Abstract
本发明的目的在于提供制备成骨细胞的方法,所述成骨细胞在对伴随各种肿瘤、外伤、手术等的骨缺损的修复、牙周病为代表的骨吸收、骨折、骨质疏松症等的治疗中能够应用,且癌变的危险性较少。作为解决该课题的办法,提供包括向哺乳动物的体细胞导入Oct9基因或其表达产物的工序,并从上述体细胞制备成骨细胞的方法。

技术领域
本发明涉及成骨细胞及其制备方法,详细而言涉及基于直接重编程的成骨细胞的制备方法。
背景技术
如果以修复伴随骨肿瘤、外伤、骨髓炎等的骨缺损或骨肿瘤等的刮治后的骨缺损为目标,向病变部移植成骨细胞,则可以促进骨形成,期待功能性和形态的预后改善。实际上,例如已经进行了将从患者的松质骨(cancellous bone)收集的骨髓细胞进行自体移植的治疗,其有效性已知。在该情况下,认为从自体骨髓细胞中包含的间充质干细胞诱导分化了成骨细胞,促进了骨形成和重建。另一方面,随着老龄化,骨质疏松症的罹患率已经增加,也存在老人如果骨折则导致长期卧床的情况。认为成骨细胞的移植可以促进治愈骨质疏松症、外伤等伴随的骨折、难治性骨折、假骨折(pseudofracture)。成骨细胞的移植也可能对以下方面有效:类风湿性关节炎、突发性股骨头坏死、变形性关节症、腰椎峡部畸形症(lumbar spondylosis deformans)、椎管狭窄症、椎间盘突出症、峡部裂(spondylolysis)、脊椎松解型滑脱症、脊柱侧弯症、脊髓型颈椎病、后纵韧带骨化症、脊髓损伤、变形性髋关节症(coxarthrosis)、变形性膝关节症(gonarthrosis)、股骨头滑脱症、骨软化症、手术后的骨的修复(心脏手术之后的胸骨的修复等)、人工踝关节手术所伴随的缺陷部的修复、骨髓炎、骨坏死(osteonecrosis)等。
另一方面,牙周病也被称为第四生活方式病,罹患率极高,并构成了各种全身性疾病的原因。随着牙周病的进行而发生牙槽骨的骨吸收,所以认为如果将成骨细胞效率良好地供应到局部的骨吸收部,则与牙槽骨的再生治疗相关。
另外,如果将成骨细胞的移植与骨移植、人工骨移植、人工关节、植入进行组合使用,则有提高治疗效果的可能性。
作为这样的移植目标的成骨细胞,迄今为止已使用了骨髓间充质干细胞、包含骨髓间充质干细胞的骨髓细胞等。但是,骨髓的获取对患者的侵害较大,也存在不能供应足够数量的骨髓细胞的情况等问题。另一方面,如果使用人胚胎干细胞(ES细胞),则不需要从患者获取骨髓,也有能够供应足够数量的成骨细胞的可能性,但有伦理问题,除此以外,存在移植后残存的ES细胞发生肿瘤化的危险性。另外,即使使用了iPS细胞不需要从患者收集骨髓,并有能够供应足够数量的成骨细胞的可能性,但存在移植后残存的iPS细胞发生肿瘤化的危险性。
在非专利文献1中进行了对人ES细胞的Osterix的慢病毒载体导入+利用成骨性(Osteogenic)培养基的向成骨细胞的诱导分化。
在非专利文献2、3中,从小鼠iPS细胞经过MSC而利用成骨性培养基进行诱导分化,得到了成骨细胞。
在非专利文献4中,向小鼠iPS细胞导入Runx2的腺病毒载体,利用成骨性培养基进行诱导分化,得到了成骨细胞。如非专利文献1~4所示那样,成骨细胞是从ES细胞、iPS细胞等多功能性干细胞进行诱导分化而制成的,因此需要长期的培养,又存在癌变的危险性。
对于向体细胞导入组织特异性的转录因子的基因群,直接诱导分化(不经过iPS细胞)为该组织细胞(直接重编程(直接转换)),例如有以下报告:
小鼠成纤维细胞→软骨细胞(导入SOX9+Klf4+c-Myc基因)
小鼠成纤维细胞→心肌细胞(导入GATA4+Mef2c+Tbx5基因)

在专利文献1中公开了用于向体细胞导入指定的基因群而有效地制备(直接转换)有功能性的成骨细胞的方法。然而,仍在追求更加优异的方法。

发明要解决的问题
