基于中间溶液的测定来计算填充到最终包装小瓶中的解冻本体溶液的量。由于(1)生物测定的变化大,以及(2)随后的无菌填充和冷冻干燥过程中的产率损失,该计算通常包含大的安全界限。产率损失是由于在该第一次冷冻、储存和解冻步骤以及随后的第二次填充、冷冻和干燥工艺中的产品压力造成的。这个计算当然是非常困难的,并且基于完整工艺的产品依赖性经验知识。
在常规工艺中,冷冻干燥通常在不具有温度控制壁的标准冷冻干燥室中进行。不幸的是,这些干燥机向放置在干燥室中的小瓶提供不均匀的热传递。尤其是那些位于边缘的小瓶由于室壁与板/架堆之间的间隙中的辐射热交换和自然对流而比位于板中心的小瓶更能集中地交换能量。这种不均匀的能量分布导致在边缘处的小瓶和在中心处的小瓶之间的冷冻和干燥动力学的变化,并且可能导致相应小瓶的活性内容物的活性变化。为确保最终产品的一致性,有必要在实验室和生产规模上都进行广泛的开发和验证工作。
出版物(Wang, D. Q., MacLean, D.和Ma, X. (2010), 标题为"生物药品冷冻干燥中的工艺稳健性(Process Robustness in Freeze Drying of Biopharmaceuticals)",于"用于制备生物药品的制剂和工艺开发策略(Formulation and Process DevelopmentStrategies for Manufacturing Biopharmaceuticals)" (F. Jameel和S. Hershenson编辑), John Wiley & Sons, Inc., Hoboken, NJ, USA中)公开了用于重组FVIII的特定冷冻干燥循环,但仍认识到根据冷冻干燥室中小瓶位置的效价变化。
WO 2010/054238 A1报道了因子VIII (FVIII)的稳定冻干药物制剂,其包含:(a)FVIII;(b) 一种或多种缓冲剂;(c) 一种或多种抗氧化剂;(d) 一种或多种稳定剂;和(e)一种或多种表面活性剂;所述FVIII包含选自以下的多肽:a) 重组FVIII多肽;b) a)的生物活性类似物、片段或变体;所述缓冲剂由约0.1 mM至约500 mM范围内的pH缓冲剂组成且所述pH范围为约2.0至约12.0;所述抗氧化剂的浓度为约0.005至约1.0 mg/ml;所述稳定剂的浓度为约0.005至约20%;所述表面活性剂的浓度为约0.001%至约1.0%;并且所述制剂不包含氯化钠(NaCl)或仅包含痕量的NaCl。
WO 2006/008006 A1涉及用于无菌制造,包括冷冻干燥、储存、测定和填充最终容器如小瓶中的微丸状生物药物产品的工艺。公开了一种生产冷冻干燥产品容器的工艺,所述工艺包括以下步骤:冷冻产品的微滴以形成微丸,冷冻干燥微丸,测定冷冻干燥微丸,并将冷冻干燥微丸装载到容器中。更具体地说,该工艺包括以下步骤:a) 冷冻产品的微滴以形成微丸,其中通过使产品的溶液穿过频率辅助喷嘴(frequency assisted nozzle)形成微滴,并且通过使所述微滴穿过低温气体的逆流从所述微滴形成微丸;b) 冷冻干燥微丸;c) 储存并均质化冷冻干燥微丸;d) 在储存并均质化冷冻干燥微丸时测定冷冻干燥微丸;和e) 将冷冻干燥微丸装载到所述容器中。
WO 2013/050156 A1描述了一种用于在封闭条件下生产冷冻干燥颗粒的生产线,所述生产线包括至少一个用于微滴产生和冷冻凝结液滴以形成颗粒的喷射室和用于冷冻干燥颗粒的大容量冷冻干燥机,所述冷冻干燥机包括用于接收颗粒的滚筒。此外,提供传送部分用于从喷射室到冷冻干燥机的产品传送。为了在端对端封闭条件下生产颗粒,每个装置和传送部分单独适于保持待冷冻干燥的产品无菌性和/或密闭的操作。
