Abstract
本发明公开了一种自荧光聚丙烯酰胺纳米粒子及其制备方法和应用,该纳米粒子由戊二醛交联处理丙烯酰胺纳米粒子制得,所述聚丙烯酰胺纳米粒子由含有多聚赖氨酸的丙烯酰胺单体溶液制备而成。本发明制备的自荧光聚丙烯酰胺纳米粒子可被高效内化到细胞中标记细胞,自体荧光在体外和体内长时间明亮稳定,具有优良的可见荧光及近红外荧光属性,且粒径均一、单分散好、带正电荷、吸水性强、亲水性好,在细胞和动物中具有极好的生物相容性。同时本发明制备方法简单方便,成本低,原料来源广泛,所制备的自荧光聚丙烯酰胺纳米粒子可用于生物检测、细胞标记、动物成像、药物递送、基因治疗等领域,具有重要应用价值。
Description
技术领域
本发明涉及纳米生物技术领域,具体涉及一种自荧光聚丙烯酰胺纳米粒子及其制备方法和应用。
背景技术
目前,根据无创、实时和高分辨率的疾病检测方式,光学成像方法使用越来越广泛。光学成像方法可以从光信号中获得疾病解剖结构,甚至疾病代谢的分子信息,在各种光学成像技术中,荧光光学成像技术具有较高的灵敏度和分辨率。用于组织成像的光可以分为紫外线,可见光和近红外(NIR)光。NIR光(700~1000nm)被称为生物体本身及其内源物质的相对低自发荧光的“生物窗”。与传统的可见荧光相比,NIR荧光(NIRF)具有一些关键优势,包括高灵敏度、高信噪比(S/N)比和深层组织穿透能力。目前,NIRF技术已逐渐应用于生物检测、细胞标记、肿瘤的成像、光疗及热疗等领域。
具有荧光属性的纳米粒子在生物检测、细胞标记、肿瘤的成像、光疗及热疗等领域具有重要应用价值。目前具有荧光属性的纳米粒子常用的制备方法为在纳米粒子表面修饰荧光分子(如FITC等)。但该修饰过程不仅需要繁琐的纳米材料表面改性及化学修饰,这种改性过程及所修饰上的荧光分子,会对纳米材料本身的物化及生物效应产生影响。例如,荧光团本身可以在荧光染料与纳米颗粒的偶联中对改变标记靶的固有行为。例如,已经报道,当迷迭香单独用作光敏剂时,单线态氧产率为0.75,而一旦将迷迭香接枝到SiO2纳米粒子上,则产率降至0.3。此外,荧光染料与纳米颗粒偶联时常发生染料的荧光淬灭。因此,有自发荧光属性的纳米材料成为发展趋势。例如,量子点(QD)就是一种具有自发荧光属性的纳米材料,且可以通过改变尺寸、形状和材料之处出可发射不同波长的荧光。然而,量子点的生物相容性不佳,必须通过分子修饰来实现更好的生物相容性,但修饰分子可能干扰量子点并影响整个检测信号。如量子点一样,许多无机纳米材料表现出较差的生物相容性,相比之下,有机纳米颗粒由于其良好的生物相容性、水溶性和可控的表面官能团而成为理想的载体。因此,构建具有良好生物相容性的自体荧光纳米材料是迫切需要的。
聚丙烯酰胺纳米颗粒(PAANP)由于其亲水性、生物相容性、生物降解性和低毒性而引起了纳米材料科学研究的特别兴趣。PAANP通常用于细胞内分析物和代谢物监测。PAANP也作为体内递送系统进行了研究。例如,PAANP已被应用于光动力疗法(PDT),用于荷载各种光敏剂。此外,PAANP的制备过程容易且成本低廉,相比之下,其他纳米材料,特别是金属和半导体量子点,具有复杂或昂贵的制造工艺。