本发明目的在于提供制备成骨细胞的方法,所述成骨细胞在对伴随各种肿瘤、外伤、手术等的骨缺损的修复、牙周病为代表的骨吸收、骨折、骨质疏松症等的治疗中能够应用,且癌变的危险性较少。
解决问题的方法
本发明发现,通过向哺乳动物的体细胞导入Oct9基因,从而不经过ES细胞、iPS细胞等多功能性干细胞,而直接(直接重编程)得到成骨细胞。
第1项,包括向哺乳动物的体细胞导入Oct9基因或其表达产物的工序,并从上述体细胞制备成骨细胞的方法。
第2项,通过向哺乳动物的体细胞导入Oct9基因或其表达产物,以及选自c-Myc基因、L-Myc基因及N-Myc基因中的至少一种或其表达产物,从而从上述体细胞制备成骨细胞的方法。
第3项,根据第1或2项中记载的方法,其中,上述体细胞为成纤维细胞。
第4项,根据第1~3项中任一项所述的方法,其进一步包括将经导入基因或其表达产物的体细胞,利用成骨细胞的诱导培养基进行培养的工序。
第5项,成骨细胞,其源自哺乳动物的体细胞并具有外源性的Oct9基因或其表达产物。
本发明包括以下成骨细胞及其制备方法。
发明效果
在本发明中,可通过直接重编程而从体细胞在短时间内提供成骨细胞。该成骨细胞由于可以从待移植的本人的体细胞容易地诱导,所以在移植了成骨细胞本身或由其制成的骨组织的情况下,也不出现免疫排斥反应等问题。另外,由于可不经过iPS细胞、ES细胞而直接从体细胞诱导成骨细胞,因此可避免癌变等由多功能性干细胞引起的问题。

具体实施方式
本发明涉及通过将哺乳动物的分化的体细胞转换为成骨细胞,从而制备成骨细胞的方法。“转换”是指将体细胞转变向目标的成骨细胞。本发明的方法的优选实施方式之一为“直接重编程”(也称为“直接转换”)的不经过以制成iPS细胞为代表的细胞的初始化的工序而将体细胞转换为成骨细胞的方法。
在本发明的方法的优选实施方式中,体细胞不进行基因的导入而被转换向成骨细胞。“不进行基因的导入”是指,体细胞的初始的基因组序列(主要指DNA的碱基序列)不变,而被转换向成骨细胞。或者,“不进行基因的导入”是指基于体细胞的初始的内在基因的功能,而将其转换向其它体细胞。
成骨细胞
本发明提供成骨细胞的制备方法。根据本发明,也可以制备前成骨细胞、不成熟成骨细胞、成熟成骨细胞、骨细胞等。在本说明书中,为简单起见,将它们全部称为成骨细胞。
可以通过以下检测确认得到成骨细胞:ALP(碱性磷酸酶)的活性的检测;基于例如mRNA的实时PCR等检测ALP、骨钙素(Osteocalcin,OC)、骨桥蛋白(Osteopontin)、Runx2等标记基因的表达;茜素红S染色、von Kossa染色(矿化的骨基质的产生)等。
Runx2在骨形成中是必要的转录因子。Runx2在活体中的从间充质干细胞向成骨细胞的分化中担任必不可少的角色。对间充质干细胞进行Runx2的强制表达,使OC(骨钙素)、BSP(Bone sialo-protein)、ALP(碱性磷酸酶)、COL1A1等成骨细胞特异性的基因升高。在Runx2敲除(knockout)小鼠中,由于成熟成骨细胞的缺失而完全不显示膜内骨化和软骨内骨化。但是,源自该小鼠的间充质干细胞有向脂肪细胞和软骨细胞的诱导能力。
ALP(碱性磷酸酶)是成骨细胞从早期向中期分化的标记。ALP多包含于成骨细胞的膜表面和由成骨细胞分泌的基质小胞,参与钙化基质产生的开始。
骨钙素(Osteocalcin,OC)为成骨细胞特异性表达,认为其有助于骨形成。
基于茜素红S的染色、von Kossa染色可以检测骨形成的重要的要素之一的矿化的骨基质的产生,即钙的沉积。