WO 2013/050161 A1公开了一种用于在封闭条件下生产冷冻干燥颗粒的生产线,所述生产线包括用于在封闭条件下批量生产冷冻干燥颗粒的冷冻干燥机,所述冷冻干燥机包括用于接收冷冻颗粒的滚筒和容纳滚筒的固定真空室,其中为了在封闭条件下生产颗粒,真空室适于在加工颗粒期间的封闭操作。所述筒与真空室开放式连通,并且提供至少一个传送部分用于在生产线的单独装置与冷冻干燥机之间的产品传送,冷冻干燥机和传送部分单独适于封闭操作,其中所述传送部分包括温度可控的内壁表面。
希望在严格与外部(这包括任何低温气体如液氮)分离的条件下生产因子VIII的冷冻干燥微丸,其中个体微丸的活性变化较小。本发明的目的是提供这种方法。
根据本发明,通过用于生产包含因子VIII的冷冻干燥微丸的方法实现了该目的,所述方法包括以下步骤:
a) 冷冻包含因子VIII的溶液的微滴以形成微丸;
b) 冷冻干燥所述微丸;
其中在步骤a)中,微滴借助于使包含因子VIII的溶液进入冷却塔形成微滴来产生,所述冷却塔具有温度可控的内壁表面和溶液的冷冻温度以下的内部温度,并且在步骤b)中,在位于真空室内的旋转容器中冷冻干燥微丸。
根据本发明的方法的操作原理具有三个明显的优点。首先,应该注意的是,在根据本发明的方法中,包含因子VIII的溶液的喷射微滴不接触逆流形式的低温气体,例如WO2006/008006 A1中所述。不需要将低温气体引入冷却塔的内部空间,因此可以省略低温气体的所有处理和消毒步骤。
其次,通过在真空室内部的旋转容器中进行冷冻干燥步骤,每个个体微丸的空间位置随时间均匀分布。这确保了均一的干燥条件,并且因此消除了架子上的冷冻干燥小瓶会存在的因子VIII活性的空间变化。
第三,通过实验发现,由于FVIII多肽暴露的总体工艺条件平均比现有技术的那些更温和,根据本发明生产的微丸显示较少的微塌陷。在显示更均匀表面的这种微丸的REM图像中,所述微丸的微塌陷发生减少是可见的,这再次导致在那些微丸的后续工艺步骤中改进的处理性能。
与WO 2006/008006 A1公开的工艺直接比较,令人惊讶地表明,本发明所得微丸的表面形态比源自WO 2006/008006 A1工艺的那些微丸的表面形态显著更均匀,本发明的工艺生产具有进一步增加的比(BET-)表面积的微丸,尽管WO 2006/008006 A1工艺的那些微丸与现有技术中已知的任何冻干工艺相比已经具有改善的均匀性和比表面积。
已经通过实验发现,冷冻干燥后微丸的实际效价为因子VIII的目标效价的86.2%至89.9%。
根据本发明的方法的所有步骤可以在无菌条件下进行并且不会损害个体步骤之间的无菌性。
冷冻微丸的产生可以用任何已知技术进行,例如来自Inotech Encapsulation,Switzerland的“封装器研究(Encapsulator Research)”,或者来自Messer-Griesheim,Germany的"低温快速造粒机(Kryogen Rapid Pelletizer)",或者来自IQFCRYOGRAN,Canada的"低温造粒机(CRYOGENIC PELLETIZER)"。然而,这些现有技术主要依靠将微滴滴入液氮中以在干燥后在其中形成微丸。
由于随后的冷冻干燥步骤,预期冷冻微丸具有窄的粒度。之后,冷冻微丸可以在无菌和低温条件下运输至冷冻干燥机。然后通过容器的旋转将微丸跨干燥室内的承载面分布。升华干燥原则上可在任何种类的适用于微丸的冷冻干燥机中进行。提供用于升华蒸气流的空间,受控的壁温和干燥室与冷凝器之间合适的横截面积的冷冻干燥机是优选的。
下面描述可用于根据本发明的方法的因子VIII变体的细节。优选地,使用衍生自幼仓鼠肾细胞而不存在额外蛋白质的重组因子VIII。