戊二醛(GTA)是一种常用的交联剂。当其用作为交联剂时,很早就发现可产生荧光。其产生机理为戊二醛自身聚合产生的双碳键(C=C)以及戊二醛醛基与被交联分子的氨基交联而产生的希夫碱(C=N)。荧光来自不同轨道之间的电子跃迁,包括C=C键中的π-π*跃迁和C=N键中的n-π*跃迁。近年来的研究表明,GTA交联可用于生产制造自发荧光纳米粒子和微球,如GTA交联牛血清白蛋白(BSA)纳米粒子、壳聚糖纳米粒子、明胶纳米粒子和壳聚糖微球。但这些研究中GTA交联材料均报道仅产生可见荧光,但没有报道使用GTA交联来制造NIRF纳米材料,也未报道可产生NIRF和及其体内应用等。由于可见荧光仅可用于体外生物检测、荧光成像,而因动物体自身强荧光背景和可见荧光激发波长弱的组织穿透力而无法用于动物体内成像。NIRF却因其激发波长很强的组织穿透力以及动物体很低的近红外荧光背景而可用于动物体内成像。因此,在近年来的生物检测、活体成像、光疗等领域产生广泛应用。
发明内容
发明目的:针对现有技术存在的问题,本发明提供一种自荧光聚丙烯酰胺纳米粒子,该聚丙烯酰胺纳米粒子同时具有优异的自发可见荧光和近红外荧光,在细胞和动物中具有极好的生物相容性,使用该纳米材料可进行细胞标记及体内外荧光成像。
本发明还提供该自荧光聚丙烯酰胺纳米粒子的制备方法和应用。
技术方案:为了实现上述目的,如本发明所述的一种自荧光聚丙烯酰胺纳米粒子,由戊二醛处理聚丙烯酰胺纳米粒子制得,所述聚丙烯酰胺纳米粒子由含有多聚赖氨酸的丙烯酰胺单体溶液制备而成。
其中,所述的丙烯酰胺单体溶液由丙烯酰胺(acrylamide,AA)、甲叉双丙烯酰胺(Bis-acrylamide,MBA)和N-(3-氨基丙基)甲基丙烯酰胺N-(3-氨基丙基)甲基丙烯酰胺(APMA)溶于水中形成。
其中,所述多聚赖氨酸(poly-lysine)包括各种分子量的多聚赖氨酸。多聚赖氨酸优选ε-聚赖氨酸(ε-poly-L-lysine,ε-PL)。
所述自荧光聚丙烯酰胺纳米粒子具有多重荧光属性,包括可见荧光及近红外荧光。可见荧光可用荧光显微镜、流式细胞仪、荧光分光光度计检测;自荧光聚丙烯酰胺纳米粒子的近红外荧光可用近红外荧光成像仪(如LiCOR Odyssey)、荧光分光光度计检测。
本发明所述的自荧光聚丙烯酰胺纳米粒子的制备方法,包括如下步骤:
(1)将丙烯酰胺、甲叉双丙烯酰胺、N-(3-氨基丙基)甲基丙烯酰胺溶于超纯水中混匀;再向溶液中再加入多聚赖氨酸混匀,形成为丙烯酰胺单体溶液;
(2)通过反相微乳液法将丙烯酰胺单体溶液加入到油相中搅拌后纯化得到聚丙烯酰胺纳米粒子(PAANP);
(3)用戊二醛对聚丙烯酰胺纳米粒子进行交联处理,制备出自荧光聚丙烯酰胺纳米粒子(fPAANP)。
作为优选,步骤(1)所述丙烯酰胺单体溶液中丙烯酰胺(AA)的浓度为250~275mg/ml,甲叉双丙烯酰胺(MBA)浓度为75~90mg/ml,N-(3-氨基丙基)甲基丙烯酰胺N-(3-氨基丙基)甲基丙烯酰胺(APMA)为10~20mg/ml,多聚赖氨酸浓度为40~50mg/ml。进一步地,AA为265mg/ml、MBA为80mg/ml、APMA为15mg/ml。
作为优选,步骤(2)所述反相微乳液法中油相为含有表面活性剂AOT与Brij30的正己烷,水相为丙烯酰胺单体溶液。
更进一步地,步骤(2)所述油相中表面活性剂AOT的浓度为30~40mg/ml,Brij30的浓度为66-72mg/ml。进一步地,AOT的浓度为35.6mg/ml,Brij30的浓度为68.9mg/ml。
最为优选,步骤(2)所述丙烯酰胺单体溶液加入到油相中搅拌后再向油相中加入过硫酸铵-四甲基乙二胺(APS-TEMED)溶液以引发聚合反应,搅拌。此外,反相微乳液法制备聚丙烯酰胺纳米粒子时,控制搅拌转速,可制备出不同粒径的聚丙烯酰胺纳米粒子。
其中,步骤(2)所述纯化过程为:蒸发去除正己烷,用无水乙醇沉淀聚合物,再用无水乙醇洗涤聚合物;乙醇挥发后聚合物中加入水,超声重悬,再加水超滤;最后用水重悬聚合物成为聚合物水溶液,超声后过滤,室温放置,得到聚丙烯酰胺纳米粒子溶液。
