Abstract
The invention relates to a method for collecting oral secretion of small lepidoptera larvae, which is characterized by comprising the following steps of: (1) rolling the middle-front part of the small lepidoptera larva from back to front by using a roller type pressing device; (2) collecting oral secretion by a liquid transfer device when the small lepidoptera larva spits out the oral secretion; (3) and finally, blowing the oral secretion in the pipettor into the collection container by using a blowing device. The method is easy to operate, easy in material obtaining, low in cost and high in collection efficiency, can be used for collecting oral secretion of the small lepidoptera larva by one person, and has wide application prospect.
Description
Technical Field
The invention relates to a method for collecting oral secretion of small lepidoptera larvae.
Background
Oral secretions of lepidopteran insects are also called regurgitation, and often contain various components such as insect saliva and digestive juice of the midgut. When lepidopteran larvae feed on plants, elicitors in oral secretions can elicit a defense response in the plant. The mutual relationship between insects and plants needs to be deeply researched, the action mechanism of insect induced plant defense reaction is clarified, and the acquisition of oral secretion of lepidoptera larvae is an important precondition.
The method for obtaining oral secretion of lepidoptera insect larvae mainly comprises the following steps of (1) pressing larva bodies to obtain oral secretion; (2) freezing the larva for several minutes, and then placing the larva at normal temperature until the larva recovers the secretion of the spitting cavity; (3) the head of the insect is collided to cause the secretion of the cavity of the outlet of the insect. In the above methods, only oral secretions from larger lepidopteran larvae are typically collected, for example: and Bombyx mori and the like. But for smaller lepidopteran larvae, for example: the above methods cannot obtain a large amount of oral secretion from diamondback moth. (1) rolling the middle front part of the small lepidoptera larva 2 from back to front by using the roller type pressing device 1;
(2) when the small lepidoptera larva 2 spits out the oral secretion 4, collecting the oral secretion 4 by using a liquid transfer device 3;
(3) finally, the oral secretion 4 in the pipettor 3 is blown into the collection container 6 by the blowing device 5.

(1) rolling the middle-front part of the small lepidoptera larva from back to front by using a roller type pressing device;
(2) collecting oral secretion by a liquid transfer device when the small lepidoptera larva spits out the oral secretion;
(3) finally, blowing the oral secretion in the pipettor into a collection container by using a blowing device;
