本发明公开了一种具有缓解青春痘作用的茶枯复合面膜及其制备方法,按体积比包括:纯化水75‑85份;丁二醇2‑4份;甘油2‑4份;透明质酸0.1‑0.3份;甲基纤维素0.4‑0.6份;β‑葡聚糖1‑2份;甘油聚醚‑26 0.5‑2份;烟酰胺1‑2份;胶原蛋白0.5‑2份;金缕梅提取物1‑3份;芦荟提取物1‑3份;茶枯提取物2‑5份;四胜肽0.4‑0.6份;六胜肽0.4‑0.6份;九胜肽0.4‑0.6份。本发明面膜对青春痘具有较好的预防和缓解作用。

一种具有缓解青春痘作用的茶枯复合面膜及其制备方法和 用途
技术领域
本发明涉及原料制备领域,尤其涉及一种具有缓解青春痘作用的茶枯复合面膜及其制备方法和用途。
背景技术
痤疮,俗称青春痘,是毛囊皮脂腺单位的一种慢性炎症性皮肤病,主要好发于青少年,对青少年的心理和社交影响很大,但青春期后往往能自然减轻或痊愈。临床表现以好发于面部的粉刺、丘疹、脓疱、结节等多形性皮损为特点。痤疮的发生主要与皮脂分泌过多、毛囊皮脂腺导管堵塞、细菌感染和炎症反应等因素密切相关。
真菌感染的治疗至今仍完全依赖于药物治疗。目前,临床使用的抗真菌药物结构类型有限(主要为多烯类两性霉素B及其衍生物、唑类药物、棘白菌素类、尼可霉素等),作用机理和靶点相对单一,且这些抗真菌药物存在种种的毒副作用和日益严峻的耐药性问题。因此,人们仍急切需要新型抗真菌药物,尤其是具有新结构类型、新作用机理或(和)作用靶点的抗真菌药物。
油茶(Camellia oleifera)是山茶科(Theaceae)山茶属植物,主要分布于热带、亚热带地区。我国的油茶资源丰富,福建、江西、湖南、广西等省均有大面积种植。茶枯为油茶籽榨油后剩下的渣饼,资源非常丰富。近年来,部分企业开始对茶枯衍生产品进行开发生产,但与其它国家的综合利用相比,在产品的品种、质量和市场占有率远不如发达国家,产品单一,产业链延伸不够。
发明内容
本发明的目的在于提供一种具有缓解青春痘作用的茶枯复合面膜及其制备方法。
本发明提供的技术方案如下:
一种具有缓解青春痘作用的茶枯复合面膜,按体积比包括:

在一实施例中,所述的复合面膜还包括防腐剂以及适量香精,所述的防腐剂为羟苯甲酯0.01-0.03份和甲基异噻唑啉酮0.001-0.005份。
作为优选,一种具有缓解青春痘作用的茶枯复合面膜,按体积比包括:

在较佳的实施例中,该复合面膜还包括羟苯甲酯0.02份;甲基异噻唑啉酮0.001份和香精适量。
在本发明中,所述茶枯提取物,由包括如下步骤的制备方法制备而得:40-80℃烘干0.5-2h,过10-30目筛,得到茶枯粉末;加入粉末5-10倍体积的石油醚加热回流2-4h,除去残留的茶油;抽滤后,残渣继续烘干;使用3-8倍粉末体积的70%乙醇,60-85℃加热回流提取2-4次,每次0.5-2h,合并提取液,减压回收溶剂至无醇味,得茶枯流体总浸膏;总浸膏用1.5-2.5倍量水混悬,使用HP20大孔树脂柱层析,依次用8-15L纯水、8-15L 30%乙醇水、8-15L 50%乙醇水、8-15L 70%乙醇水、8-15L 95%乙醇洗脱,减压回收溶剂得30%乙醇子馏分、50%乙醇子馏分、70%乙醇子馏分、95%乙醇子馏分。
由上述描述可知,本发明提供了一种具有缓解青春痘作用的茶枯复合面膜及其制备方法,以茶枯的70%乙醇馏分为主要有效成分,对青春痘具有较好的预防和缓解效果。

