本发明涉及一种补肾壮阳的中药组合物及其制备方法和用途,特别涉及一种由肉苁蓉、海马、蛇床子以及韭菜籽组成的具有补肾壮阳功效的中药组合物及其制备方法和用途。本发明属于医药技术领域。
背景技术
中医所说“肾气”,包括肾阴与肾阳两个方面,即“肾之精气”。肾有阴阳,肾精属阴,肾命门火属阳。肾藏精主生殖,肾阳除了温煦全身,增强各脏腑的功能活动外,还与泌尿和性功能有关。因此,肾阳虚就会出现骨软无力,全身怕冷,夜尿频数,性功能低下等征象。肾阳虚证是中医临床常见证型之一。对于肾阳虚证的论述可追溯到《内经》,如《素问·厥论篇》曰“阳气衰于下则为寒厥,阴气衰于下则为热厥”,“少阴厥逆,虚满呕变,下泄清”;《素问·至真要大论篇》曰“诸寒收引,皆属于肾”。以上是对肾阳虚证的症候和病机特点的概括性描述。目前尚缺少治疗阳虚的药物,因此补肾壮阳药物的研发成为阳虚患者的迫切需求。
有较多的中药有补肾壮阳的功效,其中也包括肉苁蓉、海马、蛇床子、韭菜籽。中药肉苁蓉为列当科植物肉苁蓉的干燥带鳞叶的肉质茎。性温、味甘、咸。归肾、肠经。为补肾壮阳药。始载于《神农本草经》,有补肾阳、益精血之功效。主治阳屡、腰膝酸软、筋骨无力等,有较好的药用价值,素有“沙漠人参”之美誉。海马,归肾经,味甘,性温,可补肾壮阳,《本草纲目》记载“海马暖水脏,壮阳道”。同时,《本草新编》也提及“海马入肾经命门,专善兴阳,功不亚于海狗,谁知海马不论雌雄,皆能勃兴阳道”;蛇床子系伞形科蛇床属植物蛇床的果实,性温,味辛、苦,归肾经。具有温肾壮阳、燥湿、祛风之功效,用于阳萎、宫冷、湿痹腰痛等症。蛇床子辛苦而温,温肾壮阳之功甚佳,在《千金方》30余治疗肾虚阳痿精冷方中,用蛇床子方达半数以上。韭菜籽为百合科植物韭菜的种子,性温,味微甘。韭菜籽始载于《名医别录》,具有固精、助阳、补肾、暖腰膝的功效,临床上可用于治阳痪、早泄、遗精、多尿等症。
补肾壮阳药有增强男性体质、抗疲劳、提高生殖能力等作用。而且,目前治疗男性的“阳虚”也是以补肾壮阳为主。但一个补肾壮阳的方剂并非药物的简单堆砌,也不是单纯将药效相加,而是根据病情需要,在辨证基础上,配伍适当药物。本发明通过合理搭配4中补肾壮阳的中药组合物能有效改善阳虚症状。
发明内容
本发明所要解决的技术问题是提供一种补肾壮阳的中药组合物及其制备方法和用途。
为了达到上述目的,本发明采用了以下技术手段:
本发明人将由肉苁蓉、海马、蛇床子、韭菜籽四种中药组成的组合物分成低、高剂量(0.02ml/g和0.04ml/g),分别给小鼠灌胃,通过大量的实验证该组合物可有效增重抗疲劳;同时,能够显著延长小鼠在密闭容器中的存活时间,增强小鼠耐缺氧能力;并且,给予阳虚小鼠中低、高剂量的该组合物后,药物在延长小鼠耐冻时间的同时可以明显缩短“阳虚”小鼠阴茎勃起潜伏期。与此同时,低、高剂量组合物可有效提高阳虚小鼠精液囊-前列腺、睾丸和包皮腺的脏器系数,并且低、高剂量组合物有效增加了“阳虚”小鼠的精子数量。
在上述研究的基础上,本发明提出了一种补肾壮阳的中药组合物,所述的中药组合物由以下重量百分比的各原料药组成:肉苁蓉40-50%、海马5-10%、蛇床子 20-30%以及韭菜籽20-30%。
其中,优选的,所述的中药组合物由以下重量百分比的各原料药组成:肉苁蓉45%、海马7.5%、蛇床子25%以及韭菜籽22.5%。
进一步的,本发明还提出了一种制备所述的中药组合物的方法,包括以下步骤:
(1)按照本发明所述的重量百分比称取各原料药;
(2)将称取的各原料药分别粉碎成粗粉,加水煎煮3次,每次1-2小时,合并 3次的过滤液,浓缩即得。
其中,优选的,浓缩至每毫升相当于1g生药。
更进一步的,本发明还提出了一种用于补肾壮阳的药物制剂,即,向本发明所述的中药组合物中加入制剂成型所需的辅料,按照药物制剂常规方法做成临床上适宜的各种制剂。
其中,优选的,所述的制剂为水煎剂、胶囊剂、丸剂、颗粒剂、片剂或口服液。
更进一步的,本发明还提出了所述的中药组合物在制备补肾壮阳的药物或保健品中的用途。
本发明的中药组合物具有明显的补肾壮阳作用,可有效增强体质和治疗阳虚。因此,本发明为改善阳虚症状,提供了一种新的药物选择。