作为优选,步骤(2)所述反相微乳液法制备聚丙烯酰胺纳米粒子过程中向油相溶液持续充入氩气。持续充入氩气,以排除制备过程中的氧气,防止氧化。
其中,步骤(3)所述交联处理具体过程为向步骤(2)获得的聚丙烯酰胺纳米粒子溶液中加入戊二醛溶液,超声反应;超滤水洗去除多余戊二醛;将戊二醛处理后的聚丙烯酰胺纳米粒子溶于水,超声处理;过滤聚丙烯酰胺纳米粒子水溶液以除菌,室温避光保存,得到自荧光聚丙烯酰胺纳米粒子成品。
作为优选,所述聚丙烯酰胺纳米粒子溶液中加入戊二醛溶液后的戊二醛浓度为0.5~3mg/ml。进一步地,戊二醛的浓度为1.5mg/ml。
本发明所述的自荧光聚丙烯酰胺纳米粒子在生物检测、细胞标记、动物成像、药物递送和基因治疗等领域中的应用。
GTA交联产生的荧光来自两种不同的双键,即C=N和C=C。本发明通过增加PAANP中的氨基改善PAANP的NIRF性质。通过研究了富含氨基的化学物质,最终发现通过加入多聚赖氨酸(如无毒且已经用于体内如食品添加剂ε-PL)到丙烯酰胺单体溶液中,采用反相微乳液法制备出聚丙烯酰胺纳米粒子(PAANP);之后用戊二醛对聚丙烯酰胺纳米粒子进行交联处理,制备出出荧光属性和体内外生物相容性良好的自荧光聚丙烯酰胺纳米粒子,发现GTA处理的聚丙烯酰胺凝胶显示很强的NIRF。
有益效果:与现有技术相比,本发明具有如下优点:
(1)本发明制备的自荧光聚丙烯酰胺纳米粒子粒径约16nm,Zeta电位约+16mV,可被高效内化到细胞中,标记细胞;同时自荧光聚丙烯酰胺纳米粒子的自体荧光在体外和体内长时间明亮稳定,具有优良的可见荧光及近红外荧光属性,且粒径均一、单分散好、带正电荷、吸水性强、亲水性好,在细胞和动物中具有极好的生物相容性。
(2)本发明制备方法简单方便,成本低,原料来源广泛,所制备的自荧光聚丙烯酰胺纳米粒子可用于生物检测、细胞标记、动物成像、药物递送、基因治疗等领域,具有重要应用价值。

具体实施方式
以下结合附图和实施例对本发明作进一步说明。
试剂和材料:二辛基磺基琥珀酸钠盐(AOT)、聚氧乙烯(4)月桂基醚(Brij30)、丙烯酰胺(AA)、N-(3-氨基丙基)甲基丙烯酰胺盐酸盐(APMA)及戊二醛购自Sigma Aldrich(MO,USA)。N,N'-亚甲基双丙烯酰胺(MBA)、过硫酸铵(APS)及N,N,N',N'-四甲基乙烷-1,2-二胺(TEMED)购自Biosharp(中国合肥)。ε-聚-L-赖氨酸(ε-poly-L-lysine,缩写ε-PL)购自上海世丰生物技术(中国上海)。硼氢化钠(NaBH4)购自国药集团化学试剂(中国上海)。DAPI购自Beyotime。
实施例1
(1)将530mg AA、160mg MBA、30mg APMA中溶于2ml超纯水混匀,超声获得均一溶液;再向溶液中再加入90mgε-PL混匀,形成为丙烯酰胺单体溶液;
(2)通过反相微乳液法得到聚丙烯酰胺纳米粒子:首先将1.6g AOT和3.1g Brij30加入到含有45mL正己烷的反应烧瓶中,搅拌40分钟形成油相;之后,向油相中逐滴加入步骤(1)制备的丙烯酰胺单体溶液,搅拌20分钟,最后再加入80μL新鲜制备的浓度为100mg/ml的APS水溶液和80μL TEMED以引发聚合,并剧烈搅拌2小时。聚合完成后,通过旋转蒸发去除正己烷,并通入无水乙醇沉淀聚合颗粒物,用无水乙醇沉淀洗涤聚合物数次除去表面活性剂和残余单体,乙醇挥发干净后,聚合物沉淀中加入适量水,超声重悬;聚合物用100kDa超滤管水洗超滤5次;最后将聚合物重悬于50mL超纯水中,成为聚合物水溶液,超声5min;再用0.22μm滤膜过滤聚合物水溶液,得到聚丙烯酰胺纳米粒子(PAANP)溶液;PAANP溶液室温放置。