本文中,术语“因子VIII”或“FVIII”或“rAHF”是指任何FVIII分子,其具有至少一部分的完整B结构域并且表现出与天然FVIII相关的生物学活性。在本公开的一个实施方案中,FVIII分子是全长FVIII。FVIII分子是由能够与编码FVIILC的DNA杂交的DNA序列编码的蛋白质。这种蛋白质可以包含在结构域A1-A2-B-A3-C1-C2之间或之内的各种位点的氨基酸缺失(美国专利号4,868,112)。FVIII分子还可以为天然FVIII类似物,其中已通过定点诱变替换一个或多个氨基酸残基。

如本文所用,“内源性FVIII”包括来源于意欲接受治疗的哺乳动物的FVIII。该术语还包括从存在于所述哺乳动物中的转基因或任何其他外源DNA转录的FVIII。如本文所用,“外源性FVIII”包括不来源于所述哺乳动物的FVIII。
FVIII分子天然地存在并作为由单一基因产物产生的不均匀分布的多肽存在于治疗性制剂中(参见例如Andersson等人,Proc. Natl. Acad. Sci. 本公开的起始材料是FVIII,其可以源自人血浆或通过重组工程技术产生,如专利美国专利号4,757,006;美国专利号5,733,873;美国专利号5,198,349;美国专利号5,250,421;美国专利号5,919,766;EP 306 968所述。
可用于本公开的FVIII分子包括全长蛋白质,蛋白质的前体,蛋白质的生物活性或功能性亚基或片段,及其功能性衍生物,以及其如下文所述的变体。提及FVIII是指包括这种蛋白质的所有潜在形式并且其中每个FVIII形式具有至少一部分或全部的完整的天然B结构域序列。
编码本公开的rFVIII的多核苷酸包括但不限于那些:(1)在严格杂交条件下与编码本文所述的参考氨基酸序列的核酸及其保守修饰变体特异性杂交;(2)具有在至少约25、约 50、约 100、约 150、约 200、约 250、约 500、约 1000或更多个核苷酸(高至成熟蛋白质的6996个核苷酸的全长序列)的区域内,与本文所述的参考核酸序列具有大于约95%、约96%、约97%、约98%、约99%或更高的核苷酸序列同源性的核酸序列。
多肽的变体(或类似物)包括插入变体,其中将一个或多个氨基酸残基添加到本公开的FVIII氨基酸序列中。插入可以位于蛋白质的任一端或两端,和/或可以放置在FVIII氨基酸序列的内部区域内。在任一端或两端具有额外残基的插入变体包括例如融合蛋白和蛋白质,其包括氨基酸标签或其他氨基酸标记。在一个方面,FVIII分子可以任选地含有N端Met,特别是当在细菌细胞如大肠杆菌中重组表达分子时。
在缺失变体中,除去如本文所述的FVIII多肽中的一个或多个氨基酸残基。缺失可以在FVIII多肽的一端或两端实现,和/或在FVIII氨基酸序列内除去一个或多个残基。因此缺失变体包括FVIII多肽序列的所有片段。
在取代变体中,除去FVIII多肽的一个或多个氨基酸残基并用替代残基置换。在一个方面,取代本质上是保守的,并且这种类型的保守取代在本领域中是公知的。或者,本公开包括也是非保守的取代。示例性的保守取代描述于Lehninger, [Biochemistry, 第二版; Worth Publishers, Inc., New York (1975), pp.71-77],并且在下面立即提出。
下面将描述本发明的实施方案和附加方面。除非上下文另有明确说明,否则他们可以自由组合。
对于本发明,可以加工FVIII多肽的任何缺失、取代和/或其他变体而不需要进一步改变工艺本身。然而,与工艺有关的是加工FVIII多肽,因为它对所得FVIII制剂具有特异性,与其中组分的总量相比所得FVIII制剂仅包含较少部分的实际FVIII多肽。