作为使用根据本发明得到的成骨细胞(移植材料)治疗的对象的疾病,可列举:骨肿瘤、外伤、骨髓炎等伴随的骨缺损或骨肿瘤等刮治后的骨缺损、骨折、骨质疏松症、牙周病、牙槽骨吸收、类风湿性关节炎、突发性股骨头坏死、变形性关节症、腰椎峡部畸形症(lumbar spondylosis deformans)、椎管狭窄症、椎间盘突出症、峡部裂(spondylolysis)、脊椎松解型滑脱症、脊柱侧弯症、脊髄型颈椎病、后纵韧带骨化症、脊髓损伤、变形性髋关节症(coxarthrosis)、变形性膝关节症(gonarthrosis)、股骨头滑脱症、骨软化症、下颌骨重建等因复杂骨折而受到破坏的骨折部位的重建术、手术后的骨的修复(心脏手术后的胸骨的修复等)、人工踝关节手术伴随的缺陷部的修复、骨髓炎、骨坏死(osteonecrosis)等。另外,如果移植成骨细胞,则有与骨移植、人工骨移植、人工关节、植入组合使用而提高治疗效果的可能性。另外,也可以将成骨细胞用三维的支架等进行培养而在体外制作各种形态的骨组织,通过移植该骨组织,进行上述疾病的治疗。除此以外,也将与成骨细胞的缺陷、不足或功能降低相关的各种疾病作为对象。
在本说明书中,如果没有特别说明,则术语“治疗”意指患者在患有特定的疾病或障碍的期间进行的处理,由此而减轻疾病或障碍的严重程度、或减轻一个或多个其症状、或延迟或减速疾病或障碍的发展。本说明书中,在术语“治疗”中包括“预防”。
另外,在本发明中得到的成骨细胞不限于疾病的治疗,也可以用于美容目的。例如,可以通过在由于事故、手术等而缺陷的部位移植成骨细胞或由其制成的骨组织,从而使骨基质产生而修复缺陷部位,使其丰满而变得不明显。为本说明书中便利起见,将此时对人的处理也称为治疗,可以将“患者”替换为“健康的人”或“人”,将“疾病”替换为“美容”。
另外,本发明不仅可用于人,也可以用于包括犬、猫等宠物动物、牛、马、猪、绵羊、鸡等家畜的哺乳动物的疾病的治疗。在该情况下,将“患者”替换为“患畜”或“哺乳动物”。
移植材料是指为了骨组织的修复、重建而导入活体内的含有成骨细胞的材料。移植材料包括在体外部分地或完全使骨组织再生,移植到同一或其他个体的材料。在本发明中得到的成骨细胞可以用于移植材料的制作。成骨细胞本身也构成移植材料。因此,可以将成骨细胞作为细胞制剂而移植给患者,也可以与包括羟基磷灰石、活体吸收性陶瓷等人工材料的基体材料(支架(scaffold))一起进行移植,或与支架一起培养后进行移植。在这些情况下,支架可以根据移植目标而做成各种三维的形状。
体细胞
作为本发明的方法的对象的哺乳动物的已分化的体细胞,只要源自哺乳动物,不是成骨细胞本身及在活体内不具有分化向成骨细胞的能力的细胞即可,没有特别限定。
体细胞只要源自哺乳动物即可。在将成骨细胞移植到活体的情况下,由于减少了感染、排斥反应等危险而优选使用源自被移植的受试者的体细胞(自体细胞)。然而,在以对突然的骨折等进行移植等为目标的情况下,可以将不是自体细胞的,从他人、其它动物的体细胞预先准备好的成骨细胞用于移植。也可以从预先准备好的他人、其它动物的体细胞制备成骨细胞而用于移植。即,可以预先制作成骨细胞银行或成骨细胞前体细胞的银行,以供于移植目的。在这种情况下,为了减少排斥反应等危险,可以预先将MHC进行分型。另外,也可以预先确认成骨细胞的性质、致瘤性等。
在本说明书中,作为哺乳动物,可列举:小鼠、大鼠、仓鼠、人、犬、猫、猴、兔、牛、马、猪等,特别是人。
本发明也可以用于使用了成骨细胞的各种研究、技术开发等。对例如骨的发育和老化、形态发生、重建的机制,对它们的力学应激、营养、免疫、神经、激素的影响的分析等基础研究有用。另外,对在锶90等放射性物质的内部接触辐射的对骨的影响的分析和从骨除去锶90的技术的开发等也有用。
使用本发明时,可从具有各种疾病、遗传背景的人、动物简便、迅速、廉价地建立成骨细胞,所以能够通过生物化学、分子生物学、免疫学的等手段对与疾病、遗传背景相关的成骨细胞的异常进行分析,可以由此有助于疾病的发病机制的阐明等研究、诊断法的开发。另外,如果使用这种成骨细胞来进行药品的开发、药品的毒性试验等,则可以有助于针对各种疾病的新型治疗法的开发。