(3)将60μL浓度为250mg/ml的戊二醛水溶液加入步骤(2)得到的10mL PAANP溶液中,37℃超声反应1小时;通过超滤管超滤水洗经戊二醛处理的聚丙烯酰胺纳米粒子数次,去除多余戊二醛;最后将聚丙烯酰胺纳米粒子重悬于10mL超纯水中,超声10分钟;再用0.22μm滤膜过滤聚丙烯酰胺纳米粒子溶液以除菌;过滤除菌后的聚丙烯酰胺纳米粒子溶液室温避光保存,得到自荧光聚丙烯酰胺纳米粒子(fPAANP)成品溶液。
实施例2
实施例2与实施例1的原料和制备方法相同,不同之处在于,步骤(1)中采用500mgAA、150mg MBA、20mg APMA,80mgε-PL;步骤(2)中采用1.4g AOT和3.0g Brij30;步骤(3)戊二醛溶液用量为20μL。
实施例3
实施例3与实施例1的原料和制备方法相同,不同之处在于,步骤(1)中采用550mgAA、180mg MBA、40mg APMA,100mgε-PL;步骤(2)中采用1.8g AOT和3.2g Brij30;步骤(3)戊二醛溶液用量为120μL。
试验例1fPAANP的表征研究:
使用透射电子显微镜(JEM-2100)(JEOL Ltd.)评估实施1制备的fPAANP的尺寸和分散性。用Zetasizer Nano(Malvern Instruments)分析fPAANPs的流体动力学尺寸分布和Zeta电位。用NanoDrop 2000c分光光度计(Thermo Scientific)观察UV-Vis光谱。荧光发射光谱由Hitachif-7000荧光光谱仪(Hitachi High-Technologies)在488nm和543nm激发下检测,分别得到绿色和红色荧光。通过Nicolet 5700FTIR光谱仪(Thermo Scientific)观察FTIR光谱。通过Odyssey红外成像系统(Li-COR Bioscience)扫描720和820nm发射的NIRF图像。
本发明实施例1中通过向丙烯酰胺聚合中加入ε-PL制备了新的PAANP。同时对比例采用实施例1相同的方法,不同之处在于不加入ε-PL。对比研究发现没有ε-PL时GTA交联的PAANP显示出浅黄色,而具有ε-PL时GTA交联的PAANP显示为深黄色,说明具有ε-PL的PAANP显示出强烈的NIRF(称为fPAANP)(图1a),表明ε-PL的丰富氨基在GTA交联中产生丰富的C=N键,从而促成了强烈的NIRF。另外,试验发现fPAANP在720和820nm的发射波长处均显示NIRF(图1a);但发射波长为720nm的NIRF比820nm处的NIRF更强(图1a)。PAANP水溶液呈浅蓝色,经GTA交联纯化后,fPAANP水溶液呈黄色(图1a)。PAANP和fPAANP的冻干粉末分别也显示白色和黄色。
透射电子显微镜(TEM)观察表明,fPAANP的平均尺寸为16.04±2.05nm,而PAANP为16.38±2.36nm,表明GTA交联对粒子尺寸没有明显的影响(图1b)。实际上,GTA被用作交联剂,它不仅产生了荧光,而且稳定了纳米粒子。荧光光谱分析显示fPAANP分别具有470nm/520nm和487nm/732nm的两个激发/发射峰(图1c和1d)。这些结果表明fPAANP具有可见荧光和NIR荧光的双重属性。