由此明显的是,该加工必须使得避免对作为制剂的一部分的冷冻干燥的FVIII多肽的任何潜在损害,因为最终冷冻干燥制剂中FVIII多肽活性的轻微降低导致从百分比上看对最终产品活性的巨大影响。
在根据本发明方法的一个实施方案中,所述方法进一步包括步骤b)之后的步骤c)、d)和e):
c) 储存并均质化冷冻干燥微丸;
d) 在储存并均质化冷冻干燥微丸时测定冷冻干燥微丸;
e) 将冷冻干燥微丸装载到容器中。
储存和均质化步骤c)也可以在用于冷冻干燥的真空室内的旋转容器中进行。提取统计学相关数量的样品用于进行测定。之后,将任选的单独储存容器包装为无菌密闭的(sterile containment)。在测定每个储存容器的内容物之后,所有必要的性质例如活性物含量是已知的。然后可以开始使用户定义数量的微丸进入最终容器的填充过程。将储存容器传送到隔离的填充线并停靠在无菌的转接站(docking station)。将容器的内容物在隔离器内传送到充填机的存储器。
在根据本发明方法的另一实施方案中,在步骤a)中,微滴借助于使溶液穿过频率辅助喷嘴形成微滴来产生。优选地,振荡频率≥1000 Hz至≤2000 Hz。
独立于频率辅助的喷嘴,喷嘴直径可以在100 μm至500 μm的范围内,优选在200 μm至400 μm的范围内,非常优选在350 μm至450 μm的范围内。所述喷嘴直径导致微滴尺寸在约200 μm至约1000 μm的范围内,优选在约400 μm至约900 μm的范围内,非常优选在约700μm至900 μm的范围内。
在这种情况下,“约”的尺寸意指所得微滴的尺寸与所提及的尺寸的差距不大于±30%,就分布的d90值而言。例如,约400 μm的所得微滴尺寸被理解为产生的微滴的尺寸在280 μm和520 μm之间变化,就分布的d90值而言。
所形成的微滴在中值附近显示某一微滴尺寸分布,其应该大约为上面提及的。
在喷嘴是频率辅助喷嘴的本发明的实施方案中,中值附近的变化较小。在那些实施方案中,“约”的含义可以被限制为微滴与所提及的尺寸的差距不大于±30%。
在上面刚刚提到的实例中,所得到的约400 μm的微滴尺寸则可以被理解为产生的微滴的尺寸在280 μm和520 μm之间变化,就分布的d90值而言。鉴于下面描述的效果,所以使微滴穿过频率辅助喷嘴进一步有利于进一步降低对最终冷冻干燥微丸的潜在负面影响。
通常上面给出的尺寸的微滴是有利的,因为发现后续步骤b)至e)可以以更高的FVIII活性产率进行。
不受此限制,假设由于过大的表面积体积比,较小的微滴在冷冻干燥步骤b)中过快地冷冻,并且脆弱的FVIII多肽由此被部分毁坏。此外,较小的微滴导致具有增加的变为带静电的倾向的较小微丸,而后者对随后处理这种微丸造成损害。较大的微滴不能均匀冷冻,导致部分破坏在冷冻干燥微丸的外壳上的FVIII多肽,和不完全冷冻干燥微滴的内部腔室,导致在储存期间部分破坏FVIII多肽。
在根据本发明方法的另一实施方案中,在步骤a)中,冷却塔的内表面的温度不高于-120 ℃,优选≥-150 ℃至≤-120 ℃。温度优选≥-140 ℃至≤-130 ℃。
对于在约≥ 700 μm至约≤ 900 μm的范围内的微滴尺寸,上述提及的≥ −140℃至≤ −130℃的温度是最佳的,所述微滴在下降3m至4m的距离时被冷冻。
只要冷却塔内表面上的温度保持在-120℃以下,本发明的有益效果就可以通过调整下落距离和微滴尺寸来获得。
在根据本发明方法的另一实施方案中,通过使冷却剂穿过与内表面热接触的一个或多个管冷却冷却塔的内表面。冷却剂可以是期望温度的液氮或氮蒸气。
