Abstract

The invention relates to a coconut shell extract and application thereof in enhancing rutin antiallergic capability, and a preparation method of the coconut shell extract comprises the following steps: (1) taking dried coconut shells, crushing the dried coconut shells to 20-100 meshes, adding 80% ethanol with the volume fraction of 8-15 times of the mass of the coconut shells, performing reflux extraction for 6-10 hours, filtering while hot, removing filter residues, and performing reduced pressure concentration on the filtrate to obtain an extract; (2) and (2) adsorbing the extract obtained in the step (1) by using a macroporous resin column, eluting by using 60-70% ethanol in volume fraction, collecting eluent with the volume of 4-6 times that of the column, concentrating under reduced pressure, and drying in vacuum to obtain the coconut shell extract. The coconut shell extract has the effect of enhancing the inhibition rate of rutin on hyaluronidase, namely enhancing the anti-allergic capability of rutin, can be developed and used for preparing an anti-allergic agent, and can be used in the fields of medicines, cosmetics and the like.

Description

An antiallergic agent prepared from coconut shell extract and rutin
Technical Field
The invention belongs to the field of whitening and sun protection, and particularly relates to a coconut shell extract and application thereof in enhancing rutin antiallergic capability.
Background
Coconut (coconut nucifera L.) is a monocotyledonous plant of the genus Cocos belonging to the family Palmae, and is a very valuable crop of tropical economy. Coconut meat is edible or can be used for expelling oil, and coconut water (endosperm) can be used as beverage. Coconut shell (i.e. coconut endocarp) and coconut chaff are byproducts of deep processing of coconut, the coconut shell is black brown and firm, at present, the coconut shell is mainly used for preparing coconut shell carbon through carbonization and further preparing activated carbon, and no report is provided for other applications. The invention adopts organic solvent to extract the coconut shells, and finds that the extract has the capability of enhancing rutin to clear free radicals, so that the coconut shell extract can be developed and used for preparing antioxidants to be used in the fields of medicines, foods, cosmetics and the like.
Disclosure of Invention
The invention provides a coconut shell extract, which is characterized in that the preparation method of the coconut shell extract comprises the following steps:
(1) taking dried coconut shells, crushing the dried coconut shells to 20-100 meshes, adding 80% ethanol with the volume fraction of 8-15 times of the mass of the coconut shells, performing reflux extraction for 6-10 hours, filtering while hot, removing filter residues, and performing reduced pressure concentration on the filtrate to obtain an extract;
(2) and (2) adsorbing the extract obtained in the step (1) by using a macroporous resin column, eluting by using 60-70% ethanol in volume fraction, collecting eluent with the volume of 4-6 times that of the column, concentrating under reduced pressure, and drying in vacuum to obtain the coconut shell extract.
The macroporous resin in the step (2) is selected from D101 type or AB8 type.
Another embodiment of the present invention provides a method for preparing the coconut shell extract, which comprises the following steps:
(1) taking dried coconut shells, crushing the dried coconut shells to 20-100 meshes, adding 80% ethanol with the volume fraction of 8-15 times of the mass of the coconut shells, performing reflux extraction for 6-10 hours, filtering while hot, removing filter residues, and performing reduced pressure concentration on the filtrate to obtain an extract;
Another embodiment of the present invention provides the use of the coconut shell extract as described above for the preparation of an antiallergic agent. The antiallergic agent can be used in medicine, cosmetics, etc.
Another embodiment of the invention provides the application of the coconut shell extract in enhancing rutin antiallergic effect.
An antiallergic agent characterized by comprising the coconut shell extract of the present invention.
An antiallergic agent is characterized by comprising the coconut shell extract and rutin.
An antiallergic agent is characterized by comprising the coconut shell extract and rutin, wherein the amount of the coconut shell extract is 6-8% of the weight of the rutin.
Compared with the prior art, the invention has the advantages that: the invention develops another new application of coconut shells which are byproducts of deep processing of coconuts, namely, the coconut shell extract can enhance the inhibition rate of rutin on hyaluronidase, namely enhance the anti-allergic capability of rutin, can be developed and used for preparing an anti-allergic agent, is used in the fields of medicines, cosmetics and the like, and greatly enriches the application of coconut shell wastes.
Detailed Description
In order to facilitate a further understanding of the invention, the following examples are provided to illustrate it in more detail. However, these examples are only for better understanding of the present invention and are not intended to limit the scope or the principle of the present invention, and the embodiments of the present invention are not limited to the following.
Example 1
(1) Pulverizing dried coconut shell (1.0kg) to 100 mesh, adding 8.0kg ethanol solution with volume fraction of 80%, reflux extracting for 6 hr, filtering while hot, removing filter residue, and concentrating the filtrate under reduced pressure to obtain extract;
(2) and (2) adsorbing the extract obtained in the step (1) by using a macroporous resin column, eluting by using 60-70% ethanol in volume fraction, collecting eluent with the volume of 4-6 times that of the column, concentrating under reduced pressure, and drying in vacuum to obtain a coconut shell extract (16.8 g, hereinafter referred to as a product A).
Example 2
(1) Pulverizing dried coconut shell (1.0kg) to 20 mesh, adding 15.0kg ethanol solution with volume fraction of 80%, reflux extracting for 10 hr, hot filtering, removing residue, and concentrating the filtrate under reduced pressure to obtain extract;
(2) and (2) adsorbing the extract obtained in the step (1) by using a macroporous resin column, eluting by using 60-70% ethanol in volume fraction, collecting eluent with the volume of 4-6 times that of the column, concentrating under reduced pressure, and drying in vacuum to obtain a coconut shell extract (19.5 g, hereinafter referred to as a product B).
Example 3 anti-allergic test
Samples tested at concentrations of 30. mu.g/mL and 60. mu.g/mL, respectively, were: rutin, a product A, a product B, a product A and rutin (the mass of the product A is 6 percent of the mass of the rutin), and a product B and rutin (the mass of the product B is 8 percent of the mass of the rutin) have the inhibition rate on hyaluronidase, and the following test results are according to the Chinese patent application numbers: 201310531168.0, see table 1.
TABLE 1
Figure RE-BDA0001442453970000031
"-" indicates that the sample inhibited hyaluronidase by less than 10%.
The experimental results show that the product A, B has almost no inhibition effect on hyaluronidase, but when the product is mixed with rutin, particularly when the amount of the rutin is 6-8% of the weight of the rutin, the inhibition of the rutin on the hyaluronidase can be obviously enhanced; the experimental result shows that the coconut shell extract can be used for enhancing the antiallergic effect of rutin, and further used as an additive for preparing an antiallergic agent in the fields of medicines, cosmetics and the like.
