Abstract

本发明公开了一种细胞外基质神经修复膜,所述神经修复膜是由异种动物周围神经处理得到多孔膜。本发明还公开了一种细胞外基质神经修复膜的制备方法,通过将异种动物周围神经进行脱细胞处理后,最大限度的保留促进神经再生的有益成分,制得的细胞外基质神经修复膜安全性好,能特异性促进损伤神经的再生。

一种细胞外基质神经修复膜及其制备方法
技术领域
本发明涉及一种生物医用技术领域的修复膜,特别涉及一种用于周围神经损伤修复的细胞外基质神经修复膜及其制备方法。
背景技术
神经系统是人体中最重要的器官,由中枢神经和周围神经两大部分组成,中枢神经是指脑和脊髓,周围神经则是指离开脑和脊髓的神经纤维由结缔组织连接、包裹组成粗细不等的神经束的基础上,再由结缔组织进一步包裹而形成的(肉眼可见的)神经束的集合体。周围神经损伤表现为各种原因引起的受该神经支配的区域出现感觉障碍、运动障碍和营养障碍,只有在受损神经得到修复的情况下,器官的正常功能才能得到完全的恢复。
周围神经离断损伤后,能以约1毫米/天的速度从近端向远端生长,临床上常通过对离断神经吻合的方法就行修复,然而神经吻合后仍然会存在如下问题:局部缺乏良好的促进神经纤维生长的微环境,难以形成有利于神经纤维再生的“再生室”效应;吻合口暴露,神经生长因子外流,亦会降低神经的修复效果;纤维组织侵入吻合口形成瘢痕,阻碍再生纤维通过吻合口长入神经远端;生长受阻的再生纤维极易在局部形成神经纤维瘤子。寻找一种有效的修复材料,提高神经再生的速度和质量,是当前急需解决的问题。
专利(CN201210397806.X)公开了一种促神经修复的材料,通过将雪旺细胞接种于小肠黏膜下层上,共培养获得复合了雪旺细胞的小肠黏膜下层,经反复冻融后冻干,得到含有神经生长因子的小肠黏膜下层支架。神经修复再生是一个复杂的过程,通过材料提供生长因子,并不能有效促进神经再生。专利(CN201510042570.1)公开了一种神经修复导管其制备方法,该神经修复导管有内外两层结构,内层为胶原、壳聚糖及其衍生物中的一种或多种复合材料制得,外层由胶原、壳聚糖、丝素蛋白及其衍生物中的一种或者多种复合材料制得,通过这种方式获得的神经修复导管不具有特异性引导神经再生的功能。专利(CN201510364908.5)公开了一种周围神经修复膜及其制备方法,由壳聚糖、胶原蛋白、透明质酸、几丁质、海藻糖、明胶、醋酸纤维素、乙二醇、乙酸、聚磷酸钙、双蒸水按照一定的比例制备而成。此种方法制备的神经修复膜不具有特异性引导神经再生的功能。
细胞外基质是由细胞合成并分泌到细胞外间质中的大分子物质,主要由三类成分组成:1.结构蛋白:如胶原、弹性蛋白和基膜粘连蛋白等;2.多糖:如糖胺聚糖、蛋白聚糖、透明质酸和硫酸软骨素等;3.粘连蛋白:如纤粘连蛋白和层粘连蛋白等,此外,还有一些脂质和生长因子等。将同种或异种组织进行脱细胞等处理后可获得细胞外基质,将细胞外基质应用于组织修复,为组织细胞提供再生环境,可有效促进组织修复。
发明内容
本发明的目的是提供一种细胞外基质神经修复膜,通过将同种或异种动物周围神经进行处理后,获得含有促进神经再生的丰富的结构蛋白、多糖、粘连蛋白及生长因子的神经修复膜,应用于患者周围神经损伤修复,能够特异性促进损伤周围神经的再生。
本发明的另一目的是提供上述细胞外基质神经修复膜的制备方法。
为了实现上述目的,本发明采用以下技术方案:
一种细胞外基质神经修复膜是由异种动物周围神经处理得到的膜。作为本发明的一种优选方案,取材周围神经选自猪、牛、羊、兔、鼠的周围神经,制得的脱细胞神经基质修复膜为多孔膜,膜厚度为0.05mm~0.5mm。
一种细胞外基质神经修复膜的制备方法,包括以下步骤:
(1)取新鲜动物周围神经组织,去除附着在神经组织上的筋膜、脂肪等,剪碎成小块,去离子水清洗3次;
(2)依次使用低温冻融、曲拉通X-100、过氧乙酸溶液进行脱细胞处理,去除周围神经组织中的细胞成分;
(3)10倍PBS溶液漂洗至中性,沥干水分。
(4)将步骤(3)中获得的固体成分置于盐酸-氯化钙溶液中,高速剪碎搅拌均匀后,倒入不锈钢平面盘中,低温烘干得到膜。
(5)将步骤(4)中获得的膜漂洗数次,至中性,冷冻干燥,获得多孔细胞外基质神经修复膜。
作为本发明的一种优选技术方案,步骤(2)中使用的低温冻融冷冻温度为-70℃,冷冻时间大于24h,融化温度为25℃,循环3次。
作为本发明的一种优选技术方案,步骤(2)中使用的曲拉通X-100质量浓度为1%~5%,过氧乙酸溶液质量浓度为0.05%~2%。
作为本发明的一种优选技术方案,步骤(6)中使用的盐酸-氯化钙溶液,盐酸浓度为0.001N~0.1N,氯化钙的质量浓度为5%~30%。
作为本发明的一种优选技术方案,步骤(6)中低温烘干的温度为25℃~35℃。
与现有技术相比,本发明具有以下优点:
(1)本发明制得的细胞外基质神经修复膜为多孔结构,其孔径接近半透膜,应用于周围神经损伤修复时,一方面成功预防了周围组织与损伤神经组织的粘连,另一方面,营养物质可透过膜,从而加速神经再生。
(2)本发明制得的细胞外基质神经修复膜制备过程中,使用的试剂温和,有效保留了细胞外基质中有活性的结构蛋白、多糖、粘连蛋白及生长因子,可特异性引导损伤神经的再生。
本发明对组织进行去细胞处理过程中使用的化学试剂水溶性好,易通过多次水洗去除,且试剂不与细胞外基质反应,不会产生试剂黏附在材料上不易去除的现象。
具体实施方式
实施例1:
取新鲜猪坐骨神经去除附着在神经组织上的筋膜、脂肪等,剪碎成小块,去离子水清洗3次,置于-70℃超低温冰箱中,冷冻24h,25℃解冻,循环冻融3次。依次使用2%曲拉通X-100溶液漂洗2h,0.05%过氧乙酸溶液漂洗1h,10倍PBS溶液漂洗至pH值中性,沥干水分后,称取2g投入到100mL盐酸(0.001N)-氯化钙(5%)溶液中,高速均质处理形成溶液,倒入不锈钢平面盘中,30℃热风烘干得到膜,膜经漂洗数次,至pH值中性,冷冻干燥,获得多孔细胞外基质神经修复膜,厚度为0.07mm。
实施例2:
取新鲜牛坐骨神经去除附着在神经组织上的筋膜、脂肪等,剪碎成小块,去离子水清洗3次,置于-70℃超低温冰箱中,冷冻24h,25℃解冻,循环冻融3次。依次使用5%曲拉通X-100溶液漂洗2h,2%过氧乙酸溶液漂洗0.5h,10倍PBS溶液漂洗至pH值中性,沥干水分后,称取2g投入到100mL盐酸(0.01N)-氯化钙(10%)溶液中,高速均质处理形成溶液,倒入不锈钢平面盘中,25℃热风烘干得到膜,膜经漂洗数次,至pH值中性,冷冻干燥,获得多孔细胞外基质神经修复膜,厚度为0.1mm。
实施例3:
取新鲜兔坐骨神经去除附着在神经组织上的筋膜、脂肪等,剪碎成小块,去离子水清洗3次,置于-70℃超低温冰箱中,冷冻24h,25℃解冻,循环冻融3次。依次使用3%曲拉通X-100溶液漂洗2h,1%过氧乙酸溶液漂洗1h,10倍PBS溶液漂洗至pH值中性,沥干水分后,称取3g投入到100mL盐酸(0.01N)-氯化钙(15%)溶液中,高速均质处理形成溶液,倒入不锈钢平面盘中,35℃热风烘干得到膜,膜经漂洗数次,至pH值中性,冷冻干燥,获得多孔细胞外基质神经修复膜,厚度为0.5mm。