在根据本发明方法的另一实施方案中,针对因子VIII建立目标剂量,步骤d)中的测定确定冷冻干燥微丸中因子VIII的有效含量,并将容器装载一定量的冷冻干燥微丸,其提供等于目标剂量的剂量,或超过目标剂量≤25%的剂量。优选超过目标剂量≤10%,更优选超过目标剂量≤5%。
本发明的直接结果是允许FVIII多肽的温和冷冻,其允许在填充后不显著超过目标剂量,因为该工艺不降低FVIII多肽的活性。
在根据本发明方法的另一实施方案中,步骤a)中获得的微丸具有约≥ 200 μm 至约≤ 1500 μm的最大粒度分布d50。优选约≥ 700 μm至约≤ 900 μm的最大粒度分布d50。
尺寸小于200 μm的微丸不太有利,因为在那些微丸中,冷冻会更快,这可能导致冷冻干燥多肽的损害,并因此导致效价损失,需要更高的目标剂量。此外,所得粉末的静电影响在低于200 μm的尺寸显著增加,导致本工艺产品的操作性能较差,并且可预期由于微丸在水蒸气中的截留而导致的产率损失。
将微丸尺寸增加至超过1500 μm可能会危及所述装备中微丸的完全冷冻,从而损害后续产品的总体功效。
在根据本发明方法的另一实施方案中,步骤a)中包含因子VIII的溶液具有≥ 8重量-%至≤ 12 重量-%的溶解固体含量。优选的溶解固体含量为≥ 9重量-%至≤ 11重量-%。
原则上,在本文涉及的种类的工艺中,约15至20重量-%的溶解固体的较高负载将被认为是有利的,因为通常预期得到的微丸将被更容易地冷冻并且更强烈地干燥。
然而,在目前的情况下,发现上述范围更好,因为处理的固体(包含FVIII)后来需要被重构并注射到人体内,其(在较高负载下)会导致注射溶液的不相容克分子渗透压浓度,导致潜在的组织损伤和/或注射疼痛,或另外需要显著更高的重构体积,以避免事实上使溶液不再可实际注射。

该空间130容纳管道形式的冷却装置140。冷却剂可以如图中箭头所示进入和离开冷却装置140。
流过冷却装置140的冷却剂导致内壁110的冷却并因此导致冷却塔100内部的冷却。在冷冻微丸(低温微丸(cryopellet))的生产中,经由喷嘴150将液体喷射到冷却塔中。根据参照数字160来代表液滴。

实施例:
通用–rFVIII药物产品的效价测定
使用CoatestTM FVIII试剂盒通过使用显色测定来测量效价。显色测定法由两个连续的步骤组成,其中颜色的强度与因子VIII活性成比例。在第一步中,通过因子IXa及其辅因子因子VIIIa在存在最佳量的钙离子和磷脂的情况下并在37℃孵育5分钟将因子X激活为因子Xa。存在过量的因子X,使得因子X的激活速率仅取决于因子VIII的量。在第二步中,因子Xa水解发色底物以产生发色团,并且在405 nm处通过光度计读取颜色强度。使用线性回归统计方法针对已建立的效价标准确认测定的有效性,并计算未知样品的效价。以国际单位/mL (IU/mL)报道效价。如果效价在下文中被称为[%]的值,则该值始终被理解为以UI/ml计的“目标效价”的百分比(标准化值)。显然,更大的%-效价值是优选的。
通用–尺寸排阻色谱法(SEC)来确定产物片段/聚集体的分布
将rFVIII制剂的主要组分基于其在TSK凝胶G4000SWXL柱上的流体动力学体积或分子大小分成多个区域,所述柱尺寸为7.8 mm ID x 30 cm,粒径为8 mm;孔径为450埃。
然后基于它们在276 nm激发后在340 nm处的荧光发射对它们进行定量。定量结果表示为这些区域的相对%峰面积。该程序报告色谱图具体区域的结果,并用于测量rFVIII聚集体和链的完整性。
确定了三个区域(区域1,区域2和区域3),而期望区域2(以总样品的百分比计)是最大的,因为总结了其中所有rFVIII分子未被聚集或碎片化。
通用–根据BET的比表面积
在多点测量下通过BET测定比表面积(在77开尔文下的氮气吸附),并且对于每个样品,将两个独立量的材料填充到BET容器中并分别进行分析。将容器用塞子密封,转移至样品制备站,抽空并在30℃在真空(< 0.2,巴)中预处理16 h以除去挥发性组分。随后将氮气通入样品,称重并根据DIN ISO 9277使用氮气测量。
通用-扫描电子显微镜(SEM)测量
样品的制备在氮气氛下在手套袋中进行,每个样品单独制备。将样品放在支架上并用金溅射。随后进行扫描电子显微镜测量。
实施例1
制造Kogenate® PF溶液的低温微丸。Kogenate® PF是无血浆蛋白的重组人因子VIII。下面给出1 g溶液的制剂:

根据本发明方法将本体溶液喷射到壁式冷却的(wall-cooled)冷却塔中。喷射嘴具有一个直径为400μm的孔。这对应于800 μm的目标微滴尺寸。振荡频率为1375 Hz,偏转压力(deflection pressure)为0.2巴,泵以22 rpm运转。总共持续35分钟后,收集879.3 g冷冻微丸(96%产率)。
监测冷却塔的内部温度,并且将其随时间推移的发展描绘在图2中。曲线1000表示来自冷却塔上部的传感器读数,曲线1010表示来自冷却塔中部的传感器读数,和曲线1020表示来自冷却塔下部的传感器读数。在14:45点,当温度达到-126.0℃(上传感器),-129.7℃(中传感器)和-133.1℃(下传感器)时,开始喷射操作。这由图2中的标记1030表示。在15:20点,停止喷射操作(标记1040),记录温度为-130.7℃(上传感器),-133.5℃(中传感器)和-135.2℃(下传感器)。将冷冻微丸收集在温度在-55℃至-53℃的冷却容器中。
实施例2
该实施例涉及冷冻干燥实施例1中获得的低温微丸样品。在该步骤中使用来自Meridion的LyoMotion冷冻干燥机。该机器包括转筒,在该转筒中,搅动低温微丸并使其经受干燥。
分离出具有0.95%残留水分的总计21.3g冷冻干燥微丸(72.6%产率)。
图3显示了冷冻干燥步骤的温度和压力曲线。曲线1050表示产品温度,曲线1060表示冷冻干燥机的冷凝器温度,和曲线1070表示冷冻干燥机的真空室内的内部压力。
实施例3
该实施例涉及冷冻干燥实施例1中获得的另一低温微丸样品。在该步骤中使用来自Meridion的LyoMotion冷冻干燥机。该机器包括转筒,在该转筒中,搅动低温微丸并使其经受干燥。
分离出具有0.70%残留水分的总计21.4g冷冻干燥微丸(73.7%产率)。
图4显示了冷冻干燥步骤的温度和压力曲线。曲线1080表示产品温度,曲线1090表示冷冻干燥机的冷凝器温度,和曲线1100表示冷冻干燥机的真空室内的内部压力。
结果
对于在制造后直接从工艺采集的样品,在下表中给出了所获产品的效价测定和尺寸排阻色谱法分析。
对每个实施例的两个样品进行关于其中因子VIII的效价的分析。目标效价是250.0 mg/小瓶。预期在加工本体溶液和冷冻干燥期间效价损失。在86.2%至89.9%之间的测定实际效价在变化方面比在小瓶中常规冷冻干燥中观察到的那些低得多。此处根据干燥室中个体小瓶的位置,可以观察到80.9%至91.2%的效价范围。
作为参考,下表给出了实施例2和3的前体的分析数据(IPC值)。
此外,评估在室温(RT)和2-8℃下储存高达6个月的样品的上述性质。
从图5和6可以看出(样品由活性较低的不同起始材料制备),样品的效价或片段/聚集体组成没有发生显著变化,这加强了根据本发明工艺的稳健性,就由此制得的微丸的性质而言。
实施例4 –与现有技术的直接比较