实施例1中向丙烯酰胺聚合中加入ε-PL制备了新的PAANP,通过GTA交联的PAANP显示为深黄色(图2a),相应的也具有强的NIRF(图2a);而没有ε-PL时GTA交联的PAANP显示出浅黄色(图2a),NIRF弱,特别是820nm的NIRF(图2a)。根据动态光散射(DLS)分析,分散在水中的fPAANP的水动力平均尺寸为约300nm(图2b)。Zeta电位的检测表明,由于大量的ε-PL阳离子,fPAANP具有显着增加的正电荷,Zeta电位约为+16mV(图2c)。
试验例2GTA浓度优化
方法:
采用实施例1的制备方法。其中步骤(3)用不同终浓度的(0.5~3mg/ml)GTA交联PAANP。将浓度为250mg/ml的戊二醛溶液以不同体积加入到10mL PAANP悬浮液中,使戊二醛的终浓度分别为0.5mg/ml、1mg/ml、1.5mg/ml、2mg/ml、2.5mg/ml和3mg/ml。在37℃下超声处理1小时,将制备的fPAANP洗涤6次以通过超滤除去过量的戊二醛,最终的fPAANP溶液通过补水保持10mL,超声处理后用0.22μm膜过滤,在室温下避光保存。
结果:
由于自发荧光是由GTA交联相关的C=N和C=C键产生的,因此制备高质量fPAANP的GTA用量很重要。用不同终浓度的(0.5~3mg/ml)GTA交联PAANP。可见fPAANP溶液的颜色逐渐变化(图3a)。250~270nm内fPAANP的吸光度随着GTA浓度的增加而逐渐改变(图3b)。fPAANP的吸光度显示荧光强度的增加是由于亚胺键的形成,这显然是由较高浓度的GTA产生的。在机制中,随着更多的GTA被添加到PAANP中用于更完全的交联,将产生更多的席夫碱键,并且相应地产生更强的荧光。在0.2%的浓度下,720nm发射的均匀荧光强度仍然增加,但在820nm的发射达到顶峰(图3c)。结果表明,通过在交联中使用更多的GTA可以改善自发荧光(图3d)。由于越来越多的氨基被交联到席夫碱中,随着GTA的增加,Zeta电位降低(图3e)。当GTA达到0.3%时,fPAANP水动力半径大小急剧增加(图3f),表明纳米颗粒的聚集。在0.15%的浓度下获得最小多分散指数(PDI),显示出最佳的分散性。这些结果表明,用于交联的GTA的量显着影响fPAANP的自发荧光、电荷、大小和分散性。据透露,本发明发现1.5mg/ml的GTA是制备高质量fPAANP的最佳选择。
试验例3NaBH4还原对fPAANP荧光性能的影响
方法:
将浓度为10mg/ml的NaBH4溶液以不同体积(包括0、2、4、8、16和32μL)加入460μL实施例1制备的fPAANP溶液,再加水补充至500μL终体积。混匀后室温下反应10分钟。
结果:
NaBH4是一种温和的还原剂,可将酮、醛和席夫碱(C=N)转化为相应的还原型。为了确认fPAANP的荧光是由席夫碱衍生而成,用NaBH4处理fPAANP。如预期的那样,随着NaBH4浓度的增加,还原的fPAANP(rfPAANP)的颜色逐渐消失。因此,720和820nm发射的荧光变弱(图4a和4b)。随着NaBH4浓度的增加,520nm发射的可见荧光强度也逐渐降低(图4c)。显然,越来越多的C=N和C=C键被NaBH4还原,显然,fPAANP的自发荧光来自C=N和C=C键。类似地,随着NaBH4浓度的增加,250~270nm内rfPAANP的吸光度也逐渐降低(图4d)。为了进一步证实fPAANP的荧光来自席夫碱,用傅立叶变换红外(FTIR)光谱分析PAANP、fPAANP和rfPAANP。结果表明,与PAANP相比,rfPAANP的FTIR光谱显示出两个峰(图4b)。1665cm-1处的峰归因于C=N伸缩振动,而在1530cm-1处的峰被归属于由戊二醛聚合产生的C=C键。这与通过其他含有氨基的材料的GTA交联产生的类似的FTIR光谱是一致的。这些数据表明席夫碱键(C=N)和碳双键(C=C)给fPAANP提供了自发荧光。梯度NaBH4还原测定的结果也表明NaBH4还原可用于调节fPAANP的荧光性能。
