Abstract
本发明公开了一种抑菌、消痘止痒的组合物的制备方法和应用,该组合物的活性成份包括有天山雪莲提取物,紫草提取物,维生素B3和维生素B5,该活性成份的质量配比:天山雪莲提取物:1‑9,紫草提取物:1‑9,维生素B3:0.1‑9.9,维生素B5:0.1‑9.9。本发明将二种药材分别进行提取,再复配两种维生素B3和B5,按一定的比例制成各种化妆品剂型,发现有明显的抗菌、消炎功效。该组合物可以用作皮肤轻微感染、湿疹、小儿尿布疹和蚊虫叮咬之后的特殊用途。

技术领域
本发明涉及中药化妆品领域,特别是涉及一种抑菌、消痘止痒的中药提取物组合制备方法和应用。
背景技术
医学上,皮肤病是有关皮肤的疾病,是严重影响人民健康的常见病、多发病之一,如麻风、疥疮真菌病、皮肤细菌感染等。皮肤病是皮肤受到内外因素的影响后,其形态、结构和功能均发生变化,产生病理过程,并相应的产生表现:疱疹、酒渣鼻、脓疱疮、化脓菌感染、疥疮、疤痕、癣、鱼鳞病、腋臭、青春痘、皮肤瘙痒等。当病情严重时,多数人会到医院就诊;但病情轻微时,多数人是希望有一种毒副作用小,能有效缓解不适症状同时可以护肤的特殊化妆品——药妆。
中国古代早就有“药妆”这个概念,在《神农本草经》中记载的“红玉膜、浮萍膏、面黑令白方、三花除皱液”等就具有相当好的美容效果。为了保证其无刺激性及无毒性,“药妆品”其组成部分都是精选的各种天然植物的提取物制成。其中紫草膏已收载于《中国药典》,但要用油浸泡紫草和其他药物,加蜂蜡配制而成,制备方法比较简单。其质地油腻和草药味浓是其缺点之一。
现代医学认为湿疹是一种过敏性疾病,是由多种内外因素引起的一种具有渗出倾向的皮肤炎症反应,同时研究也表明微生物的感染因素尤其是金黄色葡萄球菌可诱发和加重本病。湿疹的特点是难治、易复发、奇痒等,临床上常用的治疗方法为激素类药物,如丁酸氢化可的松软膏、糠酸莫米松乳膏等,长期使用具有副作用,并且容易复发等缺点。
天山雪莲(Saussurea involucrata)为菊科凤毛菊属多年生一次性结实的高山植物。天山雪莲生存条件极为严酷,一般生长于3000米左右的雪线以上,此环境特征为低温(全年平均越为3~5℃)、风大、氧气稀薄、紫外辐照强等,一般植物难以生存,如此苛刻生存条件造就了雪莲的稀少和珍贵。天山雪莲作为我国传统的中药和民族药,在医药上的应用已有数百年的历史。中医认为天山雪莲温肾助阳、祛风胜湿、通经活血,临床一般用来治疗风湿性关节炎、小腹冷痛、月经不调等(中国药典,2015版第一部)。现代研究发现天山雪莲含有的化学成分较多,但主要是黄酮类、糖类、生物碱、萜类及其衍生物等,现代药理学表明其在抗炎、抗肿瘤、心血管、免疫调节、妊娠终止等方面具有明显作用(天山雪莲的研究进展),其中最明显的为抗炎镇痛,同时还兼具抗氧化、抗辐射等功效,历史上对其应用最早可以追溯到清代赵学敏所编著的《本草纲目拾遗》,由于天山雪莲的抗炎镇痛、抗氧化等对缓解某些皮肤问题具有明显作用,固已有人将其制备成天山雪莲纳米乳来应用,目的在于提高天山雪莲的透皮吸收率,增强其功效。
新疆紫草(Arnebia euchroma)又称软紫草,为新疆紫草的根。性寒、味甘咸,有凉血、活血、清热、解毒透疹等功效。其有效化学成份主要分为两大类:一类是脂溶性很强的萘醌类色素主要包括紫草素及其衍生物,另一类是水溶性成分如多糖和酚酸类。药理学研究表明新疆紫草具有杀菌抗炎、免疫调节等作用,能抑制金黄色葡萄球菌及绿脓杆菌等,同时还兼具促进创伤愈合的作用(复方紫草油治疗小儿红屁45例临床研究)。
维生素B3(烟酰胺):现代研究表明,当人体皮肤缺乏某些维生素时,也会产生皮肤病,如B族维生素缺乏就会增加患脚气的风险,维生素B3性质温和,溶解性好,并且具有良好的配伍性,是一种使用历史悠久的活性成分,大量的临床试验证明其具有诸多的肌肤美容功效,除了传统的美白作用外,人们还发现有保湿、抗皱、抑菌、强化皮肤屏障等功效,特别对痤疮的治疗有明显效果,相比使用抗菌剂,维生素B3对皮肤的刺激性更小,也不会导致细菌的抗药性等。
维生素B5(D-泛醇):一般认为维生素B5具有良好保湿、促进伤口修复及抗炎效果,能够舒缓由湿疹、蚊虫叮咬及小儿尿疹带来的瘙痒,预防抓挠带来的感染(化妆品中的营养剂)。
发明内容
本发明的目的之一是提供一种具有良好抑菌、消痘止痒效果的组合物及其制备方法和应用。
实现上述目的的技术方案如下。
一种抑菌、消痘止痒的组合物,其活性成份包括有天山雪莲提取物,紫草提取物,维生素B3和维生素B5。
在其中一个实施例中,所述组合物,其活性成份的质量配比如下:
天山雪莲提取物:1-9
紫草提取物:1-9
维生素B3:0.1-9.9
维生素B5:0.1-9.9。
在其中一个实施例中,其活性成份的质量配比如下:
天山雪莲提取物:1-5
紫草提取物:5-9
维生素B3:0.1-6
维生素B5:4-9.9。其中,天山雪莲提取物与紫草提取物在一起的量为10份,维生素B3与维生素B5在一起的量为10份。