制备具有与实施例1中提供的相同制剂组成的同一批次的因子VIII药物溶液并通过不同的干燥方法进行加工。将3000 ml溶液在根据实施例1的冷却塔中冷冻,并将冷冻微丸在批量大小为319 g和2614 g的两个不同旋转式冷冻干燥机(LyoMotion)中冻干。
根据WO 2006/008006 A1中描述的方法加工溶液的单独部分。将总共1200 ml溶液以200 ml的份量通过400 μm喷嘴喷射,并以900 Hz的频率以约16.5 g/min的速率雾化。将微滴冷冻在填充有液氮的隔离容器中,所述隔离容器位于喷嘴下方约25 cm处并在整个过程中搅拌。每个部分的喷射完成后,通过将液氮灌入预冷筛并在低温下储存他们来除去冷冻微丸。一旦收集了所有部分后,就将它们放在Virtis Advantage Pro冷冻干燥机的预冷搁板上的2个衬有塑料薄膜的架子中并冷干。初次干燥在-10℃的搁板温度下持续60小时,然后在25℃下进行二次干燥8小时。干燥完成后,将干燥的微丸立即转移到玻璃瓶中并且牢固地封闭。随后,在氮气氛下将250 mg微丸称量到10R I型玻璃小瓶中。
将溶液的第三个单独部分填充到10R I型玻璃小瓶中并在常规小瓶冷冻干燥机中冷冻干燥。以每瓶2.5 ml溶液填充总共488个小瓶(总计1241g溶液),半加塞且装载入HOF冷冻干燥机中。将该溶液冷冻至-45℃,在-20℃下进行初次干燥,然后在25℃下进行二次干燥步骤。完整的冷冻干燥过程需要约65小时。在冷冻干燥机内将小瓶用塞子塞住并在卸载后直接密封。
使其后制备的所有三个样品-来自根据实施例1首次加工的那些,如WO 2006/008006 A1中所述加工的那些和来自常规小瓶冷冻干燥过程的那些-进行根据BET的比表面积测量和扫描电子显微镜(SEM)测量。
可以看出,根据本发明生产的微丸显示出更高的比表面积-这改善了冷冻干燥固体在用于施用的液体中的重构-以及更均匀的形态,这改善了那些微丸在后面的工艺步骤中的处理性能。

Claims (7)
1.用于制备包含因子VIII的冷冻干燥微丸的方法,所述方法包括下列步骤:
a)冷冻包含因子VIII的溶液的微滴以形成微丸;
b)冷冻干燥所述微丸;
其特征在于
在步骤a)中,所述微滴借助于使包含因子VIII的溶液进入冷却塔(100)形成微滴来产生,所述冷却塔具有温度可控的内壁(110)表面和溶液的冷冻温度以下的内部温度,
在步骤a)中,所述微滴借助于使溶液穿过频率辅助喷嘴形成微滴来制备,并且振荡频率为≥1000Hz至≤2000Hz,
在步骤a)中,所述冷却塔(100)的内壁(110)表面具有≤-120℃的温度,
和
在步骤b)中,在位于真空室(200)内的旋转容器(210)中冷冻干燥所述微丸。
2.根据权利要求1的方法,其进一步包括步骤b)之后的步骤c)、d)和e):
c)储存并均质化所述冷冻干燥微丸;
d)在储存并均质化所述冷冻干燥微丸时测定所述冷冻干燥微丸;
e)将所述冷冻干燥微丸装载到容器中。
3.根据权利要求1的方法,其中通过使冷却剂穿过与所述内壁(110)表面热接触的一个或多个管(140)来冷却所述冷却塔(100)的内壁(110)表面。
4.根据权利要求2的方法,其中针对因子VIII建立目标剂量,步骤d)中的测定确定所述冷冻干燥微丸中因子VIII的有效含量,并将容器装载一定量的所述冷冻干燥微丸,其提供等于目标剂量的剂量,或超过目标剂量≤25%的剂量。
5.根据权利要求1的方法,其中步骤a)中获得的微丸具有≥200μm至≤1500μm的最大粒度分布d50。
6.根据权利要求1的方法,其中步骤a)中所述包含因子VIII的溶液具有≥8重量-%至≤12重量-%的溶解固体含量。
