶血率越低,原料的抗刺激能力越强)。
制备实施例
实施例1
一种提高皮肤耐受性的组合物:交替单胞菌发酵产物提取物8g、β-葡聚糖0.5g、野大豆(GLYCINE SOJA)籽提取物0.8g、糖鞘脂类0.5g,去离子水15g、甘油0.8g、1,3-丁二醇1.5g。
制备方法为:
(1)取β-葡聚糖、交替单胞菌发酵产物提取物,加入到野大豆(GLYCINESOJA)籽提取物,搅拌至完全均匀,然后用均质器均质1.5分钟,备用原料Ⅰ;
(2)将水、甘油、1,3-丁二醇、糖鞘脂类混合均匀,注入到高压高剪切纳米均质机中,升温至60-65℃,均质3分钟,循环6次,得到备用原料Ⅱ;
(3)将备用原料Ⅰ和备用原料Ⅱ混合均匀,得到液体制剂。
实施例2
一种提高皮肤耐受性的组合物:交替单胞菌发酵产物提取物12g、β-葡聚糖1.5g、野大豆(GLYCINE SOJA)籽提取物1.2g、糖鞘脂类1.0g,去离子水25g、甘油1.2g、1,3-丁二醇2.5g。
制备方法为:
(1)取β-葡聚糖、交替单胞菌发酵产物提取物,加入到野大豆(GLYCINESOJA)籽提取物,搅拌至完全均匀,然后用均质器均质2.5分钟,备用原料Ⅰ;
(2)将水、甘油、1,3-丁二醇、糖鞘脂类混合均匀,注入到高压高剪切纳米均质机中,升温至60-65℃,均质6分钟,循环3次,得到备用原料Ⅱ;
(3)将备用原料Ⅰ和备用原料Ⅱ混合均匀,得到液体制剂。
实施例3
一种提高皮肤耐受性的组合物:交替单胞菌发酵产物提取物10g、β-葡聚糖1g、野大豆(GLYCINE SOJA)籽提取物1g、糖鞘脂类0.8g,去离子水20g重量份、甘油1g、1,3-丁二醇2g。
制备方法为:
(1)取β-葡聚糖、交替单胞菌发酵产物提取物,加入到野大豆(GLYCINESOJA)籽提取物,搅拌至完全均匀,然后用均质器均质2分钟,备用原料Ⅰ;
(2)将水、甘油、1,3-丁二醇、糖鞘脂类混合均匀,注入到高压高剪切纳米均质机中,升温至60-65℃,均质4.5分钟,循环5次,得到备用原料Ⅱ;
(3)将备用原料Ⅰ和备用原料Ⅱ混合均匀,得到液体制剂。
实施例4
一种提高皮肤耐受性的组合物:交替单胞菌发酵产物提取物9g、β-葡聚糖0.6g、野大豆(GLYCINE SOJA)籽提取物0.9g、糖鞘脂类0.6g,去离子水18g、甘油0.9g、1,3-丁二醇1.8g。
制备方法为:
(1)取β-葡聚糖、交替单胞菌发酵产物提取物,加入到野大豆(GLYCINESOJA)籽提取物,搅拌至完全均匀,然后用均质器均质2分钟,备用原料Ⅰ;
(2)将水、甘油、1,3-丁二醇、糖鞘脂类混合均匀,注入到高压高剪切纳米均质机中,升温至60-65℃,均质4分钟,循环5次,得到备用原料Ⅱ;
(3)将备用原料Ⅰ和备用原料Ⅱ混合均匀,得到液体制剂。
实施例5
一种提高皮肤耐受性的组合物:交替单胞菌发酵产物提取物11g、β-葡聚糖1.4g、野大豆(GLYCINE SOJA)籽提取物1.1g、糖鞘脂类0.9g,去离子水24g份、甘油1.1g、1,3-丁二醇2.4g。
制备方法为:
(1)取β-葡聚糖、交替单胞菌发酵产物提取物,加入到野大豆(GLYCINESOJA)籽提取物,搅拌至完全均匀,然后用均质器均质2分钟,备用原料Ⅰ;
(2)将水、甘油、1,3-丁二醇、糖鞘脂类混合均匀,注入到高压高剪切纳米均质机中,升温至60-65℃,均质5分钟,循环4次,得到备用原料Ⅱ;
(3)将备用原料Ⅰ和备用原料Ⅱ混合均匀,得到液体制剂。
试验例:刺激性测试
面霜处方见表2:

制备方法为:
(1)将水、EDTA二钠、甘油、丙烯酰二甲基牛磺酸铵/VP共聚物加入到水相釜中,加热至80-85℃搅拌溶解完全,备用。
(2)将C14-22醇、C12-20烷基葡糖苷、辛酸/癸酸甘油三酯加入到油相釜中,加热至80-85℃搅拌溶解完全,备用。
(3)将上述(2)加入到(1)中,真空搅拌均质2分钟。然后降温至40-45℃,备用。
(4)在另一个不锈钢容器中加入1,3-丁二醇、1,2-己二醇、对羟基苯乙酮,加热至60-65℃搅拌溶解完全,然后加入到上述的(3)料体中,搅拌均匀,降温至40-45℃,备用。
(5)取实施例液体制剂,加入(4)中,加入氢氧化钠、柠檬酸调pH值至5.0-6.0,搅拌均匀,然后出料,灌装得到面霜。
试验方法:将0.1g面霜涂于约左前臂4cm2的皮肤上,早晚一次,连续使用28天。相同的方法用面霜基质作对照物(去除本发明实施例3液体制剂)。第29、30天停用产品,第31天分别涂10%乳酸溶液进行刺激性测试。
评分标准如下:0:没有刺痛感
0.5:轻微刺痛感;
1:中度刺痛感;
2:发热感、发红、明显刺激感。
试验结果见表3。

试验结论:上述试验表明,本发明组合物加入化妆品后能够明显提高皮肤对刺激物耐受性。
Claims (2)
1.一种提高皮肤耐受性的组合物,其特征在于该组合物制备成液体制剂,该液体制剂原料为:
交替单胞菌发酵产物提取物8-12重量份、β-葡聚糖0.5-1.5重量份、野大豆籽提取物0.8-1.2重量份、糖鞘脂类0.5-1.0重量份,去离子水15-25重量份、甘油0.8-1.2重量份、1,3-丁二醇1.5-2.5重量份。
2.根据权利要求1所述的一种提高皮肤耐受性的组合物,其中液体制剂的制备方法为:
(1)取β-葡聚糖、交替单胞菌发酵产物提取物,加入到野大豆(GLYCINE SOJA)籽提取物,搅拌至完全均匀,然后用均质器均质1.5-2.5分钟,备用原料Ⅰ;
(2)将水、甘油、1,3-丁二醇、糖鞘脂类混合均匀,注入到高压高剪切纳米均质机中,升温至60-65℃,均质3-6分钟,循环3-6次,得到备用原料Ⅱ;
(3)将备用原料Ⅰ和备用原料Ⅱ混合均匀,得到液体制剂。