作为本发明的方法(直接重编程)的对象的体细胞,没有特别限定,可列举例如成纤维细胞、角质细胞、口腔粘膜上皮细胞、气管粘膜上皮细胞、胃粘膜上皮细胞、肠道粘膜上皮细胞、血管内皮细胞、平滑肌细胞、脂肪细胞、龈细胞(龈成纤维细胞、龈上皮细胞)、牙髓细胞、牙周韧带细胞、白细胞、淋巴细胞、肌细胞、结膜上皮细胞、破骨细胞等,优选为成纤维细胞、角质细胞、口腔粘膜上皮细胞、龈细胞、白细胞、淋巴细胞、破骨细胞等。
另外,也可列举从间充质干细胞(Mesenchymal stem cell:MSC)、神经干细胞(Neural stem cell)、肝干细胞(hepatic stem cell)、肠干细胞、皮肤干细胞、毛囊干细胞、色素细胞干细胞等体细胞干细胞而诱导分化或使其去分化或重编程而制成的体细胞。另外,也可列举从各种体细胞而诱导分化、或使其去分化或重编程而诱导成的其他体细胞。另外,也可列举从生殖系列的细胞进行诱导分化或使其去分化或重编程而诱导成的体细胞。
另外,也可列举从胚胎干细胞(Embryonic stem cell:ES细胞)、人工多功能性干细胞(induced pluripotent stem cell:iPS细胞)进行诱导分化或重编程而诱导成的体细胞。
另外,对ES细胞、iPS细胞、或生殖系列的细胞而言,严密来讲并非体细胞,但也包括于本发明的“体细胞”中(此时,将术语“体细胞”替换为“ES细胞”、“iPS细胞”或“生殖系列的细胞”)。
另外,可列举培养细胞,也可列举从培养细胞进行诱导分化、或使其去分化或重编程而诱导成的体细胞。
基因
本发明的方法包括向体细胞导入Oct9基因或其表达产物的工序。这里,“基因”只要是编码基因信息的那些即可,包括基因群DNA(通常为双链DNA)、cDNA(正链的单链DNA,或双链DNA)。另外,作为“表达产物”,可列举基因的mRNA或蛋白质。
Oct9(也称为POU class 3homeobox 4,POU3F4、Brain-specific homeobox/POUdomain protein 4、Brain-4或Brn-4)是属于POU结构域家族的3类的转录因子。认为其与八聚体基序(Octamer motif)(ATGCAAAT)特异性结合。Oct9在脑(视丘下部、海马)、内耳、胰腺等中表达。据称Oct9参与神经细胞的分化,重要参与神经干细胞的分化(文献A)。另外,据称Oct9对内耳的发育发挥重要的功能,Oct9的缺陷造成听力衰减(文献B)。但是,Oct9在成骨细胞中的功能尚属未知。
在文献C中示出了:可通过将Runx2、Osterix、Oct3/4和L-myc这4个因子导入成纤维细胞而诱导为成骨细胞,另外,通过将Oct4和L-myc这2个因子导入成纤维细胞,也可诱导为成骨细胞样的细胞。因此,在文献C中得到了Oct3/4是期望的基因的结论。但是,有报告Oct3/4在某种癌中表达(文献D、E),存在其参与肿瘤形成的可能性。
在文献C中对于Oct9没有任何提及,但这次发现,Oct9也与Oct3/4同等(或超过其)地通过单独或与myc家族的基因共导入而使成纤维细胞转换为成骨细胞样。由于Oct9与Oct3/4不同,不参与致癌,所以从该方面出发,有与Oct3/4比更优选Oct9的可能性。
另外,已知N-myc基因参与神经母细胞瘤等神经起源(neural origin)及神经内分泌(neuroendocrine)类的肿瘤的发病,其对骨肿瘤的参与尚属未知(文献F),所以存在共导入Oct9和N-Myc所诱导而成的成骨细胞的形成肿瘤的危险性低而优选的可能性。

从体细胞转变向成骨细胞的效率较高的观点出发,除了导入Oct9基因以外,可以导入选自c-Myc基因、L-Myc基因及N-Myc基因中的至少一种或其表达产物。即,作为本发明的实施方式的一部分,可列举Oct9和c-Myc的组合、Oct9和L-Myc基因的组合、及Oct9和N-Myc的组合。