Claims (9)

1.一种细胞外基质神经修复膜,其特征在于,所述的神经修复膜是由异种动物周围神经处理得到多孔膜。
2.根据权利要求1所述一种细胞外基质神经修复膜,其特征在于,所述膜厚度为0.05mm~0.5mm。
3.根据权利要求1所述一种细胞外基质神经修复膜,其特征在于,所述异种动物周围神经选自猪、牛、羊、兔、鼠的周围神经。
4.根据权利要求1所述细胞外基质神经修复膜,其特征在于,所述脱细胞神经基质修复膜为多孔膜。
5.一种细胞外基质神经修复膜的制备方法,其特征在于包括以下步骤:
(1)取新鲜动物周围神经组织,去除附着在神经组织上的筋膜、脂肪等,剪碎成小块,去离子水清洗3次;
(2)依次使用低温冻融、曲拉通X-100、过氧乙酸溶液进行脱细胞处理,去除周围神经组织中的细胞成分;
(3)10倍PBS溶液漂洗至中性,沥干水分;
(4)将步骤(3)中获得的固体成分置于盐酸-氯化钙溶液中,高速剪碎搅拌均匀后,倒入不锈钢平面盘中,低温烘干得到膜;
(5)将步骤(4)中获得的膜漂洗数次,至中性,冷冻干燥,获得多孔细胞外基质神经修复膜。
6.根据权利要求4所述,其特征在于,所述低温冻融冷冻温度为-70℃,冷冻时间大于24h,融化温度为25℃,循环3次。
7.根据权利要求4所述,其特征在于,所述曲拉通X-100质量浓度为1%~5%、过氧乙酸溶液质量浓度为0.05%~2%。
8.根据权利要求4所述,其特征在于,所述盐酸-氯化钙溶液中,盐酸浓度为0.001N~0.1N,氯化钙的质量浓度为5%~30%。
