本发明属于白蚁防治技术领域,具体涉及一种用于杀灭白蚁的诱杀虫培养方法。本发明首先将金龟子绿僵菌接种到自制的种子培养基中培养,得到金龟子绿僵菌种子液,再挑选金龟子绿僵菌的单菌落接种至发酵培养基进行培养,得到金龟子绿僵菌液,接着将刨花与蜂蜜混合后,喷洒蛋白胨溶液后灭菌,随后喷洒金龟子绿僵菌液,室温下放置干燥后,将白蚁放入培养箱中培养,再移入含有阿维菌素、阿维菌素等物质的培养箱中培养即可得到诱杀虫。将本发明培养的诱杀虫放归蚁巢,携带灭杀剂的诱杀虫可以将白蚁巢中的白蚁全部杀死,灭杀效果好,同时避免了化学药剂带来的环境污染问题。

本发明属于白蚁防治技术领域,具体涉及一种用于杀灭白蚁的诱杀虫培养方法。
背景技术
白蚁是世界五大害虫之一,属营巢居生社会性昆虫,习性怕光,长期生活于所筑巢及蚁路的黑暗环境中,组织严谨,分工精细,破坏力十分强大,能蛀食全部植物,部分动物来源物质,同时,其体内分泌的蚁酸,还会腐蚀钢筋,使混凝土变性。其危害性主要表现在:1、对建筑物的破坏,特别是对砖木结构、木结构建筑的破坏,白蚁隐蔽在木结构内部破坏或破坏其承重部位,往往造成房屋突然倒塌,导致财产损失和人员伤亡,由于其扩散力强,群体大,破坏迅速,在短期内既能造成巨大损失。2、对堤坝的破坏:在我国南方,许多江河、水库上建有土坝,这些土坝往往栖居着白蚁群体,它们在堤坝内,密集营巢,迅速繁殖,蚁道四通八达,有些蚁道甚至穿通堤坝的内外坡,当汛期水位升高时,常常出现管漏险情,甚至造成塌堤垮坝。这就是所谓的“千里之堤,溃于蚁穴”。3、对农作物、林木及园艺作物的危害:白蚁可以危害甘蔗、花生等农作物,尤其对甘蔗的危害十分严重。白蚁对林木、果树等的危害也具有相当的广泛性和严重性。另外,白蚁还能危害图书、文物、织物、武器、账册、地下电缆等,造成重大损失。
传统杀灭白蚁主要使用化学药剂、真菌、重金属、激素等,这些防治方法或者药物对人畜及环境的危害较大,尤其系化学药剂。化学药剂防治白蚁的效果虽然好,但是使用之后难以降解,有些甚至属于持久性有机污染物,不仅对人和动物具有高毒性,而且可以借由生物富集特性和挥发特性从局部影响到区域乃至全球环境。我国中草药资源丰富,人们利用中草药杀虫已经有很长的历史。目前已有利用纯中药制成杀蚁剂的研究,也有利用中药与化学药剂组合制成杀蚁剂的研究,但是上述杀灭白蚁的药物组合物均需要施放到白蚁活动频繁的区域或白蚁的巢穴之中。白蚁的巢穴隐蔽,生存繁衍能力强,灭杀剂只能灭杀一部分白蚁,很难彻底杀灭白蚁,容易留下隐患,并且化学药剂防治白蚁的毒性高,污染环境。因此迫切寻找一种彻底杀灭白蚁,容易找到蚁穴,且不会污染环境的白蚁灭杀方法。
发明内容
本发明所要解决的技术问题:针对白蚁的巢穴隐蔽,生存繁衍能力强,灭杀剂只能灭杀一部分白蚁,很难彻底杀灭白蚁,容易留下隐患,并且化学药剂防治白蚁的毒性高,污染环境的问题,提供了一种用于杀灭白蚁的诱杀虫培养方法。
为解决上述技术问题,本发明采用的技术方案是:
(1)按重量份数计,分别称取10~15份葡萄糖、10~25份玉米粉、3~10份硝酸钾、20~25份大豆蛋白粉、0.1~0.3份磷酸二氢钾和1000~1200份去离子水,搅拌混合均匀后在紫外灯下灭菌后得种子培养基;
(2)按重量份数计,分别称取15~25份葡萄糖、5~10份酵母粉、5~10份蛋白胨、10~15份琼脂、20~25份可溶性淀粉、0.5~1.0份磷酸二氢钾、2~4份硝酸钾、1~3份氯化钠和800~1000份去离子水中,搅拌混合后灭菌得发酵培养基;
(3)用接种环接种一环金龟子绿僵菌到种子培养基中,培养后得金龟子绿僵菌种子液,再使用接种环从金龟子绿僵菌种子液接种10~12个金龟子绿僵菌的单菌落到发酵培养基中,发酵后得金龟子绿僵菌液;