在本发明的方法中,只要不破坏本发明的效果即可,也可以导入除了上述基因或其表达产物以外的其它基因。作为被追加导入的基因,可列举Oct4(也称为Oct3、Oct3/4)、Oct1A、Oct6、Klf家族(KLF1、KLF2、KLF3、KLF4、KLF5、KLF6、KLF7、KLF8、KLF9、KLF10、KLF11、KLF12、KLF13、KLF14、KLF15、KLF16、KLF17)、Lin-28、Sox1、Sox2、Sox3、Sox7、Sox15、Sox17、Sox18等重编程基因。另外,也可列举Runx2、Osterix、Dlx5等骨相关基因。这些基因可以一种单独追加或追加两种以上使用。
在本发明的方法中,被导入体细胞的基因或其表达产物可以是包括Oct9基因的一种或两种以上。从简便性的观点出发,被导入体细胞的基因或其表达产物可以是一种(仅Oct9基因或其表达产物)~4种左右(除了Oct9基因或其表达产物以外有3种左右),优选为一种、两种(除了Oct9基因或其表达产物以外有一种)或3种(除了Oct9基因或其表达产物以外有两种)。作为具体实施方式,可例示:仅Oct9基因或其表达产物;Oct9基因或其表达产物,以及选自c-Myc基因、L-Myc基因及N-Myc基因中的至少一种或其表达产物这共计两种。
上述基因均是在脊椎动物中高度保守的基因,在本说明书中,只要没有特别示出动物名,即作为表示包括同源的基因。另外,上述基因还包括与野生型的基因产物有同样的作用的基因,即使所述基因有含有多态性(polymorphism)的突变。

具体而言,优选将用于转变为成骨细胞的导入基因(骨相关基因和重编程相关基因的组合、或仅重编程相关基因)组装到表达载体,向作为对象的体细胞导入表达载体,使其在细胞内表达。
作为导入基因的方法,有感染逆转录病毒载体、腺病毒载体、慢病毒载体、腺相关病毒载体、疱疹病毒载体、仙台病毒载体等病毒性载体的方法,除此以外,在导入基因和其表达产物的情况下,也可以使用利用阳离子脂质体、阳离子聚合物、电穿孔法等非病毒载体,来转染质粒载体、游离型载体、基因的表达产物(mRNA,蛋白质)的方法。另外,也可以导入mRNA。将这些在基因导入中使用的办法全部包括在内,在本说明书中称为载体。
另外,可以与治疗目标的基因一起导入作为药品选择标记的基因(嘌呤霉素抗性、杀稻瘟菌素S抗性、新霉素抗性、潮霉素抗性等),之后通过进行药物选择而选择出表达治疗用基因的细胞后使用。

被导入体细胞的基因(基因组DNA、cDNA等)可以在适当的启动子的控制下进行转录。例如,可以利用逆转录病毒载体的LTR(long terminal repeat)启动子进行转录,也可以由载体内部的另外的启动子进行表达。可以利用例如CMV启动子、EF-1α启动子、CAG启动子等组成型表达启动子,或期望的诱导性启动子。另外,也可以利用将LTR的一部分置换为其它启动子的嵌合启动子。
培养
在本发明的方法中,可以将哺乳动物的已分化的体细胞在导入基因后在培养基中进行培养。
培养可以在用于容纳细胞及培养基的适当的容器中进行。作为优选的进行培养的手段,可例示在约37℃左右及二氧化碳浓度约5%左右的条件下进行培养的手段,但并不限定于此。在上述条件下的培养例如可以使用公知的CO2培养箱进行。
对进行培养的时间而言,在不损伤本发明的效果的范围内即可,没有特别的限定。进行培养的时间可以设为例如从24小时到60天左右。
在培养中,必要时可以进行传代。在进行传代的情况下,在达到融合状态前或达到后立即回收细胞,将细胞接种于新的培养基。另外,在本发明的培养中,可以适当更换培养基。
培养基
在本发明的方法中所使用的培养基没有特别限定。可以使用DMEM(Dulbecco'sModified Eagle's Medium)、EMEM(Eagle's minimal essential medium)等普通的液体培养基。根据需要,可以添加血清成分(胎牛血清(FBS)、人血清(HS))、链霉素、青霉素等抗菌药、非必需氨基酸等成分。