上述紫草提取物,是以丙酮为溶剂回流提取,回收丙酮之后的样品,以的丁二醇复溶得到的。
一种抑菌、消痘止痒的组合物的制备方法,包括以下步骤:
取天山雪莲,粉碎成粗粉,加其8-12倍量的水浸泡0.5±0.05h,加热提取,浓缩(100mg/ml),得天山雪莲提取物;
取紫草,切碎,加10-30倍量的丙酮回流提取,回收丙酮,残渣用1,3-丁二醇溶解(50mg/ml),得紫草提取物;
按照上述比例将所述天山雪莲提取物、新疆紫草提取物以及维生素B3(烟酰胺)和维生素B5(D-泛醇)按照化妆品配方配置在一起,即得。
上述抑菌、消痘止痒的组合物在制备治疗皮肤病的药物或护肤品中的应用。
在其中一个实施例中,所述皮肤病为皮肤轻微感染、湿疹、小儿尿布疹和蚊虫叮咬。
本发明的另一目的是提供一种抑菌、消痘止痒的药制剂物或护肤品。
一种抑菌、消痘止痒的药物制剂或护肤品,其活性成份包括有上述抑菌、消痘止痒的组合物以及合适的辅料。
在其中一个实施例中,所述药物制剂或护肤品是啫喱剂,或喷雾剂,慕斯丝滑日霜剂,或油膏剂。
本发明将天山雪莲、紫草提取物、以及维生素B3和维生素B5按一定的比例组合,加到护肤品体系中,该组合物具有良好的抗菌消炎、保湿及重建皮肤屏障功效。本发明可以应用于化妆品,可以缓解一些皮肤不适症状。
本发明所述紫草的提取方法,回收丙酮提取之后的样品,以作为化妆品保湿剂的丁二醇复溶,不需要二次添加溶剂直接用到配方中。由于提取物浓缩之后,紫草色素很难被常用的酒精、石油醚等充分溶解,而且许多有机溶剂不可以用于化妆品配方。我们选取丙酮提取,然后以化妆品常用的保湿剂丁二醇直接溶解,可以不仅可减少样品溶解过程中精华的损失,同时还增加紫草的抗菌功效。
天山雪莲与新疆紫草均属于新疆地区的特产药材,本发明将二种药材分别进行提取,再复配两种维生素B3和B5,按一定的比例制成各种化妆品剂型,发现这几种物质,相互配合,具有明显的协同作用,在抗菌、消炎、祛痘止痒方面具有明显的功效。该组合物可以用作皮肤轻微感染、湿疹、小儿尿布疹和蚊虫叮咬之后的药剂或者护肤品。

实施例1:
天山雪莲提取物的制备
取天山雪莲药材100克,粉碎成粗粉,加10倍量的水浸泡半个小时,加热提取2小时,过滤,浓缩成100mg/ml的天山雪莲提取物,备用。
新疆紫草提取物的制备
新疆紫草药材10克,切碎,加20倍量的丙酮回流提取2小时,过滤,回收丙酮,残渣用1,3-丁二醇溶解,50mg/ml的提取物,备用。
将天山雪莲提取物、新疆紫草提取物、维他命B3和B5按不同比例配比,具体见表一。