试验例4细胞标记和细胞活力研究
RAW264.7、Hepa1-6和HepG2细胞获自中国类型培养物保藏中心(中国上海)。裸鼠购自南京大学模范动物研究中心(中国南京)。
方法:
将细胞在DMEM细胞培养基中培养,培养基含有10%(v/v)新生小牛血清(NBCS)、100单位/mL青霉素、100μg/mL链霉素、2.4g/L NaHCO3和4.8g/L HEPES(Hepa1-6细胞培养基含1mM丙酮酸钠)。细胞在5%(v/v)CO2培养箱中37℃培养。为了用fPAANP处理细胞,将细胞以3×104/孔的浓度接种到24孔培养板中。培养24小时后,取出培养基,500μL/孔加入无NBCS和抗生素的新鲜培养基,孵育2小时。将实施例1制备的fPAANP溶液冷冻干燥处理,溶解在超纯水中,加入不同浓度的fPAANP(使加入培养基的fPAANP终浓度为0.5~4mg/mL)以处理细胞4小时。然后在室温下用4%(g/ml)多聚甲醛溶液固定20分钟后,用PBS洗涤细胞3次,并用DAPI染色溶液染色。再次用PBS洗涤三次后,用装有冷CCD DP71的IX51倒置显微镜观察细胞并拍照。
细胞流式细胞分析和活力测定:根据上述方法,通过fPAANP处理细胞。用PBS洗涤3次后,用胰蛋白酶消化细胞,离心收集,然后再用PBS洗涤3次,用BD Accuri C6进行流式细胞分析。通过CCK8测定法在96孔板中测量细胞活力。将细胞以1×104/孔接种到培养板中。用fPAANPs处理4小时后,用PBS洗涤细胞3次。每孔加入100μL培养基和10μL CCK8混合。细胞在37℃孵育2小时。通过使用酶标仪BioTek Synergy HT在450nm处测定吸光度。经处理的细胞的活力以未经处理的对照细胞的百分比表示,未经处理的对照细胞的活力定义为100%。
结果:
因为fPAANP也显示出可见荧光,所以通过普通的荧光显微镜检查细胞就可以非常容易地研判纳米颗粒对细胞的内化效果。首先通过含有和不含有ε-PL的GTA交联的PAANP初步处理RAW264.7细胞。结果显示,用不含ε-PL的GTA交联的PAANP处理的细胞显示非常弱的荧光;但用含有ε-PL的GTA-交联PAANP(即fPAANP)处理的细胞显示出强荧光。该结果表明,fPAANP容易内化到细胞中,可用于标记细胞,表明在PAANP中添加ε-PL对于成功制备fPAANP是至关重要的。这是本发明的关键技术。
为了进一步研究fPAANP标记细胞的效率,将RAW264.7、HepG2和Hepa1-6细胞用不同浓度(终浓度0.5~4mg/mL)的fPAANP同时处理4小时。处理细胞和对照细胞用显微镜成像以检测三个通道的可见荧光,包括用于成像DAPI染色的核的蓝色荧光、在488nm激发下的绿色荧光和在543nm激发的红色荧光。合并不同通道的图像以确定细胞内分布。结果表明,细胞被纳米颗粒有效地标记。此外,细胞标记效率取决于细胞处理中使用的fPAANP的剂量。因此,用最高浓度的纳米颗粒(4mg/mL)获得最高的标记效率。在处理的细胞中,RAW264.7细胞总是显示比其他两个肝细胞更高的标记效率(图5a)。原因是细胞是巨噬细胞,其通常比其他种类的细胞具有更强的纳米颗粒吸收能力。使用流式细胞术进一步定量测量细胞标记效率。分别检测APC标记、FITC标记和PE-CY7标记的荧光(图5b)。结果与荧光显微镜成像相匹配。由fPAANP标记的细胞在可见光和红外荧光下均显示出非常好的成像效果。通过不同浓度的fPAANP处理的三种细胞的NIRF成像也显示细胞被纳米颗粒有效地标记。