具体实施方式
实施例1
茶枯适当粉碎,60℃烘干1h,过20目筛,得到茶枯粉末(1.5kg)。石油醚(10L)加热回流3h,除去残留的茶油。静置后除去上层溶液,残渣继续烘干。使用70%乙醇(5L)80℃加热回流提取3次,每次1h,合并提取液,减压回收溶剂至无醇味,得茶枯流体总浸膏。总浸膏用2倍量水混悬,使用HP20大孔树脂(8L)柱层析,依次用纯水(15L)、30%乙醇水(15L)、50%乙醇水(15L)、70%乙醇水(15L)、95%乙醇洗脱,减压回收溶剂得30%乙醇子馏分、50%乙醇子馏分、70%乙醇子馏分。
实施例2体外抗真菌活性测试
本实验以白色念珠菌敏感菌株作为指示菌,采用纸片法体外药敏实验确定活性单体化合物,再用直接目测比浊法确定活性各子馏分的最小抑菌浓度MIC值及最小杀菌浓度MFC值。
1、仪器与实验材料
1.1主要的仪器与设备
AIRTECH超净工作台(苏州苏洁净化设备有限公司);BS200电子分析天平(德国赛多利斯公司);LDZX-40SC型立式自控电热压力蒸汽灭菌锅(上海申安医疗器械厂);电磁炉(中山市诺洁仕电器有限公司);96孔细胞培养板(美国CONING COSTAR公司);精密移液枪(法国Gilson公司);BDS200倒置生物显微镜(重庆奥特光学仪器有限责任公司)。
1.2主要的实验材料
培养基:江门市凯林贸易有限公司
两性霉素B药敏片(10μg):广州瑞泰生物技术有限公司
两性霉素B:美国SIGMA公司
DMSO:广东光华化学厂有限公司
甲醇:山东禹王实业有限公司
1.3实验菌株
白色念珠菌敏感菌株:ATCC 10231,购于北纳联创生物技术有限公司。
2、实验方法
2.1菌株的活化:将保存在-80℃冰箱白色念珠菌在培养基平板上,置于34℃恒温箱培养48h,使其活化。
2.2纸片法体外药敏实验
2.2.1样品溶液的制备:分别取化合物约2mg,精密称重。用适量甲醇溶解使其浓度为20mg/mL。
2.2.2用移液器吸取20μL上述待测溶液于纸片上(直径为5mm),待溶剂挥干后,将滤纸片置于制备好的含有白色念珠菌的沙氏固体培养基(含菌量约为2×106个/μL)中,每个质量重复做三次。以甲醇作为空白对照,34℃恒温培养24h,取出测量抑菌圈直径,记录数据。
2.2.3目测比浊法测定MIC及MFC的测定
A.菌液制备:用无菌1640培养液将活化后的白色念珠菌稀释至约1×106个/μL。
B.样品及对照品溶液的制备:分别取化合物约2mg,用少量DMSO(用量约为4‰)溶解成一定浓度,加PBS配制成浓度为400μM、200μM、100μM、50μM、25μM、12.5μM、6.25μM、3.125μM的样品溶液。对照品溶液的制备与之相同。
C.分别吸取化合物各浓度的样品溶液100μL置于96孔板中(每个浓度三个复孔),再往每个样品孔中加入100μL的菌液,混匀,另三个对照孔分别取200μL的菌液,200μL PBS,200μL含2‰DMSO的菌液。对照品做法与之相同。
C.MIC及MFC值的确定:将种好的板置34℃培养箱培养24h,目测比浊法确定MIC值,以MIC的浓度为界线,将MIC浓度及以上浓度的孔板液吸取100μL,置于沙氏固体培养基中,涂布均匀,34℃培养24h,以不长菌的浓度视为MFC值。