实施例2:抗菌活性试验
1.试验菌株
ATCC 6538金黄色葡萄球菌(Staphylococcus aureus,SA)、ATCC35657肺炎克雷伯氏菌(Klebsiella pneumoniae,KP)、ATCC 8739大肠杆菌(Escherichia coli,EC)、ATCC9027铜绿假单胞菌(Pseudomonas aeruginosa,PA)、ATCC6633枯草芽孢杆菌(Bacillussubtilis,BS)、ATCC 10231白色念珠菌(Candida albicans,CA)、ATCC 16404黑曲霉菌(Aspergillus niger,AN),以上菌株购自广东省微生物菌种保藏中心
2.培养基配制
各培养基按产品说明书所写比例加入去离子水溶解(LB营养琼脂培养基40g/L,孟加拉红培养基36.6g/L,LB肉汤培养基25g/L,YPD液体培养基50g/L,霉菌液体培养基21.6g/L)。包好防水膜,121℃灭菌15min(注:孟加拉红培养基灭菌20min)。液体培养基灭菌后自然冷却至室温备用,固体培养基在灭菌后可趁热倒入培养皿,每个培养皿倒入培养基5~10ml,摇晃培养皿铺平培养基。自然冷却至室温,放入4℃冰箱保存。
3.抗菌试验
微生物接种
调节含细菌的液体培养基的OD595至0.3.0~0.35(细菌密度为108cfu)。用无菌生理盐水稀释100倍(106cfu)。每种细菌吸取100μl加入LB营养琼脂培养基涂匀。真菌通过血球计数板计数,稀释到105cfu,每种真菌吸取100μl加入孟加拉红培养基涂匀。
样品制备
根据实验样品不同通过生理盐水、无水乙醇(醇提物)按比例稀释至所需要的浓度,加入灭菌干燥后的滤纸片,浸泡3~4h。1.4.3.3样品添加
将浸泡好的滤纸片放置到接种过菌的培养基上,牛津杯中加入样品20μl,并静止30min,后放入培养箱恒温湿培养(细菌为36℃,24h;真菌为30℃,24h)。
抗菌活性检测
将达到培养时间的平皿取出,用游标卡尺测量滤纸片周围透明抑菌圈的直径,并记录。
结果如表二:不同样品的组成的抗菌试验结果
综合以上实验数据,样品4和5,对实验所用的四种细菌和两种真菌的效果相对较好。其中以天山雪莲提取物:新疆紫草提取物:维生素B3:和维生素B5(2:8:2:8)为最优。