(4)选取刨花放入培养箱1中,向培养箱中加入蜂蜜,用蛋白胨溶液喷洒在刨花上,使刨花的含水量为5~10%,搅拌混合后将培养箱1放置紫外灯下灭菌,灭菌后喷洒金龟子绿僵菌液,使刨花中的含水量为25~35%,置于室温下自然干燥至含水量为5~7%,得处理后的培养箱1;
(5)按质量比1:1分别称取苹果核和雪梨核加入料理机中,处理后得核渣,按重量份数计,分别称取100~120份核渣、1~3份伊维菌素、20~30份水和150~200份甘蔗渣混合,搅拌混合后放入培养箱2中;
(6)选取白蚁加入处理后的培养箱1中进行培养,培养后将白蚁移入培养箱2中,自由活动后即可得到用于杀灭白蚁的诱杀虫。
步骤(3)中所述的接种环的规格为1μL,培养时间为3~4天,培养温度为30~35℃。
步骤(3)中所述的发酵温度为30~35℃,发酵时间为3~5天。
步骤(4)中所述的蜂蜜的质量为刨花质量10~16%,蛋白胨溶液的质量分数为0.1%。
步骤(6)中所述的培养5~10h。
步骤(6)中所述的所述的活动时间为8~10h。
步骤(5)中还可以加入1~3份阿维菌素。
本发明与其他方法相比,有益技术效果是:
(1)本发明首先将金龟子绿僵菌接种到自制的种子培养基中培养,得到金龟子绿僵菌种子液,再挑选金龟子绿僵菌的单菌落接种至发酵培养基进行培养,得到金龟子绿僵菌液,由于白蚁对金龟子绿僵菌腐蚀过的培养基,有聚集和取食偏好性,所以本发明对金龟子绿僵菌进行富集培养,不仅可以对白蚁有着显著的引诱效果,而且还可作为白蚁诱饵的微生物毒剂;
(2)本发明将刨花与蜂蜜混合后,喷洒蛋白胨溶液后灭菌,随后喷洒金龟子绿僵菌液,室温下放置,可以将金龟子绿僵菌培养在刨花中,并且刨花为白蚁的诱饵材料,将白蚁放入中进行培养后使白蚁体中携带有微生物毒剂,随后将培养后的白蚁放入培养箱2总,由于培养箱2中含有伊维菌素,经过伊维菌素培养后诱杀白蚁放归蚁巢,携带灭杀剂的诱杀虫可以将白蚁巢中的白蚁全部杀死,灭杀效果好;
(3)在本发明的培养箱中还可以加入阿维菌素,目的是利用本发明培养的诱杀虫灭杀白蚁,白蚁与阿维菌素接触后,不会立即死亡,会经过2~4天后死亡,这时候白蚁已回到白蚁巢中,可以促使接触携带药的白蚁回巢后传染全巢,从而达到彻底杀灭白蚁的目的,并且对阿维菌素可以被微生物分解,因而在环境中无累积作用,避免了化学药剂带来的环境污染问题。
具体实施方式
按重量份数计,分别称取10~15份葡萄糖、10~25份玉米粉、3~10份硝酸钾、20~25份大豆蛋白粉、0.1~0.3份磷酸二氢钾和1000~1200份去离子水,搅拌混合均匀后在紫外灯下灭菌10~15min,得种子培养基;按重量份数计,分别称取15~25份葡萄糖、5~10份酵母粉、5~10份蛋白胨、10~15份琼脂、20~25份可溶性淀粉、0.5~1.0份磷酸二氢钾、2~4份硝酸钾、1~3份氯化钠和800~1000份去离子水中,搅拌混合均匀后在121℃下灭菌10~15min,得发酵培养基;用1μL接种环接种一环金龟子绿僵菌到种子培养基中,在30~35℃下培养3~4天后得金龟子绿僵菌种子液,再在显微镜下使用1μL接种环从金龟子绿僵菌种子液接种10~12个金龟子绿僵菌的单菌落到发酵培养基中,在30~35℃下发酵3~5天,发酵后得金龟子绿僵菌液;选取200~300g刨花放入培养箱1中,向培养箱中加入刨花质量10~16%蜂蜜,用质量分数0.1%蛋白胨溶液喷洒在刨花上,使刨花的含水量为5~10%,搅拌混合20~30min后将培养箱1放置紫外灯下灭菌处理5~10min,灭菌后喷洒金龟子绿僵菌液,使刨花中的含水量为25~35%,随后置于室温下自然干燥至含水量为5~7%,得处理后的培养箱1;按质量比1:1分别称取苹果核和雪梨核加入料理机中,处理5~10min后得核渣,按重量份数计,分别称取100~120份核渣、1~3份伊维菌素、1~3份阿维菌素、20~30份水和150~200份甘蔗渣混合,搅拌混合10~15min后放入培养箱2中;选取100~200只白蚁加入处理后的培养箱1中进行培养,培养5~10h后将白蚁移入培养箱2中,自由活动8~10h后即可得到用于杀灭白蚁的诱杀虫。