从根据本发明的方法可制备成骨细胞的效率较高的观点出发,作为培养基优选使用用于使成骨细胞分化的诱导分化培养基。“用于使成骨细胞分化的诱导分化培养基”是指包含可以使多功能性干细胞(ES细胞、iPS细胞等)向成骨细胞分化的成分的培养基。
作为用于使成骨细胞分化的诱导分化培养基,没有特别限定。可以使用例如:在普通培养基中添加了抗坏血酸(L-抗坏血酸)或其盐、β-甘油磷酸(β-Glycerophosphate)或其盐、地塞米松等肾上腺皮质激素而成的培养基。
具体而言,作为用于使成骨细胞分化的诱导分化培养基(骨诱导培养基),可例示将选自下组的成分(一种或两种以上)添加于普通液体培养基而成的那些:抗坏血酸(例如:浓度0.1~1000μg/ml左右,优选为1~100μg/ml左右);β-甘油磷酸(例如:浓度0.1~1000mM左右,优选为1~100mM左右);地塞米松(1nM~10mM左右,优选为10~1000mM左右)及氢化可的松等糖皮质激素。作为一个实施方式,可列举将50μg/ml左右的抗坏血酸、10mM左右的β-甘油磷酸、100nM左右的地塞米松(均为最终浓度)添加于添加了10%FBS或5%HS的DMEM等普通培养基而成的那些。但不限于这些。
从而,体细胞被转变为成骨细胞,制备了成骨细胞。
可以通过上述ALP的活性的检测,针对标记基因的实时RT-PCR(Reversetranscription polymerase chain reaction)、RT-PCR、茜素红S染色、von Kossa染色来确认得到了成骨细胞。
在某种实施方式中,制备而成的成骨细胞具有外源性的Oct9基因或其表达产物。这里,“外源性”主要是指作为上述导入办法的结果的所导入的基因或其表达产物的形态,指与天然的形态不同的形态。可列举例如:被除了天然的启动子以外的启动子所控制表达的基因、存在于除了天然以外的染色体上的位置、或者染色体外的基因的形态等。
实施例
虽然在以下示出了实施例,但本发明不仅限定于该实施例。
实施例1
利用PCR扩增了Oct1、Oct2、Oct5/7、Oct6、Oct8、Oct9、Oct11及N-Myc的各自的编码序列全长,插入于逆转录病毒载体质粒pMXs(Cell Biolabs Inc.、San Diego、CA、USA;catno.RTV-012)的EcoRI位点。已将Oct3/4、c-Myc及L-Myc的编码序列插入于pMXs的EcoRI位点的逆转录病毒载体质粒,来自日本京都大学的山中教授提供。以5.5×106/平皿的浓度将封装细胞PLAT-GP(Cell Biolabs Inc.、San Diego、CA、USA;cat no.cat、VPK-305)接种于10cm培养皿。次日,将上述各个质粒载体与pCMV-VSV-g质粒(Cell Biolabs Inc.、SanDiego、CA、USA;cat no.RV-110)使用X-treme Gene 9transfection reagent(RocheAppliedScience、Penzberg、Germany)共导入于该细胞。在24小时后,吸除培养上清液并加入不含有抗菌剂的新鲜培养基。再经过24小时后,回收培养上清液,使用孔径大小0.45μm的过滤器进行过滤,制成病毒悬浊液。
将人正常皮肤成纤维细胞(NHDF)重悬于添加了10%胎牛血清(FBS)、0.1mM非必需氨基酸、100μg/mL链霉素、100U/mL青霉素的Dulbecco's minimum essential medium(DMEM)培养基(完全培养基)中,接种于35mm平皿。在5%CO2/95%加湿的空气中,于37℃进行培养。次日,吸除培养上清液,加入包含具有图中记载的基因的逆转录病毒载体和4μg/mL的聚凝胺的培养基。在24小时之后吸除培养上清液,加入已添加了50μg/mL抗坏血酸、10mMβ-甘油磷酸酯和100nM地塞米松的完全培养基(成骨培养基)。每隔2~3天,用新鲜培养基更换培养基而进行培养。