本实验选取体重20-30g雄性昆明小鼠,随机分为3组,每组10只并进行小鼠体重记录;空白对照组小鼠每日灌胃生理盐水0.04ml/g,用药低剂量组小鼠每日同时灌胃实施例1制备的中药组合物0.02ml/g和0.02ml/g生理盐水,用药高剂量组小鼠每日灌胃中药组合物0.04ml/g。灌胃2周后再次对小鼠进行称重,计算小鼠体重增加值。随后,将小鼠至于水深20cm,水温20℃,水表面积2000cm2的游泳池中,实验前按实际重量的5%负重,记录小鼠开始游泳到沉入水底不再向上挣扎的游泳时间。结果如表1所示,说明中低、高剂量中药组合物有效增加小鼠体重,并有抗小鼠疲劳作用。

实施例6、中药组合物延长小鼠缺氧生存时间
小鼠给药方法与实施例5相同。实验时将小白鼠放入含有5g钠石灰的250ml 广口瓶内密闭,记录存活时间。结果如表2所示,说明中低、高剂量中药组合物有效延长小鼠缺氧生存时间。

实施例7、中药组合物延长阳虚小鼠耐冻时间
选取体重20-30g雄性昆明小鼠,随机分为5组,正常对照组、肾阳虚模型组、阳性对照组、中药组合物低剂量组和中药组合物高剂量组,每组10只,正常对照组腹腔注射25mg/kg生理盐水,其余各组每天均腹腔注射氢化可的松25mg/kg,共注射10天,构建阳虚模型。造模同时,正常对照组和阳虚模型组每天灌胃生理盐水0.04ml/g,阳性对照组每天腹腔注射丙酸睾酮5mg/kg,用药低剂量组小鼠同时灌胃实施例1制备的中药组合物0.02ml/g和0.02ml/g蒸馏水,用药高剂量组小鼠每日灌胃中药组合物0.04ml/g。
将铁皮制成大盒,提前在-10℃冰箱预冷,铁皮盒内含小室,每室放入1只小白鼠,置于-10℃恒温冰箱,每5分钟观察1次,记录小鼠存活时间。结果如表3所示,说明低、高剂量中药组合物均可明显延长小鼠耐冻时间。

实施例8、中药组合物缩短阳虚小鼠阴茎勃起潜伏期,并提高阳虚小鼠性器官系数。
小鼠处理方法与实施例7相同。利用药理生理多用仪,电刺激小鼠阴茎部位 0.5s,频率8Hz,电压10V。记录自刺激开始至阴茎完全勃起的时间,为阴茎勃起潜伏期。结果如表4所示,说明低、高剂量中药组合物均可缩短阳虚小鼠阴茎勃起潜伏期。
小鼠按实施例7相同方法处理后称重并处死,即刻称取包皮腺、睾丸、前列腺-精囊腺等器官重量,计算生殖器官脏器系数。结果如表4所示,肾阳虚模型组精子数量显著减少,阳性药物丙酸睾酮可使精子数量显著增加,低、高剂量中药组合物均可使阳虚小鼠附睾精子数量增加。
分离左侧附睾,置于20ml PBS溶液中剪碎并反复吹打,使精子游离出来,37℃孵育30分钟后,用细胞计数仪计算精子数量。结果如表4所示,说明肾阳虚模型组精子数量显著减少,阳性药物丙酸睾酮可使精子数量显著增加,低、高剂量中药组合物均可使附睾精子数量增加。

Claims (7)
1.一种补肾壮阳的中药组合物,其特征在于,所述的中药组合物由以下重量百分比的各原料药组成:肉苁蓉40-50%、海马5-10%、蛇床子20-30%以及韭菜籽20-30%。
2.如权利要求1所述的中药组合物,其特征在于,所述的中药组合物由以下重量百分比的各原料药组成:肉苁蓉45%、海马7.5%、蛇床子25%以及韭菜籽22.5%。
3.一种制备权利要求1或2所述的中药组合物的方法,其特征在于,包括以下步骤:
(1)按照权利要求1或2所述的重量百分比称取各原料药;
(2)将称取的各原料药分别粉碎成粗粉,加水煎煮3次,每次1-2小时,合并3次的过滤液,浓缩即得。
4.如权利要求3所述的方法,其特征在于,浓缩至每毫升相当于1g生药。
5.一种用于补肾壮阳的药物制剂,其特征在于,向权利要求1或2所述的中药组合物中加入制剂成型所需的辅料,按照药物制剂常规方法做成临床上适宜的各种制剂。
6.如权利要求5所述的药物制剂,其特征在于,所述的制剂为水煎剂、胶囊剂、丸剂、颗粒剂、片剂或口服液。
7.权利要求1或2所述的中药组合物在制备补肾壮阳的药物或保健品中的用途。