实例1
按重量份数计,分别称取15份葡萄糖、25份玉米粉、10份硝酸钾、25份大豆蛋白粉、0.3份磷酸二氢钾和1200份去离子水,搅拌混合均匀后在紫外灯下灭菌15min,得种子培养基;按重量份数计,分别称取25份葡萄糖、10份酵母粉、10份蛋白胨、15份琼脂、25份可溶性淀粉、1.0份磷酸二氢钾、4份硝酸钾、3份氯化钠和1000份去离子水中,搅拌混合均匀后在121℃下灭菌15min,得发酵培养基;用1μL接种环接种一环金龟子绿僵菌到种子培养基中,在35℃下培养4天后得金龟子绿僵菌种子液,再在显微镜下使用1μL接种环从金龟子绿僵菌种子液接种12个金龟子绿僵菌的单菌落到发酵培养基中,在35℃下发酵5天,发酵后得金龟子绿僵菌液;选取300g刨花放入培养箱1中,向培养箱中加入刨花质量16%蜂蜜,用质量分数0.1%蛋白胨溶液喷洒在刨花上,使刨花的含水量为10%,搅拌混20min后将培养箱1放置紫外灯下灭菌处理10min,灭菌后喷洒金龟子绿僵菌液,使刨花中的含水量为35%,随后置于室温下自然干燥至含水量为7%,得处理后的培养箱1;按质量比1:1分别称取苹果核和雪梨核加入料理机中,处理10min后得核渣,按重量份数计,分别称取120份核渣、3份伊维菌素、3份阿维菌素、30份水和200份甘蔗渣混合,搅拌混合15min后放入培养箱2中;选取200只海南土白蚁加入处理后的培养箱1中进行培养,培养10h后将海南土白蚁移入培养箱2中,自由活动10h后即可得到用于杀灭白蚁的诱杀虫。

按重量份数计,分别称取10份葡萄糖、10份玉米粉、3份硝酸钾、20份大豆蛋白粉、0.1份磷酸二氢钾和1000份去离子水,搅拌混合均匀后在紫外灯下灭菌10min,得种子培养基;按重量份数计,分别称取15份葡萄糖、5份酵母粉、5份蛋白胨、10份琼脂、20份可溶性淀粉、0.5份磷酸二氢钾、2份硝酸钾、1份氯化钠和800份去离子水中,搅拌混合均匀后在121℃下灭菌10min,得发酵培养基;用1μL接种环接种一环金龟子绿僵菌到种子培养基中,在30℃下培养3天后得金龟子绿僵菌种子液,再在显微镜下使用1μL接种环从金龟子绿僵菌种子液接种10个金龟子绿僵菌的单菌落到发酵培养基中,在30℃下发酵3天,发酵后得金龟子绿僵菌液;选取200g刨花放入培养箱1中,向培养箱中加入刨花质量10%蜂蜜,用质量分数0.1%蛋白胨溶液喷洒在刨花上,使刨花的含水量为5%,搅拌混合20min后将培养箱1放置紫外灯下灭菌处理5min,灭菌后喷洒金龟子绿僵菌液,使刨花中的含水量为25%,随后置于室温下自然干燥至含水量为5%,得处理后的培养箱1;按质量比1:1分别称取苹果核和雪梨核加入料理机中,处理5min后得核渣,按重量份数计,分别称取100份核渣、1份伊维菌素、20份水和150份甘蔗渣混合,搅拌混合10min后放入培养箱2中;选取100只台湾乳白蚁加入处理后的培养箱1中进行培养,培养5h后将台湾乳白蚁移入培养箱2中,自由活动8h后即可得到用于杀灭白蚁的诱杀虫。

按重量份数计,分别称取12份葡萄糖、20份玉米粉、7份硝酸钾、22份大豆蛋白粉、0.2份磷酸二氢钾和1100份去离子水,搅拌混合均匀后在紫外灯下灭菌15min,得种子培养基;按重量份数计,分别称取20份葡萄糖、7份酵母粉、7份蛋白胨、12份琼脂、22份可溶性淀粉、0.7份磷酸二氢钾、3份硝酸钾、2份氯化钠和9000份去离子水中,搅拌混合均匀后在121℃下灭菌12min,得发酵培养基;用1μL接种环接种一环金龟子绿僵菌到种子培养基中,在32℃下培养3天后得金龟子绿僵菌种子液,再在显微镜下使用1μL接种环从金龟子绿僵菌种子液接种11个金龟子绿僵菌的单菌落到发酵培养基中,在33℃下发酵4天,发酵后得金龟子绿僵菌液;选取250g刨花放入培养箱1中,向培养箱中加入刨花质量14%蜂蜜,用质量分数0.1%蛋白胨溶液喷洒在刨花上,使刨花的含水量为7%,搅拌混合25min后将培养箱1放置紫外灯下灭菌处理7min,灭菌后喷洒金龟子绿僵菌液,使刨花中的含水量为30%,随后置于室温下自然干燥至含水量为6%,得处理后的培养箱1;按质量比1:1分别称取苹果核和雪梨核加入料理机中,处理7min后得核渣,按重量份数计,分别称取110份核渣、2份伊维菌素、25份水和170份甘蔗渣混合,搅拌混合12min后放入培养箱2中;选取150只黑翅土白蚁加入处理后的培养箱1中进行培养,培养7h后将黑翅土白蚁移入培养箱2中,自由活动9h后即可得到用于杀灭白蚁的诱杀虫。

上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
Claims (7)
1.一种用于杀灭白蚁的诱杀虫培养方法,其特征在于具体培养步骤为:
(1)按重量份数计,分别称取10~15份葡萄糖、10~25份玉米粉、3~10份硝酸钾、20~25份大豆蛋白粉、0.1~0.3份磷酸二氢钾和1000~1200份去离子水,搅拌混合均匀后在紫外灯下灭菌后得种子培养基;
(2)按重量份数计,分别称取15~25份葡萄糖、5~10份酵母粉、5~10份蛋白胨、10~15份琼脂、20~25份可溶性淀粉、0.5~1.0份磷酸二氢钾、2~4份硝酸钾、1~3份氯化钠和800~1000份去离子水中,搅拌混合后灭菌得发酵培养基;
(3)用接种环接种一环金龟子绿僵菌到种子培养基中,培养后得金龟子绿僵菌种子液,再使用接种环从金龟子绿僵菌种子液接种10~12个金龟子绿僵菌的单菌落到发酵培养基中,发酵后得金龟子绿僵菌液;
(4)选取刨花放入培养箱1中,向培养箱中加入蜂蜜,用蛋白胨溶液喷洒在刨花上,使刨花的含水量为5~10%,搅拌混合后将培养箱1放置紫外灯下灭菌,灭菌后喷洒金龟子绿僵菌液,使刨花中的含水量为25~35%,置于室温下自然干燥至含水量为5~7%,得处理后的培养箱1;
(5)按质量比1:1分别称取苹果核和雪梨核加入料理机中,处理后得核渣,按重量份数计,分别称取100~120份核渣、1~3份伊维菌素、20~30份水和150~200份甘蔗渣混合,搅拌混合后放入培养箱2中;
(6)选取白蚁加入处理后的培养箱1中进行培养,培养后将白蚁移入培养箱2中,自由活动后即可得到用于杀灭白蚁的诱杀虫。
2.根据权利要求1所述的一种用于杀灭白蚁的诱杀虫培养方法,其特征在于:步骤(3)中所述的接种环的规格为1μL,培养时间为3~4天,培养温度为30~35℃。
3.根据权利要求1所述的一种用于杀灭白蚁的诱杀虫培养方法,其特征在于:步骤(3)中所述的发酵温度为30~35℃,发酵时间为3~5天。
4.根据权利要求1所述的一种用于杀灭白蚁的诱杀虫培养方法,其特征在于:步骤(4)中所述的蜂蜜的质量为刨花质量10~16%,蛋白胨溶液的质量分数为0.1%。
5.根据权利要求1所述的一种用于杀灭白蚁的诱杀虫培养方法,其特征在于:步骤(6)中所述的培养5~10h。
6.根据权利要求1所述的一种用于杀灭白蚁的诱杀虫培养方法,其特征在于:步骤(6)中所述的所述的活动时间为8~10h。
7.根据权利要求1所述的一种用于杀灭白蚁的诱杀虫培养方法,其特征在于:步骤(5)中还可以加入1~3份阿维菌素。