导入基因后,在第28天从细胞提取总RNA。使用针对Runx2基因的特异性探针和引物,及针对β肌动蛋白基因的特异性探针和引物(Applied Biosystems)进行了实时RT-PCR。算出Runx2基因mRNA的水平相对于β肌动蛋白基因mRNA的水平,将非导入基因群中的值设为1,计算相对的Runx2mRNA水平。
将结果(平均±标准偏差,n=3)示于图1。可知在导入了Oct3/4、Oct6、Oct9、Oct1+L-myc、Oct2+L-myc、Oct3/4+L-myc、Oct5/7+L-myc、Oct6+L-myc、Oct9+L-myc的组中,显著诱导了Runx2的mRNA表达。[*p<0.05vs.非转换对照;+p<0.05vs.仅L-myc]。特别是导入了Oct3/4+L-myc和Oct9+L-myc的细胞,强烈表达了Runx2。进一步,导入了Oct9+L-myc的细胞最强烈地表达了Runx2。
实施例2
将人正常皮肤成纤维细胞(NHDF)重悬于添加了10%胎牛血清(FBS)、0.1mM非必需氨基酸、100μg/mL链霉素、100U/mL青霉素的Dulbecco’s minimum essential medium(DMEM)培养基(完全培养基)中,接种于35mm平皿。在5%CO2/95%加湿的空气中,于37℃进行培养。次日,吸除培养上清液,加入包含具有图中记载的基因的逆转录病毒载体和4μg/mL的聚凝胺的培养基。在24小时之后吸除培养上清液,加入已添加了50μg/mL抗坏血酸、10mMβ-甘油磷酸酯和100nM地塞米松的完全培养基(成骨培养基)。每隔2~3天,用新鲜培养基更换培养基而进行培养。
导入基因后,在第28天,利用4%多聚甲醛固定细胞后,添加抗Runx2抗体(Abnova、Taipei、Taiwan)进行培养。之后加入利用Alexa fluor 488结合的二次抗体,再用DAPI染色了核。作为对照,也同样地染色了成纤维细胞。以荧光显微镜于倍率100倍进行了观察。
将结果示于图2(比例尺=200μm)。可知成纤维细胞完全不表达Runx2蛋白,但导入了Oct3/4+L-myc及Oct9+L-myc的细胞表达了Runx2蛋白。可知特别是导入了Oct9+L-myc的细胞,表达了比导入了Oct3/4+L-myc的细胞更强的Runx2。
实施例3

实施例5
将人正常皮肤成纤维细胞(NHDF)重悬于添加了10%胎牛血清(FBS)、0.1 mM非必需氨基酸、100μg/mL链霉素、100 U/mL青霉素的Dulbecco’s minimum essential medium(DMEM)培养基(完全培养基)中,接种于35 mm平皿。在5%CO2/95%加湿的空气中,于37℃进行培养。次日,吸除培养上清液,加入包含具有图中记载的基因的逆转录病毒载体和4μg/mL的聚凝胺的培养基。在24小时之后吸除培养上清液,加入已添加了50μg/mL抗坏血酸、10 mMβ-甘油磷酸酯和100 nM地塞米松的完全培养基(成骨培养基)。每隔2~3天,用新鲜培养基更换培养基而进行培养。
导入基因后,在第28天从细胞提取总RNA。使用针对骨钙素基因的特异性探针和引物,及针对β肌动蛋白基因的特异性探针和引物(Applied Biosystems)进行了实时RT-PCR。算出骨钙素基因mRNA相对于β肌动蛋白基因mRNA的水平,将非导入基因群中的值设为1,计算相对的骨钙素mRNA水平。
可知导入了Oct3/4、Oct9、c-myc、N-myc、Oct3/4+c-myc、Oct9+c-myc、Oct3/4+N-myc及Oct9+N-myc的细胞表达了骨钙素基因mRNA。特别是导入了Oct9+N-myc的细胞最强烈地表达了Runx2。
实施例6
将人正常皮肤成纤维细胞(NHDF)重悬于添加了10%胎牛血清(FBS)、0.1 mM非必需氨基酸、100μg/mL链霉素、100 U/mL青霉素的Dulbecco’s minimum essential medium(DMEM)培养基(完全培养基)中,接种于35 mm平皿。在5%CO2/95%加湿的空气中,于37℃进行培养。次日,吸除培养上清液,加入包含具有Oct9和N-myc基因的逆转录病毒载体和4μg/mL的聚凝胺的培养基。在24小时之后吸除培养上清液,加入已添加了50μg/mL抗坏血酸、10mMβ-甘油磷酸酯和100nM地塞米松的完全培养基(成骨培养基)。每隔2~3天,用新鲜培养基更换培养基而进行培养。
在基因导入28天之后,从培养皿吸除培养液,用PBS洗涤2次,用95%乙醇进行固定。用灭菌蒸馏水洗涤后加入茜素红S染色液,于室温静置15分钟。作为对照,也同样地染色了成纤维细胞。
在图6上部,通过平皿的裸眼图像(倍率x1倍)及相位差显微镜图像(倍率x40倍)(比例尺=500μm)的照片来示出结果。被染色的是钙化骨基质(实际上被染色为红色)。
进一步,从所有孔除掉茜素红S染色液,用灭菌蒸馏水洗涤后加入10%Triton X,于室温反应1小时。从各孔收集液体而移至96孔板。将使用微读板器测定的反应液的吸光度(550nm)的结果示于图6下部的图。图的纵轴为吸光度,吸光度越高表示产生了越大量的钙化骨基质。
从这些结果可知,共导入Oct9基因和L-myc基因的细胞产生了较多的钙化骨基质,以高效率转换为功能性成骨细胞。
实施例7
将人正常皮肤成纤维细胞(NHDF)重悬于添加了10%胎牛血清(FBS)、0.1mM非必需氨基酸、100μg/mL链霉素、100U/mL青霉素的Dulbecco’s minimum essential medium(DMEM)培养基(完全培养基)中,接种于35mm平皿。在5%CO2/95%加湿的空气中,于37℃进行培养。次日,吸除培养上清液,加入包含具有Oct9和N-myc基因的逆转录病毒载体和4μg/mL的聚凝胺的培养基。在24小时之后吸除培养上清液,加入已添加了50μg/mL抗坏血酸、10mMβ-甘油磷酸酯和100nM地塞米松的完全培养基(成骨培养基)。每隔2~3天,用新鲜培养基更换培养基而进行培养。
基因导入28天之后,利用4%多聚甲醛固定细胞后,加入抗骨钙素抗体(AbDSerotec、Kidlington、UK)进行培养。之后加入结合了Alexa fluor 488的二次抗体,再用DAPI染色了核。作为对照,也同样地染色了成纤维细胞。以荧光显微镜于倍率100倍进行了观察。
将结果示于图7。可知成纤维细胞完全不表达骨钙素蛋白,而导入了Oct9+N-myc的细胞强烈地表达了骨钙素。
Claims (4)
1.通过向哺乳动物的体细胞导入Oct9基因或其表达产物,以及选自c-Myc基因、L-Myc基因及N-Myc基因中的至少一种或其表达产物,从而从所述体细胞制备成骨细胞的方法,其中所述体细胞为成纤维细胞,且所述方法为体外方法。
2.根据权利要求1所述的方法,其进一步包括将导入了基因或其表达产物的体细胞,利用成骨细胞的诱导培养基进行培养的工序。
3.Oct9基因或其表达产物和至少一种选自c-Myc基因、L-Myc基因和N-Myc基因或其表达产物的基因在制备用于将哺乳动物的体细胞转化为成骨细胞的药剂中的用途,其中所述体细胞为成纤维细胞。
4.Oct9基因或其表达产物和至少一种选自c-Myc基因、L-Myc基因和N-Myc基因或其表达产物的基因在制备治疗与成骨细胞的缺陷、不足或功能降低相关的疾病的药剂中的用途。
