一种羊精子室温保存稀释液及其制备方法,属于动物繁殖技术领域,将Tris、柠檬酸、果糖、海藻糖、维生素E、青霉素、链霉素、纯净水混合均匀,调整PH值至6.60±0.05,使用0.22微米的滤器过滤,取卵黄20ml,放入混合液体中,将液体放用磁力搅拌器进行搅拌2小时,2500g离心60min,吸上清。本发明的优点是:解决羊原精在室温下保存时间短,活力下降快的问题,而提供一种可较长时间保存,并不影响精子的活力,有效提高种公羊精液室温保存的有效存活时间,成分简单、便于操作。

羊精子室温保存稀释液及其制备方法
技术领域
本发明属于动物繁殖技术领域,具体涉及一种将羊原精稀释后在室温长期保存的羊精子室温保存稀释液及其制备方法。
背景技术
与牛相比,羊精虽然已经开发出冷冻保存技术,但是生产费用高、冻后精子活力差、受胎率低,使得液态保存成为羊精保存的主要方式。液态保存分为低温保存与常温保存。低温保存的优点在于精液保存时间长,缺点为需要专门的低温设备,为了方便运输,设备体积不宜过大,但也导致设备低温维持时间有限,不方便远距离运输;常温精子保存时间短,但不需要特别的低温设备,比较适合短距离的精子运输。稀释液主要包括Tris-柠檬酸-糖类、EDTA-柠檬酸钠-糖类、柠檬酸盐-糖类三大类。糖类主要葡萄糖、果糖、蔗糖等,不同糖类在动物品种、各种配方、稀释方法、储存温度时会有不同效果,有些研究将几种糖类混合加入,低温下效果好的保存稀释液在室温时使用效果无法达到最佳。目前,几种羊精子室温保存稀释液与羊精子的稀释比例一般是在2-10倍,稀释液稀释精子后的有效存活时间(精子活力大于0.3的存活时间)一般在36h左右,精子存活时间在48h左右,因此开发出能够在室温较长时间保存精子的稀释液有重要价值。本发明的目的是开发一种能够将羊精液稀释并在常温下较长时间保存的稀释液。
发明内容
本发明解决的问题是羊原精稀释常温保存的问题,提供一种羊精子室温保存稀释液及制备方法,能够使羊原精在室温下较长时间保存,并不影响精子的活力,有效的提高了种公羊精液的利用率。
本发明的目的是通过以下技术方案实现的;一种羊精子室温保存稀释液,其特征在于:它由以下组份组成:
调整PH值至6.60±0.05。
一种羊精子室温保存稀释液的制备方法,其特征在于:它包括以下步骤:
(1)配置试剂
混合均匀,调整PH值至6.60±0.05,使用0.22微米的滤器过滤,
(2)取卵黄20ml,放入步骤(1)液体中,将液体放用磁力搅拌器进行搅拌2小时,2500g离心60min,吸上清。
本发明的优点和有益效果是:解决羊原精在室温下保存时间短,活力下降快的问题,而提供了一种可较长时间保存,并不影响精子的活力,有效提高种公羊精液室温保存的有效存活时间,成分简单、便于操作。
具体实施方式
实施例1:一种绵羊低温稀释保存液的制备方法,它包括以下步骤:
1、配制试剂
调整PH值至6.60±0.05,使用0.22微米的滤器过滤;
(2)取卵黄20ml,加到步骤(1)液体中,将液体放用磁力搅拌器进行搅拌2小时,2500g水平离心60min,吸上清;
2、精液检测
选取两只健康种公绵羊,使用假阴道法采精,精液为乳白色,无异味,镜下检测活力分别为0.65和0.6,过滤后使用密度仪检测原精实际浓度分别为羊1是10.3亿/ml,体积为3.2ml;羊2是11亿/ml,体积为1.8ml;
3、精液稀释
用步骤1制备的稀释液32.96ml将步骤2精液检测后的羊1精液稀释为1亿/ml,稀释液19.8ml将羊2精液稀释为1亿/ml,分别混匀后,然后放入22度室温保存,
精子活力检测:预先把载玻片和盖玻片放在37℃加热板上预热10分钟,用移液器离心管取15μl样品滴在载玻片上,压片后置于显微镜下镜检,选择压片均匀的样品区域,一个压片取5个视野,中间1个,四周各取1个,分别计数活精子数和死精子数,并计算活力;
活力=活精子数/活精子数与死精子数的和
羊1精子活力为0.65,精子有效存活时间为96h,羊2精子活力为0.6,精子的有效存活时间为72h。说明不同活力羊精稀释保存后存活时间有差异。但是,仍可较长时间保存,且不影响精子活力,且种公羊精液室温保存的有效存活时间有效提高。
实施例2:一种羊精子室温保存稀释液配置制备方法,它包括以下步骤:
1、配制试剂
调整PH值至6.60±0.05,使用0.22微米的滤器过滤;
(2)取20ml卵黄,加到步骤(1)液体中,将液体放用磁力搅拌器进行搅拌2小时,2500g水平离心60min,吸上清;
2、精液检测
使用假阴道法采精,精液为乳白色,无异味,镜下检测活力为0.65,过滤后使用密度仪检测原精实际浓度为12.3亿/ml,体积为2.2ml,共27.06亿精子,
3、精液稀释
将步骤2检测的原精液平均分为两份,第一份13.53亿羊原精,用步骤1配置的室温稀释液13.53ml,将羊精液稀释为1亿/ml,第二份13.53亿羊原精用步骤1配置的室温稀释液6.765ml,将羊精液稀释为2亿/ml,分别放入19度室温保存;
精子活力检测:预先把载玻片和盖玻片放在37℃加热板上预热10分钟,用移液器离心管取15μl样品滴在载玻片上,压片后置于显微镜下镜检,选择压片均匀的样品区域,一个压片取5个视野,中间1个,四周各取1个,分别计数活精子数和死精子数,并计算活力;
活力=活精子数/活精子数与死精子数的和
在室温19度下,羊精子分别稀释12倍时,精子存活时间120h,稀释6倍时,精子存活时间96h,使用该羊精子室温保存稀释液,可较长时间保存,且不影响精子活力,且种公羊精液室温保存的有效存活时间有效提高。

调整PH值至6.60±0.05。
2.一种权利要求1所述羊精子室温保存稀释液的制备方法,其特征在于:它包括以下步骤:
(1)配置试剂
调整PH值至6.60±0.05,使用0.22微米的滤器过滤,
(2)取卵黄20ml,加到步骤(1)液体中,将液体放用磁力搅拌器进行搅拌2小时,2500g离心60min,吸上清。