实施例3:紫草不同溶剂提取样品的抗菌试验比较不同溶剂提取紫草药材的抗菌活性:以溶剂为空白对照,青链霉素为阳性对照。按照实施例2中的抗菌活性实验进行。

综上实验结果,实施例一中的紫草提取方法,样品ZC III和IV为抑菌功效最佳的提取方法。但由于丁二醇粘度大,直接以丁二醇提取,紫草样品残渣很难通过离心等步骤去除,故不可取。
由于紫草提取物浓缩之后,紫草色素也即其活性成分很难被常用的溶剂如酒精、石油醚等充分溶解,而且许多有机溶剂不可以用于化妆品配方,所以我们选取丙酮提取后,再以化妆品常用的保湿剂丁二醇直接溶解,可以充分提取紫草的活性成分,减少复溶过程中活性成分的损失,从而增加护肤品的抗菌功效。
实施例4:
啫喱剂配制:本发明实施例4含本发明天然植物提取物组合物的啫喱,对比了不同组合物对应的产品外观,按质量百分比计,包括以下制备原料:
制备方法:
S1:将A相中的SP物料加水分散溶胀,溶胀完全后与A相中剩余物料一起加入乳化锅,85℃加热搅拌完全溶解,保温15min后消泡降温;
S2:将B相加水溶解,温度降至60℃时将B相的混合液加入A相中,搅拌均匀;
S3:温度降至45℃左右时将C相加入S2中的混合液,搅拌均匀,加入C、D相,混合均匀后检测合格出料,得到含本发明组合物的啫喱。
产品经过1个月50℃,3个月45℃和-22℃,1个星期光照后,颜色和性状均稳定。
实施例5:
喷雾剂配制:含本发明组合物的舒缓喷雾
制备方法:
S1:将A相中的所有物料依次加入乳化锅,85℃加热搅拌完全溶解,保温15min后消泡降温;
S2:温度降至45℃时将B相的物料加入A相中,搅拌均匀;
S3:温度降至38℃左右时将C相加入S2中的混合相,搅拌均匀,加入C相,混合均匀后检测合格出料,得到含本发明组合物的喷雾。
产品经过1个月50℃,3个月45℃和-22℃,1个星期光照后,颜色和性状均稳定。
实施例6:
慕斯丝滑日霜剂配制:含本发明组合物的慕斯丝滑日霜
制备方法:
S1:将A相和B相分别加热到85℃,使其完全溶解;
S2:将A相放入均质机中后,加入B相,85℃均质3min后,保温消泡5min,然后开始降温;
S3:温度降至65℃左右时加入预先溶解好的C相,搅拌均匀,
S4:温度降至45℃左右时加入D相搅拌均匀,最后加入E相检验合格出料,得含本发明组合物的慕斯丝滑日霜。
产品经过1个月50℃,3个月45℃和-22℃,1个星期光照后,颜色和性状均稳定。参见图3。
实施例7:
消包止痒油膏配制:含本发明组合物的消包止痒油膏
制备方法:
S1:将A相加热到85℃,使其完全溶解;
S2:将B相加热到90℃左右溶解好加入A相,搅拌均匀,保温消泡5min,然后开始降温;
S3:温度降至65℃左右时加入预先溶解好的C、D相,搅拌均匀,
S4:温度降至55℃左右时加入D、E相搅拌均匀,检验合格热出料,得含本发明组合物的消包止痒油膏。
产品经过1个月50℃,3个月45℃和-22℃,1个星期光照后,颜色和性状均稳定。参见图4。
实施例8:
由本研究团队随机选取健康自愿参加测试的人员,年龄20-55岁,自愿选取测试样品,进行止痒去痘试验。每个自愿者用之前,先将该样品涂抹在手腕部部上方3厘米处,30分钟后无发红与不适(过敏试验),才涂抹在痘痘与痕痒部位。随访结果如表:
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。

1.一种抑菌、消痘止痒的组合物,其特征在于,其活性成分由天山雪莲提取物、紫草提取物、维生素B3和维生素B5组成,该活性成分的质量配比如下:
天山雪莲提取物:1-9
紫草提取物:1-9
维生素B3:0.1-9.9
维生素B5:0.1-9.9。
2.根据权利要求1所述的抑菌、消痘止痒的组合物,其特征在于,活性成分的质量配比如下:

活性成分的质量配比如下:
天山雪莲提取物:2-3
紫草提取物:7-8
维生素B3:2-5
维生素B5:5-8。
4.根据权利要求3所述的抑菌、消痘止痒的组合物,其特征在于,
活性成分的质量配比如下:
天山雪莲提取物:2
紫草提取物:7-8
维生素B3:2
维生素B5:8。
5.一种抑菌、消痘止痒的组合物的制备方法,其特征在于,包括以下步骤:
取天山雪莲,粉碎成粗粉,加其8-12倍量的水浸泡0.5±0.05h,加热提取,浓缩,得天山雪莲提取物;
取紫草,切碎,加10-30倍量的丙酮回流提取,回收丙酮,残渣用1,3-丁二醇溶解,得紫草提取物;
按照配比将所述天山雪莲提取物、紫草提取物以及维生素B3和维生素B5 配制 在一起,即得。
6.权利要求1-4任一项所述抑菌、消痘止痒的组合物在制备治疗皮肤病的药物或护肤品中的应用。
7.根据权利要求6所述的应用,其特征在于,所述皮肤病为皮肤轻微感染、湿疹、小儿尿布疹和蚊虫叮咬。
8.一种抑菌、消痘止痒的药物制剂或护肤品,其特征在于,其活性成份包括有权利要求1-4任一项所述的抑菌、消痘止痒的组合物以及合适的辅料。
9.根据权利要求8所述的抑菌、消痘止痒的药物制剂或护肤品,其特征在于,所述药物制剂或护肤品的剂型是啫喱剂。
10.根据权利要求8所述的抑菌、消痘止痒的药物制剂或护肤品,其特征在于,所述药物制剂或护肤品的剂型是喷雾剂。
11.根据权利要求8所述的抑菌、消痘止痒的药物制剂或护肤品,其特征在于,所述药物制剂或护肤品的剂型是慕斯丝滑日霜剂。
12.根据权利要求8所述的抑菌、消痘止痒的药物制剂或护肤品,其特征在于,所述药物制剂或护肤品的剂型是油膏。