为了评估fPAANP细胞标记的持久性,将终浓度4mg/mL fPAANP处理4小时的细胞用胰蛋白酶消化收集,接种在新的大培养皿中,用于连续培养,在此期间动态检测标记细胞的荧光。结果表明,培养5d后细胞荧光逐渐降低。然而,细胞的荧光仍然是明显的,足以在5天培养后观察到。可见,该种纳米材料的荧光具有极其稳定的荧光属性,抗光漂白能力强,这种特性在体内成像上非常重要。
为了评估fPAANP对细胞的生物相容性,使用体外CCK-8测定来评价由纳米颗粒处理的细胞的活力。用不同浓度的fPAANP(终浓度0.5~10mg/mL)处理RAW264.7、HepG2和Hepa1-6细胞4小时。结果表明,对三种细胞活性影响的最大浓度分别为3mg/mL、6mg/mL和5.5mg/mL(图5c)。在三种细胞上fPAANP的半致死浓度(LC50)分别为6、9和7.5mg/mL,表明纳米颗粒具有非常优良的生物相容性。
试验例5体内分布代谢检测
方法:
动物实验由东南大学动物护理与使用委员会批准(中国南京)。使用NIRF成像仪Pearl Imager(Li-Cor)检测体内纳米颗粒的分布。裸鼠(CByJ-Cg-Foxn1nu/J)由南京示范动物研究中心提供。对小鼠静脉内或皮下注射fPAANP,并用Pearl Imager以默认参数动态成像。此外,将fPAANP标记的Hepa1-6细胞和未标记的对照细胞分别移植到小鼠的侧面和颈部,使用NIRF成像连续监测荧光。
结果:
fPAANP的体内分布和代谢对其临床应用至关重要。为此,将实施例1制备的fPAANP溶液冷冻干燥处理,溶解在超纯水中,将fPAANP分别以终浓度126mg/kg的高剂量和42mg/kg的低剂量静脉内注射入小鼠。然后用NIRF成像仪对小鼠动态成像。结果表明,注射后肝脏位置的荧光逐渐增强。在高剂量下,肝脏位置在注射后1小时已经显示出强烈的荧光,表明纳米颗粒在肝脏中的富集(被动靶向)。在任何同一时间点,给予高剂量fPAANP的小鼠的荧光显示出比低剂量fPAANP更强的荧光。注射后17小时~60小时,低剂量和高剂量纳米颗粒的小鼠荧光分别达到最高,然后纳米颗粒被逐渐代谢。然而在注射后120小时,荧光仍然可检测。在动物实验中,纳米颗粒处理的小鼠在脑中也显示出强烈的荧光(图6a),可能这种纳米可通过血脑屏障进入脑组织。
fPAANP在体内的被动吸收和分布也通过皮下注射进行了评估。发现许多纳米粒子滞留在尾部。部分纳米颗粒在缓慢吸收10小时后主要分布在肝脏中,在21小时达到最大值,40小时后下降。类似地,还有一小部分纳米颗粒富集在脑中(图6b和6c)。皮下纳米粒子的吸收能力显示出个体差异。此外,纳米粒子皮下注射后,在注射部位尾巴处,注射后25天仍然保持强烈的荧光。可见,本发明制备的fPAANP在体内具有持续的荧光属性,非常适于长时间追踪成像。而且,注射的fPAANP对研究中的小鼠的活力未产生明显影响。所有数据显示,具有强自发荧光的fPAANP在体内快速吸收,维持时间长,主要依赖肝脏代谢。然而,即使在高剂量下,fPAANP对小鼠没有显示可见的毒性,表明fPAANP在体内应用中具有高的生物相容性。
