Abstract
本发明公开了一种核酸诊断冻干试剂,包括含有2%‑10%海藻糖,2%‑5%葡聚糖,0.5%‑2%PEG8000,0.01mg‑0.02mg/ml BSA蛋白的冻干保护剂,所述冻干保护剂中含有肺炎支原体、肺炎衣原体和腺病毒三套引物和探针。本发明的优点在于冻干试剂中同时含有肺炎支原体,肺炎衣原体,腺病毒三种病原体核酸,能实现对多种病原体核酸的扩增检测,即采用一个反应体系可同时检测三个靶标,大大节约了试剂和检测耗材,检测通量高,扩大了临床应用范围。本发明的冻干试剂在冻干处理时,首先采用液氮对液态试剂进行快速预冻,缩短了后续冻干过程,提高了冻干试剂的稳定性。
Description
技术领域
本发明涉及分子生物学领域,尤其是涉及一种能同时区分三种病原体核酸的冻干试剂。
背景技术
PCR(聚合酶链式反应)是一种用于特定核酸片段的扩增放大技术,该技术广泛应用于基因检测和病原体检测,具有灵敏度高,特异性好,简便,快捷等特点,对生物和医学研究及临床诊断意义重大。
肺炎支原体(M.Pneumonia)是人类支原体肺炎的病原体。支原体肺炎的病理改变以间质性肺炎为主,有时并发支气管肺炎,称为原发性非典型性肺炎,儿童易造成重症肺炎,所以及时检测并用药对患者治疗意义重大。
人类肺炎衣原体感染呈世界性流行,不分地域、不受种族和年龄限制。人类肺炎衣原体感染一年四季均可能发生,不呈季节性特征,其流行可呈散发性或爆发性传播,尤其在空间相对封闭、人群聚集较多、空气不太流通的公共场所。尤其免疫低下者易引起肺部感染,及时检测并用药对患者治疗意义重大。
腺病毒对呼吸道、胃肠道、尿道和膀胱、眼、肝脏等均可感染,呼吸道感染的典型症状是咳嗽、鼻塞和咽炎,同时伴有发热、寒战、头痛和肌肉痛等;冬春流行季节尽量少去人员密集的公共场所,外出时戴口罩,避免接触病人,以防感染。腺病毒及时检测并用药对患者治疗意义重大。
传统用于检测肺炎支原体、肺炎衣原体、腺病毒的荧光PCR检测试剂多为液态的单重检测试剂,一个反应体系只能检测一个靶标,收费高、检测项目单一,耗材和试剂成本高,检测通量低,不能满足临床检测需求。
发明内容
本发明的目的在于提供一种能同时检测肺炎支原体、肺炎衣原体和腺病毒三种核酸的冻干试剂。
为实现上述目的,本发明可采取下述技术方案:
本发明所述的能同时区分三种病原体核酸的冻干试剂,包括含有2%-10%海藻糖,2%-5%葡聚糖,0.5%-2% PEG8000,0.01mg-0.02mg/ml BSA蛋白的冻干保护剂,所述冻干保护剂中含有肺炎支原体、肺炎衣原体和腺病毒三套引物和探针。
所述肺炎支原体引物包括肺炎支原体上游引物ccaaaaatggtcctacaccgact,肺炎支原体下游引物cgtgttcccaaaataggtttcca;所述肺炎支原体探针为ctccccgcttacggcgaggtgaatggg。
所述肺炎衣原体引物包括肺炎衣原体上游引物caactttgggagctgaattcca,肺炎衣原体下游引物ttacagagaattgcgatacgt;所述肺炎衣原体探针为cagtccaaacctaaagttgaagaacttaat。
所述腺病毒引物包括腺病毒上游引物gttgaccgggaagacaataccta,腺病毒下游引物gaatgtgctggccatgtcaag;所述腺病毒探针为cgctacacgctggctgtaggcgacaac。
所述冻干试剂的冻干过程为:
将含有2%-10%海藻糖,2%-5%葡聚糖,0.5%-2% PEG8000,0.01mg-0.02mg/ml BSA蛋白的保护剂与肺炎支原体、肺炎衣原体和腺病毒三套引物和探针按常规方法配制成液态试剂,然后将液体试剂加入涂覆有疏水材料层的预冻板凹坑内,使凹坑内的液体试剂呈现出球形液滴状,最后将预冻板浸入液氮中预冻2~5分钟,收集凝固的球形预冻试剂直接放入冻干机中进行真空冷冻干燥即可。
本发明的优点在于冻干试剂中同时含有肺炎支原体,肺炎衣原体,腺病毒三种病原体核酸,能实现对多种病原体核酸的扩增检测,即采用一个反应体系可同时检测三个靶标,大大节约了试剂和检测耗材,检测通量高,扩大了临床应用范围。本发明的冻干试剂在冻干处理时,首先采用液氮对液态试剂进行快速预冻,缩短了后续冻干过程,提高了冻干试剂的稳定性。

具体实施方式
本发明所述的能同时区分三种病原体核酸的冻干试剂,其成分含有2%-10%海藻糖,2%-5%葡聚糖,0.5%-2% PEG8000,0.01mg-0.02mg/ml BSA蛋白的冻干保护剂,Tris缓冲盐,氯化钾,dNTPs,镁离子,引物,探针,Taq酶。在冻干试剂中含有肺炎支原体、肺炎衣原体和腺病毒三套引物和探针,可同时对三种病原体核酸进行检测。
实际配制时,该冻干试剂的各成分可按下表量取:
注:表中所述“肺支”为肺炎支原体,“肺衣”为肺炎衣原体。
按上表中成分配制成液态试剂后,将液体试剂加入涂覆有疏水材料层的预冻板凹坑内(预冻板有一定厚度,上面均匀设置有多个凹坑),由于疏水材料层的作用,加入凹坑内的液体试剂会呈现出球形液滴状(如图1所示),将预冻板浸入液氮中预冻2~5分钟(注意预冻时液氮不能进入凹坑内,保证试剂的洁净度),最后收集凝固的球形预冻试剂,将其直接放入冻干机中进行真空冷冻干燥即可。
本发明冻干试剂的检测效果:
核酸诊断虽被广泛应用于各种病原体的检测上,但是核酸扩增反应试剂在常温下保存并不稳定,而且常规液态核酸检测试剂每次使用都必须要反复冻融,这对试剂的活性也有很大的影响,并且各组分不能混合在一起,用时需混合,操作繁琐,试剂盒的长期保存和长途运输将会受到很大的影响,容易因保存温度的不当而使其性能下降甚至完全失效。本发明为全预混冻干的多重PCR联检试剂可有效克服上述缺点,完全满足了临床的检测需求。
Claims (5)
1.一种能同时区分三种病原体核酸的冻干试剂,其特征在于:包括含有2%-10%海藻糖,2%-5%葡聚糖,0.5%-2% PEG8000,0.01mg-0.02mg/ml BSA蛋白的冻干保护剂,所述冻干保护剂中含有肺炎支原体、肺炎衣原体和腺病毒三套引物和探针。
2.根据权利要求1所述的能同时区分三种病原体核酸的冻干试剂,其特征在于:所述肺炎支原体引物包括肺炎支原体上游引物ccaaaaatggtcctacaccgact,肺炎支原体下游引物cgtgttcccaaaataggtttcca;所述肺炎支原体探针为ctccccgcttacggcgaggtgaatggg。
3.根据权利要求1所述的能同时区分三种病原体核酸的冻干试剂,其特征在于:所述肺炎衣原体引物包括肺炎衣原体上游引物caactttgggagctgaattcca,肺炎衣原体下游引物ttacagagaattgcgatacgt;所述肺炎衣原体探针为cagtccaaacctaaagttgaagaacttaat。
4.根据权利要求1所述的能同时区分三种病原体核酸的冻干试剂,其特征在于:所述腺病毒引物包括腺病毒上游引物gttgaccgggaagacaataccta,腺病毒下游引物gaatgtgctggccatgtcaag;所述腺病毒探针为cgctacacgctggctgtaggcgacaac。
5.根据权利要求1所述的能同时区分三种病原体核酸的冻干试剂,其特征在于:所述冻干试剂的冻干过程为:
