Description

Anti-aging skin external composition containing cyanidin-3-O-glucoside and natto gum as effective components
Technical Field
The present invention relates to an anti-aging skin external composition containing cyanidin-3-O-glucoside and natto gum as active ingredients, and more particularly to a skin external composition which exerts an inhibitory effect on collagen degrading enzymes by containing a mixture of cyanidin-3-O-glucoside and natto gum.
Background
The skin serves as a first protective film for the human body, protects various organs in the body from changes in temperature and humidity and external environmental stimuli such as ultraviolet rays and harmful substances, and plays an important role in maintaining homeostasis such as thermoregulation. However, excessive physical and chemical stimulation, stress, malnutrition, etc. from the outside may lower the normal function of the skin and promote skin aging such as loss of elasticity, keratinization, wrinkle formation, etc.
In order to prevent these phenomena and maintain healthier and more elastic skin, efforts have been made to prepare cosmetics containing bioactive substances obtained from various plants, microorganisms, etc. for effectively inhibiting skin aging by activating skin cells while maintaining the intrinsic functions of the skin. However, conventional cosmetic materials have various problems such as insufficient efficacy or induction of skin side effects.
cyanidin-3-O-glucoside (cyanidin-3-O-glucoside) is a kind of anthocyanidin, also called chrysanthemin (chrysanthemin), and is a structure formed by binding cyanidin with a glucoside bond at the third carbon atom of glucose. Recent research reports indicate that cyanidin-3-O-glucoside can effectively prevent cancer or heart disease (Cooke et al, International journel of cancer 119.9(2006): 2213-2220; Serratio et al, Life sciences 73.9(2003): 1097-1114). However, there has been no report to date on whether cyanidin-3-O-glucoside can inhibit collagen degrading enzymes, thereby having an anti-aging effect of maintaining skin elasticity and improving wrinkles.
On the other hand, natto gum (natto gum) is a highly viscous substance obtained by fermentation of soybean (soybean) with a gram-positive bacterium called Bacillus subtilis. Recently, cosmetics containing natto gum, which acts as a thickener and has a strong moisturizing effect, have been developed. However, like cyanidin-3-O-glucoside, there has been no report to date as to whether natto gum can inhibit collagen degrading enzymes and thereby have an anti-aging effect of maintaining skin elasticity and improving wrinkles, nor whether natto gum can enhance the antioxidant effect or anti-aging effect of cyanidin-3-O-glucoside.
The present inventors have conducted intensive studies in order to find a natural substance which maximizes the anti-aging effect of the skin, and as a result, have found that a composition for external application to the skin, which contains cyanidin-3-O-glucoside and natto gum, can inhibit the generation of free radicals and effectively inhibit the expression of collagen degrading enzymes, thereby achieving very excellent antioxidant and anti-aging effects, thereby completing the present invention.

Detailed Description
Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this invention belongs. Generally, the nomenclature used in this specification is that well known and commonly used in the art.
In the examples of the present invention, the inhibitory effect on DPPH oxidation was measured by mixing cyanidin-3-O-glucoside and natto gum in the same amounts, and it was found from the results that the antioxidant effect was more excellent than that when cyanidin-3-O-glucoside or natto gum alone or when delphinidin-3-O-glucoside (delphinidin-3-O-glucoside), which is another pigment in anthocyanin, and natto gum were mixed. Further, it was found that the anti-aging effect was more excellent than that obtained by using only cyanidin-3-O-glucoside or natto gum or using delphinidin-3-O-glucoside and natto gum in combination.
Accordingly, one aspect of the present invention relates to a composition for external use for skin comprising cyanidin-3-O-glucoside and natto gum as effective ingredients.
In addition, another aspect of the present invention relates to a skin elasticity-improving or wrinkle-improving skin external preparation composition containing cyanidin-3-O-glucoside and natto gum as active ingredients.
The cyanidin-3-O-glucoside has a structure shown in the following chemical formula 1, can be directly extracted from plants containing a large amount of cyanidin-3-O-glucoside by using a conventional method in the technical field, and can also be purchased and used after being extracted and refined.
[ chemical formula 1 ]
Figure GDA0001642375800000041
The present invention is characterized in that the natto gum is produced by inoculating activated Bacillus subtilis or Bacillus licheniformis (Bacillus licheniformis) into soybean and fermenting the same. The natto gum is prepared by inoculating Bacillus subtilis or Bacillus licheniformis to soybean, fermenting at a given temperature to obtain viscous substance on the surface of soybean, refining, crystallizing, and using in the composition of the invention in powder form. The type of fermented beans used for producing natto gum is most preferably soybean, but is not limited thereto.
When a composition in which natto gum is mixed with cyanidin-3-O-glucoside together is used, it is presumed that the negatively charged polyglutamic acid contained in the natto gum can interact with the positively charged cyanidin-3-O-glucoside, so that the structure of the cyanidin-3-O-glucoside is stabilized, thereby further improving the antioxidant and anti-aging effects of the cyanidin-3-O-glucoside.
The present invention is characterized in that a composition for external application to the skin, which contains cyanidin-3-O-glucoside and natto gum as active ingredients, exhibits an anti-aging effect, particularly an effect of improving skin wrinkles and an effect of improving skin elasticity, because the expression of a collagen degrading enzyme for degrading collagen in skin tissues and reducing skin elasticity is suppressed. In the present invention, "collagenase (collagenase)" and "collagenase" are used as the same meaning.
In the present invention, the ratio of the cyanidin-3-O-glucoside to the natto gum is about 1:1 to about 1:10, preferably about 1:1 to about 1:9, more preferably about 1:1 to about 1:7, still more preferably about 1:1 to about 1:5, yet more preferably about 1:1 to about 1:3, yet more preferably 3:2 to about 2:3, yet more preferably 5:4 to about 4:5, and most preferably 1: 1. When the amount of the compound is in the above range, the respective effects of cyanidin-3-O-glucoside and natto gum on the inhibition of the expression of the collagen degrading enzyme can be maximized, and the stability of the composition can be satisfied.
In addition, the present invention is characterized in that the cyanidin-3-O-glucoside is contained in an amount of 0.01 to 5 wt% based on the total weight of the composition; the natto gum is contained in an amount of 0.01 to 5 wt% based on the total weight of the composition. When the total of the above two effective components is less than 0.02% by weight, the inhibitory effect on the expression of collagen-degrading enzyme is weak; when the amount exceeds 10% by weight, the composition may have a problem of excessively high tackiness.
When the skin external composition according to the present invention is formulated into a cosmetic form, it may be formulated into a form of smoothing toner, astringent lotion, nourishing lotion, eye cream, nourishing cream, massage cream, cleansing foam, cleansing water, loose powder, essence, mask, or the like, but the formulation is not particularly limited thereto. In addition, the composition of the invention may contain adjuvants commonly used in the field of cosmetics or dermatology, such as fatty substances, organic solvents, solubilizers, thickeners, gelling agents, emollients, antioxidants, suspending agents, stabilizers, foaming agents, fragrances, surfactants, water, ionic or non-ionic emulsifiers, fillers, metal ion-blocking agents, chelating agents, preservatives, vitamins, blocking agents, moisturizers, essential oils, dyes, pigments, hydrophilic or lipophilic activators, lipid vesicles or any other ingredient commonly used in cosmetics. The above-mentioned adjuvants are added in the amounts usually used in the fields of cosmetics or skin science. The composition of the present invention may further contain a substance for promoting skin absorption so as to increase the skin-improving effect.
The composition for external application to the skin of the present invention may contain, in addition to the above-mentioned substances, other components capable of enhancing the main effects, within a range not affecting the main effects. In addition, the skin external agent composition according to the present invention further comprises a moisturizer, an emollient, an ultraviolet absorber, a preservative, a bactericide, an antioxidant, a pH adjuster, an organic pigment and an inorganic pigment, a perfume, a cooling agent, an antiperspirant, or the like. The mixing amount of the above-mentioned components can be easily selected by those skilled in the art within a range not affecting the object and effect of the present invention, and is 0.01 to 5% by weight, preferably 0.01 to 3% by weight, relative to the total weight of the composition.

[ test example 1 ] Oxidation resistance test
In order to observe the antioxidant effect of the mixture of cyanidin-3-O-glucoside and natto gum, examples 1-3, the effect of inhibiting DPPH oxidation of comparative examples 1-3 using cyanidin-3-O-glucoside alone, natto gum alone, and delphinidin-3-O-glucoside and natto gum in combination was measured and compared based on the change in absorbance caused by the reduction of the organic radical-DPPH (1, 1-diphenyl-2-picrylhydrazide).
Mu.l of 100. mu.M (in ethanol) DPPH solution and cyanidin-3-O-glucoside (reference example 1), natto gum (reference example 3), examples 1 to 3, comparative examples 1 to 3, and synthetic antioxidant-water-soluble vitamin E (Trolox) (positive control group) were prepared at the same concentration of 10000ppm, respectively, and then diluted to final reaction concentrations of 500ppm, 250ppm, 125ppm, 62.5ppm, 31.25ppm, and 15.63ppm, respectively, 10. mu.l was added to obtain reaction solutions, and after reaction at 37 ℃ for 30 minutes, absorbance was measured at 540 nm. The results of the analysis are shown in Table 2 below, in which IC is50The concentration of the sample at which the absorbance was reduced by 50% by adding the sample is shown.