为了提供使用fPAANP的细胞标记的即时实际应用,将fPAANP标记的Hepa1-6细胞分别以高浓度(终浓度126mg/kg体重)和低浓度(终浓度42mg/kg体重)皮下移植到小鼠的右臀部和颈部,而将未标记的Hepa1-6细胞移植到左侧臀部与对照相同高浓度。动态NIRF成像证实,2天后观察到肿瘤,7天后仍保持强荧光,之后随着时间的进一步延长荧光开始逐渐下降(图6d和6e)。结果进一步证实了用于细胞标记的fPAANP的高质量,表明本发明所制备的自荧光聚丙烯酰胺纳米粒子在临床医学中的潜在应用价值。
Claims (14)
1.一种自荧光聚丙烯酰胺纳米粒子,其特征在于,由戊二醛处理聚丙烯酰胺纳米粒子制得,所述聚丙烯酰胺纳米粒子由含有多聚赖氨酸的丙烯酰胺单体溶液制备而成。
2.根据权利要求1所述的自荧光聚丙烯酰胺纳米粒子,其特征在于,所述的丙烯酰胺单体溶液由丙烯酰胺、甲叉双丙烯酰胺及N-(3-氨基丙基)甲基丙烯酰胺溶于水中形成。
3.根据权利要求1或2所述的自荧光聚丙烯酰胺纳米粒子,其特征在于,所述多聚赖氨酸包括各种分子量的多聚赖氨酸。
4.根据权利要求1或2所述的自荧光聚丙烯酰胺纳米粒子,其特征在于,所述自荧光聚丙烯酰胺纳米粒子具有多重荧光属性,包括可见荧光及近红外荧光。
5.一种权利要求1所述的自荧光聚丙烯酰胺纳米粒子的制备方法,其特征在于,包括如下步骤:
(1)将丙烯酰胺、甲叉双丙烯酰胺、N-(3-氨基丙基)甲基丙烯酰胺溶于超纯水中混匀;再向溶液中再加入多聚赖氨酸混匀,形成为丙烯酰胺单体溶液;
(2)通过反相微乳液法将丙烯酰胺单体溶液制备成聚丙烯酰胺纳米粒子;
(3)用戊二醛对聚丙烯酰胺纳米粒子进行交联处理,制备出自荧光聚丙烯酰胺纳米粒子。
6.根据权利要求5所述的制备方法,其特征在于,步骤(1)所述丙烯酰胺单体溶液中丙烯酰胺的浓度为250~275 mg/ml,甲叉双丙烯酰胺的浓度为75~90 mg/ml,N-(3-氨基丙基)甲基丙烯酰胺的浓度为10~20 mg/ml,多聚赖氨酸浓度为35~55 mg/ml。
7.根据权利要求5所述的制备方法,其特征在于,步骤(2)所述反相微乳液法中油相为含有表面活性剂AOT与Brij30的正己烷,水相为丙烯酰胺单体溶液。
8.根据权利要求7所述的制备方法,其特征在于,步骤(2)所述油相中表面活性剂AOT的浓度为30~40 mg/ml,Brij30的浓度为66-72mg/ml。
9.根据权利要求5所述的制备方法,其特征在于,步骤(2)所述丙烯酰胺单体溶液加入到油相中搅拌后再向油相中加入过硫酸铵-四甲基乙二胺溶液,搅拌。
10.根据权利要求5所述的制备方法,其特征在于,步骤(2)所述聚丙烯酰胺纳米粒子纯化过程为:蒸发去除正己烷,用无水乙醇沉淀聚合物,再用无水乙醇洗涤聚合物;乙醇挥发后聚合物中加入水,超声重悬;再加水超滤;最后用水重悬聚合物,超声后过滤,室温放置,得到聚丙烯酰胺纳米粒子溶液。
11.根据权利要求5所述的制备方法,其特征在于,步骤(2)所述反相微乳液法制备聚丙烯酰胺纳米粒子过程中向油相溶液持续充入氩气。
12.根据权利要求5所述的制备方法,其特征在于,步骤(3)所述交联处理具体过程为向聚丙烯酰胺纳米粒子溶液中加入戊二醛溶液,超声反应;超滤水洗去除多余戊二醛;加水后超声反应,再过滤以除菌,室温避光保存,得到自荧光聚丙烯酰胺纳米粒子成品溶液。
13.根据权利要求12所述的制备方法,其特征在于,所述聚丙烯酰胺纳米粒子溶液中加入戊二醛溶液后的戊二醛浓度为0.5 ~ 3 mg/ml。
14.一种权利要求1所述的自荧光聚丙烯酰胺纳米粒子在制备生物检测、细胞标记或者动物成像药物中的应用。