As shown in table 2, it was found that there was almost no antioxidant effect when natto gum was used alone. However, in example 1 where cyanidin-3-O-glucoside and natto gum were mixed in a 1:1 weight ratio, the IC thereof was50The value is similar to that of water-soluble vitamin E (Trolox), but shows a particularly high antioxidant effect as compared with the case of cyanidin-3-O-glucoside alone. In addition, in example 2 in which cyanidin-3-O-glucoside and natto gum were mixed at a weight ratio of 1:3 and example 3 in which natto gum was mixed at a weight ratio of 1:9, the superior antioxidant effect as in example 1 in which the mixture was 1:1 was not exhibited, but the effect was better than that in the case where cyanidin-3-O-glucoside was used alone. In addition, examples 1 to 3 were more excellent in antioxidant effect than comparative examples 1 to 3 in which cyanidin-3-O-glucoside was replaced with delphinidin-3-O-glucoside by the same amount of delphinidin-3-O-glucoside, respectively, to be mixed with natto gum. Therefore, it can be confirmed that the composition of the present invention has an excellent antioxidant effect.
[ test example 2 ] anti-aging Performance test
In order to observe the anti-aging effects of the mixture of cyanidin-3-O-glucoside and natto gum, examples 1-3, the respective abilities to inhibit the production of collagenase (collagenase) were compared and measured with comparative examples 1-3, in which cyanidin-3-O-glucoside alone, natto gum alone, delphinidin-3-O-glucoside and natto gum were mixed, and tocopherol or EGCG. It is known that tocopherol and EGCG are substances having the property of regenerating epidermal cells of the skin, preventing aging.
Human fibroblasts (Cascade Biologics, Inc.) (Portland, OR, U.S. A.) were added to a 96-well microtiter plate (96-well microtitre plate) containing DMEM (Dulbecco's Modified Eagle's Media) medium containing 2.5 wt% fetal bovine serum, 5000 cells (wells) per well, and cultured until the coverage reached about 90%. Then, after culturing in serum-free DMEM medium for 24 hours, 10. mu.g of cyanidin-3-O-glucoside (reference example 1), natto gum (reference example 3), examples 1-3, and comparative examples 1-3 dissolved in serum-free DMEM medium at the same concentration of 50ppm, respectively, were treated for 24 hours, and after treating with tocopherol and EGCG (Sigma Aldrich Co.) at a concentration of 50. mu.M, respectively, for 24 hours, cell culture solutions were harvested.
The degree of production of collagenase in the harvested cell culture solution was measured using a collagenase measuring instrument (Amersham Pharmacia, USA). First, the harvested cell culture solution was added to a 96-well plate (96-well plate) having collagenase primary antibody uniformly applied thereto, and an antigen-antibody reaction was performed in a constant temperature bath (36 ℃) for 3 hours.
After 3 hours, a collagenase secondary antibody having a luminophore bound thereto was added to a 96-well plate (96-well plate), and the reaction was carried out for 15 minutes. After 15 minutes, a substance for inducing color development was added, color development was induced at room temperature for 15 minutes, and when the reaction was terminated by adding 1M sulfuric acid (color development), the reaction solution showed yellow color, and the intensity of the yellow color changed as the reaction proceeded.
The absorbance of a 96-well plate (96-well plate) exhibiting yellow color was measured at 405nm using a spectrophotometer, and the degree of synthesis of collagenase was calculated using the following mathematical formula 1. In this case, the reaction absorbance of the cell culture solution obtained without the treatment with the composition was used as a control group. That is, the expression level of collagenase in the group treated with the test substance was calculated with respect to the expression level of collagenase in the untreated group of 100, and the results are shown in table 3 below.

As can be seen from table 3 above, in the case of using natto gum alone, the expression of collagenase is hardly inhibited. However, example 1 in which cyanidin-3-O-glucoside and natto gum were mixed in a 1:1 weight ratio was able to inhibit the expression of collagenase more effectively in vitro (in vitro) than the case of using cyanidin-3-O-glucoside alone, and its ability to inhibit the expression of collagenase was even better than the ability of tocopherol and EGCG, which are known as antioxidants. In addition, in example 2 in which cyanidin-3-O-glucoside and natto gum were mixed at a weight ratio of 1:3 and example 3 in which natto gum was mixed at a weight ratio of 1:9, the effect of inhibiting collagenase expression as in example 1 in which 1:1 was mixed was not exhibited, but the effect was more excellent than the case where cyanidin-3-O-glucoside was used alone. Moreover, examples 1 to 3 were more excellent in the effect of inhibiting collagenase expression than comparative examples 1 to 3 in which cyanidin-3-O-glucoside was replaced with the same amount of delphinidin-3-O-glucoside, respectively, to be mixed with natto gum. Therefore, it can be confirmed that the composition of the present invention has excellent anti-aging effects, particularly skin elasticity improving effects and skin wrinkle improving effects by inhibiting collagenase expression.
As the foregoing describes the features of the present disclosure in greater detail, those skilled in the art will appreciate that the specific techniques are merely preferred embodiments and that the scope of the present disclosure is not limited in this respect. Therefore, the substantial scope of the present invention is defined by the appended claims and equivalents thereof.
